Efficacy of corticosteroid ductal irrigation in acute salivary gland inflammation induced in a rat model

  • Purpose: This study aimed to compare the therapeutic effects of corticosteroid irrigations and normal saline irrigations in the early inflammatory state of the salivary gland. Materials and Methods: Adult male Wistar rats were divided into experimental (n=6) and control (n=3) groups. Inflammation was induced in the experimental subjects on both sides of the submandibular gland with ligation. After 14 days, both sides of the glands were de-ligated and retroductal irrigation using saline (n=3) and a corticosteroid (n=3) was performed on the left sides only. The controls (n=3) were used to normalize the gland state for the effects of diet and aging. Magnetic resonance imaging was performed to confirm inflammation and post-irrigation gland recovery by measuring relative signal intensity (SI). The glands were excised for histological examination. Results: All experimental animals showed inflamed glands with increased SI and subsequent recovery of the gland with decreased SI to varying degrees. The SI of the controls showed no significant changes during the overall period. The mean SI change of the irrigated gland was higher than that of the non-irrigated side, without a significant difference. The corticosteroid-irrigated glands showed a greater change in SI than that of the saline-irrigated glands. Histology revealed that inflammation was not observed in most of the irrigated glands, while mild to moderate quantities inflammatory cells were found in non-irrigated glands. Conclusion: Corticosteroid irrigation mitigated the early stages of salivary gland inflammation more effectively than normal saline.

Irrigation Solutions in the Knee Arthroscopy - Comparison with Normal Saline and Lactated Ringer's Solution (슬관절 관절경 수술시 세척액 - 생리 식염수와 Lactated Ringer 용액의 비교)

  • Purpose: The object of this study is to compare the clinical results of noraml saline and Lactated Ringer's solution in the arthroscopic meniscus surgery on the knee. Materials and Methods: The study group included 64 patients who had an medial meniscus tear and received arthroscopic partial medial meniscectomy. The group I was normal saline group, which was used as an irrigation solution during operation. The group II was Lactated Ringer's solution group. We measured the visual analogue scale, painless active knee flexion angle, and the ratio of the knee swelling at postoperative one day for all patients. Results: The visual analogue scale and the painless active knee flexion angle were no significant differences in comparing with normal saline and Lactated Ringer's solution group. The ratio of the knee swelling in Lactated Ringer's solution group was significantly less than those of noraml saline group. Conclusion: Lactated Ringer's solution has an advantage of decreasing the knee swelling in the arthroscopic meniscus surgery on the knee.

Comparison of Oral Hygiene Effects between 0.1% Chlorhexidine and Normal Saline on the Incidence of Oral Pathogens (0.1% 클로르헥시딘을 이용한 구강간호와 생리식염수를 이용한 구강간호의 구강내 병원균 발생빈도 비교)

  • Purpose: The purpose of this study was to compare the effects of oral hygiene with 0.1% chlorhexidine or with normal saline on the incidence of pathogens in the oral cavity of patients in Intensive Care Units (ICU). Method: A quasi experimental design with non-equivalent control group and non-synchronized design was used. For the study 46 patients were recruited from a university hospital (24 for the experimental group, 22 for the control group). patients in the experimental group received mouth care with 0.1% chlorhexidine gluconate and those in the control group received mouth care with normal saline twice a day for 7 days in a row. Oral samples were taken for bacterial cultures on admission day, the 4th day and the 7th day for both groups. Results: The incidence of oral pathogens decreased in the experimental group, and increased in the control group. There was no significant difference in the incidence of oral pathogens between the two groups. However oral hygiene using 0.1% chlorhexidine gluconate decreased the incidence of oral pathogens significantly for patients who already had pathogenic bacteria in their mouths on the admission day. Conclusion: The results suggest that mouth care with 0.1% chlorhexidine is effective for decreasing the incidence of oral infection for ICU patients who have oral infections.

The Effect of Mahuang(Ephedra sinica Stapf)-Chuanwu(Aconitum carmichaeli Debx) Pharmacopuncture Treatment in Obese Patients (마황천오약침이 비만에 미치는 영향)

  • Objectives : We investigated the effects of Mahuang-Chuanwu Pharmacopuncture Treatment in Obese Patients by analysing Body mass index(BMI), waist measurement and weight. Methods : We investigated on 52 volunteers consisted of 24 subjects in experiment(Mahuang-Chuanwu Pharmacopuncture) group and 28 subjects in control(Normal Saline) group. The Study form was a single-blind and placebo-controlled clinical trial. 24 subjects in experiment group were injected Mahuang-Chuanwu Pharmacopuncture with additional common treatment and 28 subjects in control group were injected Normal Saline with additional common treatment with Dietic Theraphy and Exercise. We measured BMI, waist measurement and weight on 3 times : before 1st session, after first 5 sessions, and after 10 sessions. The SPSS 13.0 for Windows was used to analyze the data and the wilcoxon signed ranks test (in group), mann Whitney u t-test(between two groups) and ${\chi}^2$-test(description of number) were used to verify the result. Results : In experiment group. BMI, waist measurement and weight were significantly low at the period of after first 5 sessions and after 10 sessions. In control group, BMI, waist measurement and weight were significantly low at the period of after 10 sessions but weren't significantly low at the period of after first 5 sessions. The changes of BMI, waist measurement and weight in experiment group were significantly more than that in control group. Conclusions : The Results suggest that Mahuang(Ephedra sinica Stapf)-Chuanwu(Aconitum carmichaeli Debx) Pharmacopuncture is effective on the treatment of obesity, compared to Normal Saline.

How Computed Tomography Contrast Media and Magnetic Resonance Imaging Contrast Media Affect the Changes of Uptake Counts of 201Tl

  • The purpose of the study is to investigate how uptake counts of $^{201}Tl$ of radioisotopes in the human body could change, when taking computed tomography and magnetic resonance imaging right after injecting contrast media. $^{201}Tl$ radioisotope substances of iodine contrast medium, which is a computed tomography contrast medium, and paramagnetic contrast medium, which is an magnetic resonance imaging contrast medium, were used as study materials. First, $^{201}Tl$ was put into 4 cc of normal saline in test tube, and then a computed tomography contrast medium of Iopamidol$^{(R)}$ or Dotarem$^{(R)}$, was put into 2 cc of normal saline in test tube. An magnetic resonance imaging contrast medium of Primovist$^{(R)}$ or Gadovist$^{(R)}$ was also put into 2 cc of normal saline in test tube. Each contrast medium was distributed to make $^{201}Tl$ as 3 mCi, with a total of 4 cc. Gamma camera, low energy high resolution collimator, and pinhole collimator were used to obtain images. The uptake count of $^{201}Tl$ was measured with 1000 frames of images, and obtained after 10 times of repetition. This study revealed that the use of Gadovist$^{(R)}$, which is an magnetic resonance imaging contrast medium, showed the smallest number of uptake count, after measuring $^{201}Tl$ uptake count by low energy high resolution collimator. On the other hand, the use of Iopamidol$^{(R)}$, which is a computed tomography contrast medium, showed the biggest difference in uptake count, when measuring $^{99m}Tc$ uptake count by Pinhole collimator. When examining with gamma camera, using contrast medium and $^{201}Tl$, identifying the changes of uptake count is very important for improving the value of diagnosis.

Analgesic Effects of Taxus cuspidata Pharmacopuncture Using Different Extraction Methods on the Formalin-Induced Pain Model (추출 방식의 차이에 따른 주목약침이 Formalin 유도 통증 모델의 진통에 미치는 영향)

  • Objectives : This study was designed to investigate the analgesic effects of Taxus cuspidata pharmacopuncture(TCP) at KI03 on formalin-induced pain in rats and to evaluate the efficiency of different extraction methods of Taxus cuspidata(TC). Methods : 30 rats were divided into 5 groups, each consisting of 6 rats. Each of the groups was treated two times. The first treatment for its right KI03 was as follows: rats were treated with normal saline(NOR), normal saline(CON), hot-water extraction of TC(W), ethanol extraction of TC(E), and ultrasonification extraction of TC(U). The second treatment was given 35 mins after the first one. Rats in NOR were treated with normal saline at their hind-paw. All groups, except NOR, were treated with formalin(5 %, $40{\mu}{\ell}$) at their hind-paw. To evaluate pain behavior, Ultrasonic vocalization(USV) was examined to be around 18~30 kHz. After analysis of USV, blood samples were taken from the rats for analysis of Substance P, aspartate aminotransferase(AST) and alanine aminotransferase(ALT). Results : In the experimental groups, USV was significantly decreased compared with CON and similar to that of NOR in the early phase. But there were no significant differences among the three extraction methods of TC. In addition, Substance P of W was significantly decreased. AST and ALT activation showed no significant differences among the 5 groups. Conclusion : These results show that regardless of extraction methods of TC, TCP at KI03 has analgesic effects in the early phase on formalin-induced pain in rats. In particular, hot-water extraction of TC significantly reduces Substance P activation.

Olanzapine Attenuates Mechanical Allodynia in a Rat Model of Partial Sciatic Nerve Ligation

  • Background: Neuropathic pain is a global clinical problem; nevertheless, nerve injury treatment methods remain limited. Olanzapine has antinociceptive and anti-nueropathic properties; however, its preventive effects have not been assessed in nerve injury models. Methods: We prepared a partial sciatic nerve ligation (Seltzer model) or sham-operated model in male Sprague-Dawley rats under isoflurane anesthesia. In a pre-treatment study, we administered olanzapine (10 mg/kg) intraperitoneally 1 h before nerve ligation. In post-treatment and dose-dependent studies, we injected 3 different doses of olanzapine intraperitoneally 1 h after nerve ligation. Mechanical allodynia was measured before and 7 days after surgery. Immunohistochemical analysis using anti-Iba-1 antibody was used to assess the effect of olanzapine at the spinal level. Results: In the pre-treatment study, median withdrawal thresholds of the normal saline groups were significantly lower than those of the sham-operated groups; however, those of the olanzapine (10 mg/kg) and sham-operated groups were not different. In the post-treatment and dose-dependent studies, the median withdrawal thresholds of the olanzapine (2.5 mg/kg) and normal saline groups were not different; however, those of the olanzapine (10 and 50 mg/kg) groups were significantly higher than those of the normal saline groups. Olanzapine did not have a significant effect on the density of Iba-1 staining. Conclusions: Olanzapine attenuated mechanical allodynia dose-dependently in the Seltzer model. This anti-allodynic effect of olanzapine was observed even when injected 1 h after nerve ligation. This effect of olanzapine appeared to be unrelated to microglia activation in the ipsilateral dorsal horn of the lumbar spinal cord.

Deodorizing Effect of Cheonggugamrosu (청구감로수(淸口甘露水)의 구취억제효과)

  • Objective : The aim of this study was to investigate the ability of Cheonggugamrosu. a gargling agent made from herb extracts, to decrease oral malodor in vitro and in clinical research. Methods : For in vitro experiment. samples were Garglin (over-the-counter mouthwash), CheonggugamrosuA and Cheonggugamrosu B. Deodorizing activity of each sample against methyl mercaptan were analyzed by gas chromatography (GC). In clinical examination. subjects were 73 students from the oriental medicine school of Kyunghee University. The concentration of VSC in oral cavity of each subject was measured by Halimeter and subjects were divided into 3 groups that would gargle for 30 seconds with normal saline. Garglin and CheonggugamrosuA. Subsequently, concentration of VSC was measured in same way immediately, 2 hours and 4 hours after gargling. Results : During in vitro experiment by GC, deodorizing activities were 46.77% with Garglin, 80.64% with Cheonggugamrosu A and 82.28% with Cheonggugamrosu B. In clinical examination, at 2 hours after gargling, the concentration of VSC in the Cheonggugamrosu A group was significantly lower than in the normal saline group, but not in the Garglin group. 4 hours after gargling, the concentration of VSC in both CheonggugamrosuA and Garglin wasn't significantly different from the one in the normal saline group. Conclusions : Cheonggugamrosuhad better and longer deodorizing effect than Garglin in this study. Because of the safety of herbal medicine and its efficaciousness, we can use Cheonggugamrosu as a clinical prescription.

The Suppressive Effects of Daebangfung-tang against Disuse Muscle Atrophy in Gastrocnemius of Rats (대방풍탕의 흰 쥐 비복근 불용성 근위축에 대한 억제 효과)

  • Objectives : The aim of this study is to evaluate the effect of Daebangfung-tang (DBFT) on suppressing muscle atrophy followed by immobilization. Methods : Twenty young male Sprague-Dawley rats were randomized into two groups. The rats in experimental group (DBFT group) were orally administrated DBFT once a day for 2 weeks, and the rats in control group were given normal saline in the same manner. The immobilization on left leg was carried out using casting tape. The morphologies of gastrocnemius in both groups were assessed. The cross sectional area of them was measured by image analyzer. Immunohistochemical staining was implemented and the numbers of immunopositive cells implying the expression of TNF-α were measured and quantified. Results : DBFT represented the significant suppressive effects against the weight loss of the left gastrocnemius muscle and the reduction in cross sectional area of the left gastrocnemius muscle compared with normal saline. Also the treatment with DBFT significantly reduced the expression of TNF-α in gastrocnemius compared with normal saline. Conclusions : DBFT is thought to have the significant suppressive effect against muscle atrophy followed by immobilization.

Effects of intracerebroventricular injection of corticotrophin releasing factor on the gene expression of ghrelin and corticotrophin releasing factor receptors in broiler chickens

  • Objective: This study aimed to investigate the effects of corticotropin-releasing factor (CRF) on the feed intake of broiler chickens and explore its influencing mechanism. Methods: The study included two trials. In trial 1, 32 male broiler chickens (Arbor Acres, Gallus gallus domesticus) were given ventricle buried tubes, and they were allowed to recover for 3 days. At 8:00 AM, intracerebroventricular (ICV) injection with CRF or normal saline was performed in 10-day-old broiler chickens, which were divided into the 5, 10, and 20 ㎍ and control (normal saline) groups according to the dose of CRF injection. In trial 2, chickens were divided into the 10 ㎍ and control group (physiological saline) to repeat trial 1. Results: Results of trial 1 showed that the cumulative amount of feed intake in the 10 or 20 ㎍ groups was considerably lower than that of the control group after ICV injection with CRF. The lowest amount of feed intake was obtained with the addition of 10 ㎍ of CRF. In trial 2, the expression of ghrelin in the hypothalamus injected with 10 ㎍ of CRF increased significantly, but the expression of ghrelin in various sections of the small intestine considerably decreased. The expression of CRF receptor subtypes 1 (CRFR1) in the hypothalamus and some parts of the small intestine remarkably increased, and the expression of CRF receptor subtypes 2 (CRFR2) increased only in the duodenum, whereas the expression of growth hormone secretagogue receptor (GHSR-1α) in the jejunum and ileum increased considerably after ICV injection of 10 ㎍ of CRF. Conclusion: The CRF at 10 ㎍ increased ghrelin expression in the hypothalamus and CRFR1 expression in the small intestine, and this phenomenon was related to the suppressed feed intake of broiler chickens.
