Three-step PCR and RFLP Genotyping of the Swine Ryanodine Receptor Gene Using Aged Single Hair Follicles Delivered by General Mail

  • We have developed a reliable and noninvasive method for swine genotyping of single locus nuclear gene with aged single hair follicles delivered by general mail. The method is based on booster and nested PCR amplification with step-wise increase of primers and dNTPs concentrations followed by restriction endonuclease digestion. To establish this method, the ryanodine receptor (RYR 1) locus which is an economically important trait in swine industry was employed for genotyping experiment. The 3-step PCR amplication method is much less dependent on the quantity and quality of template DNA and produces enough amplification product for the detection on the ethidium bromide-stained gel such as RFLP analysis. A total of 120 pigs were subjected to the RYR 1 genotyping analysis using three-step PCR method which amplified enough quantity of PCR products from the aged single hair follicles for RFLP analysis and genotyping results were identical to the results of the corresponding ethanol-fixed skeletal muscle tissue. This approach will be a great help for porcine breeders and investigators in genotyping of swine. They can receive genotyping results later by simply plucking single hairs of their pigs at farm and sending them in general mail to the diagnostic laboratory which eliminates the inconveniences to collect ear tissue or blood cells from pigs, or the investigator's need for travel to farms in order to collect fresh hair samples.

  • While athletic abilities such as speed, endurance and recovery are important in the horse, genes related to these abilities have not been extensively investigated. Here, we characterized the horse peroxisome proliferator-activated receptor delta (PPARδ) gene and analyzed the expression of PPARδ during exercise. PPARδ is a known regulator of β-oxidation, muscle fiber transformation, and running endurance. Through evolutionary analysis using the synonymous and non-synonymous mutation ratio, it was revealed that positive selection occurred in the horse PPARδ gene. Two important domains related to nuclear hormone receptors, C4 zinc finger and ligand binding domain, were also found to be conserved well in horse PPARδ. Horse PPARδ was expressed ubiquitously in many tissues, but the expression level was various depending on the tissues. In the skeletal muscle, PPARδ increased about 2.5 folds after 30 min of exercise. Unlike in muscle, the increase of PPARδ expression was observed at 60 min but not 30 min of exercise in leukocytes. This finding might be useful for testing the endurance of horse using blood samples. Conclusively, the horse PPARδ gene is evolutionarily conserved well and can be used as a biomarker of endurance in horse.

  • Recently, as the demand for high-precision parts increases due to industrial development, a machine tool system for ultra-precision machining and polishing has been actively developed. As a result, there is an increasing demand for ultra-precision surface roughness along with dimensional processing. However, due to the increase in processing time due to the demand for ultra-precise surfaces and enormous facility investment, it is difficult to secure competitiveness. The polishing process using the abrasive film in super precision machining has been applied to machines, electronic devices, aerospace, and medical fields. Super finishing using the abrasive film which is applied in the industrial field recently can achieve high surface roughness in a short time. Also, application of industrial field is increasing due to advantages such as low noise and low dust. Recently, researches on stainless steel having strong resistance to corrosion, heat resistance, heat resistance, toughness and weldability have been actively conducted with respect to the nuclear energy industry or marine development. Therefore, in this study, surface roughness and residual stress were measured after SUS304 polishing using dynamic analysis of film polishing apparatus and polishing film.

  • Japanese low activation terrific steel(JLF) is a good material for the parts of heat exchanger such as blanket and diverter. At first, JLF was developed as a candidate for structural materials in nuclear fusion applications. However, the development of the jointing technique of JLF steel to other materials is important for wide applications of this material to the industry fields. Recently the jointing technologies including diffusion bonding, brazing, roll bonding, explosive bonding and hot iso-static pressing have been studied for the heterogeneous materials of JLF-1 steel(Fe-9Cr-2W-V-Ta) and stainless steel(STS304). Friction welding is one of the most popular welding methods for two different kinds of materials. In this paper, the JLF-1 steel was jointed to SIS304 by friction welding method and the optimal conditions of the friction welding discussed. Acoustic emission was used as a nondestructive technique to evaluate the weld quality in processing.

  • Quantifying the extent and characteristics of the excavation damage zone(EDZ) is important for the nuclear waste industry which relies on the sealing of underground openings to minimize the risk for radionuclide transport. At AECL's Underground Research Laboratory(URL) the Tunnel Sealing Experiment(TSX) was conducted and the tunnel geometry and orientation relative to the stress field had been selected to minimize the potential for the development of an EDZ. The extent and characteristics of the EDZ was measured using velocity profiling and permeability measurements in radial boreholes. The results from this EDZ characterization are used in this paper to evaluate a modeling fir estimating the extent of the EDZ. The methodology used a damage model formulated in the Universal Distinct Element Code and calibrated to laboratory properties. This model was then used to predict the extent of crack initiation and growth around the TSX tunnel and the results compared to the measured damage. The development of the damage zone in the numerical model was found to be in good agreement with the field measurements.

Britanin Suppresses IgE/Ag-Induced Mast Cell Activation by Inhibiting the Syk Pathway

  • The aim of this study was to determine whether britanin, isolated from the flowers of Inula japonica (Inulae Flos), modulates the generation of allergic inflammatory mediators in activated mast cells. To understand the biological activity of britanin, the authors investigated its effects on the generation of prostaglandin D2 (PGD2), leukotriene C4 (LTC4), and degranulation in IgE/Ag-induced bone marrow-derived mast cells (BMMCs). Britanin dose dependently inhibited degranulation and the generations of PGD2 and LTC4 in BMMCs. Biochemical analyses of IgE/Ag-mediated signaling pathways demonstrated that britanin suppressed the phosphorylation of Syk kinase and multiple downstream signaling processes, including phospholipase Cγ1 (PLCγ1)-mediated calcium influx, the activation of mitogen-activated protein kinases (MAPKs; extracellular signal-regulated kinase 1/2, c-Jun NH2-terminal kinase and p38), and the nuclear factor-κB (NFκB) pathway. Taken together, the findings of this study suggest britanin suppresses degranulation and eicosanoid generation by inhibiting the Syk-dependent pathway and britanin might be useful for the treatment of allergic inflammatory diseases.

  • As the amount of radioactive waste and related information to be managed are increasing, some organizations are trying or planning to computerize the management on radioactive waste. When we consider that information on safe management of radioactive waste should be used In association with national radioactive waste management project, standardization of data form and its protocol is required. Korea Institute of Nuclear Safety(KINS) will establish and operate nationwide integrated database in order to effectively manage a large amount of information on national radioactive waste. This database allows not only to trace and manage the trend of radioactive waste occurrence and in storage but also to produce reliable analysis results for the quantity accumulated. Consequently, we can provide necessary information for national radioactive waste management policy and related industry's planing. This study explains the database design which is the essential element for information management.

  • This study is to develop welfare programs based on the characteristics of middle-or-old-aged woman workers in manufacturing industry. 7cities and 2 counties in Kyunggi-Do were selected. for survey considering their accessibility. Total 471 woman workers were interviewed personally and the data has been analyzed using SPSS. First, the findings show that their family structure is characterized by nuclear family and they have school-aged children who need someone else's care, therefore they need child related services to help them such as after school care. Second, they have few welfare facilities in their work and they have never tried to find any outside their work. Thus, it is necessary to prepare welfare programs in their working place. The welfare programs they need most are discount stores, medical service, counseling service, and so on. In conclusion, the welfare needs of woman workers cannot be thought without considering family structure of woman workers, the degree of welfare facilities in the work, and their welfare needs.

  • Korea government aims to generate 20 percent of its electricity with clean, renewable energy by 2030, while reducing its reliance on fossil fuel and nuclear power plants. Technically, solar energy has resource potential that far exceeds the entire global energy demand. Solar energy industry has experienced phenomenal growth in recent years due to both technological improvements resulting in cost reductions and government policies for renewable energy development and utilization. Even though solar power generation has several advantages over other forms of electricity generation, the major problem is the requirement of land which is scarcely available in the local site and its cost. This study analyzes the available capacity of landing and floating solar plants for the case of chonnam province in korea. The results of design capacity show about 7.5GW for landing and 1.5GW for floating solar power plant. Also, with a purpose to comprehend intention-behaviour gap about acceptance of solar community, the solutions are suggested.

  • The Ni-base superalloy GTD111 DS is used in the first stage blade of high power land-based gas turbines. Advanced repair technologies of the blade have been introduced to the gas turbine industry over recent years. The effect of the filler metal powder on Transient Liquid Phase bonding phenomenon and tensile mechanical properties was investigated on the GTD111 DS superalloy. At the filler metal powder N series, the base metal powders fully melted at the initial time and a large amount of the base metal near the bonded interlayer was dissolved by liquid inter metal. Liquid filler metal powder was eliminated by isothermal solidification which was controlled by the diffusion of B into the base metal. The solids in the bonded interlayer grew from the base metal near the bonded interlayer inward the insert metal during the isothermal solidification. The bond strength of N series filler metal powder was over 1000 MPa. and γ phase size of N series TLP bonded region was similar with base metal by influence of Ti, Al elements. At the insert metal powder M series, the Si element fluidity of the filler metal was good but microstructure irregularity on bonded region because of excessive Si element. Nuclear of solids formed not only from the base metal near the bonded interlayer but also from the remained filler metal powder in the bonded interlayer. When the isothermal solidification was finished, the content of the elements in the boned interlayer was approximately equal to that of the base metal. But boride and silicide formed in the base metal near the bonded interlayer. And these boride decreased with the increasing of holding time. The bond strength of M series filler metal powder was about 400 MPa.
