Background: Panax ginseng Meyer (Asian ginseng) has a large nuclear genome size of > 3.5 Gbp in haploid genome equivalent of 24 chromosomes. Tandem repeats (TRs) occupy significant portions of the genome in many plants and are often found in specific genomic loci, making them a valuable molecular cytogenetic tool in discriminating chromosomes. In an effort to understand the P. ginseng genome structure, we characterized an ultrahigh copy 167-bp TR (Pg167TR) and explored its chromosomal distribution as well as its utility for chromosome identification. Methods: Polymerase chain reaction amplicons of Pg167TR were labeled, along with 5S and 45S rDNA amplicons, using a direct nick-translation method. Direct fluorescence in situ hybridization (FISH) was used to analyze the chromosomal distribution of Pg167TR. Results: Recently, we reported a method of karyotyping the 24 chromosome pairs of P. ginseng using rDNA and DAPI (4',6-diamidino-2-phenylindole) bands. Here, a unique distribution of Pg167TR in all 24 P. ginseng chromosomes was observed, allowing easy identification of individual homologous chromosomes. Additionally, direct labeling of 5S and 45S rDNA probes allowed the identification of two additional 5S rDNA loci not previously reported, enabling the refinement of the P. ginseng karyotype. Conclusion: Identification of individual P. ginseng chromosomes was achieved using Pg167TR-FISH. Chromosome identification is important in understanding the P. ginseng genome structure, and our method will be useful for future integration of genetic linkage maps and genome scaffold anchoring. Additionally, it is a good tool for comparative studies with related species in efforts to understand the evolution of P. ginseng.
This paper aimed to analyse dose sensitivity to the controllable parameters of indoor radon $(^{222}Rn)$ and its decay products (Rn-D) by applying the input~output linear system theory. Physical behaviors of $^{222}Rn\;&\;Rn-D$ were analyzed in terms of $(^{222}Rn)$ gas -generation, -migation and -infiltration to indoor environments, and the performance output-function, i. e. mean dose equivalent to Tracho-Bronchial (TB) lung region, was assessed to the following extented ranges of the controllable paramenters; a) the ventilation rate $constant({\lambda}_v)\;:\;0{\sim}50[h^{-l}].\;b)$ the attachment rate $constant({\lambda}_a)\;:\;0{\sim}500[h^{-l}].\;c)$ the unattached-deposition rate constant (${\lambda}^u_d)\;:\;0-50[h-l]$. A linear input-output model was reconstructed from the original models in literatures, as follows, which was modified into the matrices consisting of 111 nodal equations; a) indoor $^{222}Rn\;&\;Rn-D$ Behaviour; Jacobi-Porstendoerfer-Bruno model.
An excess risk of lung cancer mortality among Koreans, attributable to indoor $^{222}Rn$ daughters exposure, were quantitatively evaluated by applying a stochastic health risk projection model on the radiation exposure. The lung cancer rate in Korean males and females, based on the 1989 demographic data, were estimated to be $22.4/10^5-y\;and\;9.5/10^5-y$, respectively The lifetime baseline lung cancer risks, deduced from these rates, appeared to be 0.047 and 0.019 for males and females, respectively, and were lower than the corresponding 1984 values of 0.067 and 0.025 in the U.S.A. The excess risk coefficients, derived by modified relative risk projection model of the BEIR-IV Committee under the US National Academy of Science, per annual 1.0 WLM of exposure to indoor radon daughters were estimated to be 0.022/WLM for males, 0.009/WLM for females, and 0.017/WLM for both sexes. The resulting annual frequency of excess lung cancer mortality for the life expectancy in the Korean population appeared to be 230/10^6-WLM, which was an approximate median of $120{\sim}450/10^6-WLM$ reported so far in the world.
Kim, Jong Hyup, (Div. of Biology, Atomic Energy Research Institute.) Studies on the Cellular Metabolism in Microorganisms as influenced by Gamma-irradiation(II). On respiration rate and dehydrogenase activity of yeast cells irradiated by gamma ray from cobalt-60. 1. Oxygen uptake rate of the gamma irraiated yeast cells had been measured with Warburg's manometer, and the $O_{2}$-uptake was compared with those of normal cells. The rate of endogetious respiration increases in its $O_2$-uptake at 150, 000 rentgen dose, and at higher rentoen doses it was decreased. Exogenous respiration begin to decrease in its O_2$-uptake at 5, 000r. doses of irradiation, further decrease with increasing of doses unproportionally. 2. It appears that plasma-membrane and nuclear membrane of yeast cells have changed and denatured by gamma-irradiation, as exogenous respiration of glucose had been decreased at a dose of 200, 000r's irradiation. 3. The activity of glucose, alcoholic, lactic, succinic and glutamic deliydrogenase (G.D.H., A.D.H., L.D.H., S.D.11., and GL.D.H.) in the gamma irradaited cells had been assayed by T.T.C.(Triphenyl tetrazolium chloride) method and spectrophotometry, the obtained results were compared with those of normal cells. 4. The activity of dehydrogenase experimented exhibit a resistance against gamma irradiation at lethal dose of cells, and the activity of dehydrogenase are found to be much resistant than those of respiratory system. We may assume that the membrane substrate of mitochondria or cytoplasm had been destructed by gamma-irradiation much more than that of dehydronase system.
Background: Autophagy is an important adaptive mechanism in normal development and in response to changing environmental stimuli in cancer. Previous papers have reported that different types of cancer underwent autophagy to obtain amino acids as energy source of dying cells in nutrient-deprived conditions. However, whether or not autophagy in the process of lung cancer causes death or survival is controversial. Therefore in this study, we investigated whether nutrient deprivation induces autophagy in human H460 lung cancer cells. Methods: H460, lung cancer cells were incubated in RPMI 1640 medium, and the starved media, which are BME and RPMI media without serum, including 2-deoxyl-D-glucose according to time dependence. To evaluate the viability and find out the mechanism of cell death under nutrient-deprived conditions, the MTT assay and flow cytometry were done and analyzed the apoptotic and autophagic related proteins. It is also measured the development of acidic vascular organelles by acridine orange. Results: The nutrient-deprived cancer cell is relatively sensitive to cell death rather than normal nutrition. Massive cytoplasmic vacuolization was seen under nutrient-deprived conditions. Autophagic vacuoles were visible at approximately 12 h and as time ran out, vacuoles became larger and denser with the increasing number of vacuoles. In addition, the proportion of acridine orange stain-positive cells increased according to time dependence. Localization of GFP-LC3 in cytoplasm and expression of LC-3II and Beclin 1 were increased according to time dependence on nutrient-deprived cells. Conclusion: Nutrient deprivation induces cell death through autophagy in H460 lung cancer cells.

Assuming the $\delta$/SUP 18/O composition of the Middle Miocene seawater was about - 0.34% (SMOW), the paleotemperatures of the Middle Miocene shallow seawater in the Pohang Basin was almost equal to or slightly higher than the present summer temperature (ca 16 to 20$^{\circ}C$) near Pohang area. In this paper we proposed a new ocean radiation automatic monitoring system. The proposed system has the following characteristics: First, using NaI + PVT mixed detectors, the response speed is fast and precision analysis is possible. Second, the application of temperature compensation algorithm to scintillator-type sensors does not require additional cooling devices and enables stable operation in the changing ocean environment. Third, since cooling system is not needed, electricity consumption is low, and electricity can be supplied reliably by utilizing solar energy, which can be installed at the observation deck of ocean environment. Fourth, using GPS and wireless communications, accurate location information and real-time data transmission function for measurement areas enables immediate warning response in the event of nuclear accidents such as those involving neighboring countries. The results tested by the authorized testing agency to assess the performance of the proposed system were measured in the range of $5{\mu}Sv/h$ to 15mSv/h, which is the highest level in the world, and the accuracy was determined to be ${\pm}8.1%$, making normal operation below the international standard ${\pm}15%$. The internal environmental grade (waterproof) was achieved, and the rate of variation was measured within 5% at operating temperature of $-20^{\circ}C$ to $50^{\circ}C$ and stability was verified. Since the measured value change rate was measured within 10% after the vibration test, it was confirmed that there will be no change in the measured value due to vibration in the ocean environment caused by waves.
Purpose: The atmospheric pressure plasma jet (APPJ) has been introduced as an effective disinfection method for titanium surfaces due to their massive radical generation at low temperatures. Helium (He) has been widely applied as a discharge gas in APPJ due to its bactericidal effects and was proven to be effective in our previous study. This study aimed to evaluate the safety and effects of He-APPJ application at both the cell and tissue levels. Methods: Cellular-level responses were examined using human gingival fibroblasts and osteoblasts (MC3T3-E1 cells). He-APPJ was administered to the cells in the experimental group, while the control group received only He-gas treatment. Immediate cell responses and recovery after He-APPJ treatment were examined in both cell groups. The effect of He-APPJ on osteogenic differentiation was evaluated via an alkaline phosphatase activity assay. In vivo, He-APPJ treatment was administered to rat calvarial bone and the adjacent periosteum, and samples were harvested for histological examination. Results: He-APPJ treatment for 5 minutes induced irreversible effects in both human gingival fibroblasts and osteoblasts in vitro. Immediate cell detachment of human gingival fibroblasts and osteoblasts was shown regardless of treatment time. However, the detached areas in the groups treated for 1 or 3 minutes were completely repopulated within 7 days. Alkaline phosphatase activity was not influenced by 1 or 3 minutes of plasma treatment, but was significantly lower in the 5 minute-treated group (P=0.002). In vivo, He-APPJ treatment was administered to rat calvaria and periosteum for 1 or 3 minutes. No pathogenic changes occurred at 7 days after He-APPJ treatment in the He-APPJ-treated group compared to the control group (He gas only). Conclusions: Direct He-APPJ treatment for up to 3 minutes showed no harmful effects at either the cell or tissue level.
3D printing technology is being used in various fields such as medicine and biotechnology, and materials containing metal powder are being commercialized through recent material development. Therefore, this study intends to analyze the photon spectrum during added filtration using 3D printing material during diagnostic X-ray examination through simulation. Among the Monte Carlo techniques, MCNPX (ver. 2.5.0) was used. First, the appropriateness of the photon spectrum generated in the simulation was evaluated through SRS-78 and SpekCalc, which are X-ray spectrum generation programs in the diagnostic field. Second, photon spectrum the same thickness of Al and Cu filters were obtained for characterization of 3D printing materials containing metal powder. In addition, the total photon fluence and average energy according to changes in tube voltage were compared and analyzed. As a result, it was analyzed that PLA-Al required about 1.2 ~ 1.4 times the thickness of the existing Al filter, and PLA-Cu required about 1.4 ~ 1.7 times the thickness of the Cu filter to show the same degree of filtration. Based on this study in the future, it is judged that it can be utilized as basic data for manufacturing 3D printing additional filters in medical fields.
