Studies on the Culture of Ergot Fungus(IV) -Culture of Korean Ergot Fungus- (맥각균(麥角菌)의 배양(培養)에 관한 연구(硏究)(IV) -한국산(韓國産) 맥각균(麥角菌)의 배양(培養)-)

  • To isolate ergot strains which are capable of growing and producing alkaloids in submerged culture, strains were isolated from the sclerotia parasitizing the Graminae plants in Korea and the experiments of submerged culture of these strains yielded the following results: 1) The mycelia which were respectively isolated from the sclerotia parasitizing Agropyron semicostatum $N_{EES}$, Arundinella hirta $T_{ANAKA}$ var. ciliata $K_{OIDSUMI}$, Ischaemum anthephoroides $M_{IQ}$. var. eriostachyum $H_{ONDA}$, and Phleum pretense L. grew in Medium D by submerged culture. 2) When the strain of the ergot of Agropyron semicostatum was inoculated into six different nutrient solutions for submerged culture, its mycelium grew well in Media C, D and F, but produced alkaloids only in Medium C, indicating that Medium C is relatively suitable for the strain. 3) The extraction of the alkaloids from the culture broth by ether and T.L.C. analysis of the extract showed that it contained at least two types of alkaloids.

Growth characteristics and productivity of new Italian ryegrass (Lolium multiflorum Lam.) variation, IR 604

  • This experiment was carried out to breed a medium maturing variety of Italian ryegrass (Lolium multiflorum Lam.) in Grassland and Forage Crops Division, National Institute of Animal Science, Rural Development Administration (RDA), Cheonan from 2019 to 2020. An Italian ryegrass variety, IR 604 is a tetraploid variety with green in leaf color and has half-erect growth habit in late autumn and erect growth habit in early spring, IR 604 was on May 17 in heading date as a medium maturing variety. Also, IR 604 was longer by 11 cm in flag leaf length and longer by 1.3 cm in plant height than those of the control variety, Kowinner, respectively. IR 604 was also thicker by 1.0 mm in stem thickness and strong in winter hardness. Dry matter (DM) yield (11,036 kg/ha) of IR 604 was 16.6% higher than that of Kowinner. Total digestible nutrient (TDN), crude protein (CP) and relative feed value (RFV) of IR 604 were 60.4, 13.8% and 100.6 which are 1.4, 3.3%, 7.7 higher than those of Kowinner, respectively. Acid detergent fiber (ADF) and neutral detergent fiber (NDF) of IR 604 were 35.5 and 56.7% which are 1.3 and 3.6% lower than those of Kowinner, respectively.

Ice-Nucleation Activity of Pseudomonas syringae Isolated in Korea (한국에서 분리한 Pseudomonas syringae의 빙핵활성)

  • Cell suspensions of two isolates of Pseudomonas syringae. PS8401 from sweet persimon and PS8402 from tea plant, were active in ice nucleation at -2.5 and $-3.8^{\circ}C$, respectively. Ice nucleation at those temperature was, using micropipette method, detected in suspensions ($10^8$ olony forming unit/ml of distilled water) of cells that had been grown on nutrient agar supplemented with $2.5\%$ glycerol. Using the same method, on the other hand, the freezing temperature of distilled water only was approx. $-21.8^{\circ}C$, and those of various plant saps including corn were lower than $-11.6^{\circ}C$. Corn seedlings sprayed with cell suspensions $(10^8\;cfu/ml)$ of nutrient broth) of PS8401 began to be damaged at $-2^{\circ}C$ and were almost completely damaged at $-4^{\circ}C$, whereas seedlings sprayed with nutrient broth only were not injured until the temperature down to $-9^{\circ}C$. Amounts of frost damage measured 48 hr after application of PS8401 suspensions increased as applied bacterial cell densities were increased. Ice-nucleation activity of the cell suspensions in vitro increased with increasing the number of cells in suspension. The activity also affected by growth-medium composition or growth-temperature. Ice nucleation thus occured at -4.0, -4.4 and $-7.2^{\circ}C$ in suspensions $(10^2\;cfu/ml)$ of PS8401 that had been grown at$25\%$ nutrient agar with $2.5\%$ glycerol, nutrient agar with $2.5\%$ glucose and nutrient agar only, respectively, and occured at -4.0 and $-7.6^{\circ}C$ in suspensions $(10^2\;cfu/ml)$ of PS8401 hat had been grown on nutrient agar with $2.5\%$ glycerol at $15\~25^{\circ}C$ and $30^{\circ}C$, respectively.

  • The objectives of this study were to isolate and culture methanotrophs and to apply them for biological removal of nitrogen and phosphorous. Methanotrophs (dominant species: Methylomonas methanica) were isolated from a landfill cover soil, cultured in a NMS medium, and analyzed to reveal their characteristics of growth and nutrient removal. The methanotrophs themselves can produce substantial amount of organic substances(as COD) including methanol, formaldehyde, and formate, as carbon sources required for denitrification. For instance, the production rate for methanol was $8\;mg/L{\cdot}hr$. Moreover, the analysis of nitrogen and phosphorous in the sludge suggested that the methanotrophs assimilate nitrogen and phosphorous as growth substances.

Influence of pre-planting application of dolomite at various rates in coir-dust containing root media on the growth of red-leaf lettuce

  • This research was conducted to evaluate various levels of dolomitic lime incorporated as pre-planting fertilizers on the growth of red-leaf lettuce. To achieve this, three root media were formulated by mixing coir dust with expanded rice hull (CD+ERH, 8:2, v/v), carbonized rice hull (CD+CRH, 6:4, v/v), and ground pine bark (CD+GRPB, 6:4, v/v). During formulation, equal amounts of essential nutrients, except dolomitic lime, were incorporated into all root media and the levels of dolomitic lime were varied from 0 to $7.5g\;L^{-1}$ at 1.5 g increments. Seedlings of red-leaf lettuces at the 3rd leaf stage were transplanted into each medium treatment. Crop growths were measured 5 weeks after transplant and soil solutions were collected every week and analyzed for pH, EC, and nutrient concentrations. The treatments showing the heaviest fresh and dry weights in CD+ERH, CD+CRH, and CD+GRPB were 4.5 g, 4.5 g, and $7.5g\;L^{-1}$ of dolomite, respectively. The pHs of three root media yielding the highest crop growths were in the ranges of 6.4 to 7.1. These ECs in CD+CRH medium were around $1.0dS\;m^{-1}$ higher than those of CD+ERH and CD+GRPB when application rates of dolomitic lime were equal. $K^+$ concentrations were higher than $Ca^{+2}$ and $Mg^{+2}$ concentrations until week 2 in three root media. But $Ca^{+2}$ and $Mg^{+2}$ concentrations were higher than $K^+$ concentrations after week 3 in all root media. The concentrations of $PO_4{^{-3}}$ in all root media got abruptly lower until week 2. These results indicate that appropriate levels of dolomitic lime, as pre-planting nutrient charge fertilizers in CD+ERH and CD+GRPB media, are 4.5 and $7.5g\;L^{-1}$, respectively.

Antifungal Activity and Cultural Characteristics of the Streptomyces sp. A252 (Streptomyces sp. A252의 배양적 특성 및 항진균활성)

  • The growth rate of the A252 strain was increased in tryptic soy broth (TSB) and malt extract-yeast extract medium (ISP-2), but the antifungal activity of culture filtrate was efficient in the media of TSB and nutrient broth. The mycelial growth and the antifungal activity of culture filtrate in TSB medium were optimized at $25^{\circ}C$ and pH 6.5. The growth in 2$\%$TSB concentration was more effective than 1$\%$, but there was no difference of the antifungal activity by the TSB concentrations. The mycelial growth of A252 strain reached to maximum at 72 hr after inoculation, whereas the antifungal activity of culture filtrate was shown to have the highest level at idiophase (60 hr) after inoculation and was decreased a little after 96 hr incubation. The antifungal activity was stable in the pH range of 4 to 11 and evenly at $121^{\circ}C$. The A252 strain was characterized as Streptomyces species by the physiological properties and examination of sporophore me morphology.

Growth Characteristics of Citrobacter sp. MB2, Azo Dyes Decolorizing Bacterium (아조염료 분해균 Citrobacter sp. MB2 생육특성)

  • A Citrobacter sp. MB 2, azo dyes decolorizing bacterium, was isolated from the wastewater and soil and identified as Citrobacter sp.. It was examined that optimum conditions for culture media were 0.5% of sucrose, 1.0% of yeast extract, 0.1% of $K_2HPO_4$, 0.1% of $NaHCO_3$ per distilled water. The best efficient condition of culture was obtained at pH 7.0, $30^{\circ}C$ and aerobic shaking culture. The number of Citrobacter sp. MB2 in optimum medium was increased more than 7 fold compared to basal medium and 50 fold compared to nutrient broth. This strain was exhibited strong resistance against metal salts and antibiotics (ampicillin and penicillin G).

Effects of Particle Size of Barley on Intestinal Morphology, Growth Performance and Nutrient Digestibility in Pigs

  • A growth trial and a digestibility trial were conducted to examine the effect of feed particle size on the performance, nutrient digestibility, gastric ulceration and intestinal morphology in pigs fed barley-based diets. Barley was processed through a hammer mill to achieve four diets varying in particle size (average particle $size{\pm}standard $deviation): coarse ($1,100{\pm}2.19\;{\mu}m$), medium ($785{\pm}2.23\;{\mu}m$), fine ($434{\pm}1.70\;{\mu}m$) and mixed (1/3 of coarse, medium and fine) ($789{\pm}2.45\;{\mu}m$). Sixty-four entire male pigs were used in the growth trial and the diets were fed ad libitum between 31 kg and 87 kg live weight. Following slaughter, stomach and ileal tissues were scored for integrity (ulceration or damage) and histological measurements taken. Twenty-four entire male pigs were used in the digestibility trial, which involved total faecal collection. Over the entire growth phase, there were no differences (p>0.05) in average daily gain and feed conversion ratio between pigs fed diets of different particle size. Pigs fed the coarse and medium diets had lower (p<0.05) stomach ulceration scores (0.20 and 0.25, respectively, on a scale from 0 to 3) than those fed the mixed (0.69) or the fine diets (1.87). The stomachs of all animals fed the fine diet had lesions and stomach ulcerations were present only in this group. Pigs fed the fine diet had thicker (p<0.001) ileal epithelial cell layer with no differences (p>0.05) being observed for villous height or crypt depth. Faecal digestibility coefficients of neutral and acid detergent fibre were the highest (p<0.05) for the mixed diet, intermediate for the fine and coarse diets and the lowest for the medium diet. A similar numerical trend (p = 0.103) was observed for the apparent faecal energy digestibility coefficient. It is concluded that, with barley based diets, a variation in average particle size between $400{\mu}m$ and $1,100{\mu}m$ had no effect on pig performance but the fine dietary particle size affected the integrity of the stomach, as well as the structure of the small intestine, thus compromising overall gut health. Our data also demonstrate that changes in particle size distribution during the digestion process, rather than average particle size or particle size variation, are related to apparent faecal digestibility.

Effect of Liquid Fertilizer Contained Medium of Lactobacillus sp. and Saccharomyces sp. on Growth of Creeping Bentgrass (유산균과 효모균 배양액 함유 액비 시용이 크리핑 벤트그래스의 생육에 미치는 영향)

  • This study was conducted to investigate the effect of Lactobacillus sp. and Saccharomyces sp. on turf quality, shoot and root growth of creeping betgrass in golf course by measuring turf color index, chlorophyll content, dry weight of shoot and root, T/R ratio and root length. Fertilizer treatment was designed as follows; nonfertilizer (NF), control (CF; compound fertilizer), microorganism medium(MO; CF+MO)), microorganism medium contained Fe(MO-Fe; CF+MO-Fe) and microorganisum medium contained S (MO-S; CF+MO-S). Soil properties investigated after experiment was scarcely affected by applied fertilizers in root zone of creeping bentgrass. The turf color index and chlorophyll index of MO, MO-Fe, MO-S treatment were higher than those of NF, and similar to those of CF. The turfgrass root in MO and MO-Fe treatment was longer than others. The dry weight of shoot in MO and MO-S was higher than CF and that of root in MO and MO-Fe, and dry weight of MO was increased than that of NF and CF, by 26% and 6%, respectively. AS compared with NF, T/R ratio of CF, MO, MO-Fe and MO-S was increased, and MO and MO-Fe was similar to CF, MO-S higher. Nutrient content in CF, MO, MO-Fe and MO-S was contained more than in NF, and it was higher in shoot. These was suggested that application of MO induced the development of quality and growth of creeping bentgrass by assisting root growth and nutrients uptake.

Accumulation of Poly-$\beta$-Hydroxybutyrie Acid by Alcaligenes sp. (Alcaligenes sp.에 의한 Poly-$\beta$-Hydroxybutyric Acid의 축적)

  • Microorganisms capable of accumulating poly-p-hydroxybutyric acid(PHB) were isolated from soil by enrichment culture technique. Among them, the strain designated as FL-027 had high PHB productivity and was identified as Alcaligenes. The optimal medium composition for cell growth was 8.0 $g/\ell$ of fructose and 3.0 $g/\ell$ of $(NH_4)_2S0_4$, equivalent to C/N ratio 5.04 at pH 7.0 and $30^{\circ}C$. To investigate the optimal conditions for the PHB accumulation, we divided the process into two stages; the first stage for the growth of the cell in nutrient-rich medium and the second stage for the PHB accumulation in nutrient-deficiency medium. The optimal conditions for PHB accumulation were 8.0 $g/\ell$ of fructose and 0.25 $g/\ell$ of $(NH_4)_2S0_4$, equivalent to C/N ratio 60 at pH 6.5 and $30^{\circ}C$. PHB accumulation was stimulated by deficiency of nutrients such as $NH_4^+$, $Ca^{2+}$, $SO_4^{2+}$ in medium. Among them. $NH_4^+$ deficiency was chosen because of its effectiveness. We found the inhibition of cell growth by fructose in batch culture. In order to keep the fructose concentration at an optimal level, intermittent feeding fed-batch culture was employed, and the cell concentration as high as 10.83 $g/\ell$ whose PHB content was responsible for 43% of the dry cell weight. The purified PHB was identified as homopolymer of 3-hydroxybutyric acid by using IR and $^1H-NMR$.
