A Prolyl Endopeptidase-lnhibiting Antioxidant from Phyllanthus ussurensis

  • A prolyl endopeptidase inhibitor was isolated from the ethyl acetate soluble fraction of Phyllanthus ussurensis. The active compound was identified as an ellagitannin, corilagin. It was shown to non-competitively inhibit prolyl endopeptidase (PEP) with the $IC_{50}$ value of $1.17 \times $10^{-6}\mu$M. The Ki value was $6.70 \times 10^{-7}$ M. Corilagin was less inhibitory to other serine proteases such as chymotrypsin, trypsin, and elastase, indicating that it was relatively a specific inhibitor of PEP. Corilagin also effectively inhibited reactive oxygen species such as hydroxide and superoxide anion radical, hydrogen peroxide, and DPPH. Especially, corilagin showed potent scavel1ging activity on the superoxide anion radical in the ESR method ($IC_{50} =3.79 \times 10^{-6}$M) as well as xanthine oxidase system.

  • The purpose of this study was to investigate lower-limb joint torque of the two groups as it changed by somatosensory stimulation during the descent down three stairs of different heights and to describe the difference between the two groups, which are young people group and elderly people group. Subjects of each groups climbed down a stair at four stimulation conditions, which are non-stimulation, tibialis anterior tendon stimulation, achilles tendon stimulation, tibialis anterior - achilles tendon stimulation. Motion capture data were collected using 3D optoelectric motion tracking system that utilizes active infrared LEDs, near infrared sensor and force plate. The obtained motion capture data was used to build 3D computer simulation model. The results show that lower-limb joint torque of the two groups changed with somatosensory stimulation as they descended the stairs and the joint torque of the two groups differed from each other.

  • The theory for a new electromagnetically biased diskless combined radial and axial magnetic bearing is developed. A typical magnetic bearing system is composed of two radial magnetic bearings and an axial magnetic bearing. The axial magnetic bearing with a large axial disk usually limits rotor dynamic performance and makes assembling and disassembling difficult for maintenance work. This paper proposes a novel electromagnet biased integrated radial-axial magnetic bearing without axial disk. This integrated magnetic bearing uses two axial coils to provide the bias flux to the radial and axial air gaps of the combined bearing. The axial magnetic bearing unit in this combined magnetic bearing utilizes reluctance forces developed in the non-uniform air gaps such that the axial disk can be removed from the bearing unit. The 4-pole homopolar type radial magnetic bearing unit is also designed and analyzed. Three dimensional finite element model for the bearing is also developed and analyzed to illustrate the diskless combined magnetic bearing.

Isolation of Flavonol Rhamnosides from Loranthus tanakae and Cytotoxic Effect of Them on Human Tumor Cell Lines

  • Loranthus tanakae Fr. et Sav. (Loranthaceae) is a species of mistletoe, a semiparasitic plant growing on the branches of Quercus and Betula species as host trees. In our ongoing search for bioactive compounds from endemic species in Korea, we have investigated to isolate the chemical constituents responsible for the antitumor effect of the MeOH extract of L. tanakae. The ethyl acetate soluble part of the MeOH extract demonstrated a marginal inhibition on the proliferation of the tumor cell lines such as A549 (non small cell lung), SK-OV-3 (ovary), SK-MEL-2 (melanoma), XF498 (central nerve system), and HCT-15 (colon) in vitro. Thus, the activity-guided isolation procedure upon the ethyl acetate soluble part of the extract has been carried out and finally four flavonoid rhamnopyranosides (1-4) were isolated as active principle. The structures of 1-4 were elucidated by the physicochemical and spectral data as rhamnetin 3-O-$\alpha$-L-rhamnoside (1), quercetin 3-O-$\alpha$-L-rhamnoside (2), rhamnocitrin 3-O-$\alpha$rhamnoside (3), and kaempferol 3-O-$\alpha$-L-rhamnoside (4).

Function of Nitric Oxide in Activation-Induced Cell Death of T Lymphocytes

  • Using a murine T cell hybridoma, activation-induced cell death (AICD) was studied. As an in vitro model system for the AICD, 1 cell hybridoma expressing TCR/CD3 complex was incubated onto the immobilized purified anti-CD3 antibody. The immobilized anti-CD3 antibody induced AICD effectively up to 40%. At 1-100 $\mu$M range of SNP, an exogenous source of nitric oxide (NO), the cell proliferation was not affected, but at 1 mM SNP, cell proliferation was significantly reduced. The AICD of T cell hybridoma was inhibited by exogenous NO at non-cytotoxic concentration, In the cells undergoing AICD, the expressions of caspase-3 and FasL were detected, but not iNOS. Similar result was recognized in the apoptosis induced by dexamethasone, an apoptosis-inducing agent. However, the conversion from the inactive form of caspase-3 (32 kDa) to the active form (17 kDa) was significantly reduced in the cells in AICD induced by anti-CD3 antibody, With the result of increased PARP cleavage in the cells, we propose that another PARP cleavage pathway not involving caspase-3 may function in the anti-CD3 antibody induced AICD in the T cell hybridoma.

Validation of soy isoflavone intake and its health effects: a review of the development of exposure biomarkers

  • BACKGROUND/OBJECTIVES: It is difficult to consistently demonstrate the health effects of soy isoflavones owing to the multitude of factors contributing to their bioavailability. To accurately verify these health effects, dietary isoflavone intake should be measured using a biologically active dose rather than an intake dose. This concept has been expanded to the development of new exposure biomarkers in nutrition research. This review aims to provide an overview of the development of exposure biomarkers and suggest a novel research strategy for identifying the health effects of soy isoflavone intake. MATERIALS/METHODS: We cover recent studies on the health effects of soy isoflavones focusing on isoflavone metabolites as exposure biomarkers. RESULTS: Compared to non-fermented soy foods, fermented soy foods cause an increased concentration of isoflavones in the biofluid immediately following ingestion. The correlation between exposure biomarkers in blood and urine and the food frequency questionnaire was slightly lower than that of corresponding 24-h dietary recalls. Urinary and blood isoflavone levels did not show a consistent association with chronic disease and cancer risk. CONCLUSION: It is crucial to understand the variable bioavailabilities of soy isoflavones, which may affect evaluations of soy isoflavone intake in health and disease. Further studies on the development of valid exposure biomarkers are needed to thoroughly investigate the health effects of isoflavone.

  • At the present stage of the world economic development, a new economic system is being formed, in which non-economic values, in particular environmental and social parameters, have become widespread. A new vision of economic activity is being formed, which acquires the qualities of Smart-economy. The purpose of this paper is reveal the features of managing the development of smart cities as specific entities of the Smart-economy. New functions of economic entities are formed within the framework of the Smart-economy concept, while their role and weight in the localities' activity or formation have changed. Determining that the key trends in the Smart-economy development are such as digitalization, greening, socialization, institutionalization, and urbanization, this is necessary to note that all these trends are most active in the formation of urban ecosystems. These trends are determined by the general population growth and the urban population growth, which requires considerable attention to planning each city's development itself. Such planning could ensure the comfort of living for all its inhabitants, quality, safe, and modern life. The Smart-city's key elements and the intellectualized approach implementation planes to the decision of these or those tasks are definedIt is determined that a new ecosystem of governance is being formed.

A New Method for Coronal Force-Free Field Computation That Exactly Implements the Boundary Normal Current Density Condition

  • Previously we developed a method of coronal force-free field construction using vector potentials. In this method, the boundary normal component of the vector potential should be adjusted at every iteration step to implement the boundary normal current density, which is provided by observations. The method was a variational method in the sense that the excessive kinetic energy is removed from the system at every iteration step. The boundary condition imposing the normal current density, however, is not compatible with the variational procedure seeking for the minimum energy state, which is employed by most force-free field solvers currently being used. To resolve this problem, we have developed a totally new method of force-free field construction. Our new method uses a unique magnetic field description using two scalar functions. Our procedure is non-variational and can impose the boundary normal current density exactly. We have tested the new force-free solver for standard Low & Lou fields and Titov-Demoulin flux ropes. Our code excels others in both examples, especially in Titov-Demoulin flux ropes, for which most codes available now yield poor results. Application to a real active region will also be presented.

  • In this study aims to provide operational guidelines based on risk assessment and safety evaluation in the operation of non-traditional oil production plants, serving as alternative resources in comparison to conventional oils within the construction sector. Presently, operational facilities within these plants are active. However, integrating comprehensive management through sectional and device-specific diagnostic and evaluation systems in similar platform-based operational management systems proves to be a challenging reality. Consequently, this research introduces an evaluation methodology tailored for an operation-centric management system using an integrated management approach, and evaluates its practical applicability within the construction field.

  • Objective : This study aims to investigate the concept of mental health literacy, its evolution, and its significance in modern healthcare practices. Methods : This paper reviews the evolution of mental health literacy, examining pertinent studies both within the country and internationally. It also addresses the limitations and challenges associated with mental health diagnoses. Results : Findings from various studies indicate that public awareness and recognition of mental health issues are generally inadequate. Identified causes of mental health conditions by study participants point to personal and societal influences, while biological factors are frequently neglected. In South Korea, there is a notable preference for non-professional support over psychiatric interventions. Conclusion : This study highlights the growing importance of mental health literacy in today's society, emphasizing the need for accurate understanding of mental health issues and effective treatment approaches. Similar to physical health literacy, mental health literacy should be considered a basic human right. This perspective marks a shift from a knowledge monopoly held by experts to the inclusion and active involvement of 'people with lived experience' in the mental health service system.
