Measurement of Size of Ovaries, Follicles, and Corpus Lutea by Ultrasonography with Jeju Horse (초음파상을 이용한 제주마의 난소, 난포 및 황체의 크기 측정)

  • The purpose of the present study was to set up basic information of size and status of ovaries by using ultrasonography to retrieve in vivo matured oocytes with ovum pick- up method. Ovaries were collected from the abattoir in Jeju in May and June which is breeding season. When the size of ovaries on ultrasonography was compared with real size measured by caliper, no significant difference was shown (p<0.05). The number of preovulatory follicles (>21mm) was investigated with ultrasonography and naked eyes. Ultrasonography group had 0.83 preovulatory follicles per ovary and naked-eye group had 0.75 preovulatory follicle per ovary and their average size was 2.86 cm and 2.34 cm, respectively. The average number of follicle was 4.25 with ultrasonography and 4.38 with naked eyes. There was no significant difference considering the size of follicle and number of follicle between ultrasonography and actual size except for the size of preovulatory follicle, suggesting that information of ultrasonography is able to use for OPU or other reproductive technology of mare.

Effect of Serum and Serum Free Media on the Developmental Competence of OPU Derived Bovine IVP Embryo

  • Embryos produced with serum show the alterations in their ultrastructure, impaired compaction, abnormal blastulation, aberrant mRNA expression profiles and large calf syndrome with greater incidences of stillbirths and deaths after birth. The aim of the present study was to describe in vitro embryo production by analyzing embryo production, fetal production and pregnancy rate in free-serum medium. The OPU-IVP data used in this study from 2016. Approximately, sixteen cows (Hanwoo), which belonged to the Institute of Gyeongsang National University, were used. Two experimental group is used in this study. Serum groups were conducted in March to July and free-serum group was conducted in September to December. The recovered cumulus-oocyte complexes were morphologically classified to four grades based on the compaction of cumulus cells layers and homogeneity of the cytoplasm. The number of oocyte was significantly greater in serum groups than that in free-serum groups (29.61 ± 0.63 vs. 15.6 ± 0.62; p < 0.05). Between serum and free-serum groups indicate that average of 1st and 2nd grade oocytes were no difference (2.38 ± 1.67 vs. 2.38 ± 1.48; p > 0.05), but number of 3rd and 4th grade oocytes were greater in serum groups than that in free-serum groups (7.31 ± 7.64 vs. 5.60 ± 6.29; p < 0.05). Embryo cleaved competence was higher in rate in free-serum groups than that in serum groups (62.1% vs. 58.3; p < 0.05). However, blastocyst developmental rate was no difference between serum and free-serum groups (33.1% vs. 43.5%; p < 0.05). 986 recipients were used for embryo transfer. Pregnancy rate was indicated that between serum and free-serum group was no difference (54.6% vs. 56.3%; p < 0.05). In conclusion, we developed the free-serum system for production of in vitro bovine embryos in order to meet the developmental and qualitative requirements for large scale commercial use.

  • By adding height compensation to the usual 3 points skew adjustment device, an improved functional skew adjustment device is implemented in this paper. The proposed skew adjustment device is for traverse which will be adapted to BD and HD player system requiring more accuracy. and It can adjust not only R/T skew but also height. The system is composed of personal computer, control box, kinematic mechanism device and bar-code reader device. Application programs are implemented by visual basic.

  • In the paper, a hybrid control approach for the swing-up control of a rotary type inverted pendulum is treated using one-chip microcomputer. The control approach is composed by a scheduling logic control for swing up control and the linear state feedback control to achieve the disired inverted-state of the pendulum. The experimental cystem has been implemented by a 16-bit one-chip microcomputer with 3096 opu as the digital controller incorporating the above mentioned control approach.

Development of needle guidance system and its evaluation for ultrasound-guided follicular aspiration (초음파유도 난포채란기의 개발과 이의 검증)

  • This study was carried out to develop a newly designed ovum pick-up(OPU) instrument for ultrasound-guided transvaginal follicular aspiration in cows. This new instrument consists of out- & inner-layer stainless pipes and a grip with a trigger(hand) switch. Some gauge types of disposable needles and tubes can be attached to this inner pipe. With this instrument, while grasping an ovary with one hand, the other hand can handle in apiration and vacuum on/off with the least assitant's help. With this instrument the mean recovery rate of bovine follicular oocytes was 45.2%. In recovered oocytes, usable oocytes(Grade I & II) were 30.4% and this rate meant 1.4 oocytes per ovary. For 30 days after initial aspiration with this instrument, some adverse effects such as adhesion, hemorrhage, hematoma and other mass formation in/with ovaries were also examined by rectal examination, ultrasonographic and endoscopic images. Adhesion was found in one ovary 1 week after aspiration, and hemorrhagic lesion was found 1-2 days and petechia were found 3-5 days after aspiration and there was no remarkable adverse effects. It was found that this instrument could be applicable and safe for ovum pick-up in cows.

The effectiveness of earlier oocyte retrieval in the case of a premature luteinizing hormone surge on hCG day in in vitro fertilization-embryo transfer cycles

  • Objective: To evaluate the efficacy of earlier oocyte retrieval in IVF patients with a premature LH surge on hCG day. Methods: One hundred forty IVF patients (164 cycles) with premature LH surge on hCG day were included, retrospectively. We divided them into 2 study groups: LH surge with timed ovum pick-up (OPU) 36 hours after hCG injection (group B, 129 premature cycles), and LH surge with earlier OPU within 36 hours after hCG injection (group C, 35 cycles). Control groups were tubal factor infertility without premature LH surge (group A, 143 cycles). Results: The mean age (year) was statistically higher in group C than in groups A or B ($38.2{\pm}5.4$ vs. $36.2{\pm}4.2$ vs. $36.8{\pm}4.9$, respectively; p=0.012). The serum LH levels (mIU/mL) on hCG day were significantly higher in group B and C than in group A ($22.7{\pm}14.9$ vs. $30.3{\pm}15.9$ vs. $3.2{\pm}2.9$, respectively; p>0.001). Among groups A, B, and C, 4.9%, 31.7%, and 51.4% of the cycles, respectively, had no oocytes, and the overall rates of cycle cancellation (OPU cancellation, no oocyte, or no embryos transferrable) were 15.4%, 65.9%, and 74.3%, respectively. The fertilization rate (%) was significantly higher in group B than in group C ($73.2{\pm}38.9$ vs. $47.8{\pm}42.9$, p=0.024). The clinical pregnancy rate was significantly higher in group C than in groups A and B (44.4% vs. 27.3% vs. 9.1%, respectively, p=0.021). However, the miscarriage rate was also higher in group C than in group B (22% vs. 0%, respectively, p=0.026). Conclusion: Earlier OPU may not be effective in reducing the risk of cycle cancellation in patients with premature LH surge on hCG day. A larger scale study will be required to reveal the effectiveness of earlier ovum retrieval with premature LH surge.

Consideration on Domestic Production of Materials and Consumables for Human IVF-ET Program (체외수정 및 배아이식술 관련 재료 및 소모품의 국산화 필요성에 대한 고찰)

  • Human in vitro fertilization and embryo transfer (IVF-ET) program is a general procedure for infertile couples since first successful delivery on 1978 in UK and Korean first on 1985. Recently in Korea, more than 42,000 cases per year of IVF-ET were performed and showed good pregnancy rates compared worldwide data. The human IVF-ET procedure use many consumables, such as ovum pick-up (OPU) needles, centrifuge tubes, culture dish, ICSI pipette, culture media and ET catheters. Major of these materials are supported by the global companies. Thus, Korean IVF-ET program might be placed unstable situation by global economical risks. These uncertain problems could be overcome by the domestic production of IVF-ET materials and consumables. Two times questionnaires for Korean clinicians and researchers about the domestic production were performed and analyzed. Many of them requested domestic OPU needles, ET catheters, culture media and ICSI pipettes under good quality control and quality assurance system. This trial may be contributed to industrialization and to global competence of Korean IVF-ET program. The results of this survey can be provide a fundamental base for development and production of domestic materials and consumables for human IVF-ET program.

Translocal and Transnational Movements of Bugis and the Construction of Multiple Identities: The Case of Bugis in North Kalimantan of Indonesia and Sabah and Johor of Malaysia

  • It is widely known that the Bugis people, originally from South Sulawesi, have been migrating to many places, including both the Indonesian and Malaysian sides of the borders today. The translocal and transnational movements of the Bugis people, especially to North Kalimantan of Indonesia and Sabah and Johor of Malaysia, have occurred in several waves, particularly during the 17th century, around 1965 and from 1980 to the present. The fall of the kingdom of Somba Opu in South Sulawesi and the rise Dutch colonial power have been the triggers for the early movement of the Bugis to both the Indonesian and Malaysian borders. This was followed by the second push of the Islamic rebellion in South Sulawesi, around 1965, creating another big wave of Bugis movement. The most recent one has been mainly due to economic reasons. These different phases of the movements, as well as the dynamic interplay of various aspects, such as citizenship, ethnic, and sub-ethnic groupings, practicing of cultural traditions and keeping the language, to mention a few, have contributed to the process of the construction of the multiple identities of the Bugis. Indeed, the Bugis people are no longer identified or identify themselves as a single group, but rather have fluid and contesting identities. This paper will discuss three main issues: the history of the translocal and transnational movements of the Bugis to North Kalimantan, Sabah and Johor; the process of adaptation to these new places; and the construction of Bugis identities.

Male Factor Infertility and In vitro Fertilization-Embryo Transfer (남성불임증과 체외수정시술)

  • In vitro fertilization and embryo transfer (IVF & ET) is widely used for the males with subnormal or abnormal semen quality, as this was recommended in view of the relatively small numbers of spermatozoa required for fertilization and subsequent pregnancies could be obtained. The aim of this study is to know how the various functional parameters of spermatozoa in semen analysis affect the outcome IVF. This study was carried out between 1988-1989, with male factor patients selected on the basis of the semen quality. The selection criteria was based upon the mean values of concentration,% motility and % normal morphology from at least two semen analysis. There is a significant decrease in the fertilization and embryo transfer rates in the study group compared with control group (35.9% vs. 68% and 48.6% vs. 85.5% respectively), however, there was no significant difference in the pregnancy or delivery rates (19.6% vs. 21.4% and 60.0% vs. 62.5% respectively) per embryo transfer cycles. Fertilization rate is variously affected by the type and degree of sperm defect. No pregnancy was occurred in triple defect group and asthenoteratospermia group. There is no significant increase in the abortion rate in the male factor group. Improvement have to be made with the fertilization rate, as the pregnancy rate per OPU cycle in male factor group is still lower than that of normal group (9.5% vs. 18.3%). In conclusion, IVF can be used as a treatment for male factor infertility and the preparation of the semen sample can be modified to improve sperm recovery and obtain fertilization from abnormal semen samples.

2002년도 국내에서 실시된 가축의 수정란이식 현황

  • 2002년도 국내에서 실시된 가축의 수정란이식 현황을 파악하기 위하여 전국의 수의ㆍ축산분야의 대학, 국ㆍ공립 축산관련 연구소, 농업기술센터, 가축인공수정소 및 동물병원 등의 168개 관련기관에 2002년 1월 1일부터 12월 31일까지 소, 돼지 및 기타 동물의 수정란 생산, 이식 및 임신진단 결과에 대하여 설문서를 통하여 조사하였으며, 설문서를 작성하여 제출한 32개기관의 자료를 분석한 결과는 다음과 같다. 가축의 수정란이식을 실시하고 있는 기관은 국립기관 4개소, 지방자치단체 10개소, 대학 7개소, 생산자단체 3개소, 개인시술소 148개소로 전체 172개소였다. 한우 및 젖소 185두를 과배란처리하여 회수된 난자수는 1,334개였으며, 그 중 이식가능수정란수는 871개로 두당 평균 4.7개였다. 체외수정의 생산은 OPU유래 266개, 도축난소 유래 16,650개, 복제수정란 21,852개로 38,768개의 이식가능 체외수정란을 생산하였다. 한우에서 수정란이식은 체내수정란 475두, 체외수정란 7,515두, 복제수정란 257두 등 8,247 두가 이식되었고, 젖소에는 체내수정란 68두, 체외수정란 58두, 복제수정란 435두 등 294두가 이식되었다. 이식된 수정란의 상태별로는 신선수정란이 73.9% (6,511두), 동결수정란이 26.1%(2,297두) 이식되었고, 수란우의 품종별로는 한우 수정란을 한우 수란우에 690두, 젖소 수란우에 7,557두가 이식되었고, 젖소 수정란을 젖소 수란우 81두, 한우 수란우에 480두가 이식되었다. 수정란이식 수란우의 수태율은 임신진단이 이루어진 체내수정란이식은 33.8% (160/472두), 체외수정란이식은 34 7%(1,547/4,456두), 복제수정란이식은 10.5%(70/669두)였다. 소의 수정란이식두수는 2001년도에 비해 217%(8,808/4,052두)가 증가하였다. 돼지에서 체내수정란은 공란두 30두에서 505개의 이식가능수정란을 회수하였으며, 체외수정란은 351,777개를 생산하였고, 형질전환수정란을 수란돈 20두에 1,919개를 이식하였었다. 염소에서는 7두의 공란축으로부터 47개의 이식가능 수정란을 회수하였다.
