Effect of Environment Factors on Growth and Mortality of Cupped Oyster, Crassostrea gigas (양식장 환경이 갯벌양식 굴 (Crassostrea gigas)의 성장과 폐사에 미치는 영향)

  • In order to evaluate how the effects of aquacultural environment, such as temperature, salinity, DO, SS, Chlorophyll-a, COD and nutritive salts on Cupped oysters, an investigational study was conducted between September to December of 2011. During the study, different intermediate cage farms on the surface of the sea were used to culture and compare the growth and survival rate of the Cupped oysters in different fishing environments. The different intermediate cage farms used were Wonsando-ri, Chang-ri, and Pado-ri. In January of 2012, these oysters were transplanted to a horizontal net farm in a mudflat until July of that year. The adaptation rates of the Cupped oysters were tested at differential exposure times at varying intertidal periods. Wonsando-ri showed the highest water temperature and Chlorophyll-a levels, while the salinity was found to be within range of stable conditions among all three intermediate cage farms. Once the Cupped oysters were at the horizontal net farm, the growth was measured at distinct tidal exposure time of 1, 3, and 5 hours, whereby the growth rate was highest at 3, 1, and 5 hours, respectively. In addition, the oysters cultivated in intermediate cage farms had longer shell lengths compared to shell heights, while oysters cultured in the horizontal net farm had larger shell heights than shell lengths.

  • To investigate the variations of physico-chemical factors and microbial population, in ten stations at water region of coastal area of Chagwi-Do, Nutritive salts, water temperature, transparency, suspended solid, salinity, COD, DO, pH, heterotrophic bacteria, coliform group and Vibrio spp. were analysed three times in September, November in 2004 and February in 2005. Heterotrophic bacteria in surface water was $3.5X10^1{\sim}1.16X10^3cfu/ml,\;1.0X10^2{\sim}5.2X10^1cfu/ml\;2.0X10^1{\sim}7.6X10^1cfu/ml$ and bottom water counted $7.0X10^2{\sim}1.0X10^3cfu/ml,\;1.4X10^1{\sim}2.5X10^2cfu/ml\;2.0X10^2{\sim}4.2X10^1cfu/ml$ in September, November in 2004 and February in 2005, respectively. The cell number of total coliform bacteria in the surface water amounted to $0{\sim}4.3X10^2cfu/ml,\;0{\sim}6.0X10^1cfu/ml,\;0{\sim}1.0X10^1cfu/ml$ and bottom water amounted $0{\sim}2.2X10^2cfu/ml,\;0{\sim}5.4X0^2cfu/ml,\;0{\sim}2.0X10^1cfu/ml$ in September, November in 2004 and February in 2005, respectively. As for Vibrio spp., the cell number in the surface water was $1.0X10^1{\sim}2.5X10^2cfu/ml,\;1.0X10^1{\sim}2.0X10^1cfu/ml,\;0cfu/ml$ and bottom water counted $1.0X10^1{\sim}5.2X10^2cfu/ml,\;0cfu/ml,\;2.0X10^1cfu/ml$ in September, November in 2004 and February in 2005, respectively.

Advanced Treatment of Piggery Slurry Using Micro Ozone Bubble, UV, Ultra Sonic and Hydroxy Peroxide (미세기포화 오존과 자외선, 초음파, 과산화수소를 이용한 돈분뇨 슬러리 고도처리)

  • The aim of this study was to investigate the removal effects of the color, nutritive salts and other pollutants on piggery slurry by advanced oxidation process (AOP) system. The experimental AOP system was designed to treat 300 L of piggery slurry per hour. To enhance oxidizing power of the experimental APO system, a ultraviolet irradiation system and the ultrasonic system were attached to the AOP system. With 5 min ultrasonic treatment, COD, SS and T-N concentrations were changed from 210, 820, and 309 to 200, 760, and 262 mg/L, respectively. With 10 min ultrasonic treatment, SS and T-N concentrations tended to decrease but T-P concentration was not changed. With the treatment of both ozone and ultrasonic waves for 30 min, COD, SS, T-N and T-P decreased from 238, 900, 400, and 5 to 165, 540, 263, and 4 mg/L, respectively. With the treatment of both ozone and ultraviolet irradiation for 30 min, COD, SS, T-N and T-P decreased from 321, 340, 204, and 15 to 151, 140, 111, and 7 mg/L, respectively, and color was changed from 4,344 to 624.

The dominant species isolated from the coastal area of Chagwi-Do were identified to be Vibrio spp., Pseudoalteromonas spp. Psuedomonas spp, Bacillus spp., Alteromonas spp., Aeromonas spp., Psychrobacter spp., and Flavobacterium spp.

Analysis of the Water Quality Change Due to Water Level Control of Sayeon Dam (사연댐 수위조절시 수질변화 분석)

  • The Bangudae Petroglyphs, national treasure No. 285 is located within submerged upper districts of Sayeon dam supplying the main residential water in Ulsan. Of the many ways for the reservation of Petroglyphs located the altitude at 53~57 m, the plan that we take it out of the water lowering the water level from 60 m to 52 m has been examined mainly in case of controlling artificially the water level of the dam. In this paper, we examined expected problems from the loss of dam function and the change of water quality from water deterioration caused by the water level control of the Sayeon dam. Using the model of Vollenweider and CSTR (Continuous Stirred Tank Reactor), we analyzed the density change of BOD and COD, representative water quality index and the TP and TN, the main reason of algae growth. The result showed that the density of COD lowered a little but the density of TP and TN went up over 130% when controlling the water level from 60 m to 52 m. These changes cause a serious algae problem and if doing the water quality management as the density of TN and TP, the water quality would become worse. Water storage and supply residential water decreases, and the water quality becomes worse because of eutrophic state.

Effects of Grassed Swale Lengths on Reduction Efficiencies of Non-point Source Pollutants (식생수로 길이가 비점오염물질 저감효율에 미치는 영향)

  • Non-point pollution source is difficult to control due to uncertain outflow path and emission. So, There are many development and research to Best Management Practices(BMP) established to manage the Non-point pollution source. Besides, various methods of estimated efficiency to exact assessment of BMP is presented. In this study, the impact about length of Grassed Swale on reduction efficiency based on monitoring results of Grassed Swale by length is studied. By estimating Grassed Swale reduction efficiency in a variety of methods, the difference between the methods of estimated efficiency was compared with those that. Estimated efficiency method using ER, SOL, ROL, ROF, SOLF, and ROLF methods is analyzed. EMC analysis result is high inflow and outflow concentration distinction organic compound for nutritive salts The result of efficiency analysis along Grassed Swale length sharply increases in a Grassed Swale inlet. After this increase, the efficiency gradually decreases. This is expected that cistern installed in the end of the front. To obtain a stable reduction efficiency of Grassed Swale, minimum length 30m of Grassed Swale should be enough. Also, in order to efficiently and economically design Grassed Swale, the researches on length of Grassed Swale are needed rather than simple analysis of efficiency.

The Effect of Media Application in Aeration Tank for Aerobic Treatment of Swine Slurry (돈분뇨슬러리 폭기시 담체 설치효과)

  • This study was performed to evaluate the removal effects of nutritive salts and organic pollutants in experimental aeration reactor for treatment of piggery slurry. In this study, three types of reactors were manufactured and operated. The fibrous media was equipped in one of three reactors. Another reactor was equipped with the siliceous media and the other reactor used as a control was equipped with typical aeration system only. Treatment efficacy of three types of reactors were evaluated according to the pollutants removal rate of the piggery slurry. The results obtained in this study are as follows : 1) In the reactor containing fibrous media, the removal efficiency of BOD, T-N and T-P was 11%, 13.9% and 21.2%, respectively. 2) In the reactor containing siliceous media, the removal efficiency of BOD, T-N and T-P was 6.9%, 25.3% and 47.8%, respectively. 3) In the reactor not containing media, the removal efficiency of BOD. T-N and T-P was 6.1%, 8.1.% and 23.6%, respectively. 4) Sludge accumulation in the reactor equipped with filamentous media was lower than that of other experimental reactors.

STUDIES ON THE CYSTINE COMPONENT IN THE SERICULTURAL PROTEINS OF BOMBYX MORI L. (가잠사단백질의 각과정에서의 Cystine 성분에 대한 연구)

  • The purpose of this treatise is to prove the presence of cystine in silk fiber through wide sampling throughout all the sericultural processes of Bombyx mori.; also to show that disulfide cross linkages exist in the silk fiber. The conclusions reached were as follows: 1. Crystalline cystine was obtained from silk fibroin using Folin's Method. 2. Analytical data showing the cystine content of silk fiber and its related materials were obtained using Sullvan's Method as follows: Material Percent Cystine A. Mulberry leaf protein 0.175 B. Silkworm egg 0.33 C. Silkworm Body, matured, fat extracted, without silk gland 0.41 D. Silk gland, matured 1.23 E. Silkworm feces none F. Silkworm pupa, fat extracted 0.30 G. Silkworm moth, fat extracted 0.60 H. Raw Silk 0.22 I. Fibroin 0.175 J. Sericin 0.30 3. The presence of cystine in the silkworm was substantiated the existence of 0.175 % methionine in mulberry leaves and 0.12% methionine in the silk gland. 4. Part of the sulfhydryl compounds in the silk gland is believed to transfer to serine and methionine, with the former being secreted into the liquid silk finally as silk fiber and the latter used for nutritive purposes in the growing of silk gland tissue. 5. The cystine content is variable by mulberry species, silkworm species, sex, breeding process, and other culturing environments. 6. Hybrid silkworms require more nutritive amino acids for effective growth than the original parents, and secrete less of them as silk fiber. 7. From such an observation, the amino acid composition of silk fiber is believed to be fairly flexible. Cystine if included in the amorphous part of the fiber, especially in sericin. 8. The result from enriching the silkworm diet with pure cystine or wool cystine did not result in any advantage, therefore it is believed that the natural cystine and methionine contents in the mulberry leafaregoodenoughforsilkwormnutrition. 9. The disulfide cross linkage in silk fiber was verified by using the Harris Method. Contraction took place following the treatment of the fiber with various salts and acids. Comparisons were made with wool fiber. 10. During these experiments, the fibrious structure of silk fiber and the net-globular liquid form were photographed microscopically. It is believed that the globules of liquid silk are net-formed by the inter attraction of the OH ion of the globular peptide and the H ion of water as shown by the hair cracking behavior of the film. The net-globular protein precipitation from the mulberry protein solution showed that mulberry is a proper diet for the formation of fibrous protein in the silk fiber. 11. The significance of the presence of cystine in silk fiber as emphasized in this paper should result in modification of the general conception that cystine is absent from this fiber. Water Quality Improvement of Stagnant Water using an Upflow Activated Carbon Biofilm Process and Microbial Community Analysis (상향류 활성탄 생물막 공정을 이용한 정체 수역 수질 개선 및 공정 내 미생물 군집 해석)

  • The capacity of natural purification was limited by the interruption of natural flow and the problems such as eutrophication were occurred by nutritive salts accumulation in stagnant stream. Moreover, the inflow of non-point sources causes non-degradable materials to increase in stagnant stream. In this study, an upflow biological activated carbon (BAC) biofilm process comprised of anoxic, aerobic 1, and aerobic 2 reactors were introduced for treatment of stagnant stream and SS, $BOD_5$, $COD_{Mn}$, $COD_{Cr}$, TN, and TP were monitored in the upflow BAC biofilm reactors with continuous cycling. In order to simulate stagnant stream, the lake water of amusement park and golf course were stored as influent in a tank of $2m^3$ and hydraulic retention time (HRT) was changed into 6, 4, and 2 hours. At HRT 4hr and the lake water of amusement park as influent, the removal efficiencies of SS, $BOD_5$, $COD_{Mn}$, $COD_{Cr}$, TN, and TP showed the best water quality improvement and were 69.8, 83.0, 91.3, 74.1, 74.7, and 88.9%, respectively. At HRT 4hr and the lake water of golf course as influent, the removal efficiencies of SS, $BOD_5$, $COD_{Mn}$, $COD_{Cr}$, TN and TP were 78.5, 78.0, 80.2, 74.9, 55.6 and 97.5%, respectively. As the results of polymerase chain reaction - denaturing gel gradient electrophoresis (PCR-DGGE), microbial community was different depending on influent type. Fluorescence in situ hybridization (FISH) showed that nitrifying bacteria was dominant at HRT 4 hr. The biomass amount and microbial activities by INT-DHA test were not decrease even at lower HRT condition. In this study, the upflow BAC biofilm process would be considered to the water quality improvement of stagnant stream.

Conversion of Fermented Feed by Basidiomycetes (담자균(擔子菌)을 이용(利用)한 발효(醱酵) 사료(飼料)에 관한 연구(硏究))

  • To investigate nutritive values of a feed fermented with basidiomycetes, among the isolated strains, Lyophyllum decastes (Fr.) Sing. was found with the greatest enzyme productivity and rapid mycelial growth in rice straw medium. Optimum temperature, pH and moisture content for mycelial growth and enzyme production of the strain were $25{\sim}30^{\circ}C,\;pH\;4.0{\sim}7.0\;and\;70{\sim}75\;%$, respectively. Fifteen days of culture were required for the highest enzyme productivity. Among the sub-materials added, $30{\sim}40\;%$ of rice bran and $10{\sim}20\;%$ of defatted perilla seeds were effective for the enzyme production, but caused a reduced mycelial growth. The greatest effect of an addition of inorganic salts was obtained with $0.36{\sim}0.72\;%\;of\;(NH_4)_2HPO_4$. When 40 mesh or smaller rice straw and steam treatment at $0.5\;kg/cm^2$ were used, the mycelial growth decreased, whereas the enzyme production increased. The mycelial growth and enzyme production increased when $Ca(OH)_2$ was used as the alkali treatment, but decreased with increasing concentration of NaOH. As the fermentation proceeded, the amounts of ash, reducing sugar and total nitrogen increased, but cellulose, lignin and pentosan decreased. When the rice straw was treated with alkali, the amounts of ash, total nitrogen and lignin decreased, but reducing sugar and cellulose increased. At higher NaOH concentration, the variation become greater. The in vitro dry matter digestibility of the products increased from 55.03 % at the beginning of the fermentation to 62.72 % at 45 days after fermentation. The most effective alkali treatment on the digestibility of rice straw was KOH followed by NaOH. However, the digestibility increased with increasing concentration of NaOH. The digestibility of pretreated with alkali increased after fermentation as well.
