의료기관 영양서비스 현황 I : 영양부서 조직.인력체계 및 작업생산성 (Hospital Nutrition Services I : Organization, Personnel and Productivity of Nutrition Department)

  • To evaluate the infra structure supporting hospital nutrition services, we conducted a survey on the unit of organization, unit of dietitians work system, number of personnel engaged on nutrition services, productivity of food service, management of dietitians works, computerization of nutrition services etc. Total ninety-six hospitals were participated in the survey, and they were varied in terms of hospital classification, location, number of beds and type of food service management. All of the large hospitals with more than 400 beds conducted nutrition services under the department of nutrition, but some of the middle and small hospitals with less than 400 beds conducted nutrition services under the other department such as administration. In most of the tertiary hospitals, the work of dietitians were separated in which food services and medical nutrition services were conducted independently by different dietitians, whereas, in most of general hospitals and all the hospital, food services and medical nutrition services were conducted by the same dietitians in all time. The numbers of dietitians and cooks per 100 beds were fewer in the large hospitals with more than 400 beds than the hospitals with less than 400 beds, and the number of cooking and meal serving assistants were the just opposit. The average productivity of food service was 44.5 meals per hour for each dietitian, 84.8 meals per hour for a cook and 7.0 meals per hour for a cooking and meal serving assistant. The productivities for dietitians and cooks tend to be higher in large hospitals than middle and small hospitals, whereas the productivities for cooking and meal serving assistants were just opposite. The large hospitals seemed to solve the problem on the lack of working personnels by hiring part-time workers and by utilization of computer system for their works. The pattern of daily work management in food service area was not much different between dietitians duties, but the pattern of daily work management in medical nutrition service area was different in a way which the analysis of patients nutrient intakes was almost not conducted by dietitians handling both food services and medical nutrition services. Therefore, this study demonstrates that there are significant differences in the infra structures conducting nutrition services among hospitals, suggesting that the strategies to improve this improve this structure in relation to the improvement of service qualities need to be investigated in the future. Effects of Supplementing with Single or Multiple Trace Minerals on Growth Performance, Fecal Mineral Excretion and Nutrient Utilization in Pullets from 1 to 18 Weeks of Age

  • This study investigated whether supplemental Cu, Fe, Zn, and Mn are needed in a practical diet for pullets. Four hundred and twenty females of an egg-laying strain (1-d-old, Lohmann Brown Layer) were randomly distributed into 4 groups, consisting of 7 replicates of 15 birds each. During the 18-week experimental period, chicks were given three basal diets in sequence, each with single or multiple Mn, Zn and Cu supplementation to improve the mineral balance gradually. In the Control, no Mn, Zn, and Cu were added; in the single Mn supplemented group (sMn) Mn was added to 120, 60, and 60 mg/kg for 1-6, 7-12, and 13-18 weeks of age, respectively; in the multiple Mn and Zn supplemented group (mMnZn), Mn was added to 180, 90, and 90 mg/kg and Zn was added to 120, 105, and 105 mg/kg for 1-6, 7-12, and 13-18 weeks of age, respectively; in the multiple Mn, Zn, Cu supplemented group (mMnZnCu), Mn, Zn, and Cu were added to the same multiple of basal Fe concentration relative to NRC (1994) recommendations. Energy and protein metabolizability were determined by subtracting energy/protein intake by energy/protein excretion (from both feces and urine) and dividing by energy/protein intake. There were no statistically significant differences between groups in terms of feed intake, final body weight or tibia length throughout the experiment. Optimal growth performance was observed in the Control, while adding trace minerals to basal diets tended to result in decreased productive performance. Protein metabolizability was increased by mMnZn and mMnZnCu treatments, but energy metabolizability was not affected. Concentrations of Mn, Zn, Cu in excreta varied greatly related to dietary content, and the retentions of Cu, Fe, Zn and Mn were all increased due to the improvement of mineral balance. Based on these results, it is suggested that the concentrations of Cu, Fe, Zn and Mn in typical basal diets used in this study were adequate for normal growth for pullets from 1 to 18 weeks of age.

The Limiting Sequence and Proper Ratio of Lysine, Methionine and Threonine for Calves Fed Milk Replacers Containing Soy Protein

  • The limiting sequence and relative ratio of lysine (Lys), methionine (Met), and threonine (Thr) for calves about 2 mo of age fed milk replacers (MR) containing soy protein are not clearly defined. The objective of the study was to investigate the effect of supplementing MR containing 22% CP, half from soy protein concentrate (SPC, 40.56% CP, flour) and half from whey proteins, with Lys, Met, and Thr to estimate amino acid (AA) sequence and their relative ratio for calves about 2 mo of age. A method of partial deduction of AA was adopted. Twenty-four newborn calves (half males and half females, $40.7{\pm}0.9$ kg of BW) were fed 1 of 4 MR diets for 56 d (n = 6/diet). The diets were supplemented with all (positive control) or with 2 of the 3 AAs: Lys, Met and Thr, (i.e., PC (22% CP, 2.34% Lys, 0.72% Met and 1.80% Thr), PC-Lys (22% CP, 1.64% Lys, 0.72% Met and 1.80% Thr), PC-Met (22% CP, 2.34% Lys, 0.50% Met and 1.80% Thr), and PC-Thr (22% CP, 2.34% Lys, 0.72% Met and 1.26% Thr)). Calves were fed thrice daily; starter (20% CP, 1.03% Lys, 0.30% Met and 0.69% Thr), hay (3.23% CP, 0.29% Lys, 0.12% Met and 0.23% Thr) and water were offered free choice. Starter and hay were only offered beginning on d 36 (after 5 wk) and d 43 (after 6 wk), respectively. BW, body size and blood samples measures were taken every two weeks. Three-day total collection of feed refusals, feces, and urine were recorded starting at d 33 and d 54 of age, respectively. From the results, the limiting sequence and relative ratio between the 3 AAs in calves with different diet structures were calculated. The limiting sequence of the 3 AAs were ranked as Lys, Met and Thr; the proper ratio was 100:29:70 for MR-only diet and 100:30:60 for diets consisted of MR, starter and hay. Nitrogen digestion and utilization and nutrient digestibility were negatively affected by AA deletion treatments. From the evidence of this experiment, it did not appear that the AA limiting sequence was selectively altered by differences in diet structures such as would be encountered in practice. The relative ratio between the 3 AAs varied with the offer of starter and hay to calves, and the average ratio was 100:29.5:65 for calves during 2 to 10 wk of age.

청국장으로부터 분리한 Poly(γ-glutamic acid)를 생산하는 균주 Bacillus subtilis GS-2의 분리 및 γ-PGA의 확인 (Isolation of Bacillus subtilis GS-2 Producing γ-PGA from Ghungkukjang Bean Paste and Identification of γ-PGA)

  • Poly-${\gamma}$-glutamic acid (${\gamma}$-PGA)는 청국장이 발효할 때 생성되는 점질성 물질의 하나로, monomer glutamic acid의 ${\alpha}$-아미노기와 ${\gamma}$-카르복실기 사이의 amide linkage에 의해 결합된 D(-)와 L(-) glutamic acid repeat units로 이루어진 homopolymer이다. 주로 Bacillus sp.에 의해 생산된다. ${\gamma}$-PGA는 수용성, 음이온성, 무독성, 생분해성, 생체적 합성, 식용 등의 다양한 특성을 가지고 있기 때문에 여러 분야에서 응용되고 있다. 본 실험에서 ${\gamma}$-PGA를 생산하는 균주를 우리나라의 전통발효식품인 청국장으로부터 분리하였다. 분리균주의 형태 및 배양학적, 생리학적 특성, API kit 및 16S rRNA 서열을 사용하여 동정한 결과, 분리균주 GS-2는 B. subtilis와 가장 유사하여 B. subtilis GS-2로 명명하였다. 분리 정제된 ${\gamma}$-PGA의 동정은 TLC, HPLC, FTIR 그리고 $^1H$-NMR spectroscopy를 통하여 확인하였다.

물리·화학적 돌연변이 유도를 통한 Paracoccus haeundaensis의 astaxanthin 생산량 증대 (Enhanced Production of Astaxanthin in Paracoccus haeundaensis Strain by Physical and Chemical Mutagenesis)

  • Carotenoid는 천연 지용성 색소이며, 세균, 조류, 식물 등이 생산한다. 세계 시장의 대부분을 차지하는 합성 염료의 대안으로서 현재는 조류나 세균, 갑각류 등의 원료로부터 아스타잔틴의 생산, 정제, 이용이 주목 받고 있다. 이 연구는 UV와 EMS를 이용하여 P. haeundaensis의 돌연변이를 유도하고, 결과적으로 astaxanthin을 과잉 생산하는 돌연변이주를 선별하고 특성을 확인하기 위해 다양한 배양 및 영양 조건을 이용하여 astaxanthin 생산량을 확인하였다. 실험 결과 UV 조사 시간이 증가하거나, EMS 농도가 증가할수록 균주의 생존율이 감소하였다. Astaxanthin 과잉 생산 돌연변이 균주의 경우 400 mM EMS와 UV 20분을 순차적으로 처리한 방법에서 선별된 변이주가 가장 높은 astaxanthin 생산량을 보이는 것을 확인하였으며, 이 균주의 이름을 PUE로 명명하였다. PUE의 최적 배양 조건은 $25^{\circ}C$, pH 7-8, 3% NaCl이며, 1% raffinose, 3% potassium nitrate 첨가 시 astaxanthin 생산량이 증가하는 것으로 밝혀졌다. PUE에서는 wild type 균주에 비해 astaxanthin 생산량이 1.58배 증가함을 확인할 수 있었다. 본 연구의 실험 결과, 돌연변이 유도에 의해 선별된 변이주는 astaxanthin의 산업적 생산에 활용 가능한 후보가 될 수 있을 것으로 사료된다.

Effects of Feeding Urea and Soybean Meal-Treated Rice Straw on Digestibility of Feed Nutrients and Growth Performance of Bull Calves

  • The experiment was conducted for a period of 56 days with twelve Bangladeshi bull calves of average body weight of $127.20{\pm}11.34$ kg. The calves were divided into 3 groups having 4 animals in each. The animals were fed urea-treated rice straw designated as A) 4% urea-treated rice straw, B) 4% urea+4% soybean-treated rice straw and C) 4% urea+6% soybean-treated rice straw. In addition, all the animals were supplied 2 kg green grass, 350 g Til-oil-cake and 100 g common salt per 100 kg body weight of animals. Straw was treated with 4% urea solution and soybean meal at 4 and 6% were added to treated straw and kept for 48 h in double layer polythene bags under anaerobic condition. Urea treatment improved crude protein (CP) content of rice straw from 2.68 to 8.70% and it was further increased by 10.74 and 12.12% with the addition of 4 and 6% soybean meal. Dry matter (DM) intake (kg) was higher (p<0.05) in C (4.2) followed by B (4.1) and A (4.0). Crude protein intake was significantly higher (p<0.05) in group B and C than group A. Total live weight gains were 20.2, 24.8 and 25.6 kg for calves of group A, B and C respectively (p<0.01). The addition of soybean meal to treated rice straw did not affect the coefficients of digestibility of DM, OM, EE and NFE. However, CP and CF digestibility were significantly higher in group B and C (p<0.05). The values for digestible crude protein (DCP), digestible ether extract (DEE), digestible nitrogen free extract (DNFE) and total digestible nutrients (TDN) were significantly (p<0.05) higher in diet C and B in comparison to diet A, but there were no significant difference in digestible organic matter (DOM) and digestible crude fibre (DCF) value among the groups. It may be concluded that 4% urea treated rice straw can be fed to growing bull calves with 2 kg green grass and a small quantity of concentrate without any adverse effect on feed intake and growth. Moreover, soybean meal at 4 and 6% can be added to urea treated rice straw at the time of treatment for rapid hydrolyzing of urea, which resulted an improvement in nutrient digestibility and better utilization of rice straw for growth of growing bull calves.

Nutrient Intake, Its Utilization, Rumen Fermentation Pattern and Blood Bio-Chemical Constituents of Sheep Fed Urea Treated Mustard (Brassica campestris) Straw

  • A study was conducted to compare the feeding value of urea treated and untreated mustard straw (MS) for sheep. Treated MS was prepared by adding urea-N at 1.84% and followed by packing in a pit silo for 21 days. Two groups of six empty Avikaline ewes were fed untreated (UTMS) and treated (TMS) mustard straw along with 200 g concentrate per head daily for 90 days. Untreated MS had 0.41% N and the urea treatment increased its N value to 1.58 %. The cell wall constituents were decreased in the TMS except for cellulose which remained unaffected. Dry matter intake of TMS was consistently higher than that of UTMS. Digestibility of DM, OM and fibre fractions of MS improved by the urea treatment. Ewes in both groups were in positive N balance while % N retention was lower in UTMS (26.30%) than in TMS (52.14%). The TMS fed group on average consumed 30.2 g DM, 2.9 g digestible crude protein and $0.2MJ\;DE\;per\;kg\;BW\;day^{-1}$ and maintained their weight whereas, the UTMS fed ewes lost weight. The VFA concentration in rumen liquor was higher in TMS than in UTMS. Total-N, ammonia-N and TCA-precipitable-N were also higher in TMS fed ewes. Blood glucose concentrations in the two groups were similar at initiation of the study. However the glucose concentration of UTMS fed group was significantly (p<0.01) lower than those fed UTMS at the termination of the study. Urea-N concentration was also higher in TMS fed group after 90 days of feeding period. It is concluded that urea treatment of MS improved N value of MS from 0.41% to 1.58% along with sizable improvement in nutritive value and in conjunction with 200 g concentrate, TMS can serve as maintenance ration for sheep. Effects of Corn and Soybean Meal Types on Rumen Fermentation, Nitrogen Metabolism and Productivity in Dairy Cows

  • Twelve multiparous Holstein dairy cows in mid-lactation were selected for a replicated $4{\times}4$ Latin square design with a $2{\times}2$ factorial arrangement to investigate the effects of corn and soybean meal (SBM) types on rumen fermentation, N metabolism and lactation performance in dairy cows. Two types of corn (dry ground [DGC] and steam-flaked corn [SFC]) and two types of SBM (solvent-extracted and heat-treated SBM) with different ruminal degradation rates and extents were used to formulate four diets with the same basal ingredients. Each period lasted for 21 days, including 14 d for adaptation and 7 d for sample collection. Cows receiving SFC had a lower dry matter (DM) and total N intake than those fed DGC. However, the milk yield and milk protein yield were not influenced by the corn type, resulting in higher feed and N utilization efficiency in SFC-fed cows than those receiving DGC. Ruminal acetate concentrations was greater and total volatile fatty acids concentrations tended to be greater for cows receiving DGC relative to cows fed SFC, but milk fat content was not influenced by corn type. The SFC-fed cows had lower ruminal ammonia-N, less urea N in their blood and milk, and lower fecal N excretion than those on DGC. Compared with solvent-extracted SBM-fed cows, cows receiving heat-treated SBM had lower microbial protein yield in the rumen, but similar total tract apparent nutrient digestibility, N metabolism measurements, and productivity. Excessive supply of metabolizable protein in all diets may have caused the lack of difference in lactation performance between SBM types. Results of the present study indicated that increasing the energy degradability in the rumen could improve feed efficiency, and reduce environmental pollution.

Staphylococcus aureus Phage Type 29에 대한 오골계 난백 Lysozyme의 용균성 (Lytic Action of Egg White Lysozyme Isolated from Ogol Fowl on Staphylococcus aureus Phage Type 29)

  • 오골계 난백 lysozyme의 용균특성을 조사하고, 이를 식품보존료로 이용하기 위한 자료를 얻고자 오골계 난백으로부터 추출 분리한 lysozyme의 각종 미생물에 대한 용균성을 조사하였다. 그 결과, 공시균중 Gram 양성균인 Staphylococcus aureus phage type 29와 Bacillus subtilis ATCC 6633에 대하여는 용균성이 인정되었으나, Gram 음성균인 E. coli를 비롯한 여타의 균종과 Staphylococcus aureus phage type 57은 lysozyme에 대한 용균 감수성이 인정되지 않았다. 분리된 lysozyme은 S. aureus phage type 29를 공시균주로 하여 이를 육즙배지에 접종하고, $37^{\circ}C$에서 24시간 정치배양하여 세포를 회수한 후 540nm에서 흡광도가 0.6이 되도록 0.05M 초산완충액(pH 4.5)에 현탁시켜, 여기에 0.05%의 lysozyme을 가하여 $30^{\circ}C$, 30분간의 조건으로 반응시킬 때 용균성이 가장 좋았다. 또한 0.05%의 lysozyme은 반응액 중에 glycine(1%)을 첨가하므로서 공시균에 대한 용균효과가 양자의 상승작용에 의하여 그 단용시보다 현저히 증대(<50%) 되었으나, 기타 다른 첨가제와 금속이온 및 lysozyme과의 혼용효과는 인정되지 않았다.

Effects of Protein Supplement Sources on Digestibility of Nutrients, Balance of Nitrogen and Energy in Goats and Their In Situ Degradability in Cattle

  • The experiment was conducted to determine in situ rumen degradability of soybean meal (SM), fish meal (FM), sesame cake (SC) and Italian ryegrass hay (IRGH) and the effect of supplementing the above protein meals to IRGH on digestibility and balance of nutrients in three Saanen goats. For measuring the degradability, nylon bags containing each meal were incubated in the rumen of one fistulated dry cow for 3, 6, 12, 24, 36, and 48 hrs. Degradability revealed that SM protein was highly degradable in the rumen (99.1%), while FM protein was less degradable (76.8%) with SC protein being intermediate (91.2%) at 48 hrs of incubation (p < 0.01). Degradation rate of the potentially degradable fraction was estimated to be 12.12, 5.88 and 5.88%/hr for SM, FM and SC, respectively. In the metabolism trial, all goats were offered daily 900 g IRGH and one of the supplements, SM (100 g), FM (75 g) or SC (100 g). Intake of DM, OM, CP and GE was similar among diets. However, digestibility of OM for SC diet was significantly lower than that for diets supplemented with FM and SM (p < 0.10). Nitrogen (N) excreted in faeces (p < 0.05) and in urine (p < 0.10) was, respectively, higher and lower for SC diet than that for the other two diets. The same tendency was observed in energy losses in faeces (p < 0.10) and in urine (p < 0.05). There was no difference in energy loss in methane or in heat production among diets. Consequently, no significant difference was observed in N retention (2.13, 0.42 and -0.11 g/day for FM, SC and SM diet, respectively) or in energy retention (-1.49, -2.14 and -2.70 MJ/day for FM, SM and SC diet, respectively). These results showed that protein supplements affected the digestion of diets based on grass hay with 7.45% CP in DM in goats, although there was no significant influence on N or energy retention.
