Screening of Silkworm Breeds for Tolerance to Bombyx mori Nuclear Polyhedro Virus (BmNPV)

  • BmNPV (Bombyx mori nuclear polyhedrosis virus) causes nuclear polyhedrosis in silkworms. The tolerance of silkworms to BmNPV is controlled by polygenes. This paper reports on the relative tolerance of silkworm breeds among the germplasm maintained at Andhra Pradesh State Sericultural Research & Development Institute (APSSRDI), Hindupur, India. The silkworm larvae out of second moult were per orally inoculated with BmNPV polyhedra $(l{\times}l0^{th}//ml)$ and reared upto spinning. The response to BmNPV had been categorized into apparent tolerance, real tolerance and susceptibility. Among the 145 silkworm breeds screened, 18 bivoltines and 16 polyvoltines were found to have real tolerance to BmNPV.

  • 유용물질을 생산할 수 있는 곤충 baculovirus expression vector system의 국내 개발을 위하여, Bm-NPV의 다각체 단백질 유전자를 탐색, 클로닝 하고 그 구조를 분석한 결과는 다음과 같다. 1. AcNPV의 다각체 단백질 유전자를 포함하고 있는 EcoRI-Ⅰ fragment내의 0.93kb를 probe로 하여 southern 분석한 결과, BmNPV의 다각체 단백질 유전자는 PstⅠ-F fragment(7.4Kb)에 위치하였다. 2. BmNPV의 다각체 단백질 유전자를 포함하는 PstⅠ-F fragment를 E. coli에 클로닝시켜서 pBmP-F라 명명하고, 다시 southern분석을 통해 subcloning하여 pBmP-H라 명명하였으며 pBmP-H의 제한효소 지도를 작성하였다. 3. pBmP-H의 염기서열분석결과 구조유전자를 포함한 1,259 bp의 염기서열이 결정되었다. Iatrou 등이 보고한 BmNPV 다각체 단백질 유전자의 염기서열과 비교한 결과 10개의 염기서열에서 차이를 보였으며, 74 Val이 Ile로, 76 Asn이 Ser으로 155 Met이 Val로 아미노산 변경된 결과를 보였다.

Construction of a Baculovirus Expression System Using Hyphantria cunea Nuclear Polyhedrosis Virus for Eukaryotic Cells

  • Baculovirus transfer and expression vectors with Hyphantria cunea nuclear polyhedrosis virus (HcNPV) were constructed. An initial transfer vector, pHcEV, constructed using HcNPV was previously reported (Park et al. 1993. J. Kor. Soc. Viral. 23: 141-151). Herein, the size of the vector was properly reduced, and a functionally perfect vector was constructed and named pHcEV-IV (6.7 kb). The vector has a 2.2-kb HcNPV DNA sequence in the 5'-flanking region of the vector's polyhedrin gene promoter. The 1.8-kb HcNPV DNA sequence, poly A signal sequence, T3 primer sequence, and 13 multicloning site sequences, in order, were ligated in front of the translation start codon of the polyhedrin gene. The cloning indicating marker lacZ gene was inserted into the pHcEV-IV, named pHcEV-IV-lacZ, and transferred into the wild-type virus. Recombinant expression virus, lacZ-HcNPV, was constructed by replacing the lacZ gene in the pHcEV-IV-lacZ with the polyhedrin gene of the wild-type virus. The recombinant virus was isolated from blue plaques that produce $\beta$-galactosidase without polyhedra. The lacZ gene insertion was confirmed by Southern hybridization analysis. The expression of the lacZ gene in Spodoptera frugiperda cells infected with the lacZ-HcNPV was examined by SDS-PAGE and colorimetric assay. One 116-kDa LacZ protein band appeared on the PAGE. The production rate of the $\beta$-galactosidase was approximately 50 international units (IU) per min per ml between 2 to 5 days postinfection (p.i.). The highest activity occurred at five days p.i. was 170 IU/min/$m\ell$. The enzyme activity first appeared about 20 h p.i. as measured by colorimetric assay.

멸강나방에 대한 곤충핵다각체병바이러스의 교류감염성 (Cross Infectivity of Nuclear Polyhedrosis Viruses to the Common Armyworm, Pseudaletia separata)

  • 밤나방류에 대한 천적미생물을 중심으로 하는 신방제법의 개발을 목적으로 멸강나방에 강한 병원성을 갖는 바이러스의 선별시험을 실시하여 다음과 같은 결과를 얻었다. 멸강나방(Pseudaletia(=Leucania) separata, Walker)유충에 강한 병원성을 가진 핵다각체바이러스를 선별코자 21종(개통)의 NPV를 orchard grass의 양면에 발라서 멸강나방 2령 유충에 첨식한 결과 멸강나방 핵다각체병바이러스 4계통과 벼밤나방 핵다각체병바이러스가 병원성이 높았다. 멸강나방 핵다각체병바이러스 3계통(LsNPV-F, LsNPV-G 및 LsNPV-Y)를 공시해서, 다각체를 인공사료에 혼합하여 해서 멸강나방 1~6령의 유충에 대한 병원성 시험을 행한 결과 1~2령 유충에 대한 병원성이 높고, 4령 이후에는 낮아 약령기방제의 중요성을 시사했다.

  • HcNPV DNA genome 을 제한효소 EcoRI 으로 절단하여 그들의 일부 절편을 pUC8 vector 에 cloning 한 후 E. coli JM 83 세포에 형질 전환시켰다. 이 결과 24 개의 EcoRI 절편중 12 개의 절편이 cloning 되었다. 이들 제조합체중 4 개는 eNP-O, eNP-Q, eNP-R, eNP-S 라 명명하였다. 또한 이들 제조합체의 외래 유전자 발현을 SDS-PAGE 에 의해 단백질 패턴을 분석하였다. 그 결과 제조합체 eNP-O, eNP-Q, eNP-R 에서는 E. coli JM 83 숙주세포의 단백질 밴드와 비교하여 다른 분자량을 갖는 밴드가 나타났다.

  • Autographa californica Nuclear Polyhedrosis Virus의 Nucleocapsids의 형성과 Polyhedra Occlusion Bodies의 형성을 Spodoptera frugiperda 세포에 감염된 후 3 일째 된 것을 관찰하였다. 1. A. californica NPV는 Spodoptera 세포의 핵내에서 분열복제되어 Nucleocapsid를 형성하여, 외투막을 얻은 후에 Polyhedra Occlusion Bodies 로 함입 되었다. 2. A. californica NPV가 감염된 세포의 핵은 팽창되어 핵막이 세포질까지 거의 신장되어 있었다. 3. Polyhedra Occlusion Bodies의 형성은 NPV로 감염된 세포에서 작은 엉성한 단백질 또는 Polypeptides가 점진적으로 커지면서 바이리스들이 함입되면서 더욱 덩치가 커지게 되고, 세포질내에 NPV 바이러스들이 없어지면 Polyhedra는 외부에 매끈한 막을 형성하는 것이 관찰되었다. 4. Polyhedra Occlusion Bodies의 형태는 바이러스 다발이 함입되어 있는 상태에 따라서 다양하였다. 5. Polyhedra 에 있는 바이러스 다발들은 외투막을 가지며 바이러스사이에도 기저물질이 존재하는 것을 볼 수 있었다.

Optimization of Propagation of Anagrapha falcifera Nuclear Polyhedrosis Virus in Spodoptera Frugiperda 21 Cells

  • Propagation of Anagrapha falcifera nuclear polyhedrosis virus(AfNPV) was investigated using well-plates and split-flow air-lift bioreactors. In well-plate experiments, the effects of pH, cell density at a point of infection, serum concentration, DEAE-dextran, and lipid on virus propagation were all closely examined. The AfNPV titer in well-plates was optimal at pH 6.8 and $3{\times}10^6$ cells/$cm^2$. The virus titer was not dramatically affected when the fetal bovine serum concentration was reduced from 10% to 5%. The addition of cholesterol at AfNPV infection of Sf21 cells enhanced the virus titer, whereas the addition of DEAE-dextran did not improve the titer. The AfNPV titer ($3.8{\times}10^7$ $TCID_{50}/ml$) at optimized conditions for well-plate experiments was 2.5-fold higher than for the control. In bioreactor experiments, the AfNPV titer showed its maximum level at air flow rates of 20-40 ml/min. In a split-flow air-lift bioreactor, AfNPV titer ($2.3{\times}10^7\;TCID_{50}/ml$) was 1.5-fold higher than the control when the culture was at pH 6.8 and supplemented with 0.34 mM cholesterol.

Screening of silkworm strains for efficient recombinant protein production by Autographa californica nucleopolyhedrosis virus (AcNPV)

  • Baculoviruses base vectors come to be regarded as methods for in vivo gene delivery and transient expression to the silkworm. In the case of silkworm, B. mori, two types of baculoviruses, AcNPV (Autographa californica nuclear polyhedrosis virus) and BmNPV (Bombyx mori nuclear polyhedrosis virus), are potentially applicable as vectors. Recently, AcNPV showed promising results with some silkworm strains despite different host-specificities. We searched for a highly-permissive silkworm strain in the B. mori stocks of Kyungpook National University that could produce high levels of recombinant protein. Seventy strains were screened using the recombinant AcNPV/BmA3-Luc virus. Based on the measured luciferase activity, the strains could be divided into three groups, high-, middle-, and low-permissive strains, according to their relative recombinant protein expression levels. At 48 hours post-injection, the luciferase activity in the high-permissive strains was 500-fold greater than that of the low-permissive strains. At 72 hours post-injection, a significant elevation in luciferase activity was observed in the hemocytes of all strains. Then, based on the above results, the High Permissive Strain (HPS) S10 and the Low Permissive Strain (LPS) S39 were pick up and was carried out Dot blotting, RT-PCR and Real time PCR.

Expression of Bovine Growth Hormone Gene in a Baculovirus, Hyphantria cunea Nuclear Polyhedrosis Virus

  • Bovine growth hormone (bGH) gene was expressed in an insect Spodoptera frugiperda cell line using a Baculovirus, Hyphantria cunea nuclear polyhedrosis virus (HcNPV). The bGH gene in pbGH plasmid was sequenced and amplified by PCR technique with two primers containing NcoI sites. The bGH gene consisted of 654 bp (217 amino acid residues), the 5'-untranslated region of the cloned bGH cDNA contains 56 bp, and the 3'-untranslated region contains 145 bp and two pallindromic regions. The amplified bGH gene DNA fragment (654 bp) was inserted into the NcoI site of the pHcEVII vector, which was named pHcbGH. The pHcbGH transfer vector DNA and the wild type HcNPV DNA were cotransfected into S. frugiperda cells to construct a recombinant virus. Eight recombinant viruses were selected and named HcbGH. One clone, HcbGH-4-1 showed largest plaque size, therefore the recombinant virus was further studied. The multiplication pattern of the recombinant HcbGH-4-1 was similar to that of the wild type HcNPV. The bGH gene DNA in the HcbGH-4-1 recombinant was confirmed by Southern blot hybridization. The amount of the bGH (217 amino acid residues, 21 kDa) produced in S. frugiperda cells infected with the HcbGH-4-1 recombinant was approximately 5.5 ng per ml ($10^6$ cells) by radioimmunoassay.

  • 흰 불나방 핵 다각체바이러스(Hyphantria cunea nuclear Palyhedrosis virus: HcNPV) 다각체 단백질 유전자포함 절편의 탐색과 클로닝을 행하였다. Autographa cahfornica NPV EeoRI-I 절편 (약 7.3 kb), Bombyx mori NPV PstI-F 절편 (약 7 kb) 및 합성 oligonucleotide ( 3D-mer) 를 probe로 한 southern hybridization을 행하여 HcNPV PstI - L 절편 (5.3 kb)을 탐색하고, pUC18을 이용하여 E. coli에 형질전환시켜 클로닝하였다. 클로닝한 plasmid의 EeoRI, SaIl, Kpnl, HindIII, SacI 및 AvaI의 제한효소지도를 작성하고 pHeP-L(8.0 kb)이라 명명히였으며, 이를 다시 pHcP-Ll(4.7 kb), pHcP-L2(7.1 kb), pHcP-L3(5.3 kb), pHcP-L4(4.2 kb) 및 pHeP-L5(4.5 kb)로 subeloning 하였다.
