한반도 신귀화식물: 가는타래사초 (사초과) (A new naturalized plant in Korea: Carex molestiformis Reznicek and Rothrock (Cyperaceae))

  • 국내 미기록종 가는타래사초(Carex molestiformis Reznik and Rothrock)를 강원도 평창군 벽파령 일대에서 처음으로 발견하였다. 본 분류군의 원산지는 북아메리카 미국의 남부지역(Missouri, Oklahoma, Arkansas등 11개 주)으로, 자생지에서는 주로 개활지나 강둑, 범람원 등에서 무리지어 밀생한다. 타래사초절(sect. Ovales)에 속하며, 자웅성 소수를 갖는 다년생 초본으로, 한 화서에 달리는 소수의 수가 2-4개로 적고, 과낭의 폭이 2.6-3.4 mm로 넓으며, 과낭 배면의 맥이 6-9개로 많아 국내에 자생하는 근연종인 타래사초(C. maackii Maxim.) 및 귀화종인 한석사초(C. scoparia)와 명확히 구별된다. 국명은 본 분류군이 형태적으로 타래사초와 유사하며, 식물체가 전체적으로 가늘어 '가는타래사초'로 신칭하였고, 분류군에 대한 기재문, 화상자료 및 도해 그리고 유사종에 대한 검색표를 제시하였다. 또한, 가는타래사초와 근연종의 유전적 차이를 확인하고자 3개(chloroplast DNA matK, ndhF, nuclear ribosomal DNA internal transcribed spacer) DNA barcode 지역의 부분 염기서열을 비교하여, 총 8개의 가는타래사초의 종특이적 단일염기다형성(single nucleotide polymorphisms)을 확인하였다.

정선황기의 세포유전학적 연구 (A cytogenetic study of Astragalus koraiensis Y. N. Lee)

  • 국내에 자생하는 황기속(Astragalus L.) 식물인 정선황기(A. koraiensis)의 핵형을 분석하고, 5S 및 45S rDNA 유전자를 이용한 FISH 실험에 기초하여 세포유전학적 연구를 수행하였다. 핵형 분석 결과, 정선황기의 체세포 염색체수는 2n = 16으로, 기본 염색체수는 x = 8임을 확인하였다. 염색체 조성은 6쌍의 중부염색체(염색체 1, 3, 4, 5, 6, 8)와 2쌍의 차중부염색체(염색체 2, 7)로 구성되었다. 정선황기의 염색체상에서의 FISH 결과, 1쌍의 45S rDNA signal이 5번 염색체의 동원체 부위에서 관찰되었고, 2쌍의 5S rDNA signal이 4번 염색체의 단완 말단부위와 7번 염색체의 동원체 부위에서 관찰되었다. 이는 기존의 황기 및 제주황기, 몽골황기(A. mongholicus) 와는 전혀 다른 FISH 패턴을 보이고 있어 정선황기가 고유종임을 암시하지만, 형태학적으로 유사한 갯황기(A. sikokianus) 및 A. bhotanensis 와의 비교연구를 수행하여 정확한 분류학적 처리가 이루어져야 할 것이다.

산화네오디뮴 기도투여에 따른 폐내 활성산소종 발생 및 DNA의 산화적 손상 (The Effect of Neodymium Oxide on the Generation of Reactive Oxygen Species and DNA Oxidative Damage by Intratracheal Instillation)

  • Objectives: This study was performed to assay the effect of neodymium oxide on the generation of reactive oxygen species and DNA oxidative damage by intratracheal instillation. Methods: Two groups of rats were exposed to neodymium oxide($Nd_2O_3$) via intratracheal instillation with doses of 0.5 mg and 2.0 mg, respectively. At two days and at 12 weeks after exposure, the contents of neodymium oxide in the lung, liver, kidney, heart and brain, leukocyte, olive tail moment, ROS, RNS, lactate dehydrogenase, albumin, cytokine and MDA from BALF were measured. Results: Neodymium oxide contents in the liver, kidney, heart, and brain were detected at less than $1{\mu}g/g$ tissue concentration. However, in the lungs at four weeks the highest amount were detected and then found to be drastically reduced at 12 weeks. ROS and RNS in bronchoalveolar lavage increased in concentration dependently at two days, four weeks and 12 weeks after neodymium oxide instillation. However, ROS and RNS decreased with the passage of time. At two days the total number of WBC in BALF in the high concentration group was significantly increased, and at four weeks the total number of WBC were significantly increased in the low and high concentration groups(p<0.01). At two days after exposure, the LDH of the low and high concentration groups was significantly increased. At 12 weeks, only the LDH of the high concentration group was significantly increased compared to in the control group(p<0.01). As a result of Comet assay, after two days, damage to the DNA of the low and high concentration groups was observed. Conclusions: Intratracheal instillation of neodymium oxide induces the generation of ROS and DNA damage in rats.

핵형분석과 FISH 기술을 이용한 솔비나무와 다릅나무의 세포유전학적 연구 (Cytogenetic Study of Maackia amurensis Rupr. & Maxim. and M. fauriei (Levl.) Takeda Using Karyotyping Analysis and the FISH Technique)

  • 국내에 자생하는 Maackia속 2종(솔비나무, 다릅나무)의 염색체수 및 핵형을 분석하고 5S와 45S rDNAs를 이용한 bicolor FISH를 수행하였다. 솔비나무와 다릅나무의 체세포 염색체수는 동일하게 2n = 2x = 18로 관찰되었고, 염색체의 길이는 $3.58{\sim}5.82{\mu}m$이였다. 솔비나무의 염색체 조성은 2쌍의 중부 염색체(염색체 1과 7번), 4쌍의 차중부 염색체(염색체 4, 6, 8, 9번), 그리고 3쌍의 차단부 염색체(염색체 2, 3, 5번)로 확인되었다. 다릅나무의 염색체는 4번이 차단부 염색체, 7번이 차중부 염색체로 솔비나무와 차이를 보였으나 다른 염색체의 동원체 위치는 유사하게 관찰되었다. 5S와 45S rDNA를 이용한 FISH 결과, 45S rDNA 유전자는 솔비나무와 다릅나무에서 각각 1쌍으로 관찰되었고 2번 염색체의 2차 협착 부위에서 확인되었다. 5S rDNA유전자를 이용한 물리지도 작성에서는 두 종 사이를 구별할 수 있는 결과를 확인할 수 있었다. 솔비나무의 경우 염색체 7번과 8번의 동원체 부위에서 2쌍이 각각 관찰되었고, 다릅나무에서는 염색체 7번과 8번뿐만 아니라 3번과 4번 염색체에서도 관찰되어 모두 4쌍으로 확인되었다. 따라서, 5S rDNA유전자를 이용한 FISH방법을 통해 세포학적으로 두 종을 구분할 수 있었다.

A Checklist of the Basidiomycetous Macrofungi and a Record of Five New Species from Mt. Oseo in Korea

  • Basidiomycetous macrofungi play important roles in maintaining forest ecosystems via carbon cycling and the mobilization of nitrogen and phosphorus. To understand the impact of human activity on macrofungi, an ongoing project at the Korea National Arboretum is focused on surveying the macrofungi in unexploited areas. Mt. Oseo was targeted in this survey because the number of visitors to this destination has been steadily increasing, and management and conservation plans for this destination are urgently required. Through 5 field surveys of Mt. Oseo from April to October 2012, 116 specimens of basidiomycetous macrofungi were collected and classified. The specimens were identified to the species level by analyzing their morphological characteristics and their DNA sequence data. A total of 80 species belonging to 57 genera and 25 families were identified. To the best of our knowledge, this is the first study to identify five of these species-Artomyces microsporus, Hymenopellis raphanipes, Pholiota abietis, Phylloporus brunneiceps, and Sirobasidium magnum-in Korea.

  • The wild silkworm, Bombyx mandarina, was believed the only ancestor of B. mori, inhabits the limited area of Eastern Asia including China, Korea and Japan. However, the geographic dimorphism of B. mandarina was reported with chromosome number and arylphorin gene. In connection with those dimorphism, we studied the genetic differences of ITS-2 region in rDNA purposing the differentiation and geographic variation within the species of B. mandarina. (omitted)

Variation in the number of nucleoli and incomplete homogenization of 18S ribosomal DNA sequences in leaf cells of the cultivated Oriental ginseng (Panax ginseng Meyer)

  • Background: Wild ginseng, Panax ginseng Meyer, is an endangered species of medicinal plants. In the present study, we analyzed variations within the ribosomal DNA (rDNA) cluster to gain insight into the genetic diversity of the Oriental ginseng, P. ginseng, at artificial plant cultivation. Methods: The roots of wild P. ginseng plants were sampled from a nonprotected natural population of the Russian Far East. The slides were prepared from leaf tissues using the squash technique for cytogenetic analysis. The 18S rDNA sequences were cloned and sequenced. The distribution of nucleotide diversity, recombination events, and interspecific phylogenies for the total 18S rDNA sequence data set was also examined. Results: In mesophyll cells, mononucleolar nuclei were estimated to be dominant (75.7%), while the remaining nuclei contained two to four nucleoli. Among the analyzed 18S rDNA clones, 20% were identical to the 18S rDNA sequence of P. ginseng from Japan, and other clones differed in one to six substitutions. The nucleotide polymorphism was more expressed at the positions 440-640 bp, and distributed in variable regions, expansion segments, and conservative elements of core structure. The phylogenetic analysis confirmed conspecificity of ginseng plants cultivated in different regions, with two fixed mutations between P. ginseng and other species. Conclusion: This study identified the evidences of the intragenomic nucleotide polymorphism in the 18S rDNA sequences of P. ginseng. These data suggest that, in cultivated plants, the observed genome instability may influence the synthesis of biologically active compounds, which are widely used in traditional medicine.

Composition and Diversity of Salivary Microbiome Affected by Sample Collection Method

  • Purpose: The purpose of this study was to investigate whether various saliva collection methods affect the observed salivary microbiome and whether microbiomes of stimulated and unstimulated saliva and plaque differ in richness and diversity. Methods: Seven sampling methods for unstimulated saliva, stimulated saliva, and plaque samples were applied to six orally and systemically healthy participants. Bacterial 16S ribosomal RNA genes of 10 major oral bacterial species, namely, Aggregatibacter actinomycetemcomitans, Porphyromonas gingivalis, Tannerella forsythia, Treponema denticola, Fusobacterium nucleatum, Prevotella intermedia, Prevotella nigrescens, Streptococcus mitis, Streptococcus sobrinus, and Lactobacillus casei, were analyzed by real-time polymerase chain reaction. We comprehensively examined the dependence of the amount of bacterial ribosomal DNA (rDNA), bacterial-community composition, and relative abundance of each species on sample collection methods. Results: There were significant differences in the bacterial rDNA copy number depending on the collection method in three species: F. nucleatum, P. nigrescens, and S. mitis. The species with the highest richness was S. mitis, with the range from 89.31% to 100.00%, followed by F. nucleatum, P. nigrescens, T. denticola, T. forsythia, and P. intermedia, and the sum of the proportions of the remaining five species was less than 1%. The species with the lowest observed richness was P. gingivalis (<0.1%). The Shannon diversity index was the highest in unstimulated saliva collected with a funnel (4.449). The Shannon diversity index was higher in plaque samples (3.623) than in unstimulated (3.171) and stimulated (3.129) saliva and in mouthwash saliva samples (2.061). Conclusions: The oral microbial profile of saliva samples can be affected by sample collection methods, and saliva differs from plaque in the microbiome. An easy and rapid technique for saliva collection is desirable; however, observed microbial-community composition may more accurately reflect the actual microbiome when unstimulated saliva is assayed.

한국산 꺽지과 어류 3종의 세포유전학적 연구 (Cytogenetic Analysis of Three Centropomid Species in Korea)

  • 한국산 꺽지과 어류 3종에 대한 세포유전학적 기초 자료를 얻기 위하여 염색체 핵형, 적혈구 세포 및 핵의 크기와 DNA 함량을 조사하였다 꺽지 (Coreoperca herzi), 꺽저기 (C. kawamebari) 및 쏘가리 (Siniperca schezeri)의 핵형은 각각 2n=48 (4SM+44A, T), 2n=48 (6SM+42A, T) 2n=48 (4SM+44A, T)이었으며, 암수간 heteromorphic한 성염색체는 발견할 수 없었다. 쏘가리의 적혈구 세포 및 핵의 표면적과 체적은 다른 2종보다 작았다. DNA함량은 적혈구 세포 크기에서 같은 경향을 보여, 쏘가리가 1.47pg/cell로 꺽지 (1.83pg/cell) 및 꺽저기 (1.85pg/cell)보다 유의하게 작았다.

Genetic Relationships of Four Korean Oysters Based on RAPD and Nuclear rDNA ITS Sequence Analyses

  • Random amplified polymorphic DNA (RAPD) marker and sequence analyses of the internal transcribed spacer (ITS) region of ribosomal DNA were used to assess phylogenetic relationships of four Korean oyster species. The average number of species-specific markers identified from five universal rice primers (URPs) by RAPD-PCR was 1.8 for Crassostrea gigas, 3.2 for C. nippona, 3.6 for C. ariakensis, and 4.6 for Ostrea denselamellosa. The length of the ITS (ITS1-5.8S-ITS2) region ranged from 1,001 to 1,206 bp (ITS1, 426-518 bp; 5.8S, 157 bp; and ITS2, 418-536 bp), while the GC content ranged from 55.5-61.1% (ITS1, 56.8-61.8%; 5.8S, 56-57.3%; and ITS2, 54.1-62.2%). A phylogenetic analysis of the oysters based on our RAPD, ITS1, and ITS2 sequence data revealed a close relationship between C. gigas and C. nippona and a distant relationship between the genera Crassostrea and Ostrea. Our results indicated that RAPD and ITS sequence analysis was a useful tool for the elucidation of phylogenetic relationships and for the selection of species-specific markers in Korean oysters.
