Ultrastructure in Porcine Oocytes following Intracytoplasmic Injection of Murine Spermatozoa

  • Although successful pronuclear formation and apposition were seen in porcine oocytes following mouse sperm injection, little is known on the morphology of male and female pronuclei following sperm injection. The objective of this study is to describe the ultrastructure of porcine zygote following murine sperm injection in relation to the chronology of pronuclear S phase. At 40h ~ 44h following in vitro maturation, Cumulus cells were removed in TCM-HEPES with 0.1% hyaluronidase. Then, spermatozoa was injected into the cytoplasm of oocytes. After. injection, all oocytes were transferred to NCSU23 medium and cultured at 39$^{\circ}C$ under 5% $CO_2$ in air. Oocytes were fixed in 2% glutaraldehyde in Dulbeccos phosphate-buffered saline and observed by Transmission Electron Microscopy. Nuclear precursor bodies were observed in each pronucleus. A cluster of large and small granules was attached in the nucleolus precursor body. After the apposition of male and female chromatin, chromatin condensation was observed throughout the nucleoplasm and nucleolus precursor bodies and condensed chromatin in contact with clusters of small and large granules and the nuclear envelope were found in apposed pronuclear regions. These results suggest that non-species specific nuclear cytoplasmic interactions take place during pronuclear formation and apposition following sperm injection.

Effect on Embryogenesis and Ultrastructural Behavior of lamda-DNA Following Microinjection into Fertilized Eggs of Xenopus laevis (Xenopus 수정란에 미세주입된 ${\lambda}-DNA$의 배발생에 미치는 영향 및 미세 구조에 관한 연구)

  • In an attempt to produce transgenic amphibia, bacteriophage ${\lambda}-DNA$ was microinjected into fertilized eggs of Xenopus laevis, and the effect on early embryogenesis and the ultrastructural behavior of exogenous DNA were investigated. The effect of microinjected gene on embryonic development showed differences according to the concentration of injected DNA and the incubation temperature. Various concentrations of ${\lambda}-DNA$ were tested. Among those, microinjection of 1-2 ng DNA dissolved in 20 nl TE buffer was not shown to disturb normal embryonic development and was recorded the highest survivability to the late tadpole stage (Stage 43); however, injection of increased concentrations of DNA than above provoked irregular cleavages or abnormal appearances, which resulted in reduced survivability. When the injected embryos were incubated at low temperatures (e.g., $12^{\circ}C$), 54.5% of the embryos developed to Stage 43, whereas 42.4% survived when incubated at room temperature. The survivability showed also differences according to the injection site. 58.0% of the embryos developed to Stage 43 when microinjected into the vegetal pole, whereas 44.9% survived when microinjected into the animal pole. To understand the structural fate or behavior of injected DNA a combined light and electron microscopical study was applied. The nucleus-like structure was observed in the ${\lambda}$ DNA-injected embryos, which was quite a similar to the interphase nuclei of normal Xenopus laevis. The nucleus-like structure showed the typical double-layered nuclear membrane and nuclear complexes; however, it consisted of unusual structures such as furrows of nuclear envelope into the nucleoplasm.

Generation of Design Time History Complying With Japanese Seismic Design Standards for Nuclear Power Plants (일본 원전 내진설계 기술기준을 적용한 모의지진파(가속 도시간이력) 작성)

  • Seismic designs for Korean nuclear power plants (NPPs) under earthquakes' design basis are noticed due to the recent earthquake events in Korea and Japan. Japan has developed the technologies and experiences of the NPPs through theoretical research and experimental verification with extensively accumulated measurement data. This paper describes the main features of the design-time history complying with the Japanese seismic design standard. Proper seed motions in the earthquake catalog are used to generate one set of design time histories. A magnitude and epicentral distance specify the amplitude envelope function configuring the shape of the earthquake. Cumulative velocity response spectral values of the design time histories are compared and checked to the target response spectra. Spectral accelerations of the time histories and the multiple-damping target response spectra are also checked to exceed. The generated design time histories are input to the reactor building seismic analyses with fixed-base boundary conditions to calculate the seismic responses. Another set of design time histories is generated to comply with Korean seismic design procedures for NPPs and used for seismic input motions to the same reactor containment building seismic analyses. The responses at the dome apex of the building are compared and analyzed. The generated design time histories will be also applied to subsequent seismic analyses of other Korean standard NPP structures.

Dimensional synthesis of an Inspection Robot for SG tube-sheet

  • To ensure the operational safety of nuclear power plants, we present a Quadruped Inspection Robot that can be used for many types of steam generators. Since the Inspection Robot relies on the Holding Modules to grip the tube-sheet, it can be regarded as a hybrid robot with variable configurations, switching between 4-RRR-RR, 3-RRR-RR, and two types of 2-RRR-RR, and the variable configurations bring a great challenge to dimensional synthesis. In this paper, the kinematic model of the Inspection Robot in multiple configurations is established, and the analytical solution is given. The workspace mapping is analyzed by the solution-space, and the workspace of multiple configurations is decomposed into the workspace of 2-RRR to reduce the analysis complexity, and the workspace calculation is simplified by using the envelope rings. The optimization problem of the manipulator is transformed into the calculation of the shortest contraction length of the swing leg. The switching performance of the Inspection Robot is evaluated by stride-length, turning-angle, and workspace overlap-ratio. The performance indexes are classified and transformed based on the proportions and variation trends of dimensional parameters to reduce the number of optimization objective functions, and Pareto optimal solutions are obtained using an intelligent optimization algorithm.

Ultrastructure of the Granular Glands in the Amphibian Skin (양서루 피부 과립선의 미세구조)

  • The authors observed the ultrastructure of the granular glands in the amphibian skin with an electron microscope. The specimens from the experimental animals (Bombina orientalis, Bufo bufo gargarizans, Rana nigromaculata and Rana rugosa) were fixed in 2.5% glutaraldehyde-paraformaldehyde fixative in phosphate buffer at pH 7.2 prior to fixation in 1% osmium tetroxide, dehydrated in graded ethanol and acetone, embedded in Epon 812 mixture, and sectioned with a LKB-ultramicrotome. the ultrathin sections were contrasted with uranyl acetate and lead citrate and observed with a JEOL-100B electron microscope. The results were as follws: 1. The granular gland in the amphibian skin consisted of the glandular epithelial and the myoepithelial cells. 2. The epithelial cells of the granular gland in the amphibian skin consisted of the dark cells but the light cells were also observed in that of Bombina orientalis. 3. The granular glands of the amphibian skin were in holocrine fashion. 4. The nuclei of the epithelial cells of the amphibian cutaneous granular glands were round or oval and showed small and large inforldings of nuclear envelope. Heterochromatins were mainly distributed near the nuclear envelope. Mitochondria were mainly distributed in the perinuclear portion and rough-surfaced endoplasmic reticulums were developed in the cytoplasm but smooth-surfaced endoplasmic reticulums were not well developed. 5. Secretory granules were round or oval and electron-dense and less electron-dense granules were observed. 6. The authors infer that the differences in electron density of the secretory granules in the granular glands of the amphibian skin are due to difference in the concentrations of secretory substances as related to the processes of its formation, and that those chemical components are identical.

  • The spermatozoa of bagrid catfish, Pseudobagrus fulvidraco are approximately $76{\mu}m$ in length, and a relatively simple and elongated cell composed of a spherical head, a short middle piece and a tail. The ultrastructure of spermatozoa of P. fulvidraco is characterized by the following features. The acrosome is absent as in most teleost. The round nucleus measuring about $1.67{\mu}m$ in length and diameter is depressed with a deep nuclear fossa. The nuclear fossa, the length of which is about three-fifths of the nuclear diameter, contains the proximal and distal contrioles. The two centrioles are oriented approximately $160^{\circ}$ to each other. The filamentous materials give rise to satellite appendages arranged tangentially from the triplets of the distal centriole and the doublets of the anterior end of the axoneme toward the nuclear envelope. The mitochondria are not fused and their number is 20 or more. They are arranged in two or three layers and two rings within the cytoplasmic collar and surround the axoneme. They are separated from the axoneme by the cytoplasmic canal. The axoneme is of the 9+2 microtubular pattern and has inner but no outer dynein arms. The two lateral fins are in the same plane with the two central microtubules, the doublets 3 and 8, which are ultrastructural characteristics of the sperm tail unlike other siluroids lacking the lateral fins.

Seismic Response Analysis of NPP Containment Structures to Improve the Guidelines of Strong Motion Duration (강진지속시간 기준 개선을 위한 원전 격납구조물의 지진응답해석)

  • This paper addresses a fundamental study that is necessary to complement and improve the current domestic design specifications for the strong motion duration criterion and the envelope function of artificial accelerograms that can be applied to the earthquake-proof design of nuclear structures. The criteria for the design response spectra and strong motion duration suggested by USNRC and ASCE Standard 4-98 are commonly being used in the profession, and they are first compared with each other and reviewed. By applying 209 real strong earthquake records that are greater than 5 in magnitude at rock sites to the strong motion duration criterion in ASCE 4-98, an empirical regression model that predicts the strong motion duration as a function of the earthquake magnitude was then developed. Using synthetically generated earthquake time histories for the 10 cases whose strong motion durations varied from 6 to 20 seconds, extensive seismic analyses were finally conducted to identify the effects of the strong motion durations on the seismic responses of the nuclear power plant containment structures.

Evaluation of Gap Heat Transfer Model in ELESTRES for CANDU Fuel Element Under Normal Operating Conditions (CANDU형 핵연료봉의 정상상태 계산용 ELESTRES 코드내 간극 열전달 모델 평가)

  • The gap conductance between the fuel and the sheath depends strongly on the gap width and has a significant influence on the amount of initial stored energy. The modified Ross and Stoute gap conductance model in ELESTRES is based on a simplified thermal deformation model for steady-state fuel temperature calculations. A review on a series of experiments reveals that fuel pellets crack relocate, and are eccentrically positioned within the sheath rather than solid concentric cylinders. In this paper, the hue recently-proposed gap conductance models (offset gap model and relocated gap model) are described and are applied to calculate the fuel-sheath gap conductances under experimental conditions and normal operating conditions in CANDU reactors. The good agreement between the experimentally-inferred and calculated gap conductance values demonstrates that the modified Ross and Stoute model was implemented correctly in ELESTRES. The predictions of the modified Ross and Stoute model provide conservative values for gap heat transfer and fuel surface temperature compared to the offset gap and relocated gap models for a limiting power envelope.

Epidermal Homeostasis and Dry Skin Management (표피항상성과 건조피부의 관리)

  • Epidermis is one of the most dynamic organs in the human body. Multiple layers of keratinocytes in the epidermis continuously undergo proliferation, differentiation, and desquamation cycles, which is the bases of maintaining the epidermal homeostasis. Epidermal homeostasis eventually leads to establish and maintain permeability barrier homeostasis, the most important function of the epidermis. The permeability barrier is located in the stratum corneum. Tightly coordinated regulations are required for the sustained normal barrier function. Extensive studies have established that several nuclear hormone liposensors, including peroxisome proliferator-activated receptor a PPARa, PPARb/d, PPARg and LXRs are expressed in keratinocyte. Activation of PPARs and LXRs could provide a mechanism to coordinate the formation of the corneocytes and extracellular lipid membranes that constitute the stratum corneum. Topical application of PPAR/LXR ligands to murine skin results in the increased expression of keratinocyte differentiation-related proteins, such as involucrin, loricrin, profilaggrin, and trans-glutaminase 1, which would stimulate cornified envelope formation. In conclusion, topical application of ligands or activators of PPAR/LXR as an epidermotherapy would be a promising option to deal dry skin conditions such as atopy.

Ultrastructure of Cryptococcus neoformans in the Skin Tissue (조직내 Cryptococcus neoformans의 전자현미경적 관찰)

  • A case of systemic cryptococcosis developed in 4 year old boy was described and illustrated by light and electron microscope. Light microscopically, the upper dermis of the skin showed chronic nonspecific inflammation with numerous spherical spores surrounded by a clear halo created by the wide gelatinous capsule. Ultrastructurally, the C. neoformans showed the wide capsule containing microfibrils that appeared to radiate from the cell wall and to coil and interwine in various directions. The cell was uninucleate with a single nucleolus. Along the inner nuclear envelope, numerous small vesicles were present. In addition, C. neoformans presented membranous organelles derived from the plasma membrane and comparable to bacterial mesosomes.
