Background: The efficacy of ginseng, the representative product of Korea, and its chemical effects have been well investigated. The ginsenoside RG3 has been reported to exhibit apoptotic, anticancer, and antidepressant-like effects. Methods: In this report, the putative effect of RG3 on several cellular function including cell survival, differentiation, development and aging process were evaluated by monitoring each specific marker. Also, mitochondrial morphology and function were investigated in ultraviolet (UV)-irradiated normal human dermal fibroblast cells. Results: RG3 treatment increased the expression of extracellular matrix proteins, growth-associated immediate-early genes, and cell proliferation genes in UV-irradiated normal human dermal fibroblast cells. And, RG3 also resulted in enhanced expression of antioxidant proteins such as nuclear factor erythroid 2-related factor-2 and heme oxygenase-1. In addition, RG3 affects the morphology of UV-induced mitochondria and plays a role in protecting mitochondrial dysfunction. Conclusioin: RG3 restores mitochondrial adenosine triphosphate (ATP) and membrane potential via its antioxidant effects in skin cells damaged by UV irradiation, leading to an increase in proteins linked with the extracellular matrix, cell proliferation, and antioxidant activity.
Purpose: Large skin defect by various causes, should be covered by autologous skin graft. But, the donor site of autologous skin graft is limited and leaves permanent donor scar and contracture. There have been our trial to engineer artificial skin using allogenic dermis (AlloDerm) with basement membrane. Methods: Dermal and epidermal layer were separated by immersing in dipase solution for 30 minutes, and the separated layers were treated with 0.05% trypsin for 10 minutes. And then each layer was cultivated to fibroblasts and keratinocytes on a culture medium. Fibroblasts were first penetrated into basement membrane of allogenic dermis facing down, then allogenic dermis was flipped over to face up and keratinocytes were transplanted to allogenic dermis. Results: Observing artificial skin fabricated in vitro, we found following: 1) The artificial skin opened in air for 5 days formed epidermal layer. In dermal layer, fibroblast was distributed evenly among all. 2) The artificial skin opened in air for 30 days formed thicker and thicker, and it formed basement membrane, spinous and granular layers. PAS stain to confirm existence of basement membrane showed positive reaction. 3) Cytokeratin 10 stain to confirm the formation of epidermal layer showed positive reaction. 4) The formation of thick keratin, lamellar body and desmosome similar to human skin were observed in result of an electron micrograph. Conclusion: As a result of research, the structure seen in normal skin such as rete ridge, is found in reproduced artificial skin. This type of artificial skin can be used as a useful model for investigating skin disease and for clinical application also.

Protection of skin cells from chronic infrared-A (IRA) irradiation is crucial for anti-photoaging of the skin. In this study, we investigated the protective activity of Rg3(S) and Rg3(S)-incorporated anionic soybean lecithin liposomes (Rg3/Lipo) with a size of approximately 150 nm against IRA-induced photodamage in human fibroblasts. The formulated Rg3/Lipo showed increased solubility in aqueous solution up to a concentration of 200 ㎍/ml, compared to free Rg3(S). In addition, Rg3/Lipo exhibited superior colloidal stability in aqueous solutions and biocompatibility for normal human dermal fibroblasts (NHDFs). After repeated IRA irradiation on NHDFs, elevated levels of cellular and mitochondrial reactive oxygen species (ROS) were greatly reduced by Rg3(S) and Rg3/Lipo. In addition, cells treated with Rg3/Lipo exhibited noticeably reduced apoptotic signals following IRA irradiation compared to untreated cells. Thus, considering aqueous solubility and cellular responses, Rg3/Lipo could serve as a promising infrared protector for healthy aging of skin cells.
In this study, the effects of verapamil on growth rate, apoptosis, production of transforming growth factor (TGF-${\beta}$) and fibronectin were evaluated in keloid and normal human dermal fibroblasts. Both fibroblasts were primarily cultured from earlobe keloids of three female patients and treated with various concentrations of verapamil. Cell toxicity was assessed by MTT assay, growth rate and apoptosis by FACS, and the production of TGF-${\beta}$ and fibronectin by ELISA and Western blot, respectively. In the $MTT_{50}$, the cell growth was more suppressed in keloid fibroblasts. In the $MTT_{90}$, cell growth was more stimulated in normal fibroblasts. No significant effect appeared on TGF-${\beta}$ expression but an increase in extracellular fibronectin secretion was found in keloid fibroblasts. Keloid fibroblasts responded to verapamil more sensitively, and the percentage of apoptosis was higher at the $MTT_{50}$l. In brief, verapamil had growth-inhibitory effect with inducing apoptosis at the $MTT_{50}$, but rather growth-stimulatory effect at the $MTT_{90}$. The biphasic effect of verapamil depending on the dose might explain one of the reasons of relapse after keloid treatment with verapamil. Clinical application with high concentration (2.5 mg/ml) is advised unless excessive dosage is used.
인삼(Panax ginseng C. A Meyer)의 뿌리는 전통적인 항-노화 및 항-주름제로 동양에서 사용되어 왔다. 그러나 인삼의 어떤 성분이 주름 형성을 억제하는데 효과적인지는 아직 밝혀지지 않았다. 최근 인삼의 주요 활성 성분으로 생각되는 ginsenosides가 20가지 이상 분리되었다. 이들 중 본 연구원들은 인삼에 의한 항-주름의 작용기작을 밝히기 위해 세포간질(extracellular matrix, ECM) 물질대사에 있어 ginsenoside Rg3의 진피에서의 효과를 시험하였다. 본 연구에서, ginsenoside Rg3의 항-주름 효과를 연구하기 위해 진피의 세포간질 구성 성분과 성장 인자를 ELISA (enzyme-1in14ed immunosorbent assay) 측정법으로 평가하였다. Ginsenoside Rg3은 human dermal fibroblasts 배양에서 type I procollagen과 fibronectin(FN) 생합성을 농도 증가에 비례하여 촉진시키고(p < 0.05, n=3), 농도에 비례하여 TGF-$\beta$1 수준을 증가 (p < 0.05, n=3) 시키는 것으로 밝혀졌다. RT-PCR 분석에서 AP-1 전사 인자(transcription factor)의 일부인 c-Jun의 mRNA 수준이 human dermal fibroblasts에서 ginsenoside Rg3에 의해 감소되었다. 이들 결과들은 ginsenoside Rg3이 fibroblasts에서 TGF-$\beta$1과 AP-1의 발현을 변화시킴으로써 type I collagen과 FN합성을 촉진시킴을 보여준다.
쿠메스트롤은 식물이 스트레스에 대항해 합성하는 phytoalexins의 일종으로, 알팔파 새싹, 클로버, 콩나물에서 일반적으로 발견된다. 본 연구에서는 쿠메스트롤의 자외선에 의해 유도되는 피부 진피세포 광노화 예방 효능에 관한 연구를 실시하였다. 쿠메스트롤 전처리는 자외선 B 조사에 의해 감소된 Sirt1 단백질 발현 및 활성과 하위 미토콘드리아 생합성 관련 유전자인 PGC-$1{\alpha}$, NRF1, TFAM의 발현 변화를 감소시켰다. 또한, ATP 및 ROS 생성량을 정상화시키고 피부 노화를 유도하는 최종당화산물 생성을 억제하였다. 이상의 결과에서 쿠메스트롤은 자외선 조사에 의해 발생하는 진피 세포 내 미토콘드리아 손상 및 이에 따른 당화 단백질 생성을 감소시킴으로써 피부 광노화 현상으로부터 보호할 수 있음을 확인하였다.

Skin aging appears to be the result of overlapping intrinsic (including genetic and hormonal factors) and extrinsic (external environment including chronic light exposure, chemicals, and toxins) processes. These factors cause decreases in the synthesis of collagen type I and elastin in fibroblasts and increases in the melanin in melanocytes. Collagen Type I is the most abundant type of collagen and is a major structural protein in human body tissues. In previous studies, many products containing collagen derived from land and marine animals as well as other sources have been used for a wide range of purposes in cosmetics and food. However, to our knowledge, the effects of human collagen-derived peptides on improvements in skin condition have not been investigated. Here we isolate and identify the domain of a human COL1A2-derived protein which promotes fibroblast cell proliferation and collagen type I synthesis. This human COL 1A2-derived peptide enhances wound healing and elastin production. Finally, the human collagen alpha-2 type I-derived peptide (SMM) ameliorates collagen type I synthesis, cell proliferation, cell migration, and elastin synthesis, supporting a significant anti-wrinkle effect. Collectively, these results demonstrate that human collagen alpha-2 type I-derived peptides is practically accessible in both cosmetics and food, with the goal of improving skin condition.
While adipose-derived stem cell-conditioned medium (ADSC-CM) has been demonstrated to promote skin wound healing, the mechanism regulating this effect remains unelucidated. In this study, we aimed to investigate the role of Ell3 in the wound healing activity of ADSC-CM. In vitro analysis revealed that Ell3 suppression in ADSCs impairs the promotive activity of ADSC-CM on the proliferation and migration of mouse embryonic fibroblasts (MEF) and normal human dermal fibroblasts (NHDF). Consistently, the expression of MMP family genes, which regulate cell proliferation and migration, was significantly suppressed in MEF and NHDF treated with siEll3-transfected ADSC-CM. Proinflammatory cytokines, such as interleukin-1 and interleukin-6, were highly expressed in MEF treated with siEll3-transfected ADSC-CM. The wound healing activity of siEll3-transfected ADSC-CM was significantly lower than that of the control in vivo. Our results suggest that Ell3 may contribute to the inhibition of inflammatory response during skin wound healing.
노화 과정 중에 일어나는 extracellular matrix (ECM)의 변성은 피부의 주름과 탄력 감소를 유발한다. 현재까지 항노화의 주요 타겟은 metalloproteases 혹은 콜라겐이나 엘라스틴 같은 구조 단백질에 집중되어 있지만, 최근 세포와 ECM 단백질(콜라겐, 피브릴린, 파이브로넥틴) 간의 상호작용이 세포의 생존과 증식, 조직의 재건에 중요한 역할을 한다고 알려졌다. 파이브로넥틴은 다른 ECM 단백질이나 인테그린 같은 세포 표면 수용체와 결합할 수 있는 부위를 가진 부착 단백질이다. 최근 보고에 따르면 세린 프로티아제들에 의해 분해된 파이프로넥틴 조각이 골아세포에서 MMPs 발현을 증가시킨다. 그러나 파이브로넥틴 조각의 사람 피부에서의 역할은 보고된 바 없다. 본 연구에서는 노인의 피부에서 파이브로넥틴 조각이 현저히 증가되어 있으며, 섬유아세포에 파이브로넥틴 조각을 처리하였을 시, MMP-1의 발현과 MMP-2의 활성이 증가한다는 것을 입증하였다. 이 결과는 파이브로넥틴 조각이 피부 노화를 유발하는 새로운 인자일 가능성을 제시하고 있다.
Sunlight, and in particular its UV component, is the major environmental trigger that underlies the major signs of human skin and skin cancer in general. Therefore, this study was carried out to investigate the UV protection effects of R. coreanus. R. coreanus was extracted by ultra high pressure extraction process at 500 MPa and $30^{\circ}C$ for 5 and 15 minutes. The cytotoxicity of the extracts extracted by ultra high pressure process on human dermal fibroblast cell CCD-986sk, human kidney normal cell HEK293, and human lung normal cell HEL299 was measured as 17.5%, 16.5% and 14.0%, respectively in adding $1.0\;mg/m{\ell}$ of the samples, which was much lower than that from conventional water extraction method at $100^{\circ}C$ as 23.2%, 22.5%, 21.2%. The secretion of $NO^-$ from macrophage showed $15.9\;{\mu}M$ on the R. coreanus extract from this process, which was higher than others. Prostaglandin $E_2$ ($PGE_2$) production from UV-induced human skin cells was also greatly decreased down to $510\;pg/m{\ell}$, compared to the control. From the results, we considered that the extracts from R. coreanus could be potent natural materials for skin anti-inflammation agent, and could be used as a potential anti-aging for the photo-damaged skin.
