Isolation of Bacteria from Jeotgal Using High-salt-content Media and Their Growths in High-salt Condition (고염에서 생장하는 젓갈 유래 Bacteria의 분리 및 고염에서의 생육 특성)

  • Proteolytic bacteria were isolated from Myeolchi-jeotgal and Saeu-jeotgal using high-salt-content media and their growths in the media containing 25% NaCl were monitored to draw the role of bacteria in the ripening of jeotgal. The most populous genus in Myeolchi-jeotgal detected on agar media with 15% NaCl was Bacillus and its relatives, while the most populous in Saeu-jeotgal was Staphylococcus. Among the isolates, Virgibacillus halodenitrificans from Myeolchi-jeotgal and Halobacillus trueperi from Saeu-jeotgal showed proteinase activities. The species from Myeolchi-jeotgal showed proteinase activity on the agar media with 8% NaCl were similar to those isolated from the media with 15% NaCl. The dominant of Myeolchi-jeotgal isolated at the 15% NaCl concentration may be involved in the proteolysis. The proteolytic species from Saeu-jeotgal on the agar media with 8% NaCl were the genera Bacillus, Salinicoccus, and Salimicrobium those were not the dominants at 15% NaCl condition. The dominant isolates from Saeu-jeotgal on agar media with 15% NaCl may not be involved in the proteolysis of Saeu-jeotgal. Vb. halodenitrificans and Staphylococcus equorum, the dominant species from Myeolchi-jeotgal and Saeu-jeotgal, showed growths at the nutrient broth containing 25% NaCl. They may play a significant role in the ripening of jeotgal and have a high possibility to be used as the starter.

Effect of Saline Soil and Crop Growth with Bottom Ash from Biomass Power Plant Based Wood Pellet (우드펠릿 기반 바이오매스 발전소로부터 배출된 저회를 활용한 염류토양 및 작물성장에 미치는 영향)

  • BACKGROUND: The salt in soil interrupts crop growth. Therefore, water resources are used to remove any salt found in the soil. However, water resources have been reduced by global warming; thus, a new study is required into reducing the salt in soil. Recently, the bottom ash (BA) of a biomass power plant was found to be similar to biochar. Hence, it can be used to remove heavy metals and wastewater through the adsorption characteristics of BA. The objective of this study was to evaluate the improvement effects on crop growth in saline soil containing the BA from biomass power plants. METHODS AND RESULTS: The effect on crop growth in the saline soil supplemented with BA was studied with the crop-planted pots, which were packed by reclaimed greenhouse soils collected from Byolyang, Suncheon. The BA application level was 25, 50, 100, 200, and 400 kg/10a (referred as BA25, BA50, BA100, BA200, and BA400, respectively). The BA increased the fresh weights of the leaf and root, while nitrogen uptake increased by approximately 24-102% and 54-77%, respectively for the lead and root. The phosphorous uptake increased by 38%, although only in the leaf of the lettuce. In the case of soil, BA increased water content, pH, EC, CEC, and NH4+ and the SAR of the soil decreased by 5-15%. The bottom ash increased the contents of Ca2+ and Mg2+, and fixed the amount of Na+. CONCLUSION(S): It was confirmed the bottom ash of a biomass power plant, based on wood pellets, improved crop growth, and increased the nutrient uptake of crops in saline soil. In addition, bottom ash, which has a wide range of porosity and high values of pH and EC, improved properties of the saline soil. However, the BA has a large amount of B, As, and heavy metals. Finally, it may require a study on the safety and contamination of heavy metals contained in the bottom ash, which would be applied in soil for a long time.

Effect of Graded Levels of Rice Mill Feed (RMF) Supplementation on Intake, Nutrient Digestibility, Microbial N Yield and Growth Rate of Native (Bos Indicus) Bulls Fed Rice Straw Alone

  • Rice bran commonly available in Bangladesh is a mixture of rice hulls (60%), bran (35%) and polishing (5%), referred here as rice mill feed (RMF). Dose response effect of RMF supplementation to a straw diet including a zero level was measured on the intake, digestibility, nitrogen balance, microbial N yield and growth rate of growing native (Bos indicus) bulls. Twelve bulls of 33 months old and $272{\pm}31.5kg$ weight were randomly allocated to diets having 0 (T1), 1 (T2) and 2 (T3) kg RMF in addition to 200 g wheat bran, 200 g molasses, 60 g salt and 30 g oyestershe\l powder. Concentrate intake was 5.5, 19.2 and 29.5% of the dietary intake for the T1, T2 and T3 treatment respectively. RMF supplementation had no significant effect on the straw DM intake. However, with the increasing levels of RMF supplementation, total DM & digestible OM intake and the whole gut digestibilities of DM, OM, N & ADF increased but in deminishig return. Total microbial N yield estimated from the urinary purine excretion were 15.35, 26.56 and 38.44 g/d for the treatment T1, T2 and T3 respectively. Both the N intake and the N balance increased linearly in response to increasing level of RMF. Supplementation of RMF linearly increased the energy intake and dietary energy concentration. Growth rate in the T1, T2 and T3 treatments were 112, 125 and 250 g/d respctively. The basal N excretion and the maintenance energy requirement of the experimental animals were estimated to be 615 mg/kg $W^{0.75}/d$ and 447 kJ/kg $W^{0.75}/d$ respectively. The estimated efficiency on N utilization was 0.83 mg/mg of N intake ($r^2=0.997$) while the efficiency of metabolizable energy utilization for growth was 0.15. Since animal refused higher levels of RMF, inclusion up to 2 kg level (about 25% of the total DM intake) appears to have no depressing effect on the performances of animal. However, RMF itself fail to meet the critical nutrient need of the rumen microbes. Therefore response of supplementing RMF after correcting the critical nutrient deficiency need to be studied.

Studies on the RK-temperate phage of bacillus cereus (Bacollis cereis의 RK-용원파아지에 관한 연구)

  • The RK-temperate phage which infected with Bacillus cereus was isolated and the characters were investigated. The induction of RK-temperate phage from host bacterium attained by ultraviolet light irradiation (15W, 30cm, 30-120sec) and mitomycin C treatment (0.2-2 ug/ml). The host range of RK-temperate phage was not revealed with lysogenic and related strains of B. cereus. But B. cereus(PS) 352 which obtained by N-nitrosoguanidine treatment (1,000{$\mu}g/ml)$ to phage infected with host bacteria was sensitive bacteria of RK-temperate phage. RK-temperate phage was stabilized at the condition of nutrient broth (pH 7-8), Tris-buffer (pH 7-8) and ammonium buffer (pH 8-9) and Sorensen's phosphate buffer (pH 6-7), but unstabilized at other salt solutions and pH range. Also, thermostability was to $45^{\circ}C$ but unstabilized at above $50^{\circ}C$. At RK-temperate phage, the measurment values of head, neck, mid tail and end tail were 59nm, $9{\times}16nm,\;10{\times}189nm,\;and\;10{\times}14nm$ respectively. The morphology of head was regular polyhedron, and the end tail was coneate form. On the one hand, the number of capsid protein layer of tail were consist of 4, 35, and 1 at neck, mid tail, and end tail, respectively. RK-temperate phage was identified with DNA phage and G+C contents were 38.63. The latent time of RK-temperate phage was 30 minutes and the burst size was 70-80. And the host bacteria was lysed in case of multi-infection, above moi 1.

A Follow-up Study on the Nutritional Status of Korean Fatty Liver Patients with Different Conditions (한국인 지방간 환자의 병세변화에 따른 영양상태에 관한 Follow-up연구)

  • The purpose of this study is to investigate dietary factors which influence the conditions of Korean fatty liver patients. The subjects were 83 fatty liver patients living in city, ages of 10 to 60 yrs old, and they were the same patients studied 9 months ago in previous study. The patients were groupsed into 3 groups(improved, similar, worsened groups) according to the changes in disease condition. Nutrient and alcohol intakes and health status were investigated. In improved group, the patients tended to lose weight, the number of exercising patients was increased, the consumptions of calorie, carbohydrate, calcium, phosphorus were decreased. And alcohol intake was decreased markedly and serum total amino acids(AA), EAA, NEAA, BCAA and AMAA contents, EAA/NEAA and BCAA/AMAA ratios were increased. In worsened group, patients tended to gain weight, the number of exercising patients decreased, the consumptions of calorie, protein, carbohydrate, calcium, phosphorus and niacin decreased but vitamin. A intake increased. And also, in this group, alcohol intake of male patients decreased and that of female slightly increased, and serum EAA, BCAA and AMAA contents, EAA/NEAA and BCAA/AMAA ratios, and total fatty acid contents increased and total AA and NEAA contents decreased. In conclusion, it is desirable to reduced weight, stress, alcohol, salt and animal fat consumptions, and to exercise, and to take adequate amount and quality of protein to improve conditions of fatty liver patients.

Effect of Diluent Salt Concentration and pH on the Enumeration of Vibrio parahaemolyticus by Direct Plating on Selective Agar

  • The maintenance of physiological activity during dilution is very critical for the accurate enumeration of Vibrio spp. in marine samples. We investigated the effect of various diluents on the recovery of Vibrio parahaemolyticus using the direct plate counting and most probable number (MPN) methods. The effects of NaCl (0.85 and 3%) and pH (from 6.6 to 7.4) in diluents based on distilled water or phosphate buffered saline (PBS) were evaluated with three V. parahaemolyticus strains. PBS-3% NaCl (pH 6.6), as opposed to PBS, was the most effective diluent at maintaining viable cell numbers up to 2 log CFU/g during dilution for direct plate counting using on thiosulfate citrate bile salts sucrose (TCBS) selective agar, as well as minimizing the difference in cell numbers between TCBS and non-selective nutrient agar. It also increased counts of V parahaemolyticus inoculated into oysters relative to PBS (p<0.01), suggesting that PBS-3% NaCl (PH 6.6) can reduce the problem of underestimating V. parhaemolyticus counts using PBS alone.

American Television: A Source of Nutrition Education and Information

  • Television is a powerful and persuasive teacher. It has the potential to influence perceptions, knowledge, beliefs, and behaviors, thus nutritionists need to be aware of the nutrition-related information (NRI) in television programming and the effect this information has on viewers. The purpose of this article was to review research published in peer-reviewed journals between 1988 and 2003 that examined the NRI embedded in American television programming, which is exported to over 125 nations, and its impact on nutrition knowledge, attitudes, and/or behaviors. This review revealed that, for the past 15 years, NRI was commonly included in both television advertisements and shows. Advertised foods were mainly high in fat, sodium, and/or sugar. In addition, the NRI embedded in food advertisements tended to be misleading or inaccurate. Prime-time television shows included numerous NRI containing scenes every hour, with situation comedies having the most and real-life re-enactment shows the least. Overall, low nutrient density foods accounted for approximately 40 percent or more of all food references on prime-time television shows. In television shows, foods were mostly consumed as snacks rather than meals and children often ate more nutritious foods than adults. Although relatively few studies have examined the impact of television programming on viewers, those that do exist indicate that as children watch more television, nutrition knowledge and understanding declines while misconceptions about nutrition increase. Advertising influences children's food purchase requests and subsequent purchases by adults, with the most requested and purchased foods being high in sugar, fat, and/or salt foods. Existing research indicates that television must be acknowledged as a major source of NRI and a potentially powerful influence on dietary practices.

Effect of Reclaimed Sewage Irrigation on Paddy Rice Culture and Soil Characteristics (오수처리수의 관개가 벼 생육 및 토양 특성에 미치는 효과)

  • Effect of reclaimed sewage on the paddy rice culture was examined by field experiment for two consecutive years. The domestic sewage was treated by the constructed wetland and the effluent of the treatment wetland was used for irrigation water. The reclaimed sewage was diluted before irrigation in the first year and it was used without dilution in the second year experiment. Growth components and yields were compared against the control plot where conventional method was applied. And also soil characteristics of the plots before and after reclaimed sewage irrigation were analyzed Generally addition of the reclaimed sewage irrigation didn't affect paddy rice culture adversely and even enhancement was observed. Fertilization was thought to be important factor for rice culture rather than irrigation water quality. Conventional fertilization and reclaimed sewage irrigation which contained high nutrient concentration resulted in better growth and more yield. Unlike widespread concern lodging did not happen even in the case of irrigation with average 90mg/L of T-N and conventional fertilization. Soil characteristics changed after irrigation and significant EC increasing was observed for the reclaimed sewage irrigation plots. From soil analyses salt accumulation could be a more potential problem than nutrients like nitrogen and phosphorus in the use of reclaimed sewage irrigation. Overall reclaimed sewage irrigation was thought to be one of practical alternatives for the ultimate disposal of sewage in rural area.

A Study on Customized Nutrition Intervention Program Design and Application for the Low-Income Elderly (저소득층 노인을 위한 맞춤영양관리 프로그램의 개발과 시범 적용 연구)

  • This study aimed to plan nutrition support programs for the elderly living alone whose nutrition status were seriously concerned, conducted seven stages nutrition intervention program on a trial basis, and evaluated the effectiveness of the program of the Elderly Nutrition Support Project. Subjects were selected for personalized nutrition management based on nutritional risk score and nutrition intervention were tailored to the problems occurred. The elderly nutrition support program targets were 44 senior citizens who lived alone with low income. The 33 (as Type 1) of the subjects with whom milk, tofu, seaweed, eggs, black beans have been supported, and also provide nutrition education, and the rest 11 persons (as Type 2) to whom food was not supported but provide nutrition education programs. As a result, all subjects showed that compared with pre and post program implementation, their daily exercise time and milk and protein consumption level were increased and some improvement was observed regular meals consumption and low-salt diets. Their nutrient intake level such as calories, protein, calcium, iron improved after implementation. In addition, NSL DETERMINE scores significantly improved from 13.21 to 7.24 in Type 1 and 11.27 to 9.91 in Type 2. As positive dietary behavioral changes were observed as in that they purchased more protein and calcium rich foods.
