황복(Takifugu obscurus)의 정액특성과 정자보존

  • 정액의 ml당 정자수는 $1.13 \pm 0.34 \times 10^{10}$이었고 spermatocrit는 $64.8 \pm 1.4$였으며, 정장의 삼투질농도는 $266 \pm 2$ mOsm/kg이었다. 총 단백질 함량과 총 지질 함량은 정장에 비해 정자에서 높은 값을 보였고, glucose는 검출되지 않았다. 황복 정자를 16일간 $0 \pm 0.5 \circ C$에서 냉장보존 하였을 때, 수정률은 0-0.7%로 보존효과가 저조한 것으로 나타났다. 황복 정자의 냉동보존을 위한 희석액으로는 marine fish Ringer's solution, 동해방지제로는 5% DMSO가 적합하였으며, 이때의 수정률은 $79.3 \pm 7.5$%였다.

The effect of high potassium adaptation on hybridoma cell growth

  • Hybridoma cells were adapted in media containing up to 80 mM $K^+$. The adapted cells obtained tolerance at high osmotic pressure and low pH. The adapted cells showed a maximum viable cell density of $1.1{\times}10^6$ cells/ml when a batch culture was progressed in a nutrient-fortified medium with Erlenmeyer flask at 450 $mOsm/kgH_2O,$ as compared to $4.8{\times}10^5$ cells/ml for non-adapted cells grown under the same conditions. The adapted cells also showed a maximum viable cell density of $7.8{\times}10^5$ cells/ml with the same method at initial pH 7.0 as compared to $5.3{\times}10^5$ cells/ml for non-adapted cells. Adaptation of animal cells at high $K^+$ levels may therefore lead to an improvement of their performance at limited conditions.

Physico-chemical Properties and Cold Storage of River Puffer (Takifugu obscurus) Milt (황복(Takifugu obscurus) 정액의 물리$\cdot$화학적 성상과 냉장보존)

  • To obtain the basic data for the preservation of river puffer (Takifugu obscurus) sperm, experiments were carried out on the physico-chemical properties and cold storage of milt. The average number of sperm and spermatocrit in milt stripped were $1.13\pm0.34\times10^{10}/ml$ and 64.8$\pm$1.4, respectively. Osmolality of seminal fluid was 266$\pm$2 mOsm/kg, Total protein and total lipid from sperm were higer than that from seminal fluid. $Ca^{2+}$ and $Na^{+}$ concentrations were higher in the seminal fluid than in the sperm, while $Mg^{2+}$ and $K^{+}$ concentrations were lower in the seminal fluid. When sperm of river puffer were preserved in $0\pm0.5^{\circ}C$ with various diluents for 16 days, fertilization rate was $0\~0.7\%$. It suggested that cold storage of river puffer sperm was detrimental to sperm fertility.

Zinc may increase bone formation through stimulating cell proliferation, alkaline phosphatase activity and collagen synthesis in osteoblastic MC3T3-E1 cells

  • Zinc is an essential trace element required for bone formation, however not much has been clarified yet for its role in osteoblast. We hypothesized that zinc would increase osteogenetic function in osteoblasts. To test this, we investigated whether zinc treatment enhances bone formation by stimulating osteoblast proliferation, bone marker protein alkaline phosphatase activity and collagen synthesis in osteoblastic MC3T3-E1 cells. MC3T3-E1 cells were cultured and treated with various concentrations of zinc (0, 1, 3, 15, 25 uM) along with a normal osteogenic medium (OSM) as control for 1, 5, 10 days. As measured by MTT assay for mitochondrial metabolic activity, cell proliferation was stimulated even at low zinc treatment (1-3 ${\mu}M$) compared to OSM, and it was stimulated in a zinc concentration-dependent manner during 5 and 10 days, with the most pronounced effect at 15 and 25 uM Zn. Cellular (synthesized) alkaline phosphatase (ALP) activity was increased in a zinc concentration-dependent manner, so did medium (secreted) ALP activity. Cellular collagen concentration was increased by zinc as time went by, therefore with the maximum zinc stimulatory effect in 10 days, and medium collagen concentration showed the same pattern even on 1 and 5 day. This zinc stimulatory effect of collagen synthesis was observed in cell matrix collagen staining. The study results imply that zinc can increase osteogenic effect by stimulating cell proliferation, ALP activity and collagen synthesis in osteoblastic cells.

Studies on Transfer of In Vitro Fertilized Mouse Embryos Following Ultrarapid Freezing II. Effect of Treatment of pH, Osmolality and Sperm Preincubation on Development of In Vitro Fertilization Mouse Embryos (생쥐 체외수정난의 초급속동결 및 이식에 관한 연구 II. pH, 삼수압 및 정자 전배양처리가 생쥐 처리수정난의 발달에 미치는 영향)

  • These studies were carried out to overcome 2-cell block and in vitro development to blastocysts in vitro fertilization of mouse embryos. The unfertilized ova were obtained by superovulation in ICR mice of 4 to 6 weeks old. Tyrode's 280 solution was used as basal media, and the pH range of media examined was designed from 6.5 to 7.5 with 0.2 interval and the range of osmolality from 250 to 370 mOsm with 20 interval, and the period of sperm preincubation examined was 30, 60, 120, and 180 minutes. The ova developed to 2-cell embryos after 26hrs of incubation with preincubated sperm were evaluatated as in vitro fertilized ones. The results obtained were summarized as follows: 1. The optimal ranges of pH and osmolality of culture media and of sperm preincubation time for in vitro development of in vitro fertilized ova to blastocyst were pH 7.1 to 7.3, 250 to 350 mosmol and 60 to 180 min, respectively. 2. With the media of pH 7.1, 310 mOsm and sperm preincubation period of 120min in another experiment of large sample size, the in vitro fertilized ova was found 66.5% and the in vitro development of in vitro fertilized ova to blastocyst was found 35.8%. From the above results it was concluded that the optimal conditions of pH and osmolality of the media for mouse IVF and embryo culture, and the period of sperm preincubation might be 7.1, 310 mOam and 120min, respectively.

Effects of Thawing Conditions on the Viability and Acrosomal Morphology of Cryopreserved Boar Semen (동결보존한 돼지정액의 융해조건이 정자의 생존율과 첨체변화에 미치는 효과)

  • This experiment was carried out to investigate the effects of osmolarity of thawing diluents, seminal plasma added in thawing diluents on the sperm viability and the effects of thawing temperature, the temparature of the thawing diluents on the sperm viability and acrosomal morphology of boar spermatozoa by the straw method. The result obtained were summarized as follows: 1. The sperm viablilty after thawing of the frozen semen was shown greater in the high osmolarity(392~492mOsm) than low osmolarity(300mOsm) in thawing diluent. The added levels of seminal plasma in thawing diluent did not affect the viability of frozen-thawed boar semen. 2. In terms of thawing temperature, the sperm viability was shown higher in the frozen semen thawed at 5$0^{\circ}C$ for one min. (p<0.01) than those thawed at 2$0^{\circ}C$ or 37$^{\circ}C$ for one min. The sperm viability was not significant at the diluent temparature of 2$0^{\circ}C$or 37$^{\circ}C$ after thawing: but the sperm viability was higher in thawing diluent at 2$0^{\circ}C$ than in that at 37$^{\circ}C$. However, the effects of thawing temperature and diluent solution on normal acrosomal rate were not significant. 3. Cleavage rates of oocytes fertilized with frozen semen were 46.4% and 43.3%, respectively, which were thawed at 5$0^{\circ}C$ for one min. and then diluted in mBTS medium at 2$0^{\circ}C$or 37$^{\circ}C$. To sum up, the sperm viability was shown greater at the high of thawing diluents of frozen boar semen. In terms of thawing conditions, the sperm viability was shown greater, when semen was thawed at a high temperature for a short time and then diluted at the same temperature as that in the straw.

  • This study compared the results of Importance-Performance Analysis (IPA) on trail managerial conditions by trail section in Mudeung-Mountain Provincial Park. The study also examined the validity of using overall survey method (OSM: analyzing IPA from the data collected from all the trails without separating trails by section) by comparing the OSM results with those obtained from the survey by section. A total of 258 users who visited the Park was surveyed using questionnaire in the four designated trail sections during the September of 2000. The result of IPA showed that there are differences in twenty three attributes out of the 26 selected attributes (88.5%) among four trail sections. The overall IPA results obtained from the total of four trail sections were not the same with the IPA results from each trail section in twenty three attributes (88.5%) out of the 26 selected attributes. It suggests that we need to use a survey segmented by trail sections rather than a survey for overall trails to obtain better information for more efficient trail management.

  • In this paper, we propose a spatial query processor, SQUERY, which can derive rich query results through spatial reasoning on the initial knowledge base, as well as, process both qualitative and quantitative queries about the topological and directional relationships between spatial objects. In order to derive richer query results, the query processor expands the knowledge base by applying forward spatial reasoning into the initial knowledge base in a preprocessing step. The proposed query processor uses not only qualitative spatial knowledge describing topological/directional relationship between spatial objects, but also utilizes quantitative spatial knowledge including geometric data of individual spatial objects through geometric computation. The results of an experiment with the OSM(Open Street Map) spatial knowledge base demonstrates the high performance of our spatial query processing system.

Effects of UK 14,304, An ${\alpha}_2$-Adrenergic Agonist, on Renal Function in Dog (${\alpha}_2$-아드레날린 효능약인 UK 14,304의 신장기능에 미치는 영향)

  • The effects of UK 14,304, an ${\alpha}_2$-adrenergic agonist, on renal function were investigated in dogs. UK 14,304, when given intravenousely($15.0{\mu}g/kg,\;50{\mu}g/kg$), produced the increase of urine flow accompanied with the marked augmentation of free water clearance ($C_{H_2O}$) and reabsorption rates of sodium in renal tubules ($R_{Na}$), and the remarkable decrease of osmolar clearance ($C_{osm}$) and the amounts of sodium excreted in urine ($E_{Na}$). UK 14,304 given into a renal artery($1.5{\mu}g/kg,\;5.0{\mu}g/kg$) elicited the increase of urine flow with the augmentation of $C_{H_2O}$ in both kidney. UK 14,304, when administered into carotid artery($3.0{\mu}g/kg,\;10.0{\mu}g/kg$). exhibited the same aspect as shown in intravenous UK 14.304 at smaller dose than the intravenous dose. Diuretic action of intravenous UK 14,304 were produced together with increase of $C_{H_2O}$ in situation of water diuresis too, changes of renal function in this state were the increase of $C_{osm},\;E_{Na},\;and\;E_K$ (excreted amounts of potassium in urine), and the decrease of $R_{Na}\;and\;R_K$, these were different appearances from situation of saline diuresis. Diuretic action of intravenous UK 14,304 were blocked completely by post or pretreatment of yohim-bine, ${\alpha}_2$-adrenergic blocking agents, and inhibited by pretreatment of vasopressin, antidiuretic hormone. Above results suggest that UK 14,304 produces the diuretic action by the inhibition of vasopressin secretion and suppression of electrolytes reabsorption of electrolytes in renal tubules mediated with central ${\alpha}_2$-adrenoceptor in dog.

Effect of evaporation-induced osmotic changes in culture media in a dry-type incubator on clinical outcomes in in vitro fertilization-embryo transfer cycles

  • Objective: This study investigated whether adding outer-well medium to inhibit osmotic changes in culture media in a dry-type incubator improved the clinical outcomes of in vitro fertilization-embryo transfer (IVF-ET) cycles. Methods: In culture dishes, the osmotic changes in media (20 µL)-covered oil with or without outer-well medium (humid or dry culture conditions, respectively) were compared after 3 days of incubation in a dry-type incubator. One-step (Origio) and G1/G2 (Vitrolife) media were used. Results: The osmotic changes in the dry culture condition (308 mOsm) were higher than in the humid culture conditions (285-290 mOsm) after 3 days of incubation. In day 3 IVF-ET cycles, although the pregnancy rate did not significantly differ between the dry (46.2%) and humid culture (51.0%) groups, the rates of abortion and ongoing pregnancy were significantly better in the humid culture group (1.5% and 49.5%, respectively) than in the dry culture group (8.3% and 37.8%, respectively, p<0.05). In day 5 IVF-ET cycles, the abortion rate was significantly lower in the humid culture group (2.2%) than in the dry culture group (25.0%, p<0.01), but no statistically significant difference was observed in the rates of clinical and ongoing pregnancy between the dry (50.0% and 25.0%, respectively) and humid culture groups (59.5% and 57.3%, respectively) because of the small number of cycles. Conclusion: Hyperosmotic changes in media occurred in a dry-type incubator by evaporation, although the medium was covered with oil. These osmotic changes were efficiently inhibited by supplementation of outer-well medium, which resulted in improved pregnancy outcomes.
