Effects of Nonylphenol on Early Development of Olive Flounder, Paralichthys olivaceus (Nonylphenol이 넙치, Paralichthys olivaceus의 초기 발생에 미치는 영향)

  • Effects on nonylphenol (NP) treatment on early development of fertilized eggs and survival of larvae in olive flounder, Paralichthys olivaceus were investigated. Fertilized eggs and hatched larvae were exposed to aqueous solutions of NP at nominal concentrations of 50, 100, 150 and 200 $\mu\textrm{g}$/L. In the control I and II (methanol-carrier control) groups, the hatchability of fertilized eggs were 90.7$\pm$7.02 and 90.0$\pm$5.29% (P <0.05), respectively. However, treatments of NP concentrations at 50, 100, 150 and 200 ug/L were reduced to 78.7$\pm$4.16, 46.0$\pm$9.17, 48.0$\pm$3.46 and 33.3$\pm$11.02% (P<0.05), respectively. The time to hatching of fertilized eggs was delayed in high-dose NP treatment groups rather than control groups. Fertilized eggs of NP immersed group could not hatch normally, and also vertebra of the larvae observed as abnormal shape. Cumulative mortality of hatched larvae after 26hr NP treatment was 38.3, 78.3, 88.3 and 100% in NP 50, 100, 150 and 200 ug/L treatment groups, respectively. Whereas, control I and II were 11.7, 16.7% (P<0.05). Hatched larvae showed most death at the embryonic development stage with abnormalities. These results suggest that NP treatment in fertilized eggs and larvae was inhibited as normally development for fertilized egg and growing of larvae.

Transcriptome analysis of the livers of ducklings hatched normally and with assistance

  • Objective: "Hatchability" is an important economic trait in domestic poultry. Studies on poultry hatchability focus mainly on the genetic background, egg quality, and incubation conditions, whereas the molecular mechanisms behind the phenomenon that some ducklings failed to break their eggshells are poorly understood. Methods: In this study, the transcriptional differences between the livers of normally hatched and assisted ducklings were systematically analyzed. Results: The results showed that the clean reads were de novo assembled into 161,804 and 159,083 unigenes (${\geq}200-bp$ long) by using Trinity, with an average length of 1,206 bp and 882 bp, respectively. The defined criteria of the absolute value of log2 fold-change ${\geq}1$ and false discovery rate${\leq}0.05$ were differentially expressed and were significant. As a result, 1,629 unigenes were identified, the assisted ducklings showed 510 significantly upregulated and 1,119 significantly down-regulated unigenes. In general, the metabolic rate in the livers of the assisted ducklings was lower than that in the normal ducklings; however, compared to normal ducklings, glucose-6-phosphatase and ATP synthase subunit alpha 1 associated with energy metabolism were significantly upregulated in the assisted group. The genes involved in immune defense such as major histocompatibility complex (MHC) class I antigen alpha chain and MHC class II beta chain 1 were downregulated in the assisted ducklings. Conclusion: These data provide abundant sequence resources for studying the functional genome of the livers in ducks and other poultry. In addition, our study provided insight into the molecular mechanism by which the phenomenon of weak embryos is regulated.

Inheritance of Feeding Response of the Silkworm, Bombyx mori, to MP-O Artificial Diet. (MP-O 인공사료에 대한 누에의 섭식상과 유전현상)

  • Differences of feeding response of 280 silkworm genetic stocks and 71 breeding lines to MP-O artificial diet and the mode of the inheritance were investigated. Feeding response to MP-O diet varied markedly between the silkworm varieties, and the non-normally distributed curve for the response was observed. From the genetic analysis, the high feeding response to MP-O diet was recessive to the low feeding response. Therefore it is considered that the high feeding response of the newly hatched silkwrom larvae to MP-O diet is controlled by a recessive gene.

Embryonic Development and Metamorphosis of the Ascidian Halocynthia aurantium (붉은멍게(Halocynthia aurantium)의 배발생과 변태)

  • The ascidian Halocynthia aurantium (sea peach), which belongs to the phylum Chordata, is thought to be a valuable organism of aquaculture like H. roretzi (sea pineapple), but its biological characteristics such as development and ecology are not well known. In this study, in order to obtain basic data for H. aurantium farming, the development processes of H. aurantium inhabiting the east coast of Gangwon-do were investigated and compared with those of H. roretzi, a related species. As a result, the morphology and developmental stages of the fertilized eggs, embryos and larvae of H. aurantium were very similar to those of H. roretzi. Fertilized eggs of H. aurantium took about 42.1 hours to hatch at 11℃, almost similar to 40.9 hours of H. roretzi. The time required for larvae to metamorphose into juveniles after hatching was very similar between the two species. The hatched larvae of the two species became juveniles with oral and atrial siphons after 23 days at 11℃. Both types of embryos developed slowly in seawater at low temperatures and rapidly developed at high temperatures. Fertilized eggs of H. aurantium hatched in an average of 62.3 hours at 9℃, 42.1 hours at 11℃, and 36.3 hours at 13℃, whereas those of H. roretzi hatched in an average of 60.4 hours, 40.9 hours, and 35.2 hours. Most of H. aurantium embryos did not develop normally above 15℃, so it is thought that attention is needed in the seed production processes.

Production of Transgenic Homozygous Diploid in Mud Loach(Misgurnus mizolepis) I. Transfer of Luciferase Gene and Evaluation of Mud Loack Expression Vector

  • Validities of several gene transfer methods including microinjection, electroporation and lipo-fection with luciferase gene (pRSVL), and effectiveness of mud loach expression vector which contains ARS from mud loach on production of transgenic mud loach were evaluated. Microiniection revealed the $0\~8\%$ of transgene incidence in 2-week-old fish with significant mosaicism. Electroporation and lipofection of mud loach sperm also successfully introduced the transgene into sperm cells, and transferred the foreign DNA into zygote. Gene transfer by electroporation and lipofection showed a range of $0\~28\%$ and $0\~48.1\%$ of transgene incidence, respectively in newly hatched larvae, altough most DNA introduced were gradually degraded with the development of fish. Microinjections of mud loach expression vector caused a significantly reduced survival rate of mud loach embryos with severe teratogenic effects, and ARS/Luc transgene could not be detected in normally developed fish after microinjection.

The Effect of Temperature on the Colony Development of Bombus ignitus Smith (호박벌(Bombus ignitus Smith)의 봉군발육에 미치는 영향)

  • The queens of Korean native bumblebee species, Bombus ignitus were collected from the field in the spring of 1997 and reared under the various temperatures as 15$^{\circ}C$$\pm$1$^{\circ}C$, 2$0^{\circ}C$$\pm$1$^{\circ}C$, $25^{\circ}C$ $\pm$1$^{\circ}C$, 3$0^{\circ}C$$\pm$1$^{\circ}C$, 60%$\pm$5% of relative humidity and 14L : 10D, to investigate their ecological characteristics, as comparing with control(29$^{\circ}C$$\pm$1$^{\circ}C$, 60%$\pm$5% of relative humidity and red light). As a result, eggs were laid by foundation queen at 15$^{\circ}C$, but they didn't hatched out while at 2$0^{\circ}C$ eggs were laid by them and they hatched out. When a foundation queen laid eggs at $25^{\circ}C$ and they all were hatched out and normally grown. Finally they were developed to the normal colony. B. ignitus worker survived for 77days at 2$0^{\circ}C$, about 69days at $25^{\circ}C$, about 68days at 3$0^{\circ}C$ and about 63days at 29$^{\circ}C$(control). The first brood workers emerged in 25-27days from the egg which a foundation queen laid. At the various temperatures, the captive queens stand to lay eggs in about 18days at 15$^{\circ}C$, 15days at 2$0^{\circ}C$, 11days at $25^{\circ}C$, 11days at 3$0^{\circ}C$ and 4days in control. The first worker and the first drone from the egg cell of the collected queens appeared in about 27days and 72days at 2$0^{\circ}C$, 26days and 88days in $25^{\circ}C$, 24days and 65days at 3$0^{\circ}C$, 25days and 71days in control, respectively. In colony foundation, 33% of the collected queen at 2$0^{\circ}C$ and $25^{\circ}C$, 100% of the disposed queen at 3$0^{\circ}C$ and in control, and 67% of them at 3$0^{\circ}C$ and in control both produced new queens in 66days and 88days, respectively. The life span of the colony founded covered fro about 3 months at $25^{\circ}C$, 3$0^{\circ}C$ and in control. At lower temperatures, the life span of queen is shorter; 2 months at 15$^{\circ}C$ and 3 months at 2$0^{\circ}C$, respectively. A colony which normally developed, varied in size with rearing temperatures; about 20heads at 2$0^{\circ}C$, 482heads at $25^{\circ}C$, 330heads at 3$0^{\circ}C$ and 452heads in control. A foundation queen monthly oviposited 1.5egg cells at 15$^{\circ}C$, 3.0egg cells at 2 $0^{\circ}C$, 21.7egg cells at $25^{\circ}C$, 42.3egg cells at 3$0^{\circ}C$ and 47.0egg cells in control. As a colony developed in June and July, egg cells as well as daily average cells increased in number, as compared to those in May and August. Also, in June and July, interval(days) of egg laying is shorter than in May and August. Number of nectar pots during the colony development varied with rearing temperatures; 31pots at $25^{\circ}C$, 39pots at 3$0^{\circ}C$, 23pots in control. The emergence of new queen showed a distinctively different two patterns; early emerging type and late emerging type. Workers are rapidly increased from early in July to early in August at $25^{\circ}C$, but at 3$0^{\circ}C$ and in control, emergence of workers are remarkably increased from the middle of June and last until July. No new queen emerged at 15$^{\circ}C$, 2$0^{\circ}C$ and $25^{\circ}C$. New queens at 3$0^{\circ}C$ and in control emerged between late in June and early in July. Thus emergence of new queen was temperature dependent.

  • Effects of intraperitonial injections of human chorionic gonadotropin (HCG) on ovulation and spawning of the loach, Misgurnus mizolepts, were investigated. Matured females spawned successfully by a single dose of 6 IU (HCG) per gram body weight. Spawning usually occured 13 to 25 hours after hormone injection. Most of the eggs were fertilized and hatched normally. Fertilization rate, hatching percentage and Pseudo-gonadosomatic index were not correlated with increasing HCG doses. For the study of multiple spawning of this species, fatness, hepatosomatic and gonadosomatic indices of artificially spawned females were checked for 42 days after first spawning. According to this result, females could spawn again 40 days after first spawning.

Study on The Usability of Mouse Hatched Blastocysts in Embryos Transfer (수정란 이식시 생쥐 완전탈출 배반포기배의 유용성 검토)

  • This study was carried out to investigate the in vivo developmental potential of mouse zona-hatched blastocysts (HBs). The HBs were cultured in vitro until day 5 and day 6 from zygotes produced in vivo and classified to small (S-HBs), medium (M-HBs) and large (L-HBs) on the basis of embryo diameters. The results obtained in these experiments were summarized as follows ; 1) when the blastocysts at day 4 were further cultured for $24\sim48hr$, HBs obtained at day 5 and day 6 culture in vitro were 29.1% and 22.8%, respectively. 2) Also, when the total cell number of HBs were counted, cell numbers of classified HBs on day 5 and day 6 to small ($77.3\pm5.3$, $59.6\pm4.4$), medium ($83.7\pm4.0$, $66.8\pm3.5$) and large ($100.7\pm2.6$, $88.9\pm3.8$) were increased as their size increases. Especially, there were significantly different between S-HBs and L-HBs (p<0.01). 3) In addition, when the classified HBs were transferred into when the classified HBs were transferred into day 3 pseudopregnant recipients, the pregnancy and implantation rates of S-HBs (28.6%, 15.7%), M-HBs (44.4%, 30.9%) and L-HBs (62.5%, 49.1%) at day 5 were increased as their size increases. However, this pattern was not showed in embryo transfer of day 6 HBs. But, when the live fetuses formation against total implantation rates were observed, the result (87.5%) of S-HBs of day 5 was significantly higher than that of the others (p<0.01). Therefore, this study demonstrates that in vitro cultured healthy HBs can not only be developed normally with good pregnancy rates, implantation rates and live fetuses formation, but also served as a fundamental data for utility of supernumerary HBs in human blastocyst transfer.

Effects of $Ca^{++}$ Inhibitors on Compaction of Mouse 8-cell Embryos (생쥐 8세포배의 Compaction에 미치는 칼슘 Inhihitor의 영향)

  • In mammalian development, the embryo which is in the process of compaction, involves a progressive flattening of blastomeres against each other with the consequence that the embryo assumes a spherical shape. This stage happens in the first differentiation. The present study was aimed to examine the involvement of other metal ions in compaction by treating with various divalent cations in the absence of $Ca^{++}$. When 8-cell embryos were cultured in $Ca^{++}$-free medium for 24hrs, they developed to 16-cell stage but did not compaction, and degenerated after 48hrs of culture. Embryos were cultured in $Ca^{++}$-free medium for 24hrs and then transferred to the control medium showed the normal compaction afterwards. When 8-cell embryos were cultured in the presence of $Ni^{++}$, known as a $Ca^{++}$ inhibitor, they cleaved to 16-cell stage but did not compact in the absence of $Ca^{++}$. On the other hand, embryos cultured in the media containing both $Ca^{++}$ and $Ni^{++}$ developed normally so that they underwent compaction during culture for 48hrs. However, they failed to hatch during further 24hrs in the same medium, indicationg that $Ni^{++}$ may exert some harmful effects. Embryos grow in the control medium that contained $Ca^{++}$ but not $Ni^{++}$, developed to the hatched blastocysts. The treatment with $Cd^{++}$ $10^{-1}$,$10^{-2}{\mu}M$, $Mn^{++}$ or $Ba^{++}$ 10,100, $1000{\mu}M$ in $Ca^{++}$-free medium, respectively, inhibited compaction and embryonic degeneration began as in $Ca^{++}$-free medium. When 3, 5, 10mM of $Sr^{++}$, known as a substitute for $Ca^{++}$ in cell, was added to $Ca^{++}$-free medium, respectively, compaction was induced unlike the above metal ions. Embryos were cultured in $Sr^{++}$ developed to blastocysts, but failed to hatch after 72hrs and degenrated. On the other hand, when embryos were cultured in 3, 5, 10mM of $Sr^{++}$ but in $Ca^{++}$-free medium for 24hrs respectively and then transferred to the control, they showed the similiar development as that in the control.

Biology of Three Species of the Genus Tipula (Diptera: Tipulidae) in Korea (한국산 Tipula속(파리목 : 각다귀과) 3종의 생태)

  • This study was conducted to investigate the biology of three species, Tipula latemarginata Alexander, T. nova Walker and T. aino Alexander, of the Genus Tipula in Korea. Field surveys for the biology of the species were carried out at the three sites of Neaseong Stream area in Bongwha County from January 2001 to December 2005. Also many individuals of the species were reared in laboratory to investigate the biology. The larval habitats of the species were streams, rivers, paddy fields, damp earth or leaky revetments. The Tipula larvae were herbivorous. Newly-hatched first instar larvae fed on soft algae, and the larvae gradually fed on leaves, stems and roots of a variety of plants or hard leaf litters, as they grew. During the molting of the larvae their body kept moving from left to right and up and down. Molting usually took not more than 2 hours, and about four hours after molting, the larvae started to eat heavily. The Tipula larvae in aquatic places moved to drier land for pupation and went through short prepupal stage lasting 1-2 days for pupation at fourth instar larval stage. When emerging, the Tipula pupae placed their head and thorax on the earth, but the other parts in the earth. Emergence from the pupal case required about 20 min. to one hour. Mating of Tipula adults took place within 5-7min. after emergence and the duration of mating was about 40 min. The female adults of the species laid eggs by walking with 3 pairs of legs over the damp earth or algal beds. Their body was positioned vertically on the ground with their wings spread $120^{\circ}$ and legs landed on the surface. The oviposition usually took place from one day to 4 days after emergence and the number of eggs carried by female adults were an average of 501-760 per individual. Tipula adults didn't normally feed, and drank water only occasionally. For a few days after emergence, the adults reared in the laboratory rarely drank water. As they neared to death, however, they frequently drank water. The longevity of adults reared in the laboratory with only water during the summer was ca. 4-9 days and males usually survived a little longer than females. The longevity of T. nova was increased 3 times or more as much by feeding them 3% sugar water. Male adults of T. latemarginata outnumbered female adults by 2.6 : 1 in the fields.
