  • Background: Lung adenocarcinoma (LUAD) with ground-glass opacity (GGO) can become aggravated, but the reasons for this aggravation are not fully understood. The goal of this study was to analyze the genetic features and causes of progression of GGO LUAD. Methods: LUAD tumor samples and normal tissues were analyzed using an Illumina HiSeq 4000 system. After the tumor mutational burden (TMB) was calculated, the identified mutations were classified as those found only in GGO LUAD, those present only in nonGGO LUAD, and those common to both tissue types. Ten high-frequency genes were selected from each domain, after which protein interaction network analysis was conducted. Results: Overall, 227 mutations in GGO LUAD, 212 in non-GGO LUAD, and 48 that were common to both tumor types were found. The TMB was 8.8 in GGO and 7.8 in non-GGO samples. In GGO LUAD, mutations of FCGBP and SFTPA1 were identified. FOXQ1, IRF5, and MAGEC1 mutations were common to both types, and CDC27 and NOTCH4 mutations were identified in the non-GGO LUAD. Protein interaction network analysis indicated that IRF5 (common to both tissue types) and CDC27 (found in the non-GGO LUAD) had significant biological functions related to the cell cycle and proliferation. Conclusion: In conclusion, GGO LUAD exhibited a higher TMB than non-GGO LUAD. No clinically meaningful mutations were found to be specific to GGO LUAD, but mutations involved in the epithelial-mesenchymal transition or cell cycle were found in both tumor types and in non-GGO tissue alone. These findings could explain the non-invasiveness of GGO-type LUAD.

  • Non-syndromic hearing loss (NSHL) is a common hereditary disorder. Both clinical and genetic heterogeneity has created many obstacles to understanding the causes of NSHL. The present study has attempted to ravel the genetic aetiology in NSHL progression and to screen out potential target genes using computational approaches. The reported NSHL target genes (2009-2020) have been studied by analyzing different biochemical and signaling pathways, interpretation of their functional association network, and discovery of important regulatory interactions with three previously established miRNAs in the human inner ear as well as in NSHL such as miR-183, miR-182, and miR-96. This study has identified SMAD4 and SNAI2 as the most putative target genes of NSHL. But pathogenic and deleterious non-synonymous single nucleotide polymorphisms discovered within SMAD4 is anticipated to have an impact on NSHL progression. Additionally, the identified deleterious variants in the functional domains of SMAD4 added a supportive clue for further study. Thus, the identified deleterious variant i.e., rs377767367 (G491V) in SMAD4 needs further clinical validation. The present outcomes would provide insights into the genetics of NSHL progression.

  • Based on latest research, the parametric quality statistics cannot be used in non-normal process with demand pattern of many-variety and small-volume, since it involves extremely small sample size. The research proposes nonparametric quality statistics according to the number of lot or batch in the non-normal process. Additionally, the nonparametric Process Capability Index (PCI) is used with 14 identified non-normal distributions.

  • Usnea longissima has a long history of use as a traditional medicine. Several bioactive compounds, primarily belonging to the polyketide family, have been isolated from U. longissima. However, the genes for the biosynthesis of these compounds are yet to be identified. In the present study, three different types of non-reducing polyketide synthases (UlPKS2, UlPKS4, and UlPKS6) were identified from a cultured lichen-forming fungus of U. longissima. Phylogenetic analysis of product template domains showed that UlPKS2 and UlPKS4 belong to group IV, which includes the non-reducing polyketide synthases with an methyltransferase (MeT) domain that are involved in methylorcinol-based compound synthesis; UlPKS6 was found to belong to group I, which includes the non-reducing polyketide synthases that synthesize single aromatic ring polyketides, such as orsellinic acid. Reverse transcriptase-PCR analysis demonstrated that UlPKS2 and UlPKS4 were upregulated by sucrose; UlPKS6 was downregulated by asparagine, glycine, and alanine.

  • The goal of this study is to investigate the effect of non-synchronous sensing when using wireless sensors on structural identification and to attempt correcting such errors in order to obtain a better identification result. The sources causing non-synchronous sensing are discussed first and the magnitudes of such synchronization errors are estimated based on time stamps of data samples collected from Imote2 sensors; next the impact of synchronization errors on power spectral densities (PSDs) and correlation functions of output responses are derived analytically; finally a new method is proposed to correct such errors. In this correction method, the corrected PSDs of output responses are estimated using non-synchronous samples based on a modified FFT. The effect of synchronization errors in the measured output responses on structural identification and the application of this correction method are demonstrated using simulation examples. The simulation results show that even small synchronization errors in the output responses can distort the identified modal and stiffness parameters remarkably while the parameters identified using the proposed correction method can achieve high accuracy.

  • Objective: The adulteration of raw beef (BMr) with dog meat (DMr) and pork (PMr) becomes a serious problem because it is associated with halal status, quality, and safety of meats. This research aimed to develop an effective authentication method to detect non-halal meats (dog meat and pork) in beef using metabolomics approach. Methods: Liquid chromatography-high resolution mass spectrometry (LC-HRMS) using untargeted approach combined with chemometrics was applied for analysis non-halal meats in BMr. Results: The untargeted metabolomics approach successfully identified various metabolites in BMr DMr, PMr, and their mixtures. The discrimination and classification between authentic BMr and those adulterated with DMr and PMr were successfully determined using partial least square-discriminant analysis (PLS-DA) with high accuracy. All BMr samples containing non-halal meats could be differentiated from authentic BMr. A number of discriminating metabolites with potential as biomarkers to discriminate BMr in the mixtures with DMr and PMr could be identified from the analysis of variable importance for projection value. Partial least square (PLS) and orthogonal PLS (OPLS) regression using discriminating metabolites showed high accuracy (R2 >0.990) and high precision (both RMSEC and RMSEE <5%) in predicting the concentration of DMr and PMr present in beef indicating that the discriminating metabolites were good predictors. The developed untargeted LC-HRMS metabolomics and chemometrics successfully identified non-halal meats adulteration (DMr and PMr) in beef with high sensitivity up to 0.1% (w/w). Conclusion: A combination of LC-HRMS untargeted metabolomic and chemometrics promises to be an effective analytical technique for halal authenticity testing of meats. This method could be further standardized and proposed as a method for halal authentication of meats.

  • Analyzing the aftermath of events at domestic nuclear power plants brings in the question: "Why do workers not comply with the prescribed procedures?" The current investigation of nuclear power plant events identifies their reasons considering the factors affecting the workers' behaviors. However, there are some complications to it: in addition to confirming the action such as an error or a violation, there is a limit to identifying the intention of the actor. To overcome this limitation, the study analyzed and examined the reasons for non-compliance identified in nuclear power plant events by Reason's rule-related behavior classification. For behavior analysis, I selected unit behaviors for events that are related to human and organizational factors and occurred at domestic nuclear power plants since 2017, and then I applied the rule-related behavior classification introduced by Reason (2008). This allowed me to identify the intentions by classifying unit behaviors according to quality and compliance with the rules. I also identified the factors that influenced unit behaviors. The analysis showed that most often, non-compliance only pursued personal goals and was based on inadequate risk appraisal. On the other hand, the analysis identified cases where it was caused by such factors as poorly written procedures or human system interfaces. Therefore, the probability of non-compliance can be reduced if these factors are properly addressed. Unlike event investigation techniques that struggle to identify the reasons for employee behavior, this study provides a new interpretation of non-compliance in nuclear power plant events by examining workers' intentions based on the concept of rule-related behavior classification.

  • In this paper, the design of PID controller using Neural networks for the control of non-linear system is presented. First, non-linear system is identified using BPN(Backpropagation Network) algorithm. This identified model is connected to the PID controller and the parameters of PID controller are updated to the direction of reducing the difference between the identified model output and model reference output in arbitrary input signal. Therefore, identified model output tracks the model reference output in an acceptable error range and the parameters of controller are updated adaptively. The output of the system has a good performance in case of both noisy and noiseless model reference and we can control the system stable in off-line when the dynamics of the system is changed.
