Localization of Anti-Actin-Gold Particles (10 nm) Labeled to Nuclear Actin of Urechis Sperm and Spermatids (항-액틴-금 입자 표지에 의한 개불(Urechis unicinctus) 정자 및 정세포 핵 Actin의 분포)

  • Urechis unicinctus spermatogenic cells, sperm and spermatids, prepared from testis are investigated to identify nuclear actin using amoeba monorlonal anti-actin as the first Ab and gold particles (10 nm) conjugated mouse IgG (immunogold) as the Ab marker. The Ag-Ab reactions analyzed the localization of nuclear actin of the spermatogenic cells and the immunogold particles incorporated mainly with nuclear matrices. A few immunogold particles are merged into the acrosomes and the other architectures of spermatogenic cells, such as mitochondrion and centrioles. It is often observed and there is a tendency in which the incorporated immunogold particles are increased in number in the nuclear matrices of sperm compared with that of spermatids The increments and decrements of the incorporated immunogold particles according to developmental stages and the spermatogenic architec-tures are interpreted and discussed in aspect of acrosomal function and of nuclear condensation of spermatids.

The Performance of Ictal Brain SPECT for Localizing Epileptogenic Foci in Temporal Lobe Epilepsies (측두엽 간질에서 발작기 뇌관류 SPECT의 간질병소 국소화 성능)

  • Anterior temporal lobectomy has become a widely used resective surgery in patients with medically intractable temporal lobe epilepsies. Prerequisites of this resection include the accurate localization of the epileptogenic focus and the determination that the proposed resection would not result in unacceptable postoperative memory or language deficits. The purpose of this study was to evaluate the performance of ictal SPECT compared to MRI findings for localization of epiletogenic foci in this group of patients. 11 patients who had been anterior temporal lobectomy were evaluated with ictal $^{99m}Tc$-HMPAO SPECT and MRI. MRI showed 8/11(73%) concordant lesion to the side of surgery and ictal SPECT also showed 8/11(73%) concordant hyperperfusion. In 3 cases with incorrect or nonlocalizing findings of MRI, ictal SPECT showed concordant hyperperfusion. In 2 cases confirmed by pre-resectional invasive EEG, MRI showed bilateral and contralateral lesion but ictal SPECT showed concordant hyperperfusion. 3 delayed injection of ictal SPECT showed discordant hyperperfusion. Thus, ictal SPECT was a useful method for localizing epileptogenic foci in temporal lobe epilepsis and appeared complementay to MRI.

  • Through a project "Development of Nuclear Safety Parameter Monitoring System", a nuclear data link system was established between Kori nuclear unit 2 and Nuclear Safety Center. We present in this paper the selected parameter sets, a description of the developed pseudo-network software and the functional descriptions of the equipments involved. We also include the conceptual design of the Kori four unit ERF/SPDS system, along with the localization direction for the related software and hardware. hardware.

Subcellular Localization of Novel Stress Protein VISP (새로운 스트레스 단백질인 VISP의 세포내 위치)

  • Previously we demonstrated that virus-inducible stress protein (VISP) is induced in fish cells by the infection of a fish rhabdovirus. In this paper, we investigated the subcellular localization of the VISP and determined the region of VISP responsible for the subcellular localization. The CHSE-214 cells were stained with monoclonal antibody raised against VISP and observed with confocal microscope to detect the endogenous VISP. The results showed that the VISP localizes to the perinuclear region as spots. A plasmid expressing VISP fused to enhanced green fluorescent protein (EGFP) was constructed. The transient expression of full-length VISP fused to EGFP in CHSE-214 cells confirmed the spot formation of the VISP at perinuclear region. To determine the region responsible for the perinuclear localization of the VISP, we constructed a series of deletion mutants and, by using these deletion mutants, we found that C-terminal region of the VISP (aa 612-710) is essential for the perinuclear distribution of VISP and that this region contained nuclear receptor binding motif (691-TLTSLLL-697). Our results suggest that VISP localizes to the perinuclear region and C-terminal regions are important for this localization. Further studies on the role of the perinuclear localization of VISP in IHNV growth mali reveal the novel mechanism of IHNV pathogenecity.

Host Cell Nuclear Localization of Shigella flexneri Effector OspF Is Facilitated by SUMOylation

  • When Shigella infect host cells, various effecter molecules are delivered into the cytoplasm of the host cell through the type III secretion system (TTSS) to facilitate their invasion process and control the host immune responses. Among these effectors, the S. flexneri effector OspF dephosphorylates mitogen-activated protein kinases and translocates itself to the nucleus, thus preventing histone H3 modification to regulate expression of proinflammatory cytokines. Despite the critical role of OspF, the mechanism by which it localizes in the nucleus has remained to be elucidated. In the present study, we identified a potential small ubiquitin-related modifier (SUMO) modification site within OspF and we demonstrated that Shigella TTSS effector OspF is conjugated with SUMO in the host cell and this modification mediates the nuclear translocation of OspF. Our results show a bacterial virulence factor can exploit host post-translational machinery to execute its intracellular trafficking.

Functional Neuroimaging in Epilepsy: FDG-PET and SPECT (간질에서의 기능적 뇌영상:양전자방출단층촬영과 단일광전자방출 단층촬영)

  • Finding epileptogenic zone is the most important step for the successful epilepsy surgery. F-18 fluorodeoxyglucose positron emission tomography (FDG-PET) and single photon emission computed tomography (SPECT) can be used in the localization of epileptogenic foci. In medial temporal lobe epilepsy, the diagnostic sensitivity of FDG-PET and ictal SPECT is excellent. However, detection of hippocampal sclerosis by MRI is so certain that use of FDG-PET and ictal SPECT in medial temporal lobe epilepsy is limited for some occasions. In neocortical epilepsy, the sensitivities of FDG-PET or ictal SPECT are fair. However, FDG-PET and ictal SPECT can have a crucial role in the localization of epileptogenic foci for non-lesional neocortical epilepsy. Interpretation of FDG-PET has been recently advanced by voxel-based analysis and automatic volume of interest analysis based on a population template. Both analytical methods can aid the objective diagnosis of epileptogenic foci. Ictal SPECT was analyzed using subtraction methods and voxel-based analysis. Rapidity of injection of tracers, ictal EEG findings during injection of tracer, and repeated ictal SPECT were important technical issues of ictal SPECT. SPECT can also be used in the evaluation of validity of Wada test.

Specific Localization of DNMT1 in Mouse and Bovine Preimplantation Embryos

  • DNA methylation is a covalent modification of DNA that can modulate gene expression and is now recognized as a major component of the epigenome. During evolution, the dinucleotide CpG has been progressively eliminated from the genome of higher eukaryotes and is present at only 5% to 10% of its predicted frequency. Approxymately 80% of the remaining CpG sites contain methylated cytosines in most vertebrates and they are distributed in a pattern that is unique in each tissue and is inversely correlated with gene expression. The pattern of methylation is faithfully maintained during cell division by the enzyme Dnmt1, the maintenance DNA methyltransferase, which catalyzes the transfer of a methyl group from S-adenosyl-methionine to the 5'-position of the cytosine ring. We have been identified bovine Dnmt1 cDNA full-length recently (AY173048) Little is known on the functions of Dnmt1 in bovine preimplantation embryos. Thus, we analyzed the specific pattern of Dnmt1 in in vitro derived/nuclear transfer bovine and in vivo derived mouse embryos to monitor the epigenetic reprogramming process. We investigated these process by using indirect immunofluresence with an antibody to Dnmt1. According to other studies, Dnmt1 accumulates in nuclei of early growing oocytes but is sequestered in the cytoplasm of mature oocytes. In 2-cell and 4-cell embryos, Dnmt1 is cytoplasmic, but at the 8-cell stage, it is present only in the nucleus. By the blastocyst stage, Dnmt1o is again found only in the cytoplasm. Thus, nuclear localization of Dnmt1o in preimplantation embryos is limited to the 8-cell stages After implantation, Dnmt1 is localized in the nucleus in mouse. However, we have found different patterns of Dnmt1 nuclear localization. Though we used the common antibody, immune-localization data revealed that Dnmt1 antibody have been detected at the nucleus in 1-cell to blastocyst embryos. Therefore, maybe we think that the functions of Dnmt1 between bovine and mice are different. In order to Identify the mechanisms that regulate DNA methylation in bovine preimplantation embryo, we have plans on using bovine oocyte and somatic specific Dnmt1 antibodies.

  • Finding the exact location of faults in a program requires enormous time and effort. Several fault localization methods based on control flows of a program have been studied for decades. Unfortunately, these methods are not applicable to programs based on data-flow languages. A recently proposed mutation-based fault localization method is applicable to data-flow languages, as well as control-flow languages. However, there are no studies on the effectiveness of the mutation-based fault localization method for data-flow based programs. In this paper, we provided an experimental case study to evaluate the effectiveness of mutation-based fault localization on programs implemented in Function Block Diagram (FBD), a widely used data-flow based language in safety-critical systems implementation. We analyzed several real faults in the implementation of FBD programs of a nuclear reactor protection system, and evaluated the mutation-based fault localization effectiveness for each fault.

  • We present a case in which a patient with acute hemorrhagic gastritis demonstrated abnormal gastrointestinal accumulation of radiotracer during $^{99m}Tc$-methylene diphosphonate(MDP) skeletal scintigraphy. A hemorrhagic gastritis was subsequently demonstrated by endoscopy. The mechanism for the intestinal localization of $^{99m}Tc$-MDP in this patient is not clear, but we guess that the extravasated blood containing the radiopharmaceutical cannot recirculate and stays at the bleeding site, so we can see the intestinal activity.

Design and performance prediction of large-area hybrid gamma imaging system (LAHGIS) for localization of low-level radioactive material

  • In the present study, a large-area hybrid gamma imaging system was designed by adopting coded aperture imaging on the basis of a large-area Compton camera to achieve high imaging performance throughout a broad energy range (100-2000 keV). The system consisting of a tungsten coded aperture mask and monolithic NaI(Tl) scintillation detectors was designed through a series of Geant4 Monte Carlo radiation transport simulations, in consideration of both imaging sensitivity and imaging resolution. Then, the performance of the system was predicted by Geant4 Monte Carlo simulations for point sources under various conditions. Our simulation results show that the system provides very high imaging sensitivity (i.e., low values for minimum detectable activity, MDA), thus allowing for imaging of low-activity sources at distances impossible with coded aperture imaging or Compton imaging alone. In addition, the imaging resolution of the system was found to be high (i.e., around 6°) over the broad energy range of 59.5-1330 keV.
