  • This experiment was carried out to investigate the effects of liming, manure application, and inoculation with Rhizobium on the early growth, nodulation, seedling establishment, winter survival, and forage yield and quality of alfalfa(Medicago sativa L.) at the Experimental Livestock Farm, College of Agriculture, Seoul National University, Suweon in 1985 and 1986. The results obtained are as follows: 1. Seedling vigor and nodule formation of alfalfa at early stage were the best in the treatments of liminginoculation( L1) and liming-manure-inoculation(LM1). 2. Seedling establishment was not significantly affected by the treatments of liming, manure application, nodulation and the combination of these. 3. The LI and LMI treatments showed the highest winter survival of 94.3 and 83.5%, respectively, but the lowest winter su.rvival of 55.9 and 58.5% was found in no treatment(None) and inoculation( I ), respectively. 4. No differences in crude protein(CP1, acid detergent fiber(ADF) and neutral detergent fiber(NDF) contents were found among the treatments. 5. There were significant differences in the total dry matter yield of alfalfa among the treatments. The total dry matter yields of 10,618, 11,830, 11,855 and 13,456 kg/ha from the treatments of L, LM, LI and LMI, respectively, were significantly higher than those of None, M and I treatments. 6. Based on the results of the experiment, it appears that the dry matter yield of alfalfa could be enhanced by the treatments of liming with Rhizobium inoculation and also liming, manure application and Rhizobium inoculation as a practical method.

Effect of chitosan on bone matrix expression and mineralization in primary rat calvarial cell (키토산이 백서 태자 두개관세포의 세포외기질 발현과 석회화에 미치는 영향)

  • Periodontal therapy has dealt primarily with attempts at arresting progression of disease, however, more recent techniques have focused on regenerating the periodontal ligament having the capacity to regenerate the periodontium. The effect of chitosan, a carbohydrate biopolymer extracted from chitin, on periodontal ligament regeneration is of particular interest. The purpose of this study was to evaluate the effect of chitosan on the expression of extracellular matrix proteins in primary rat calvarial cells in Vitro. In the control group, cells was cultured with BGjb media. In the experimental groups, cells were cultured with chitosan in concentration of 0.01, 0.1, 1.0 and 2.0 mg/ml. Then each group was characterized by examining alkaline phosphatase activity at 3 and 7 days, and the ability to produce mineralized nodules of rat calvarial cells at 14 and 21 days. Synthesis of type I collagen (COL-I), osteocalcin (OCN), bone sialoprotein (BSP) was evaluated by RT-PCR at 14 days. The results were as follows: 1. Alkaline phosphatase activity was significantly higher in the concentration of chitosan 0.01mg/ml, 0.1mg/ml and 1.0mg/ml compared to control (p<0.05). 2. The percentage of mineralized bone nodule was more in the concentration of chitosan 0.1mg/ml and 1.0mg/ml than the control. 3. At 14 day culture, the expression of OCN was increased by chitosan in concentration of 1.0 mg/ml and 2.0 mg/ml. These results suggested that chitosan in concentration of 0.1 and 1,0 mg/ml stimulate the extracellular matrix of primary rat calvarial cells and may facilitate the formation of bone.

Effect of Culture Method and Medium Composition on Shoot Regeneration from Sporophytes of Cyrtomium caryotideum var. coreanum Nakai. (참쇠고비(Cyrtomium caryotideum) 포자체로부터의 식물체 재생에 미치는 배양방법 및 배지구성물질의 영향)

  • This study was conducted to investigate the efficient propagation method of Cyrtomium caryoptideum var. coreanum by sporophyte culture. The influence of origin of explant sources (rhizome, blade, or stipe) and homogenization of culture materials on shoot regeneration were investigated. As a result, only rhizome explant exhibited the organogenic capacity and the shoot regeneration was promoted by homogenization of culture material. Vigorous and excellent growth of multiple shoots was induced on the half-strength of inorganic salts containing MS medium. It was appeared that optimum nitrogen content of shoot regeneration was half-strength of nitrogen containing MS medium (30mM) and optimum sucrose concentration was 1%. Addition of $NaH_2PO_4$ to culture medium generally enhanced shoot multiplication and promoted growth of the regenerants. The organogenic capacity of homogenized rhizomes was especially promoted on medium supplemented with $5{\mu}M$ kinetin plus $5{\mu}M$ IBA. The incorporation of $0.1\sim0.2%$ activated charcoal on medium supplemented with growth regulators prevented the formation of multiple bud primordia - nodule-like bud clusters and improved the normal morphogenesis of sporophytes.

Hepatoprotective Effect of Coccinia indica Against $CCl_4$ Induced Hepatotoxicity

  • The hepatoprotective effect of the ethanolic extract of Coccinia indica fruits in rats treated with carbon tetrachloride. In hepatotoxic rats, liver damage was studied by assessing parameters such as aspartate aminotransferase (AST), alanine aminotransferase (AlT), alkaline phosphatase (AIP) and gamma glutamyl transpeptidase (GGT) in serum, and concentrations of total proteins, total lipids, phospholipids, triglycerides and cholesterol in both serum and liver. The effect of co-administration of ethanolic extract on the above parameters was further investigated. Histopathological study of the liver in experimental animals was also undertaken. Hepatic damage as evidenced by a rise in the levels of AST, AIT, AIP and GGT in serum, and also changes observed in other biochemical parameters In serum and liver showed a tendency to attain near normalcy in animals co-administered with the extract. The normal values for AST (IU/L), AIP (IU/I), protein (g/100 ml) and total lipids (mg/100 ml) in serum (i.e.,20.24, 70.04, 5.72 and 135.54 respectively) were found to alter towards values 32.61, 127.11, 3.83 and 265.91 in hepatotoxic rats. These parameters Attained near normal values (I.e.,22.82, 79.30, 5.22 and 151.24 for AST, AIP protein and total lipids respectively) in ethanolic extract co-administered rats. Profound steatosis, ballooning degeneration and nodule formation observed in the hepatic architecture of $CCl_4$ treated rats were found to acquire near-normalcy in drug co-administered rats, thus corroborating the biochemical observations. Thus the study substantiates the hepatoprotective potential of ethanolic extract of Coccinia indica fruits.

Response of Nodulation and Leaf Nitrate Reductase Activity of Alfalfa to Exogeneous Nitrate Supply (질산태 질소 공급이 알팔파의 뿌리혹 형성 및 엽중 Nitrate Reductase 활성에 미치는 영향)

  • A full understanding of the interdependence of leaf nitrate (($No_3$ ̄) metabolism and symbiotic nitrogen($N_2$) fixation in legume crops is needed to help maximize the use of both N sources as well as to improve forage quality through the inhibition of leaf nitrate accumulation. The present work examines the effects of added nitrate, the level of which are 0,2,4,8 and 12mM, on the nodule formation and leaf nitrate utilization and on the possibility of inducing nitrate-toxicity to livestocks in two alfalfa varieties, ' Vernal ' of grazing type and ' Victoria ' of hay type. Higher level of exogeneous nitrate resulted in the increased above-ground dry weight. Nodulation was inhibited severely when more than 8mM NO$_3$ ̄ was supplied to alfalfa plants, and leaf nitrate reductase reached a maximunm at 4mM nitrate supply. The $V_{max}$of nitrate reductase in leaves of Vernal was similar to that of Victoria, whereas the $K_m$ of Vernal was higher than that of Victoria. High accumulation of leaf nitrate, $4{\times}10^{-5}$ g/g leaf fresh weight, was shown at 12mM nitrate supply, which was thought to be not enough to induce nitrate-toxicity to livestocks.icity to livestocks.

  • Some nodules occur in the Milyang pyrophyllite deposit which are hydrothermal alteration products by Late Cretaceous andesitic tuff. These nodules are divided into two types on the basis of mineral assemblages; diaspore and pyrophyllite nodules. The diaspore nodules consist mainly of diaspore, kaolinite, pyrophyllite and pyrite with a small amounts of wavellite and tourmaline. They are light purplish grey in color, ellipsoid in shape and range 1 cm to 15 cm in size. A small or large diffuse band exists in some nodules. The platy coarse-grained diaspore is intergrown with the fine-aggregated kaolinite in the central part of the nodule. It appears that the grain size become fine from center to margin. The pyrophyllite nodules, which have the same shape with diaspore nodules, consist dominantly of pyrophyllite accompanied by small amounts of quartz, kaolinite, svanbergite, wavellite, tourmaline and apatite. Chemical compositions of alteration zones and nodules show that the wall rock alteration involved mainly the removal of large quantities of silica and alkalies and small quantities of Ca, Mg and Fe. The sharp increase in the Al content of the nodules is the result of residual concentration of alumina by the leaching of the mobile components. The pyrophyllite nodules were formed in the fluid saturated with quartz as ${\mu}_{HK_{-1}}$ and ${\mu}_{H_{2}O}$ increase. Under this condition, the pyrophyllite-kaolinite-quartz assemblage was stable. Diaspores formed from pyrophyllites in the fluid undersaturated with quartz as ${\mu}_{H_{2}O}$ increases (decreasing temperature). Under this condition, diaspore-pyrophyllite-kaolinite assemblage become stable. The formation temperature of the nodules on the basis of mineral assemblage is estimated as $275{\sim}340^{\circ}C$.

AN EXPERIMENTAL STUDY FOR ESTABLISHMENT OF ORTHOTOPIC SALIVARY TUMOR MODELS IN MICE (마우스에서 타액선암 동위종양 모델 제작을 위한 실험적 연구)

  • Purpose: Adenoid cystic carcinoma (ACC) is a relatively rare tumor that arises in glandular tissues of the head and neck region and sometimes has a protracted clinical course with perineural invasion and delayed onset of distant lung metastasis. Treatment failure of salivary ACC is most often associated with perineural and hematogenous tumor spread. However, very little has been known about the cellular and molecular mechanisms of perineural invasion and hematogenous distant metastasis of parotid ACC. This study was designed to develop an orthotopic tumor model of parotid adenoid cystic carcinoma in athymic nude mice. Experimental Design: A melanoma cell line was injected into the parotid gland of athymic mice to determine whether such implantation was technically feasible. A parotid ACC cell line was then injected into the parotid gland or the subcutaneous tissue of athymic mice at various concentrations of tumor cells, and the mice were thereafter followed for development of tumor nodule. The tumors were examined histopathologically for perineural invasion or regional or distant lung metastasis. We used an oral squmous cell carcinoma cell line as control. Results: Implantation of tumor(melanoma) cell suspension into the parotid gland of nude mice was technically feasible and resulted in the formation of parotid tumors. A parotid ACC cell line, ACC3 showed no significantly higher tumorigenicity, but showed significantly higher lung metastatic potential in the parotid gland than in the subcutis. In contrast, mucosal squmous cell carcinoma cell line doesn’t show significantly higher lung metastatic potential in the parotid gland than in the subcutis. The ACC tumor established in the parotid gland seemed to demonstrate perineural invasion of facial nerve, needs further study. Conclusion: An orthotopic tumor model of salivary ACC in athymic nude mice was successfully developed that closely recapitulates the clinical situations of human salivary ACC. This model should facilitate the understanding of the cellular and molecular mechanisms of tumorigenisis and metastasis of salivary ACC and aid in the development of targeted molecular therapies of salivary ACC.

TERT mRNA Expression is Up-Regulated in MCF-7 Cells and a Mouse Mammary Organ Culture (MMOC) System by Endosulfan Treatment

  • Endosulfan is one of the organochlorine pesticides, which are well-known endocrine disruptors (EDs), and it acts as an estrogen agonist. Estrogen is a group of hormones that play an important role in mammary gland function and are implicated in mammary carcinogenesis. In the present study, we studied the effects of endosulfan on nodule like alveolar lesion (NLAL) formation in mouse mammary gland development using a mouse mammary gland organ culture (MMOC) system. Although endosulfan-treated mammary glands did not form NLALs, more alveolar buds were formed in this group than in the negative control (vehicle-treated) group. In addition, telomerase reverse transcriptase (TERT) mRNA expression levels were increased in endosulfan-treated mammary glands in a dose-dependent manner. Telomerase can be activated by estrogen, therefore, we examined the effects of endosulfan on telomerase activity, and found that the telomerase activity in estrogen receptor-positive MCF-7 cells was up-regulated by endosulfan treatment. Moreover, this activation was accompanied by the up­regulation of the TERT mRNA expression. Also, transient expression assays using CAT reporter plasm ids containing various fragments of the TERT promoter showed that this imperfect palindromic estrogen-responsive element is almost certainly responsible for the transcriptional activation by endosulfan. These results may help elucidate the endocrine disrupting mechanism of endosulfan.

Marker-Exchange Mutagenesis of Pectate Lyase Gene in Rhizobium fredii (Rhizobium fredii Pectate Lyase 유전자의 Marker-Exchange 변이)

  • Rhizobium fredii USDA193 is one of the causal organism for root nodule formation in soybean (peking). Previously we cloned the pectate lyase gene (SY1) of R. fredii USDA193. The $pel^-$ mutants (SY1$\Omega$ and SY1$\Omega$1) of SY1 were obtained using the in vitro insertional omega mutagenesis of RpelB (of Rhizobium pel) and fill-in reaction of RpelE (of Rhizobium pel) gene respectively, and we constructed two mutants (R, fredii USDA193$\Omega$ and R. fredii USDA193$\Omega$1) in pectate lyase function by marker-exchange with pe1B::$\Omega$ and R. fredii USDA193 strain (rif). The pectate lyase activity of two pel- mutant of R. fredii USDA193 was determined by spectrophotometric method. However, all pectate lyase activity of these mutants was not lost upon the mutagenesis by marker-exchange. This suggests that other pectate lyase genes may be present on the plasmid or the chromosome of R. fredii. As yet we do not have evidence linking RpelB and RpelE genes of R. fredii directly to the early nodulation process.

The Effects of Deer(Cervus nippon) Antler Extracts on Differentiation of MC3T3 Cells (녹용 추출물에 의한 MC3T3세포의 분화 촉진)

  • Deer antler has been widely prescribed in Chinese and Korean pharmacology. Although there have been several reports concerning the effects of deer antler, such as anti-aging action, anti-inflammatory activity, antifungal action and regulatory activity of the level of glucose, the effect on bone has not determined yet. The purpose of this study was to examine the effect of deer antler on osteoblast differentiation. Hexane extract(CN-H) and chloroform extract(CN-C) were acquired from deer antler(Cervus nippon) and MC3T3-E1 pre-osteoblasts were cultured in the presence or absence of each extract. Osteoblast differentiation was estimated with the formation of mineralized nodules and the mRNA expression of alkaline phosphatase(ALP), osteocalcin(OC) and bone sialoprotein(BSP) which are markers of osteoblast differentiation. Non-treated group did not show mineralized nodule. CN-C or CN-H-treated group showed minerlaized nodules in 16 days. In northern blot analysis, CN-C or CN-H-treated group showed the elevated expression of ALP, BSP and OC in 16 days. These results suggest the possibility to develop deer antler as a bone regenerative agent in periodontal therapy by showing the stimulating activity of deer antler on differentiation of osteoblast.
