Current status on the occurrence and management of disease, insect and mite pests in the non-chemical or organic apple orchards (무농약 유기재배 사과원의 병해충 발생 및 관리 실태)

  • Current status on the occurrence and the management of the major disease, insect and mite pests were investigated in the organic or non-chemical pest control orchards from 2005 to 2009. Numbers of certified organic or non-chemical apple orchards were increased from 14 in 2005 to 78 in 2008. Severe damages on leaves and fruits occurred by the several diseases such as marssonina blotch, bitter rot, white rot, sooty blotch and flyspeck, and the several insect pests such as apple leaf-curling aphid, woolly apple aphid, oriental fruit moth and peach fruit moth on the almost certified organic or non-chemical pest control orchards. About 10 and 18 environmental-friendly materials were used to control diseases and insect or mite pests respectively. But, lime sulfur and bordeaux mixture to diseases and machine oil, plant oil mixed with egg yolk, and pheromone mating disruptions to insect pests were effective to control under the adequate conditions. At present, it is extremely difficult to produce organic apples in Korea. Growers must consider about and solve so many conditions on the cultivar, weather, local site, marketing and so on, before when they decide to change from conventional or IPM(Integrated Pest Management) to organic or non-chemical pest control orchards.

An Analysis Regarding Trends of Dualism in Korean Agriculture (농업생산 양극화 추이에 대한 연구)

  • Purpose - The structural changes of Korean agriculture are complex due to heterogeneous production processes and farms' features. This study analyzed trends of dualism in Korean agriculture over the period 2000-15 based on farm-level data to clarify the specific trends of dualism in terms of farm income, farm-size, and farm operators' age. From the results of this study, we would be able to understand the features of structural changes in Korean agriculture more profoundly. Research design, data, and methodology - We incorporated farm-level data in South Korea: Agricultural census and Farm household economy survey. As measures of inequality, we used size-weighted quantiles, and normalized Gini coefficients as well as mean and conventional quantiles. The size-weighted quantiles are more robust to changes in the number of small farms, but they are more sensitive to changes in the distribution of farm-size. Thus, they would be more useful to identify trends of dualism of Korean agriculture. Results - The results show that the farmland distribution of crop farms became more skewed and dispersed. However, the herd distribution of livestock farms became more concentrated. To be specific, their mean and 1st quantile increases more rapidly than their size-weighted 2nd quantile and size-weighted 3rd quantile. Gini coefficients of livestock farms regarding their herd distribution decreased by 0.1 on average. In the case of income distribution, the results indicate that the polarization regarding farm household/agricultural/non-agricultural income became more severe. However, we also found that the distribution of transfer income became concentrated continuously. The results imply that transfer income including subsidies would decrease farm income polarization. Lastly, during the study periods, Korean farms were aging over time, and age distribution of them more concentrated. Conclusions - The structure of Korean agriculture has been changing, even though the absolute size of it decreased over time. Land (herd) distribution became more dispersed (concentrated). Inequality regarding agricultural income became more severe, and it made farm household income more polarized even though transfer income would decrease income gaps among farms. Lastly, farms continue to age regardless of farm types and this might affect the structural changes in Korean agriculture in the future.

Identification of Candidate Porcine miRNA-302/367 Cluster and Its Function in Somatic Cell Reprogramming

  • MicroRNAs (miRNAs) are approximately 22 nucleotides of small noncoding RNAs that control gene expression at the posttranscriptional level through translational inhibition and destabilization of their target mRNAs. The miRNAs are phylogenetically conserved and have been shown to be instrumental in a wide variety of key biological processes including cell cycle regulation, apoptosis, metabolism, imprinting, and differentiation. Recently, a paper has shown that expression of the miRNA-302/367 cluster expressed abundantly in mouse and human embryonic stem cells (ESCs) can directly reprogram mouse and human somatic cells to induced pluripotent stem cells (iPSCs) efficiently in the absence of any of the four factors, Oct4, Sox2, c-Myc, and Klf4. To apply this efficient method to porcine, we analyzed porcine genomic sequence containing predicted porcine miRNA-302/367 cluster through ENSEMBL database, generated a non-replicative episomal vector system including miRNA-302/367 cluster originated from porcine embryonic fibroblasts (PEF), and tried to make porcine iPSCs by transfection of the miRNA-302/367 cluster. Colonies expressing EGFP and forming compact shape were found, but they were not established as iPSC lines. Our data in this study show that pig miRNA-302/367 cluster could not satisfy requirement of PEF reprogramming conditions for pluripotency. To make pig iPSC lines by miRNA, further studies on the role of miRNAs in pluripotency and new trials of transfection with conventional reprogramming factors are needed.

Understanding the Protox Inhibition Activity of Novel 1-(5-methyl-3-phenylisoxazolin-5-yl)methoxy-2-chloro-4-fluorobenzene Derivatives Using Holographic Quantitative Structure-Activity Relationship (HQSAR) Methodology (홀로그램(H) QSAR 방법에 따른 1-(5-methyl-3-phenylisoxazolin-5-yl)methoxy-2-chloro-4-fluorobenzene 유도체들의 Protox 저해 활성에 관한 이해)

  • Holographic quantitative structure activity relationships (HQSAR) as 2D QSAR between the herbicidal activities against root and shoot of rice plant (Orysa sativa L.) and barnyardgrass (Echinochloa crus-galli), and structures of A=3,4,5,6-tetra-hydrophthalimino, B = 3-chloro-4,5,6,7-tetrahydro-2H-indazolyl and C = 3,4-dimethylmaleimino substituents in 1-(5-methyl-3-phenylisoxazolin-5-yl)methoxy-2-chloro-4-fluorobenzene derivatives were studied and discussed. The statistical results of four HQSAR models for the herbicidal activities against root and shoot of the two plants showed the best predictability of the herbicidal activities based on the cross-validated $r^2\;_{cv}\;(q^2=\;0.760{\sim}0.924)$, non cross-validated conventional coefficient $(r^2\;_{ncv}\;=\;0.868{\sim}0.970)$ and PRESS values $(0.123{\sim}0.261)$. The results indicated that the qualities of HQSAR models for barnyardgrass were slightly higher than that of rice plant. And also, the predictability of HQSAR models were higher $(q^2\;=\;HQSAR\;>\;CoMFA)$ than CoMFA but the conventional coefficients of HQSAR models lower $(r^2\;=\;HQSAR\;<\;CoMFA)$ than CoMFA. Moreover, from the contribution maps, it was founded that the selectivity between the two plants depends upon the 2-fluoro-4-chloro-5-alkoxyanilino and $R_3$ substituent on the C-phenyl ring. These features suggest where to modify a molecular structure in order to improve its selective of herbicidal activities against barnyardgrass.

Effects of ambient temperature and dietary glycerol addition on growth performance, blood parameters and immune cell populations of Korean cattle steers

  • Objective: This study was performed to evaluate whether ambient temperature and dietary glycerol addition affect growth performance, and blood metabolic and immunological parameters, in beef cattle. Methods: Twenty Korean cattle steers ($405.1{\pm}7.11kg$ of body weight [BW], $14.2{\pm}0.15$ months of age) were divided into a conventional control diet group (n = 10) and a 2% glycerol- added group (n = 10). Steers were fed 1.6% BW of a concentrate diet and 0.75% BW of a timothy hay diet for 8 weeks (4 weeks from July 28th to August 26th and 4 weeks from August 27th to September 26th). Blood was collected four times on July 28th, August 11th, August 27th, and September 26th. Results: The maximum indoor ambient temperature-humidity index in August (75.8) was higher (p<0.001) than that in September (70.0), and in August was within the mild heat stress (HS) category range previously reported for dairy cattle. The average daily gain (ADG; p = 0.03) and feed efficiency (p<0.001) were higher in hotter August than in September. Glycerol addition did not affect ADG and feed efficiency. Neither month nor glycerol addition affected blood concentrations of cortisol, triglyceride, or non-esterified fatty acid. Blood concentrations of cholesterol, low-density lipoprotein, high-density lipoprotein, glucose, and albumin were lower (p<0.05) on August 27th than on September 26 th, and blood phosphorus, calcium and magnesium concentrations were also lower on August 27th than on September 27th. Glycerol addition did not affect these blood parameters. Percentages of $CD4^+$ T cells and $CD8^+$ T cells were higher (p<0.05) on July 28th than on August 27th and September 26th. The blood $CD8^+$ T cell population was lower in the glycerol supplemented-group compared to the control group on July 28th and August 27th. Conclusion: Korean cattle may not be significantly affected by mild HS, considering that growth performance of cattle was better in hotter conditions, although some changes in blood metabolic and mineral parameters were observed.

Variations of N2O by no tillage and conventional-tillage practices under the different kinds of fertilizer applications on the cultivation of soybean in Korea

  • Anthropogenic activities have increased the concentrations of greenhouse gases, such as $CO_2$, $CH_4$, $N_2O$, HFCs, $SF_6$, and PFCs, in the atmosphere. Among others, $N_2O$ is well known as an important greenhouse gas accounting for 7.9% of the total greenhouse effect and the effect of its emission is 310 times greater than that of $CO_2$. Agricultural $N_2O$ emissions are now thought to contribute to about 60% of the global anthropogenic $N_2O$ emission, which have been increased primarily due to fertilizer N consumption and manure management. Therefore, the reduction of $N_2O$ emissions in agriculture is being required. This study was conducted to determine the variation of $N_2O$ emissions by no-tillage (NT) and conventional tillage (CT) practices in the cultivation of soybean from the sandy loam soils under the different kinds of fertilizer treatments June through September 2016 in Cheong-ju, Republic of Korea. An experimental plot, located in the temperate climate zone, was composed of two main plots that were NT and CT, and were divided into four plots, respectively, in accordance with types of fertilizers (chemical fertilizer, liquid pig manure, hairy vetch and non-fertilizer). Among all the treatments, $N_2O$ emission was the highest in August and the lowest in June. When $N_2O$ emissions were evaluated during the growing season (June to September) in all fertilizer treatments, NT with hairy vetch treatment emitted the highest $N_2O$ emission in August, whereas, $N_2O$ emissions was the lowest in NT with non-fertilizer treatment in June, respectively (p = 0.05). Based on the cumulative amount of $N_2O$ emissions during the growing season of soybean, NT had lower $N_2O$ emission than CT by 0.01 - 0.02 kg $N_2O$, although NT had higher $N_2O$ emission than CT by 0.03 kg $N_2O$ in only the chemical fertilizer treatments. As a result, it seems that the applications of liquid pig manure and hairy vetch rather than chemical fertilizer could decrease the $N_2O$ emission in NT, compared to CT.

Phylogeny of Marine Yeasts Isolated from Coastal Seawater in the East Sea of Korea (동해 연안해역에서 분리한 해양효모의 분자계통학적 해석)

  • BACKGROUND: Yeasts are used in a variety of industries. However, most industries are biased toward Saccharomyces cerevisiae ; so we sought to explore non-conventional yeasts (NCY). This study aimed to isolate yeasts from seawater collected from the East Sea of Korea and to analyze the NCY. METHODS AND RESULTS: We first collected seawater and performed pure isolation using four kinds of medium (GPY, DOB + CSM, DG18, and SCG). In total, 314 strains and 17 genera were isolated by ITS sequencing, including Aureobasidum pullulans (236 strains), Cryptococcus (19 strains), Cystobasidium (18 strains), and Rhodotorula (9 strains). Upon in-depth analysis, A. pullulans, the most dominant genus (236 strains), was divided into Group II (147 strains), Unknown I (8 strains), and Unknown II (49 strains). CONCLUSION: In this study, a total of 314 strains were isolated from seawater; many of these yeasts have been found and reported in seawater previously. In-depth analysis of A. pullulans, showed the dominance of Group I (21 strains) and Group II (147 strains) We also discovered Unknown I (8 strains) and Unknown II (49 strains), which have not been reported previously.

Evaluation of Organic Sudangrass for Feed Value, Silage Quality and Palatability in Korea (유기 수단그라스의 사료가치, 사일리지 품질 및 기호성 평가)

  • The main nutritional problem of sorghum${\times}$sudangrass hybrid (Sorghum bicolor${\times}$Sorghum sudanese) silage is low quality and palatability. This experiment was conducted to evaluate whether organic com grain and crushed rice addition of sorghum${\times}$sudangrass hybrid silage increases forage quality of the silage. The sorghum${\times}$sudangrass hybrid silages with added com grain and crushed rice were similar to conventional com silage in moisture content. However organic sorghum${\times}$sudangrass hybrid silage had low pH values. Silage added with gain and byproduct had higher crude ash, acid detergent fiber (ADF) and neutral detergent fiber (NDF) contents than control com silages, while its non-fiber carbohydrate (NFC) and total digestible nutrients (TDN) showed the opposite results. Lactic acid, butyric acid and lactic percentage of total organic acid (L/T) of sorghum${\times}$sudangrass hybrid silages were higher than those of com silage, but acetic acid was higher than control. In vitro dry matter digestibility (IVDMD) of com silage was higher than other sorghum silages. Feed intake of sorghum${\times}$sudangrass hybrid silage added with crushed rice was highest among silages. Therefore, these data indicate that crushed rice and com grain added sorghum${\times}$sudangrass hybrid silage could be recommended as the most effective treatment for increasing silage quality and palatability.

Evaluation of benzene residue in edible oils using Fourier transform infrared (FTIR) spectroscopy

  • The use of food grade hexane (FGH) for edible oil extraction is responsible for the presence of benzene in the crude oil. Benzene is a Group 1 carcinogen and could pose a serious threat to the health of consumer. However, its detection still depends on classical methods using chromatography which requires a rapid non-destructive detection method. Hence, the aim of this study was to investigate the feasibility of using Fourier transform infrared (FTIR) spectroscopy combined with multivariate analysis to detect and quantify the benzene residue in edible oil (sesame and cottonseed oil). Oil samples were adulterated with varying quantities of benzene, and their FTIR spectra were acquired with an attenuated total reflectance (ATR) method. Optimal variables for a partial least-squares regression (PLSR) model were selected using the variable importance in projection (VIP) and the selectivity ratio (SR) methods. The developed PLS models with whole variables and the VIP- and SR-selected variables were validated against an independent data set which resulted in $R^2$ values of 0.95, 0.96, and 0.95 and standard error of prediction (SEP) values of 38.5, 33.7, and 41.7 mg/L, respectively. The proposed technique of FTIR combined with multivariate analysis and variable selection methods can detect benzene residuals in edible oils with the advantages of being fast and simple and thus, can replace the conventional methods used for the same purpose.

Nutritional Improvement of Masoor(Lens esculenta) by Supplementation with Different Kinds of Meat

  • The study was conducted to determine the nutiritional value of Masoor (Lens esculenta) in raw and cooked forms. Supplement value of various types of meat i.e. poultry, mutton and beef at 10, 15, and 20 percent levels for dite containing cooked Masoor was also assessed. Nutritional value of Masoor was determined by chemical analysis as well as through rat assay. Masoor contained an average of 23.18 percent protein and less than two percent fibre. Conventional method of cooking resulted in about 2 per cent increase in Masoor protein. Masoor had 0.83 percent of lysine and cooking destroyed 18 percent of it. Other amino acids in Masoor also showed losses on cooking. Protein efficiency ratio (PER) of diets containing raw Masoor was 1.49 and was reduced to 1.44 by cooking. Cooking of Masoor did not alter true digestibility (TD) percentage. However, net protein utilization (NPU) was improved from 44.60 in raw to 47.77 in cooked. Diets containing cooked Masoor and supplemented with different types of meat significantly improved PER (1.45 to 1.65), TD 76.03 to 87.84 percent and NPU 42.84 to 50.72 percent over non supplemented diets. 20 percent level of supplemented meat showed comparatively better results than other levels in case of improvement in PER, TD and NPU.
