대두의 조리 가공에 따른 Phytate 함량 및 단백질 소화율 (Effect of Cooking and Processing on the Phytate Content and Protein Digestibility of Soybean)

  • 대두식품의 가공 및 발효에 따른 phytate 함량의 변화, 그 함량이 pepsin과 pancreatin에 의한 단백질 소화에 미치는 영향, 그리고 전기영동법에 의한 단백질 분획의 변화를 조사하였다. 황색콩의 phytate 함량은 2.4%이었고 가공 및 발효에 따라 감소하여 두유 0.2%, 두부 0.7%, 비지 0.4%로, 청국장 0.9%, 메주 1.4%, 간장 0.2%, 된장 1.0%로 나타났다. Phytate/protein 비율은 pepsin에 의한 단백질 소화율과 상관성이 없었으나 pancreatin에 의한 단백질 소화율과는 부(負)의 상관관계를 나타냈다(p<0.01, r= -0.73). 전기영동 결과 콩을 물에 불리면 수용성 분획이 변화되었고 두유 제조시 비지에는 주로 저분자량 밴드들이 새로이 나타났다. 발효 과정에서는 단백질이 분해되어 밴드가 거의 나타나지 않았다.

Nicotinic acid changes rumen fermentation and apparent nutrient digestibility by regulating rumen microbiota in Xiangzhong black cattle

  • Objective: The aim of this study was to investigate the impact of dietary nicotinic acid (NA) on apparent nutrient digestibility, rumen fermentation, and rumen microbiota in uncastrated Xiangzhong black cattle. Methods: Twenty-one uncastrated Xiangzhong black cattle (385.08±15.20 kg) aged 1.5 years were randomly assigned to the control group (CL, 0 mg/kg NA in concentrate diet), NA1 group (800 mg/kg NA in concentrate diet) and NA2 group (1,200 mg/kg NA in concentrate diet). All animals were fed a 60% concentrate diet and 40% dried rice straw for a 120-day feeding experiment. Results: Supplemental NA not only enhanced the apparent nutrient digestibility of acid detergent fiber (p<0.01), but also elevated the rumen acetate and total volatile fatty acid concentrations (p<0.05). 16S rRNA gene sequencing analysis of rumen microbiota revealed that dietary NA changed the diversity of rumen microbiota (p<0.05) and the abundance of bacterial taxa in the rumen. The relative abundances of eight Erysipelotrichales taxa, five Ruminococcaceae taxa, and five Sphaerochaetales taxa were decreased by dietary NA (p<0.05). However, the relative abundances of two taxa belonging to Roseburia faecis were increased by supplemental 800 mg/kg NA, and the abundances of seven Prevotella taxa, three Paraprevotellaceae taxa, three Bifidobacteriaceae taxa, and two operational taxonomic units annotated to Fibrobacter succinogenes were increased by 1,200 mg/kg NA in diets. Furthermore, the correlation analysis found significant correlations between the concentrations of volatile fatty acids in the rumen and the abundances of bacterial taxa, especially Prevotella. Conclusion: The results from this study suggest that dietary NA plays an important role in regulating apparent digestibility of acid detergent fiber, acetate, total volatile fatty acid concentrations, and the composition of rumen microbiota.

卵白단백질에 대한 감마선 조사의 영향 (Effect of Gamma Irradiation on Egg White Protein)

  • The ovalbumins obtained from hen's and duck's egg white were irradiated with up to 100 kGy at room temperature. The purified proteins were evaluated for their in vitro digestibility by incubating successively with pepsin and pancreatin conjugate. Amino acid compositions and SDS-PAGE pattern in these proteins were also analyzed. The obtained results indicated that gamma irradiation within the tested dose range(up to 100kGy) produced no statistically significant changes in the in vitro digestibility an amino acid compositions. Analysis of gamma-irradiated ovalbumins by SDS-PAGE revealed radiolysis of ovalbumin into proteins or peptides of low molecular weights.

Effects of Extrusion Variable on Functional and Nutritional Properties of Extruded Oat Products

  • The purpose of this research was to study the effects of initial moisture levels and extrusion temperatures on dietary fiber, nitrogen solubility index, available lysine, and the in vitro protein digestibility of extruded oat productes. The dehulled grains were ground in a Brabender quadrumat Senior mill and the coarse fraction, with higher crude protein, lipids and dietary fiber were conditioned on various mositre levels (15.5~25.5%) and extruded in a Brabender single-screw laboratory extruder. The extrudates showed a higher amount of soluble dietary fiber (8.14%) than in the raw material . However, the extrusion process affected the nutritional value of the protein due to a decrease in available lysine with increased temperature . The in vitro protein digestibility was unaffected by initial moisture levels and the extrusion temperatures examined.

토끼고기의 지방질과 단백질에 관한 연구 -II. 단백질을 중심으로- (Studies on Lipids and Proteins of Rabbit Meat -II. Emphasis on quality of rabbit meat protein-)

  • The present study was carried out to evaluate the nutritional quality of rabbit meat protein. The composition of amino acids contained in rabbit meat was compared with those of other animal meats such as beef, pork and chicken. Also included in this study was the question whether the cooking and storage conditions affect the amino acid composition and the pepsindigestibility of rabbit meat protein. The results are summarized as follows: 1. The large variation observed from sample to sample of EAA (essential amino acid) composition in rabbit meat was found to be an interesting but peculiar property of rabbit meat protein. The most limiting amino acid of rabbit meat protein was phenylalanine, whereas methionine was the first limiting amino acid of both beef and pork proteins. Chemical scores of various meat proteins were 68, 65, 66, and 74 for rabbit meat, beef, pork, and chicken respectively. 2. In pan roasting, the EAA damaged most by heat was methionine (15%). When cooked after two months of frozen storage, lysine decreased most. 3. Higher pepsin digestibility was obtained by cooking rabbit meat after seasoned in alcohol, ginger juice, and other spices compared with various other cooking conditions without seasoning. The pepsin digestibility value was even higher for the seasoned meat than for the raw meat. 4. Among various meats tested the rabbit meat showed the lowest pepsin digestibility. 5. A simple measurement of released methionine could be used to determine relative digestibility instead of measuring $NH_2-N$ content after pepsin digestion. From all the results obtained in this study it can be concluded that rabbit meat is a good Protein food item when used fresh and stored properly to prevent rancidity problems. It is suggested to study further the peroxidation effect of unsaturated fatty acids on protein quality. This study was supported by the Ministry of Science and Technology in Korea.

Nutritional Evaluation of Fodder Tree Leaves with Goats

  • Two experiments were conducted to evaluate the nutritional value of common fodder tree leaves with goats fed at 50% of total ration. In experiment 1, leaves from six fodder tree species i.e., Ailanthus aitissima, Elaeagnus angustifolic, Morus alba (Mulberry), Populus spp, Robina pseudoacacia and Salix babylonia were harvested in spring and winter from northern areas of Pakistan. Chemical composition and apparent in situ dry matter digestibility (DMD) of fodder tree leaves were measured. Results showed that crude protein (CP) values were higher (p<0.05) in all the species during spring compared to winter (17.9% vs 12.0%). The concentration of NDF in Elaeagnus and Robinia was higher in spring, whereas no seasonal difference was found in other species. In situ DMD was higher (p<0.05) in Ailanthus and Populus at spring while it was higher (p<0.05) in Elaeagnus, Mulberry and Robinia at winter. There was no (p<0.05) seasonal effect on in situ DMD of Salix. In experiment II, four iso-nitrogenous and iso-caloric rations viz., A, B, C and D were prepared containing 50% (winter harvested) sun dried leaves of Salix, Robinia, Mulberry and Elaeagnus, respectively and 50% concentrate. Dry matter and crude protein intakes were higher (p<0.05) given ration A (Salix) whereas DM and CP digestibility was lowest (p<0.05) given ration B. Nitrogen retention was higher (p<0.05) given ration A. Goats fed on fodder tree leaves and concentrate showed moderate intake and digestibility.

Effects of xylanase supplementation to wheat-based diets on growth performance, nutrient digestibility and gut microbes in weanling pigs

  • Objective: This study was designed to investigate the effects of an Aspergillus sulphureus xylanase expressed in Pichia pastoris on the growth performance, nutrient digestibility and gut microbes in weanling pigs. Methods: A total of 180 weanling pigs (initial body weights were $8.47{\pm}1.40kg$) were assigned randomly to 5 dietary treatments. Each treatment had 6 replicates with 6 pigs per replicate. The experimental diets were wheat based with supplementation of 0, 500, 1,000, 2,000, and 4,000 U xylanase/kg. The experiment lasted 28 days (early phase, d 0 to 14; late phase, d 15 to 28). Results: In the early phase, compared to the control, average daily gain (ADG) was higher for pigs fed diets supplemented with xylanase and there was a quadratic response in ADG (p<0.05). In the entire phase, ADG was higher for the pigs fed 1,000 or 2,000 U/kg xylanase compared to the control (p<0.05). The gain to feed ratio was higher for pigs fed diets supplemented with 1,000 or 2,000 U/kg xylanase compared to the control (p<0.05). Increasing the amount of xylanase improved the apparent total tract digestibility of dry matter, crude protein, neutral detergent fiber, calcium, and phosphorus during both periods (p<0.05). Xylanase supplementation (2,000 U/kg) decreased the proportion of Lachnospiraceae (by 50%) in Firmicutes, but increased Prevotellaceae (by 175%) in Bacteroidetes and almost diminished Enterobacteriaceae (Escherichia-Shigella) in Proteobacteria. Conclusion: Xylanase supplementation increased growth performance and nutrient digestibility up to 2,000 U/kg. Supplementation of xylanase (2,000 U/kg) decreased the richness of gut bacteria but diminished the growth of harmful pathogenic bacteria, such as Escherichia-Shigella, in the colon.

Ruminal Characteristics, Blood pH, Blood Urea Nitrogen and Nitrogen Balance in Nili-ravi Buffalo (Bubalus bubalis) Bulls Fed Diets Containing Various Levels of Ruminally Degradable Protein

  • Four ruminally cannulated Nili-ravi buffalo bulls were used in a $4{\times}4$ Latin Square design to determine the influence of varying levels of ruminally degradable protein (RDP) on ruminal characteristics, digestibility, blood pH, blood urea nitrogen (BUN) and nitrogen (N) balance. Four isonitrogenous and isocaloric diets were formulated (NRC, 2001). The control diet contained 50% RDP. The medium (MRDP), high (HRDP) and very high (VHRDP) ruminally degradable protein diets had 66, 82 and 100% RDP, respectively. Increasing the level of dietary RDP resulted in a linear decrease in ruminal pH. A quadratic effect of RDP on ruminal pH was also observed with quadratic maxima at the 66% RDP diet. Dietary RDP had a quadratic effect on total bacterial and protozoal count with maximum microbial count at the 82% RDP diet. Increased microbial count was due to increasing level of ruminal ammonia nitrogen ($NH_3-N$). Increasing dietary RDP resulted in a linear increase in dry matter digestibility. Provision of an adequate amount of RDP caused optimum microbial activity, which resulted in improvement in DM digestibility. Increasing the level of dietary RDP resulted in a linear decrease in crude protein (CP) and neutral detergent fiber digestibility. Blood pH remained unaltered across all diets. A linear increase in ruminal $NH_3-N$ and BUN was noted with increasing level of dietary RDP. The increase in BUN was due to increased ruminal $NH_3-N$ concentrations. A positive N balance was noted across all diets. The results are interpreted to suggest that buffalo bulls can utilize up to 82% RDP of total CP (16%) with optimum results.

Nutrient Intake and Utilization by Range Managed Sheep in Critical Physiological Stages Maintained on Grazing with Concentrate Supplementation in a Hot Semi-Arid Environment

  • The reported study was conducted on range managed Malpura ewes that were non-breeding empty, were at an advanced stage of pregnancy, and were in early lactation, under a protocol of free grazing with concentrate supplementation at 1.00, 1.25 and 1.50% of their body weight to assess their plane of nutrition and nutrient intake. The biomass yield of pasture plots was 1689, 1820 and 2912 kg/ha in pregnancy, lactation and empty phases, respectively. In addition to natural shrubs and forbs, Cenchrus ciliaris (36.4%) and dead litter (31.6%) were the major component of pasture vegetation during pregnancy. The dead litter disappeared during the lactation and empty phase with a concomitant increase in distribution of Cenchrus ciliaris to 73.0 and 87.2% respectively. The daily dry matter consumption from supplemental concentrate and free grazing was 70.1, 57.3 and 63.5 g/kg $W^{0.75}/d$ with concentrate to roughage ratio of 40:60, 47:53 and 33:67 in pregnancy, lactation and empty phases respectively. Digestibility of DM and OM were similar in the three phases while CP digestibility was higher (p<0.0l) during lactation than other two phases. Digestibility of NDF, ADF and cellulose were higher (p<0.0l) in empty than pregnancy and lactation, while hemicellulose digestibility was similar in lactation and empty and lower in pregnancy phase. The ewes in ~hases of pregnancy, lactation and empty consumed 7.1, 7.7 and 6.1 g DCP and 197.2, 214.6 and 232.5 kcal DE/kg $W^{0.75}/d$ respectively. It is concluded that ewes maintained on semi-arid Cenchrus dominated pasture with concentrate supplementation during pregnancy, lactation and empty phases consumed 45.2, 45.1 and 35.2 g DCP/Mcal ME respectively.

Comparative analysis of the energy values of oat hay, tall fescue, annual ryegrass, and barnyard millet using in vivo digestibility results and predicted values

  • Accurate measurements of feed energy values have a positive effect on the control of feed intake for animals, the formation of prices, and on economic profits. However, few studies have attempted to measure the energy values of domestic roughages and to validate an energy value prediction model. The purpose of this study is to build a database through measurements of the nutrient digestibility and digestible energy results of roughages used in Korea and to evaluate the domestic applicability of the National Research Council (NRC) estimation model. Oat hay, annual ryegrass, tall fescue, barnyard millet, and concentrates for beef cattle were used in the test. As a result of an in vivo digestibility trial, the total digestible nutrients (TDN) of oat hay, annual ryegrass, tall fescue, and barnyard millet were found to be 68.73, 55.02, 55.71, and 52.89%, respectively. As a result of comparing the estimated values using the NRC equation with the in vivo results, there was a difference of 3.55 to 6.84%P in the TDN. Inferred from this result, it is considered that TDN calculations using the NRC equation can be reasonable for the test feeds used in this study. These results can be utilized when revising the Korean Standard Tables of Feed Composition, which provides a comprehensive overview of Korean feed.
