Screening of the antiparasitic activity of gold nanoparticles on hydatid cysts protoscolices in vitro

  • Many scolicidal agents have been used to destroy fertile protoscolices, but these scolicidal agents have side effects, highlighting the need for research on effective and non-toxic replacement scolicidal agents. Gold nanoparticles (AuNPs) are biocompatible and non-toxic. The current study examined the effects of AuNPs in killing the protoscolices of Echinococcus granulosus in vitro using eosin staining. The protoscolices were treated with 0.2, 0.4, 0.8, or 1.0 mg/mL of AuNPs for 15, 30, 45, or 60 minutes. A concentration of 1.0 mg/mL was the most efficient in killing the protoscolices after 60 minutes exposure, reaching 96%, followed by 0.8 mg/mL (84.5%), whereas 0.4 and 0.2 mg/mL of AuNPs achieved a death rate of 76.8% and 68.5%, respectively. The loss of the protoscolices was lower at shorter exposure times with the same concentration of AuNPs and increased as the AuNP concentration was increased at the same exposure time. Significant differences were found between the different groups compared to the control group.

Risk Assessment of Airborne Toxic Metals in Thejon Industrial Complex (대전공단지역의 대기중 독성금속에 대한 위해도 평가)

  • The research centers on the concentration profile and risk assessment of toxic metals for ambient air in Taejon industrial complex. Airborne concentrations of each toxic metal for risk assessment were obtained from 2-year sampling by high volume air sampler and analysis by ICP-MS and ICP-AES in the complex. The long-term arithmetic mean of human carcinogen, arsenic, hexavalent chromium and nickel subsulfide was 5.53, 2.16 and $3.46ng/m^3$ while the mean of probable human carcinogen, beryllium, cadmium and lead was 0.08, 2.35, $293.29ng/m^3$, respectively. And the long-term arithmetic mean concentration of non-carcinogenic metal, manganese was $55.91ng/m^3$. The point risk estimate for the inhalation of carcinogenic metals was $3.6{\times}10^{-5}$, which was higher than a risk standard of $10^{-5}$. About 75% of the cancer risk was to the inhalation of human carcinogen, arsenic. Thus, it is necessary to properly manage arsenic risk in Taejon industrial complex. The point hazard index by the inhalation of manganese was 1.1. Therefore, an investigation into Taejon industrial complex is needed to obtain more fine long-term concentration data for airborne non-carcinogenic metals including manganese.

Is N-acetylcysteine Treatment Based on Ingestion Amount Valid in Acute Acetaminophen Overdose Patients? (급성 Acetaminophen 중독시 복용량에 의한 N-Acetylcysteine의 사용은 적절한가?)

  • Purpose: In many Korean hospitals, serum acetaminophen concentrations in cases of overdose cannot be measured initially because of inadequate laboratory facilities. Under these circumstances, physicians base the administration of the antidote, N-acetylcysteine, on ingestion amounts as determined by initial history taking. We therefore examined the correlated between ingested amounts and serum acetaminophen concentrations. Methods: Medical records were reviewed retrospectively for patients who presented to the ED with acetaminophen overdose between January 2002 and March 2006. Fifty-nine patients were recruited and sixteen patients were excluded. The forty-three remaining patients were placed into either the high-risk or low-risk group based on their ingested amount (140 mg/kg), and were separately categorized into the toxic or non-toxic group based on their serum acetaminophen concentrations, according to the Rurnack-Matthew nomogram. Results: Ten patients (83.3%) among twelve in the high-risk group were found to have non-toxic serum concentrations, and just one patient (3.2%) among thirty-one in the low-risk group fell into the toxic group based on their serum concentrations. The sensitivity and specificity of risk stratification of the ingested amount as a predictor of intoxication requiring antidote therapy were 66.7% and 75.0%, respectively. Conclusion: This study suggests that the therapeutic decision for acetaminophen overdose should not be based solely on ingested amount only, but requires assessment of acetaminophen concentration.

A HISTOMORPHOMETRIC STUDY OF BONE APPOSITION TO NEWLY DEVELOPED TI-BASED ALLOYS IN RABBIT BONE (가토의 경골에 이식된 새로운 티타늄계 합금 주위의 골형성에 관한 형태학적 연구)

  • Research advances in dental implantology have led to the development of several different types of materials and it is anticipated that continued research will lead to advanced dental implant materials. Currently used pure titanium has relatively low hardness and strength which may limit its ability to resist functional loads as a dental implant. Ti-6Al-4V also has potential problems such as corrosion resistance. osseointegration properties and neurologic disorder due to aluminium and vanadium, known as highly toxic elements, contained in Ti-6Al-4V. Newly developed titanium based alloys(Ti-20Zr-3Nb-3Ta-0.2Pd-1In, Ti-20Zr-3Nb-3Ta-0.2Pd) which do not contain toxic metallic components were designed by the Korea Institute of Science and Technology (KIST) with alloy design techniques using Zr, Nb, Ta, Pd, and In which are known as non-toxic elements. Biocompatibility and osseointegration properties of these newly designed alloys were evaluated after implantation in rabbit femur for 3 months. The conclusions were as follows : 1. Mechanical properties of the new designed Ti based alloys(Ti-20Zr-3Nb-3Ta-0.2Pd-1In, Ti-20Zr-3Nb-3Ta-0.2Pd) demonstrated close hardness and tensile strength values to Ti-6Al-4V. 2. New desinged experimental alloys showed stable corrosion resistance similar to the pure Ti but better than Ti-6Al-4V. However, the corrosion rate was higher for the new alloys. 3. Cell culture test showed that the new alloys have similar cell response compared with pure Ti and Ti-6Al-4V with no cell adverse reaction. 4. New designed alloys showed similar bone-metal contact ratio and osseointegration properties compared to pure Ti and Ti-6Al-4V after 3 months implantation in rabbit femur. 5. Four different surface treatments of the metals did not show any statistical difference of the cell growth and bone-metal contact ratio.

Biological Parameters for Evaluating the Toxic Potency of Petroleum Ether Extract of Wattakaka volubilis in Wistar Female Rats

  • Objectives: The present study investigated the toxic properties of petroleum ether extract of Wattakaka (W.) volubilis in Wistar female rats. Methods: An in vitro brine shrimp lethality bioassay was studied in A. Salina nauplii, and the lethality concentrations were assessed for petroleum ether extract of W. volubilis. A water soluble portion of the test extract was used in different concentrations from $100-1000{\mu}g/mL$ of 1 mg/mL stock solution. A 24-hours incubation with a 1-mL aliquot in 50 mL of aerated sea water was considered to calculate the percentage rate of dead nauplii with test extract administration against a potassium-dichromate positive control. The acute and the sub-acute toxicities of petroleum ether extract of W. volubilis were evaluated orally by using gavage in female Wistar rats. Food and water intake, body weight, general behavioral changes and mortality of animals were noted. Toxicity or death was evaluated following the administration of petroleum ether extract for 28 consecutive days in the female rats. Serum biochemical parameters, such as alanine aminotransferase (ALT), alkaline phosphatase (ALP), bilirubin, total cholesterol, triglyceride, total protein, glucose, urea, creatinine, sodium, potassium and ${\alpha}$-amylase levels, were measured in the toxicity evaluations. Pathological changes in isolated organs, such as the liver, kidneys, and pancreas, were also examined using hematoxylin and eosin dye fixation after the end of the test extract's administration. Results: The results of the brine-shrimp assay indicate that the evaluated concentrations of petroleum ether extract of W. volubilis were found to be non-toxic. In the acute and the sub-acute toxicity evaluations, no significant differences were observed between the control animals and the animals treated with extract of W. volubilis. No abnormal histological changes were observed in any of the animal groups treated with petroleum ether extract of W. volubilis. Conclusion: These results suggest that petroleum ether extract of W. volubilis has a non-toxic effect in Wistar female rats.

  • Environmental toxicants such as toxic metals can alter epigenetic regulatory features such as DNA methylation, histone modification, and non-coding RNA expression. Heavy metals influence gene expression by epigenetic mechanisms and by directly binding to various metal response elements in the target gene promoters. Given the role of epigenetic alterations in regulating genes, there is potential for the integration of toxic metal-induced epigenetic alterations as informative factors in the risk assessment process. Here, we focus on recent advances in understanding epigenetic changes, gene expression, and biological effects induced by toxic metals.

Characterization of Toxic Pollutants in Ash and Flue Gas from Gasification Incinerator of Waste Tires (폐타이어 건류 소각에서 발생되는 재와 배기 가스에서의 독성 오염 물질의 정량)

  • The problem of disposing of huge quantities of used tires is of growing concern to every country. As an economical solid waste management, a gasification followed by incineration process was applied to scrap tires to recover heat and to reduce waste volume for final landfill disposal. The gasification temperature, combustible and non-combustible gasified products and possibly produced air pollutants were predicted by changing equivalent mole ratios of carbon to oxygen by a chemical equilibrium model. For a risk assessment of ash toxic pollutants including heavy metals and toxic organics were thoroughly analyzed. Gasification bottom ash contained much more toxic organic compounds than fly ash, whereas fly ash contained higher concentration of heavy metals such as Pb and Cd. Pretreatment or secure landfill technology is suggested for a safe management of ash produced from the gasification incinerators.

Toxicity Reduction of Wastewater from a Rubber Products Manufacturing Factory by Gamma-ray Treatment (감마선 처리를 이용한 고무공장 폐수의 생물독성 저감)

  • Both raw wastewater and effluent from a rubber products manufacturing factory were found to be toxic to Daphnia magna though the effluent satisfied current water quality standards. Thus, in order to reduce toxicity, advanced oxidation processes (AOPs) such as gamma-ray (${\gamma}-ray$) treatment and ozonation ($O_3$) were applied. A combined ${\gamma}-rays/O_3$ treatment at 20 kGy after coagulation significantly reduced toxicity of raw wastewater, changing 48-h toxic unit (TU) value from 201.21 to 23.92. However, toxicity of treated water was higher than that of effluent (TU = 12.15). This shows limitation of gamma-ray treatment to remove toxicity of raw wastewater. In case of effluent, the combined ${\gamma}-rays/O_3$ treatment at 20 kGy efficiently decomposed toxic compounds down to non toxic level. This work strongly supports the necessity of toxicity reduction evaluation as well as toxicity-based effluent management.

Data intercomparison and determination of toxic and trace elements in Algae using Instrumental Neutron Activation Analysis (중성자방사화분석에 의한 Algae중의 독성미량원소의 정량 및 실험실간 비교검증)

  • For the non-destructive multi-elemental analysis of environmental and biological materials, instrumental neutron activation analysis (INAA) was applied for the determination of toxic and trace elements in a set of three Algae samples provided by the International Atomic Energy Agency (IAEA). The analytical quality control was evaluated by comparing the analytical results of two standard reference materials of the National Institute of Standards and Technology (NIST); Oyster Tissue (SRM 1566a) and Citrus Leaves (SRM 1572). According to given analytical procedure, the concentration of 15-25 elements including spiked elements such as As, Cd, Cr and Hg in Algae samples were determined. To identify and validate these results, a data intercomparison program using more than 35 analytical methods in 150 laboratories was carried out and the estimated statistical data are summarized. Result of INAA is favorable, therefore, it is illustrated that can be applied for routine analysis of essential and toxic elements in algae samples as well as analytical quality assurance.

Toxic Elements in Edible Mollusks from Igneada Coasts of the Black Sea, Turkey

  • In this present work, the amounts of toxic elements were determined in the soft tissues of mollusks (Mytilus galloprovincialis and Rapana venosa) collected from Igneada shores of the Black Sea where suggested as Marine Protection Area (MPA). M. galloprovincialis accumulated the highest amounts of Cd in winter, while the highest amounts of Cd and Hg were detected in R. venosa in winter and autumn. The concentrations of toxic elements found in the soft tissues of mollusks varied for Cd: 0.07-0.14, Hg: 0.03-0.44 and Pb: 0.09-0.21 mg/g dry wt. The estimated levels of all non-essential metals in the present work were lower than the limits permitted by European Community Regulation (EU) and the Turkish Food Codex (TFC). The estimated weekly intakes (EWI) and daily intakes (EDI) of all the through consumption of these seafood by Turkish people in the Igneada coasts of the Black Sea were quite below the permissible tolerable weekly/daily intakes for 70 kg person (PTWI / PTDI) set by FAO/WHO. As results, it can be concluded that no hazard effects on people health would be raised at present from the consumption of these mollusks' species.
