The role of porous graphite plate for high quality SiC crystal growth by PVT method (고품질 4H-SiC 단결정 성장을 위한 다공성 흑연 판의 역할)

  • The present research is focused on the effect of porous graphite what is influenced on the 4H-SiC crystal growth by PVT method. We expect that it produces more C-rich and a change of temperature gradient for polytype stability of 4H-SiC crystal as adding the porous graphite in the growth cell. The SiC seeds and high purity SiC source materials were placed on opposite side in a sealed graphite crucible which was surrounded by graphite insulator. The growth temperature was around $2100{\sim}2300^{\circ}C$ and the growth pressure was 10~30 Torr of an argon pressure with 5~15 % nitrogen. 2 inch $4^{\circ}$ off-axis 4H-SiC with C-face (000-1) was used as a seed material. The porous graphite plate was inserted on SiC powder source to produce a more C-rich for polytype stability of 4H-SiC crystal and uniform radial temperature gradient. While in case of the conventional crucible, various polytypes such as 6H-, 15R-SiC were observed on SiC wafers, only 4H-SiC polytype was observed on SiC wafers prepared in porous graphite inserted crucible. The defect level such as MP and EP density of SiC crystal grown in the conventional crucible was observed to be higher than that of porous graphite inserted crucible. The better crystal quality of SiC grown using porous graphite plate was also confirmed by rocking curve measurement and Raman spectra analysis.

The Outbreak, Maintenance, and Decline of the Red Tide Dominated by Cochlodinium polykrikoides in the Coastal Waters off Southern Korea from August to October, 2000 (2000년 여름 남해안에 나타난 Cochlodinium polykrikoides 우점 적조의 발생 특성)

  • We investigated the outbreak, maintenance, and decline of the red tide dominated by C. polykrikoides in the coastal waters off Southern Korea from August to October, 2000, by combining field data and NOAA satellite images. In general, the C. polykrikoides blooms, which have occured annually in Korean coastal waters from 1995 to 1999, initiate between late August and early September around Narodo Island and expand to the whole area of the southern coast. However, initiation and short-term change of the bloom of 2000 were quite different from the pattern observed previously. In mid-August, thermal fronts in sea surface temperature(SST) were formed: 1) between the Tsushima Warm Current Water (TWCW) and the Southern Korean Coastal Waters (SKCW), 2) between the jindo cold water mass and the southwestern coastal waters, and 3) between the upwelled cold waters in the southeast coast and the offshore warm waters. Free-living cells of C. polykrikoides were concentrated in these frontal regions. In late August, the thermal front TWCW-SKCW approached the mouth of Yeosuhae Bay where Seomjin River water and anthropogenic pollutants from the Industrial Complex of Gwangyang Bay are discharged. In the blooms of 2000 initiated in Yeosuhae Bay in late August, the dominant species, C. polykrikoides, co-occured with Alexandrum tamarense, Gymnodinium mikimotoi, Skeletonema coastatum, and Chaetoceros spp. Two typhoons, 'Prapiroon' and 'Saomai' during and the C. polykrikoides bloom probably affected the abundance of this species. After the former typhoon passed the Korean Peninsula, cell growth of C. polykrikoides was maximal, but after the latter typhoon, the C. polykrikoides bloom disappeared (20 September). On 5 October, the blooms dominated by C. polykrikoides broke out within the coastal waters of Jinhae Bay and Hansan-Keoje Bay. NOAA satellite images showed that the isothermal line of 22$^{\circ}C$ extended into Jinhae Bay. In this bloom, C. polykrikoides also occurred simultaneously with Akashiwo sanguinea(=Gym-nodinium sangunium), a common red tide-forming dinoflagellate species in fall and winter in these coastal bays.

The Effects of the Fruits of Foeniculum vulgare on Skin Barrier Function and Hyaluronic Acid Production in HaCaT Keratinocytes (HaCaT 세포에서 회향 열매의 피부장벽기능과 hyaluronic acid 생성에 미치는 영향)

  • Foeniculum vulgare (FV) has long been used in traditional medicine for the treatment of inflammatory diseases. In addition, it is usually known as an important medicinal and aromatic plant widely used as a carminative, digestive, lactogogue, and diuretic, and for treating respiratory and gastrointestinal disorders. The skin barrier protects against the invasion of pathogens, fends off chemical and physical assaults, and protects against extensive water loss. In this study, the effects of solvent-fractionated FV fruits on strengthening the skin barrier and maintaining moisture, as well as their antifungal activity, were investigated in human keratinocyte (HaCaT) cells. The expression of involucrin, loricrin, filaggrin, hyaluronic acid synthase, human β defensin, and cathelicidin genes and proteins was measured by reverse transcription polymerase chain reaction (RT-PCR) and western blotting. The production of hyaluronic acid was determined by enzyme-linked immunosorbent assay (ELISA). The butanol fraction increased the expression of involucrin and filaggrin. Both the ethyl acetate and the butanol fractions increased hyaluronic acid production by promoting the expression of hyaluronic acid synthase-1. Although the antimicrobial peptides were increased by FV crude extract and its fractions, the samples did not show a significant effect compared to the normal group. These results suggest that the butanol fraction of FV could be very useful in cosmetics for the treatment of dermatological diseases.

Optogenetics: a New Frontier for Cell Physiology Study (광유전학: 세포 생리 연구를 위한 새로운 frontier)

  • Optogenetics is the combination of optical and molecular strategies to control designated molecular and cellular activities in living tissues and cells using genetically encoded light-sensitive proteins. It involves the use of light to rapidly gate the membrane channels that allows for ion movement. Optogenetics began with the placing of light-sensitive proteins from green algae inside specific types of brain cells. The cells can then be turned on or off with pulses of blue and yellow light. Using the naturally occurring algal protein Channelrhodopsin-2 (ChR2), a rapidly gated light-sensitive cation channel, the number and frequency of action potentials can be controlled. The ChR2 provides a way to manipulate a single type of neuron while affecting no others, an unprecedented specificity. This technology allows the use of light to alter neural processing at the level of single spikes and synaptic events, yielding a widely applicable tool for neuroscientists and biomedical engineers. An improbable combination of green algae, lasers, gene therapy and fiber optics made it possible to map neural circuits deep inside the brain with a precision that has never been possible before. This will help identify the causes of disorders like depression, anxiety, schizophrenia, addiction, sleep disorder, and autism. Optogenetics could improve upon existing implanted devices that are used to treat Parkinson’s disease, obsessive-compulsive disorder and other ailments with pulses of electricity. An optogenetics device could hit more specific subsets of brain cells than those devices can. Applications of optogenetic tools in nonneuronal cells are on the rise.

Storage Quality of Ready-to-Eat Campbell Table Grapes as Affected by Active Modified Atmosphere Packaging (기체충진 포장조건에 따른 신선편이 캠벨 포도의 저장 중 품질변화)

  • The storage quality of ready-to-eat Campbell table grapes which were packaged under modified atmospheres was investigated in order to examine the effect of high $O_2$ and $CO_2$ on the fruit. Fresh table grapes with 10-15 berries were packed into polypropylene (PP) trays and were top-sealed with polythylene terephthalate/PP film. The initial gas compositions inside the packages were air, 20% $O_2$/10% $CO_2$/70% $N_2$, and 40% $O_2$/60% $N_2$. Sealed packages with low density polyethylene film bags and perforated PP trays were also used as a further treatment and control, respectively. The quality attributes were assessed during storage at $5^{\circ}C$ for 28 days. Fruit packaged in high $CO_2$ concentration showed the lowest viable cell counts of inherent microorganisms among all samples, although they suffered from severe off-flavors. High levels of $O_2$ significantly lowered flesh weight loss and maintained the flavor of grape. In an overall sensory aspect, the high $O_2$ and $CO_2$ packages exhibited greater scores than the air and control at the end of the storage period. Other quality attributes showed no significant differences among treatments. Results suggest that packaging with an appropriate combination of high $O_2$ and $CO_2$ can be used as an effective processing treatment for improvement of the storability of ready-to-eat table grapes.

The effects of continuous and intermittent compressive pressure on alkaline phosphatase activity of MC3T3-E1 cells (지속적 및 간혈적 가압력이 MC3T3-E1 세포의 Alkaline phosphatase 활성도에 미치는 영향)

  • The propose of this study was to evaluate the difference of cellular activity dependent on intermittent compressive force by determining the alkaline phosphatase activity. Alkaline phosphatase activity was measured on control and experimental groups every 24, 48, 72hours. Experimental groups consisted of continous and intermittent compressive group which were compressed by $300gm/cm^2$ of diaphram pump. The intermittent compressive group was connected by timer which was worked on 10 minutes an off 10 minutes. The results were as follows; 1. The alkaline phosphatase activity between control and experimental groups showed not significant difference at compressed 24 hours. 2. The alkaline phosphatase activity of experimental groups were more increased than control group at compressed 48 hours. 3. The alkaline phosphatase activity of intermittent compressive group showed significant increased to control group. Whereby continuous compressive group showed not significant difference to control at 72 hours. 4. The alkaline phosphatase activity of intermittent compressive group were stringly increased than continuous compressive groups. 5. Between experimental groups and control group no other morphologic changes were detected by microscopic findings.

Study on the Improvement of Bovine Somatic Cell Nuclear Transfer Technique (소 체세포 핵이식기술의 효율 증진에 관한 연구)

  • This study was conducted to examine the effects of oocyte maturation period, phytohemagglutinin-P (PHA-P) treatment and activation agent on the enucleation, fusion, activation or in vitro development of bovine nuclear transfer embryos. Bovine oocytes were enucleated at 16∼24 h of in vitro maturation (IVM). Adult ear skin cells treated or non-treated with PHA-P were transferred into enucleated oocytes. Reconstituted oocytes treated or non-treated with PHA-P were fused by a pulse of 1.5 kV/cm for 30 $\mu$sec. Fused oocytes were activated with a combination of calcium ionophore (A23187) and cycloheximide (CHXM) or dimethylaminopurine (DMAP), and cultured in vitro for 7∼9 days. Enucleation rate was significantly increased when oocytes were matured for 16∼18 h (70.2∼92.3%, P<0.05) compared to that of oocytes were matured for 20∼24 h (44.3∼53.4%). The location of metaphase-II plate was far off from the 1st polar body as maturation time was increased. PHA-P treatment of donor cells or reconstituted oocytes significantly improved fusion rate (P<0.05). Cleavage and blastocyst formation rates were significantly increased after activation with a combination of A23187 and DMAP (78.6% and 32.9%, respectively) compared to those of embryos activated with a combination of A23l87 and CHXM (48.5 and 15.2%, respectively). From the present result, it is suggested that high enucleation efficiency can obtained by using oocytes matured for 18 h. It also shows that PHA-P treatment can improve the fusion rate, and activation with a combination of A23187 and DMAP can enhance the embryo development.

Antimicrobial Effect of Chitosan and Chitooligosaccharides against Bacterial Diseases of Cultured Flounder (양식 넙치의 세균성 질병에 대한 키토산 및 키토올리고당의 항균효과)

  • Antimicrobial effect of chitosan and its oligosaccharides was examined on Vibrios sp., Edwardsiella tarda and Streptococcus sp., which are major pathogenic bacteria inducing bacterial diseases of acquacultured flounder. Chitosan oligosaccharides (COS ) were produced by enzymatic hydrolysis of chitosan in an ultrafiltration mombrane bioreactor system which was established with three membranes with different molecular weight cut-off (MWCO) 1,000, 5,000 and 10,000, and fractionated into three kinds of COS, based on their molecular weight sizes. The three kinds of COS were as follows : relatively high molecular weight COS [HMW-COS, molecular weight distribution of 7,000 to 24,000 Da〕, medium molecular weight COS 〔MMW-COS, 1,500 to 6,000 Da〕, and low molecular weight COS 〔LMW-COS, 1,000 to 1,500 Da). Chitosan and HMW-COS effectively inhibited the growths of Vibrio sp. and Streptococcus sp. and their antimicrobial activities were superior to the others with smaller molecular weights. This result suggested that antimicrobial effect of chitosan preparations extremely depend on their molecular weight sizes. Antimicrobial effect of chitosan and HMW-COS on E. tarda was improved by longer inoculation times. Scanning electron microscopy in morphological change of E. tarda treated with chitosan preparations showed that chitosan and HMW-COS bound to the cells and suppressed the growth of the cells. This observation appears to prove the fact that positive charged amines of chitosan electrostatically bind to negative charged compounds of cell walls.

High Voltage Performance of the Electrical Double Layer Capacitor with Various Electrolytes (다양한 전해액을 적용한 전기이중층 커패시터의 고전압 특성 연구)

  • Electric double layer capacitors (EDLC: electric double layer capacitors) have drew attention as an energy storage device for the next generation because of their outstanding power capability and durability. But their usage is somewhat limited due to low energy density over secondary batteries. One of methods to improve the energy of EDLC is expanding the voltage window of cell operation by increasing the charge cut-off voltage. In this study, $SBP-BF_4$ (spirobipyrrolidinium tetrafluoroborate), $TEA-BF_4$ (tetraethylammonium tetraflouroborate) and $EMI-BF_4$ (1-ethyl-3-methylimidazolium tetrafluoroborate) in AN (acetonitrile) were selected to evaluate the possibility of application at high voltage environment. The LSV (linear sweep voltammetry) measurements showed that the 1.5M SBP-BF4/AN electrolyte was stable over a wide potential window and showed the best electrochemical performance compared to other combinations of electrolytes at high voltage environments (over 3.0 V). Furthermore, TMSP (tris(trimethylsilyl) phosphite) was applied to 1.5M SBP-BF4/AN in order to maintain stable performance at high voltage for the long period of time. The electrolyte with TMSP additive showed the capacity retention of 93% after 10,000 cycles at 3.3 V.

A Study Regarding Bacterial Contamination of Surfaces in Dental Offices (치과진료실 내 표면 세균 오염에 관한 연구)

  • This study studied samples taken off surfaces at three sites (Unit chairs, light handles, cuspidors) of 19 dental hospitals and 28 clinics located in Gyeonggi-do and Incheon, South Korea. The bacterial contamination levels of surfaces were $44.82{\times}10^3CFU/mL$ in cuspidors, higher than in unit chairs ($5.47{\times}10^3CFU/mL$) and light handles ($16.28{\times}10^3CFU/mL$). The values were statistically higher at dental hospitals than at dental clinics, the greater number of patients being associated with the higher bacterial cell count in the cuspidors. The results of identifying the strains isolated purely from surfaces at dental clinic showed Gram positive 47.3% and Gram negative 52.7%. Among Gram positive, the most numberous bacteria were Micrococcus luteus (10.9%), Bacillus pumilus (3.6%), and Staphylococcus aureus (3.6%). Among Gram negative, the most numberous bacteria were Acinetobacter ursingii (5.5%), Brevundimonas diminuta (4.5%), Chryseobacterium (Flavo.) indologenes (CDC IIb) (4.5%), and Methylobacterium sp. (4.5%). This study measures the level of bacterial contamination and identifies the strains isolated in dental clinics. It recognizes the importance of infection control, and the results of the study may be considered as the basis for establishing specific plans for prevention of infection.
