Selection of Brevibacillus brevis B23 and Bacillus stearothermophilus B42 as Biological Control Agents against Sclerotinia Rot of Lettuce (상추 균핵병 생물적방제를 위한 Brevibacillus brevis B23과 Bacillus stearothermophilus B42의 선발)

  • Bacillus spp. isolated from mushroom medium wastes were evaluated for their biocontrol potentials on control of Sclerotinia rot of lettuce. The Bacillus isolates were more effectively obtained from waste suspension when directly added into nutrient agar(NA) medium than plating on the agar medium. Totally 42 isolates obtained from the wastes B23 and B42 showed highest antifungal activity against eight fungal pathogens such as Sclerotinia sclerotiorum, Rhizoctonia solani, Pythium ultimum, Phytophthora capsici, Fusarium oxysporum, Colletotrichum gloeosporioides, Cladosporium cucumerinum, and Botrytis cinerea and B23 and B42 were finally selected for further studies. Optimal concentration of the isolates was $10ml(10^7cfu/ml)$ to suppress the Sclerotinia rot of lettuce. Supplements such as starch, glycerol, and egg-yolk successfully maintained the bacterial population for 30 days in vitro and increased bio-control potentials against the disease. The bacterial isolate B23 alone showed 72% control value, furthermore it presented 95% control value when supplemented with 0.2% of starch, glycerol, and egg-yolk. The promising Bacillus isolates B23 and B42 were identified as Brevibacillus brevis and Bacillus stearothermophillus, respectively, based on morphological and physiological characteristics according to API database.

Effects of Lead, Copper and Cadmium on Pseudomonas cepacia KH410 Isolated from Freshwater Plant Root (담수식물 근계로부터 분리된 Pseudomonas cepacia KH410 균주에 대한 납, 구리, 카드뮴의 영향)

  • A ubiquitous bacterium,Effects of Lead, Copper and Cadmium on Pseudomonas cepacia KH410 Isolated from Freshwater Plant Root was isolated from freshwater plant root and interactions of lead, copper and cadmium with this strain was studied. Mass production of dry cell weight 2.72 g-DCW/ι-medium was obtained by cultivation in a nutrient medium containing 1% yeast extract, 1% soytone and 0.5% NaCl, pH 7.0, at temperature of 28℃ for 24 hrs under aeration. The mass of dry cell produced after exposure with 100 mg/ι of heavy metal was 1.98 g/ι for lead, 1.58 g/ι for copper and 0.20 g/ι for cadmium, respectively. The minimal inhibitory concentrations (MIC) for each heavy metal was 1.3 mM for lead,0.8 mM for copper and 0.4 mM fur cadmium, respectively. Cell aggregation occurred by each heavy metal exposure was observed from 1 day to 4 days by an optical microscope. Entrapment, precipitation effects on cell by heavy metals between 10 min and two hours were examined by an electron microscopy. Cadmium appeared to be the most toxic on cells and the order of toxicity was cadmium>copper>lead.

Distribution and Characterization of Heterotrophic Bacteria Isolated from Deodorizing Biofilters of a Nightsoil Treatment Plant (분뇨처리장의 질소성 악취물질처리를 위한 생물탈취탑내 종속영양세균의 분포 및 특성)

  • The distributions and characteristics of bacteria on the deodorizing biofilters were studied as a part of biofiltration research for odor gas removal. The odor gas originated from a currently operating nightsoil treatment plant were mainly nitrogenous compounds of ammonia and amines. The filter media were consisted various ratio of pine bark, nightsoil cast and compost, and were sampled before and after operation of 153 days. Before operation, the level of bacteria on nitrification agar medium, nutrient agar medium and thiosulfate agar medium were around $10^7{\sim}10^8cfu/g$ media. After operation, the level decreased but still high as $10^5{\sim}10^7cfu/g$ media. The bacteria from reactors before and after operations were either rod or coccus after growth on nitrification agar media. The 45 isolated heterotrophic bacteria were further characterized and Pseudomonas, Bacillus, Flavobacterium, Alcaligenes were dominant genera. Genus Alcaligenes was particularly dominant when the isolates were incubated on nitrification agar media first and then transfered to nutrient media. The portion of Alcaligenes in the biofilter increased after operation, and was higher in the media contained nightsoil cast, which showed better treatment than the media with pine bark only.

  • Selection of optima] nutrient sources and cultural methods for liquid spawn culture of Flammulina velutipes were carried out. The optimal temperature and pH for mycelial growth of F. velutipes were $20^{\circ}C$ and 6.0 to 7.5. respectively. In the 250ml ${Delta}$-flask culture. the amount of inoculum and culture period for the optimal mycelial growth of F. velutipes were 3 mycelial disks(diametcr 6mm) and 6 days, respectively. For the mass production of submerged culture. the optimal inoculum amount and aeration rate of F. velutipes were 5%(inoculum vol/medium vol.) and l.0vvm(vol of air/vol. of medium/min), respectively.

A Study on Development and Validation of Food Frequncy Questionnaire for Koreans (식품섭취도 조사지의 개발 및 타당도 검증에 관한 연구)

  • The purpose of this study was to develop and validate the Food frequency questionnaire (FFQ) for dietary studies of Koreans. One hundred and five food items for the Food frequency questionnaire were selected based on information of frequently consumed foods from National Nutrition Survey Reports and on raw data from a dietary survey on diabetic patients. Frequency of consumption was determined through nine categories ranging from more than three times a day to almost never to indicate how often the specified amount of each food item was consumed during the past month. Three portion sizes were given for each food item(small, medium or large) with respect to a stated medium portion. Seventy-three healthy women served for the validation study. They completed both the FFQ and a 3-day diet record. The FFQ estimate of mean nutrient intake was higher by 10-20% than that of the 3-days diet record and the Spearman correlation coefficients between the two methods ranged from 0.26 to 0.59 . The degree of agreement was from 36% to 38% when nutrients intake assessed by the FFQ and 3day diet record were classified within the same quintile. On the whole , the result of this study seemed to be in good agreement with other studies. Therefore the FFQ developed in this study is considered to be a reliable tool in assessing the dietary habits of Korean.

Fertility Assessment of the Piggery Wastewater Trickling Filtrate for Orchard Grass (Dactylis glomerata L.) and Soil

  • In search of a method to achieve sustainable agricultural practices, a trickling filter was employed for the piggery wastewater treatment, where rice straw was the support medium in place of more commonly used materials. The filtrate from the trickling filter were applied to a soil, on which orchard grass (Dactylic glomerata L.) was grown followed by amendment of the treated straw medium. Orchard grass was cut twice, and growth parameters and yields were measured. Soil chemical properties before and after harvesting orchard grass were analyzed. Development of the forage crop was greatly enhanced by the application of the filtrate in terms of fresh weight, dry weight, and the absorption of nutrients. Better growth and higher nutrient uptake were found in the second cutting of the orchard grass. Organic matter content, ionic intensity, exchangeable cations, and phosphate of the soil increased with the application of the filtrate. The relative high concentrations of salts in the piggery trickling filtrate, expressed in electrical conductivity and content of sodium, exerted no detrimental effect on the crop and soil.

Effect of Transinoculation of Goat Rumen Liquor on Degradation and Metabolism of Mimosine in Sheep Fed with Leucaena leucocephala Leaves

  • The effect of transinoculation of goat rumen liquor into sheep rumen on mimosine toxicity was studied. One adult Kutchi male goat having higher mimosine degradation capacity than sheep was gradually adapted to Leucaena leucocephala (Leucaena) leaves by feeding increasing level of eucaena leaves supplementation for 1 month. Six Bharat Merino rams (12-18 months of age) were divided into two equal groups with (group I) or without (group II) infusion of 200 ml of goat rumen liquor per animal. The mimosine degradation in groups I and II were 3.04 and 2.31; 3.90 and 3.73 mg per day per 10 ml rumen liquor respectively after 1 and 2 weeks of leucaena feeding leaves. Total rumen bacterial population in RGCA medium and in a selective medium containing iron showed an increasing trend in both groups, while the bacterial population growing in the presence of cellulose showed a decreasing trend. Animal performance data did not show any adverse effect. Results revealed that transinoculation of rumen liquor from leucaena leaves adapted goat to sheep rumen did not help to improve mimosine degradation in the sheep. The sheep transinoculated with goat rumen liquor displayed no in vivo improvements in nutrient utilization vis-a-vis mimosine metabolism.

  • Through enrichment on two nutrient agars, 57 Thermus isolates were recovered from 15 hot spring samples taken from the Rehai geothermal area, Tengchong, China. Unique growth characteristics were observed when the strains were transferred from YIM14 medium to Thermus medium. Phylogenetic analysis showed that the 16S rDNA sequences of the isolates and clones from the Rehai geothermal area farmed a monophyletic group on the phylogenetic tree. A secondary structure comparison showed that their 16S rRNAs have unique secondary structure characteristics.

Biosynthesis of L-Ascorbic Acid by Microorganisms in Kimchi Fermentation Process

  • Kinchi is and important source of various vitamins, minerals, dietary fiber, organic acids and other nutrients. In order to get a basic information for developing vitamins-rich funtional kimchi, we investigated microorganisms which are capable of synthesis of vitamin C in Kimchi system. Microorganisms isolated from aliquots of kimchi were screened and cultured by using MRS or nutrient agar medium. L-Ascorbic acid produced by microorganism in medium was measured with high performance liquid chromatography. As the result, we isolated two bacteria strins N7 and N5202 preducing L-ascorbic acid from the kimchi system. Morphological and Gram staining experiment showed that N7 was Gram positive bacilli, while N5202 was Gram negative. There were also several bacteria that were considered to synthesizs erythorbic acid which is an analog of ascorbic acid. These results suggested that vitamin C-rich functional food could be developed by using the kimchi microorganisms.

Application of Subirrigation Using Capillary Wick System to Pot Production

  • Alternative subirrigation way, capillary wick system (CWS) was tested to reduce labor cost, waste water, contamination of ground water, and use of fungicide compared to overhead irrigation system (OIS). CWS helped reduce remarkably the working hours for watering from 4 hours in OSI to just 5 minutes. Labor cost was saved 98% in CWS compared to OIS. By the physical characteristics of various growing media, 1 coconut coir+2 perlite (v/v) mixture was selected because it had an ideal distribution of three phase, e.g. 1 solid: 1 liquid: 2 gas phase. Medium mixture containing scoria had so high bulk and particle density to hurt root. In bark-containing medium, the liquid phase and the percent saturation of liquid phase with time elapsed was lower than that of other mixture. It meant that the mixture contained very low level of water. Application of CWS for cyclamen pot production played an important role in reducing the incident of fusarium wilt symptom from 18% in conventional over watering system to 4%. Cyclamen pot irrigated by capillary wick had shorter petiole and more leaves than those by overhead watering. As a result, this system was highly beneficial to get uniform pot products with high quality. It improved water and nutrient solution efficiency relative to conventional overhead irrigation system (OIS).
