Isolation of Cryptosporidium parvum oocysts from fecal samples - The combination of ether extraction and discontinuous sucrose gradients (분변에 함유된 Cryptosporidium parvum 오오시스트의 순수분리 -Ether extraction과 discontinuous sucrose gradients의 병용)

  • A calf and 50 mice were infected with Cryptosporidium parvum and their fecal materials were collected and treated 10 ether extraction (EE), followed by discontinuous sucrose gradients (DSG) method. EE method was to remove some of fat or lipid from feces. Sediments were washed by centrifugation ($1,500{\;}{\times}{\;}g$ for 10 min., 3 times) In phosphate-buffered saline and then these washed sediments were sleeved sequentially through stainless steel screens with a final mesh of 250 ($61{\;}{\mu\textrm{m}}$ porosity) to remove other debris. After sieving, the materials were suspended in 2.5% potassium dlchromate solution. Oocysts were counted by using a hemocytometer and the recovery rate of pure oocysts was calculated on the basis of the count. Following centrifugation ($1,500{\;}{\times}{\;}g$ for 30 min.) by DSG method, most oocysts were recovered at the interface between a gravity of 1.103 and 1.064. The recovery rates of pure oocysts from the fecal suspension of the calf ($3.8{\;}{\times}{\;}10^7/ml$) and the mouse ($3.2{\;}{\times}{\;}10^6/ml$) treated with EE method were 81.6% and 51.6%, respectively. It is suggested that the recovery rate was dependent on the number of oocysts In each suspension treated with EE method. To get the 50% recovery rate, there must be more than $2{\;}{\times}{\;}10^6$ oocysts per ml of the fecal suspension treated with EE method. By the combination of the two methods it was possible to isolate C. parvum oocysts from normal feces of the calf and mouse as well as from dlarrhelc feces.

Detection Efficiency of Microcalcification using Computer Aided Diagnosis in the Breast Ultrasonography Images (컴퓨터보조진단을 이용한 유방 초음파영상에서의 미세석회화 검출 효율)

  • Digital Mammography makes it possible to reproduce the entire breast image. And it is used to detect microcalcification and mass which are the most important point of view of nonpalpable early breast cancer, so it has been used as the primary screening test of breast disease. It is reported that microcalcification of breast lesion is important in diagnosis of early breast cancer. In this study, six types of texture features algorithms are used to detect microcalcification on breast US images and the study has analyzed recognition rate of lesion between normal US images and other US images which microcalification is seen. As a result of the experiment, Computer aided diagnosis recognition rate that distinguishes mammography and breast US disease was considerably high 70~98%. The average contrast and entropy parameters were low in ROC analysis, but sensitivity and specificity of four types parameters were over 90%. Therefore it is possible to detect microcalcification on US images. If not only six types of texture features algorithms but also the research of additional parameter algorithm is being continually proceeded and basis of practical use on CAD is being prepared, it can be a important meaning as pre-reading. Also, it is considered very useful things for early diagnosis of breast cancer.

Experimental studies of Glycine max Merr. (black bean), Triticum aestivum L. (wheat) and Oryza sativa L. (rice bran) extracts on the effects of hair growth activity and physical properties (검은콩, 밀, 쌀겨 추출물이 모발의 성장과 물리적 특성에 미치는 효과)

  • Objects : Glycine max Merr. (black bean), Triticum aestivum L. (wheat) and Oryza sativa L. (rice bran) have been widely used for treatment of relaxion of smooth muscle, gastrointestinal hemorrhage and alopecia in Korean Traditional Medicine. In this research, we examined the effect of the extracts, obtained from EtOH extracts of 3 kinds of traditional plants, on hair growing activity of the DP6 and C3H10T1 cell and physical properties. Materials and Methods : On the basis of previous studies, three traditional plants were selected and we extracted them with ethanol. We evaluated their hairy dermal papillar cell proliferation activity and mouse mesenchymal stem cell in vitro model. Also, 3 herbal extracts were added to the normal shampoo formulation in ranges of 0.1% and we validated tensile properties and physical changes using aged hair. In this research, we compared the tensile strength, shine and color appearance between the hair (general formulation) and the hair after applying shampoo with natural extracts. To analyze the luster and color image, we use the SAMBA hardware and software made by Bossa Nova Technologies. Results : In the comparative test for tensile characteristic between the hair treated general formulation(control) and the hair applying special formulation including 3 kinds of extracts, tensile distance and energy of the latter are larger than control on average. The shine and color appearance were also increased after using shampoo including natural extracts(shine : 10.9%, color appearance: 24.12%). We observed the enhancement of hair growth activity in the DP6 and C3H10T1 cell. Especially black bean extracts had the most powerful effect in the dermal papillar cell proliferation. Conclusion : These experiments suggest that extracts of Glycine max Merr. (black bean), Triticum aestivum L. (wheat) and Oryza sativa L. (rice bran) stimulate the hair growth activity and can improve physical activities of aged hair. Shampoo product, which contains 3 kinds of natural extracts, would be used for the treatment for aged hair.

  • In the high temperature situation like in a fire, the high strength of concrete (HSC) has extreme danger named explosive spalling. It is assumed that the major cause of explosive spalling is water vapour pressure inside concrete. This paper examines the effect of the compressive strength and the moisture content on the initial occurrence of explosive spalling. For the effective experiment of the initial explosive spalling, the curve of ISO834 temperature profile is applied on the basis of 15 minute and 30 minute. As a result, the more increase the compressive strength and the moisture content, the more increase the occurrence and phenomenon of explosive spalling. This paper analyzes the territory of explosive spalling depending the compressive strength and the moisture content. The explosive spalling is not examined in the case of the compressive strength 50~100 MPa and the moisture content below 3% and the compressive strength over 100 MPa and the moisture content below 1%. Also, due to the HSC, which makes it more difficult to transport vapour and moisture, very high vapour-pressure may occur close to the surface, there is a greater risk that HSC spalls compared with normal strength concrete (NSC).

Restoration planning of the Seoul Metropolitan area, Korea toward eco-city

  • In order to prepare a basis for ecological restoration of the Seoul Metropolitan area, ecological diagnoses on soil physico-chemical properties and vegetation structure were carried out. Land use patterns, actual vegetation, and biotope patterns were also investigated based on aerial photograph interpretation and field checks. I formulated landscape elements overlaying those data and evaluated the ecological value of each element. Soil pollution was evaluated by analyzing soil samples collected in each grid on the mesh map, divided by 2km $\times$ 2km intervals. Soil samples were collected in forests or grasslands escaped from direct human interference. Soil pollution evaluated from pH, and SO$_4$, Ca, Mg, and Al contents of soil was more severe in the urban outskirts than in the urban center. Those soil environmental factors showed significant correlation with each other. Vegetation in the urban area was different in species composition from that in suburban areas and showed lower diversity compared with that in the suburban areas. Successional process investigated by population structure of major species also showed a difference. That is, successional trend was normal in suburban areas, but that in urban areas showed a retrogressive pattern. The landscape ecological map of Seoul indicates that the urban center lacks vegetation and greenery space is restricted in urban outskirts. Such an uneven distribution of vegetation has caused a specific urban climate and thereby contributed to aggravation of air and soil pollution, furthermore causing vegetation decline. From this result, it was estimated that such uneven distribution of vegetation functioned as a trigger factor to deteriorate the urban environment. I suggested, therefore, a restoration plan based on landscape ecological principles, which emphasizes connectivity and even distribution of green areas throughout the whole area of the Seoul to solve this complex environmental problem. In this restoration plan, first of all, I decided the priority order for connection of the fragmented greenery spaces based on the distances from the core reserves comprised of green belt and rivers, which play roles as habitats of wildlife as well as for improvement of urban environment. Next, I prepared methods to restore each landscape element included in the paths of green network to be constructed in the future on the bases of such preferential order. Rivers and roads, which hold good connectivity, were chosen as elements to play important roles in constructing green network by linking the fragmented greenery spaces.

Neuronal injury in AIDS dementia: Potential treatment with NMDA open-channel blockers and nitric oxide-related species

  • The neurological manifestations of AIDS include dementia, encountered even in the absence of opportunistic superinfection or malignancy. The AIDS Dementia Complex appears to be associated with several neuropathological abnormalities, including astrogliosis and neuronal injury or loss. How can HIV-1 result in neuronal damage if neurons themselves are only rarely, if ever, infected by the vitus\ulcorner In vitro experiments from several different laboratiories have lent support to the existence of HIV- and immune-related toxins. In one recently defined pathway to neuronal injury, HIV-infected macrophages/microglia as well as macrophages activated by HIV-1 envelope protein gp120 appear to secrete excitants/neurotoxins. These substances may include arachidonic acid, platelet-activating factor, free radicals (NO - and O$_2$), glutamate, quinolinate, cysteine, cytokines (TNF-${\alpha}$, IL1-B, IL-6), and as yet unidentified factors emanating from stimulated macrophages and possibly reactive astrocytes. A final common pathway for newonal suscepubility appears to be operative, similar to that observed in stroke, trauma, epilepsy, and several neurodegenerative diseases, including Huntington's disease, Parkinson's disease, and amyotrophic lateral sclerosis. This mechanism involves excessive activation of N-methyl-D-aspartate (NMDA) receptor-operated channels, with resultant excessive influx of Ca$\^$2+/ leading to neuronal damage, and thus offers hope for future pharmacological intervention. This chapter reviews two clinically-tolerated NMDA antagonists, memantine and nitroglycerin; (ⅰ) Memantine is an open-channel blocker of the NMDA-associated ion channel and a close congener of the anti-viral and anti-parkinsonian drug amantadine. Memantine blocks the effects of escalating levels of excitotoxins to a greater degree than lower (piysiological) levels of these excitatory amino acids, thus sparing to some extent normal neuronal function. (ⅱ) Niuoglycerin acts at a redox modulatory site of the NMDA receptor/complex to downregulate its activity. The neuroprotective action of nitroglycerin at this site is mediated by n chemical species related to nitric oxide, but in a higher oxidation state, resulting in transfer of an NO group to a critical cysteine on the NMDA receptor. Because of the clinical safety of these drugs, they have the potential for trials in humans. As the structural basis for redox modulation is further elucidated, it may become possible to design even better redox reactive reagents of chinical value. To this end, redox modulatory sites of NMDA receptors have begun to be characterized at a molecular level using site-directed mutagenesis of recombinant subunits (NMDAR1, NMDAR2A-D). Two types of redox modulation can be distinguished. The first type gives rise to a persistent change in the functional activity of the receptor, and we have identified two cysteine residues on the NMDARI subunit (#744 and #798) that are responsible for this action. A second site, presumably also a cysteine(s) because <1 mM N-ethylmaleimide can block its effect in native neurons, underlies the other, more transient redox action. It appears to be at this, as yet unidentified, site on the NMDA receptor that the NO group acts, at least in recombinant receptors.

The Effect of Aerobic Rhythmical Exercise Program on Physical Fitness, Self-efficacy and Quality of Life in Elderly (유산소 리듬 운동프로그램이 노인의 체력, 자기효능감 및 삶의 질에 미치는 효과)

  • The purpose of this study was to determine the effects of aerobic rhythmical exercise program on $VO_2$ max, leg strength, grip strength, flexibility, exercise self-efficacy and quality of life in elderly. This quai-experimental study was designed as a non-equivalent control group pretest-posttest study. 36 subjects, aged between 65 and 73 years who have normal cognition. sensory function and cerebellum function participated in this study. 19 experimental group subjects participated aerobic rhythmical exercise program. Aerobic rhythmical exercise program was developed on the basis of calisthenics, stretching, aerobic exercise, Korean traditional dance and music by the author. The program consisted of approximately 1 hours of exercise, 3 times a week for 9 weeks. During 1 hours workout, there were 15 minutes of warm-up dancing, 35 minutes of conditioning dance and 10 minutes of cool-down dancing. The intensity for the conditioning phase was between $40\%\;and\;60\%$ of age adiusted maximum heart rates. The $VO_2$ max, grip strength, leg strength, flexibility, exercise self-efficacy and quality of life were measured prior to and following the experimental treatment. Data were analyzed with $x^2-test$, t-test, mean, standard deviation. percentage of change and ANCOVA test using SAS program. Results were obtained as follows. 1. The $VO_2$ max of the experimental group were significantly lower than those of the control group following the aerobic rhythmical exercise program(F=7.76, p=0.008). 2. The grip strength of the experimental group were significantly lower than those of the control group following the aerobic rhythmical exercise program(F=4.61, p=0.039). 3. The leg strength of the experimental group were significantly lower than those of the control group following the aerobic rhythmical exercise program(F=9.29, p=0.000). 4. The was no significantly difference of flexibility between experimental and control group 5. The self-efficacy of the experimental group were significantly lower than those of the control group following the aerobic rhythmical exercise program(F=43.40, p=0.000). 6. The quality of life of the experimental group were significantly lower than those of the control group following the aerobic rhythmical exercise program(F=8.91. p=0.005). The results suggest that aerobic rhythmical exercise program can improve $VO_2$ max, grip strength. leg strength. exercise self-efficacy and quality of life elderly. Thus. aerobic rhythmical exercise program was a useful nursing intervention for elderly.

  • Cryogenic LNG(Liquefied Natural Gas) which is stored in the cylindrical storage tanks of $100,000m^3$ has very complex flow phenomena and the changes of thermal properties with exterior conditions and operation modes. These complex thermofluid behaviors are affected by the storage, exterior conditions of LNG, design specifications and heat transfer characteristics of tanks. Also, those have influence on the stable storage and supply of LNG in the storage tanks. Thus this study peformed the analysis on the 2-D heat transfer of the tank with exterior conditions, on the Cool Down Process in order to cool down the LNG Storage Tank at the initial normal state, and on the Filling Process considered for incoming and rising of LNG. The analysis on the Mixing LNG Storage was studied too. At last, the visualized program on the complex thermofluidodynamic analysis was developed on the basis of the above analyses. The development of this program becomes to be used to the basic design of the commercial tanks as well as to assure technical skill of the analysis on the thermal stability of the stored LNG in the LNG Storage Tank.

TGF-$\alpha$, -$\beta$$_1$, and bFGF mRNA Expression of Lens Epithelial Cells in Senile and Diabetic Cataract

  • Anterior subcapsular cataract was developed by opacification with transdifferentiation and abnormal proliferation of lens epithelial cells (LECs) and pathological accumulation of extracellular matrix (ECM). After-cataract also be caused by a similar transdifferentiation of LECs remaining after surgery and the accompanying increase of ECM deposits. It is blown that prostaglandin E2 and cytokine, such as TGF-$\beta$, bFGF, and IL-1, were associated with abnormal proliferation and transdifferentiation of LECs. The aim of this study was to detect the expression of transforming growth factor-$\alpha$ (TGF-$\alpha$), transforming growth factor-$\beta_1$(TGF-$\beta_1$) and basic fibroblast growth factor (bFGF) in LECs of senile and diabetic cataract. The expressions of these growth factors in lens epithelial cells were determined. The sample for growth factor determination were collected in senile cataract patients without metabolic disorder, especially diabetes mellitus and diabetic cataract patients. The mRNA expression of growth factors was detected by semi-quantitative reverse transcription - polymerase chain reaction (RT-PCR) followed by Southern blot analysis. Statistics were analysed using Wilcoxon rank sum test. Semi-quantitative RT-PCR/southern analysis of RNA obtained from thirty surgical specimens demonstrated that the level of mRNA expression of TGF-$\alpha$, -$\beta_1$ and bFGF was increased in diabetic cataract lens tissues compared with senile cataract specimens but non-significant, bFGF and TGF-$\beta_1$ mRNA expression were detected in most patients, expression level of TGF-$\beta_1$ was most high on the basis of normal ocular concentration. Detection rate of TGF-$\alpha$ in diabetic cataract was 1.5 fold higher than in senile cataract (P=0.098). TGF-$\alpha$, TGF-$\beta_1$, and bFGF mRNA expression of LECs were detected in senile and diabetic cataract. In both patient groups, expression level of TGF-$\beta_1$, mRNA was high, so We suggest TGF-$\beta_1$ strong influence in development of senile cataract and of diabetic cataract also. TGF-$\alpha$ expression level was similar but more frequently detected in diabetic cataract than in senile cataract. In conclusion, TGF-$\alpha$ may be associated with early development of diabetic cataract.

Plant Regeneration through Somatic Embryogenesis of Leymus chinensis Trin. (양초(Leymus chinensis Trin.)의 체세포배발생에 의한 식물체 재분화)

  • Chinese leymus (Leymus chinensis Trin.) is a perennial grass that is widely distributed at high pH sodic and arid soil in the northeastern Asia. An efficient regeneration system was established through somatic embryogenesis of mature seeds to understand its high adaptability to harsh environmental conditions on the basis of molecular biology. The calli were efficiently induced (about $70\%$) from mature seeds on MS medium supplemented with $1.5\;\cal{mg/L}$ 2,4-D. Somatic embryos were formed from the surface of embryogenic callus on MS medium supplemented with $2.0\;\cal{mg/L}\;kinetin\;and\;0.5\;\cal{mg/L}$ NAA after 3 weeks of culture. Roots were induced from the shoot when transferred to MS medium without plant growth regulator for 1 week. Plant regeneration rate was $36\%$ and regenerated plantlets were grown to normal mature plants in pot. An efficient plant regeneration system in this study will be useful for molecular breeding of L. chinensis.
