AN IMPROVED ALGORITHM FOR RNA SECONDARY STRUCTURE PREDICTION

  • A ribonucleic acid (RNA) is one of the two types of nucleic acids found in living organisms. An RNA molecule represents a long chain of monomers called nucleotides. The sequence of nucleotides of an RNA molecule constitutes its primary structure, and the pattern of pairing between nucleotides determines the secondary structure of an RNA. Non-coding RNA genes produce transcripts that exert their function without ever producing proteins. Predicting the secondary structure of non-coding RNAs is very important for understanding their functions. We focus on Nussinov's algorithm as useful techniques for predicting RNA secondary structures. We introduce a new traceback matrix and scoring table to improve above algorithm. And the improved algorithm provides better levels of performance than the originals.

NanoBio-Technology for Practical Implementation in Drug Discovery

  • To date, various nanobiotechnologicalapproaches for biosensors and drug development have been explosively studied. Despite of successful demonstrations, the new technologies hardly enjoyed routine applications in practical nanobiomedicine. Here, researchers trained at the interface of basic sciences and engineering are expected to play critical roles. In this tutorial, I will introduce recent studies which harness graphene derivatives for developing bioanalytical platforms to quantitatively analyze various enzyme activities and biomarkers. The systems rely on attractive interaction between graphene oxide and nucleic acids or phospholipids. Recently, one of the graphene-based bioassay system was applied to anti-viral drug screening and potent hit compounds were identified to treat hepatitis C. This study clearly shows that a new nanobio-technology can be routinely implemented in drug discovery, providing many advantages over conventional methods.

젖산 생산을 위한 배지 최적화 (Ootimization of Mekium Components for Lactic Acid Production)

  • Medium components for lactic acid production were optimized with a strain of Lactobacillus sp., isolated by our Lab. Nitrogen source was the key component and manganese ion was also important for lactic acid production in this strain. Optimal concentration of manganese ion was 0.03 g/l as MnSO$_{4}$ 4 - 5 H$_{2}$O base. Other mineral elements, however, had little effect on it. Among the nitrogen sources we examined, yeast extract showed the highest productivity. Yeast extract, the exellent but very expensive medium component, could be partially replaced by soytone until 60% dry base with higher productivity, or by tryptone enforced with vitamines and nucleic acids. In order to replace yeast extract completely, we examined several inexpensive nitrogen sources and their enzymatic hydrolyzates. The hydrolyzate of vital wheat gluten was the best of them.

  • Urine contains protein and nucleic acid(urea, uric acid, creatinine, ammonia, amino acids), various organic and inorganic materials, vitamin, hormone, enzyme etc. The examination of gualitative or quantitative change of the above mentioned materials and picking up the abnormal materials are useful to diagnose diseases. The test strip for examination of urine is applied to the routine test, monitoring of medical therapy and recurrence, self monitoring, and screeing in preventive medicine. We have been using multitest strip for checking the bacterial infection(nitrite), PH, protein, glucose, ketone body, urobilinogen, bilirubin and occult blood. So it is possible to diagnose three groups of diseases as follows, abnormal metabolism of the carbohydrate, diseases of kidney and other urogenital system, diseases of hepatobiliary system and hemolytic disease causing abnormal metabolism of bile juice.

  • Polyalkylene phosphates - polymers that are built on the repeating unit of the diester of phosphoric acid: -[OP(O)(OH)Oalkylene]-, are known to form backbones of nucleic and teichoic acids. Various synthetic ways will be reported for the synthesis of the bare chains, where "alkylene" in the formula above means mostly two or three methylene groups. Some other structures have also been prepared. Several applications of these polymers are to be discussed, namely as liquid membranes, as components of two-block copolymers (ionic-nonionic diblock dihydrophilic) used as modifiers of CaCO3 crystallization, and as components of the inorganic-polymer hybrid materials. Some other applications in the biomedical field will also be discussed.

Adenine Inhibits B16-F10 Melanoma Cell Proliferation

  • Adenine, a purine base, is a structural component of essential biomolecules such as nucleic acids and adenine nucleotides. Its physiological roles have been uncovered. Adenine suppresses IgE-mediated allergy and LPS-induced inflammation. Although adenine is known to inhibit lymphocyte proliferation, the effect of adenine to melamoma cells is not reported. Here, we investigated the growth inhibitory effects of adenine on B16-F10 mouse melanoma cells. Adenine suppressed the proliferation of B16-F10 cells in dose-dependent manner with the maximal inhibitory dose of 2 mM. Adenine treatment induced cell death molecular markers such as PARP and caspase 3 cleavages. Pan-caspase inhibitor z-VAD dramatically rescued the cell death molecular markers, cell proliferation recovered marginally. These results provide the possibility of adenine to be used as an anti-tumor agent.

Biomarkers and genetic factors for early prediction of pre-eclampsia

  • Pre-eclampsia is known to cause considerable maternal morbidity and mortality. Thus, many studies have examined the etiopathogenesis of pre-eclampsia. While many pathophysiological factors related to pre-eclampsia have been identified, the precise etiopathogenesis of pre-eclampsia remains unclear. Numerous studies have identified factors for the early prediction for pre-eclampsia to lead to preparation and closer observation on pre-eclampsia when it occurs. This article reviews on current studies of biomarkers and genetic factors related to pre-eclampsia, which may be important for developing strategies for early prediction of pre-eclampsia.

  • Degradation of deoxyribonucleic acid(DNA) and ribonucleic acid(RNA) by cell-free extract of mixed rumen protozoa of buffalo rumen was investigated. DNA was observed to be degraded rapidly during an initial incubation period of 2 hr with simultaneous appearance of degradation products. RNA on the other hand recorded a rapid degradation during an initial incubation period of 1 hr. RNA degradation products appeared upto an incubation period of 2 hr. DNA was observed to degrade into oligo- and mononucleotides. pyrimidine nucleosides, purine nucleoside adenosine and bases xanthine, hypoxanthine and thymine. Degradation products of RNA comprised of pyrimidine nucleosides, purine nucleoside, adenosine and bases xanthine, hypoxanthine and uracil besides oligo- and mononucleotides.

배추에서 분리한 순무 모자이크 바이러스의 특성 및 역전사 중합효소 연쇄반응법(RT-PCR)을 이용한 검정 (Characterization and RT-PCR Detection of Turnip Mosaic Virus Isolated from Chinese Cabbage in Korea)

  • Turnip mosaic virus)TuMV-Ca) was isolated from a Chinese cabbage showing severe mosaic and black necrotic spots symptoms in Korea. The virus was identified as a strain of TuMV by its host range test, particle morphology, serology, double stranded RNA analysis. For detection of the virus, reverse transcription and polymerase chain reaction(RT-PCR) was performed with a set of 18-mer TuMV-specific primers to amplify a 876 bp DNA fragment The virus was rapidly detected from total nucleic acids of virus infected tissues as well as native viral RNA of purified virion particles by RT-PCR. Detection limit of the viral RNA by RT-PCR was 10 fg.

Role of tea catechins in prevention of aging and age-related disorders

  • Tea polyphenols especially catechins have long been studied for their antioxidant and radical scavenging properties. Scientists throughout the world have investigated the usefulness of the regular green tea consumption in several disease conditions. In-vitro and in-vivo experiments on catechins especially epigallocatechingallate have revealed a significant role in many ways. Reactive oxygen species have been increasingly implicated in the pathogenesis of many diseases and important biological processes. Toxic effects of these oxidants, commonly referred to as oxidative stress, can cause cellular damage by oxidizing nucleic acids, proteins, and membrane lipids. Oxidative stress has been related to aging and age related disorders. It is found that in a wide variety of pathological processes, including cancer, atherosclerosis, neurological degeneration, Alzheimer's disease, ageing and autoimmune disorders, oxidative stress has its implications. Catechins have been reported to be useful in combating aging and age related disorders like cancer, cardiovascular disorders and neurodegenerative diseases. In this mini review we will discuss such studies done across the globe.
