Changes of Cell Surface Hydrophobicity of a Serratia marcescens with Cultivation Time and Temperatures (배양온도와 시간에 따른 Serratia marcescens 표면의 소수성 성질변화)

  • S. marcescens cultured at $30^{\circ}C$ with vigorous shaking was shown to produce red-pigment, prodigiosin, in the senescent phase of growth. Also, it showed many hydrophobic characteristrics, which were tested by the adherence to noncharged surfaces of polystyrene dishes, a typical agent for the binding of hydrophobic cells and molecules. However, when the cell was cultured at $37^{\circ}C$, it no longer produced either red pigment or hydrophobic materials. Therefore, the bacteria cultured at $37^{\circ}C$ was completely washed-out from the polystyrene dishes at the copious washing step with tap water, in contrast to the cell cultured at $30^{\circ}C$ which was sticked onto the polystyrene dishes very tightly. The lipid compositions extracted from the S. marcescens cultured at $30^{\circ}C$ or $37^{\circ}C$ were very different from each other; the phospholipids, glycolipids and unidentified lipids were produced from the cell cultured at $30^{\circ}C$, whereas large amounts of serratamolide, amphipathic compound, were produced from the cell cultured at $37^{\circ}C$. The data suggest that the pronounced cell surface hydrophobicity of the S. marcescens is mediated by a combination of several surface factors that were affected by cultivation time and temperatures.

A Survey on the Actual Wearing Condition of Lab Gowns (실험 가운의 착용 실태 조사)

  • The purpose of this study was to develop functional lab gowns in the view of safety and work efficiency. As the first step of this study, we surveyed on satisfaction rates, preferences and inconvenience items in regard of lab gowns. The data was obtained from 138 graduate students specialized in science and 113 medical students. Respondents answered that the first purpose of wearing lab gown was to protect body from danger. Wearing frequency followed with ‘always’, ‘as possible as’, and then ‘sometimes’. Forty-five percentage of respondents answered that they didn't wear lab gowns often in summer because of hot weather. The contaminated or damaged parts of lab gowns followed with cuffs, abdomen, thigh, lower part, and then chest. Washing frequency followed with ‘once a month’, ‘once per six months’, and then ‘more than once a week’ There were significant differences in normal wearing frequency, wearing frequency by season and washing frequency due to major. There were no significant difference by sex and major in all questions. In regard of the protective capacity of current lab gowns, most respondents answered that ‘It's normal’(60%), ‘It's enough’(21%), so that they didn't show much dissatisfaction. Forty-three percentage of respondents were not satisfied with cuffs. Respondents answered that the cuffs were in danger and inconvenient because of broad cuffs. In regard of color, preference for white were high and followed with ivory, light sky, light green. and then light pink. Materials of current lab gowns were cotton/polyester or polyester 100%. Sixty-eight percentage of respondents were satisfied with that and others were not. The reasons for dissatisfactions were as follows; those materials were not protective to chemicals thoroughly enough. Cotton/polyester materials were heavy. Wearing sensation of polyester 100% was not good.

Wearing Evaluation of the Fall Impact Protective Pants for the Elderly Women (노년 여성을 위한 낙상충격 보호팬츠의 착의평가)

  • This study conducted a wearing evaluation of fall impact protective clothing developed in the previous study that evaluated activity, appearance, and practicality for elderly women. A total of 6 kinds of pants, which consisted of the three kinds of pads and two kinds of designs, were evaluated and the results are as follows. First, the activity of the protective clothing was tested with six women in their 60's and scored highly in most criterions that indicated satisfactory activity. The honeycomb type pad was superior in four of ten criterions for the activity evaluation results by pad types. Design B without a knee pad was evaluated as more comfortable on two motions that put pressure on the knee area. Second, the expert group considered design A to be better than design B, while the subject group preferred design B over design A in design appearance evaluation. Both the subject group and expert group evaluated that design A is better than design B in consideration of pads. Third, in the assessment of practicability, both the expert group and the subject group provided the highest score on the question of if elderly women needed protective clothing. The evaluation of washing ability indicated no noticeable change in the clothing form and size before and after washing.

Biorisk Assessment of Medical Diagnostic Laboratories in Nigeria

  • Background: The aim of this study was to assess public and private medical diagnostic laboratories in Nigeria for the presence of biosafety equipment, devices, and measures. Methods: A total of 80 diagnostic laboratories in biosafety level 3 were assessed for the presence of biosafety equipment, devices, and compliance rate with biosafety practices. A detailed questionnaire and checklist was used to obtain the relevant information from enlisted laboratories. Results: The results showed the presence of an isolated unit for microbiological work, leak-proof working benches, self-closing doors, emergency exits, fire extinguisher(s), autoclaves, and hand washing sinks in 21.3%, 71.3%, 15.0%, 1.3%, 11.3%, 82.5%, and 67.5%, respectively, of all laboratories surveyed. It was observed that public diagnostic laboratories were significantly more likely to have an isolated unit for microbiological work (p = 0.001), hand washing sink (p = 0.003), and an autoclave ($p{\leq}0.001$) than private ones. Routine use of hand gloves, biosafety cabinet, and a first aid box was observed in 35.0%, 20.0%, and 2.5%, respectively, of all laboratories examined. Written standard operating procedures, biosafety manuals, and biohazard signs on door entrances were observed in 6.3%, 1.3%, and 3.8%, respectively, of all audited laboratories. No biosafety officer(s) or records of previous spills, or injuries and accidents, were observed in all diagnostic laboratories studied. Conclusion: In all laboratories (public and private) surveyed, marked deficiencies were observed in the area of administrative control responsible for implementing biosafety. Increased emphasis on provision of biosafety devices and compliance with standard codes of practices issued by relevant authorities is strongly advocated.

Anti-microbial Effects of Washing and Chlorine Treatments on Fresh Fruits (과일류의 염소 소독 농도 및 세척 횟수에 따른 미생물 제거 효과)

  • This study examined the anti-microbiological effects of chlorine treatment on the surface of fresh fruits, in order to improve microbiological safety in school foodservice operations. Non-peeled fruit(strawberries) and peeled fruit(bananas) were treated with different concentrations of chlorinated water and rinsing numbers, followed by microbiological testing. The fruits were immersed at different concentrations of chlorinated water(0 ppm, 50 ppm, and 100 ppm) and durations(3 min and 5 min), and were then rinsed with tap water(one time, two times, or three times). The total viable cell counts of both the strawberries and bananas ranged from $10^3$ CFU/g to $10^4$ CFU/g, and coliform levels ranged from $10^2$ CFU/g to $10^3$ CFU/g. As the chlorine concentration, immersion time, and rinsing number increased, anti-microbiological activity increased. The largest microbial reduction was shown with immersion for 5 min in 100 ppm chlorinated water and three rinsings. In the strawberries, this treatment reduced the initial population of total viable cells and coliforms by 3.29 log CFU/g and to an undetectable level, respectively, no total viable cells or coliforms were detected on the banana surface following this treatment. However, after a sterilization treatment with immersion for 5 min in 50 ppm chlorinated water and three rinsings, the total viable cell counts and coliform counts of the strawberries and bananas decreased to acceptable levels, based on the microbiological standards for ready-to-eat foods. Overall, it was shown that the sterilization treatment of 50 ppm chlorinated water, soaking for 5 min, and three rinsings provided an effective reduction in surface microbes, and enhanced the microbiological safety of the fruit.

Formative Application Using Korean Traditional Paper (한지패션소재의 조형적 활용)

  • It was proved the practical use of the unique traditional Korean paper through the previous researches. The purpose of this study was to suggest the adequate application which is making various applied patterns in the fashion products so as to contribute to textile, fashion, and Korean paper industry. Six kinds of vests with the same fashion material using the Korean traditional paper were manufactured in order to investigate the images of materials in case of wearing them on the actual human bodies. As the results of image assessment for the materials and the factor analysis, 'classical', 'natural', 'Korean traditional', 'rural' and 'luxury' images were presented as the representative image of the Korean traditional paper material. Making repeated patterns of stripes and zigzag by sewing-machine stitches showed the natural effect on Korean traditional paper. Well-matched harmony was shown between holes made by the needle in sewing and slits torn by the tension to the needle and the thread. Especially, transparency by irregular holes formed by artificial frictional washing could shown special formative arts harmoniously with semi-transparency of Korea traditional paper and touch of sewing yarn. After the unripe persimmon dyeing, holes made by the needle became more hard and tight. The changes were no longer shown by the artificial frictional washing and then fixed. As the result of applying the Korean traditional paper fashion materials with various patterns for the basic formed garments, it was shown that the common and simple garments were changed the garments with special formation which could be found on the art garments due to the creative touch and formative arts using the Korea traditional paper fashion material. A variety of formative application by means of the developed samples was suggested to enlarge the practicality. As the result of this study, it was discovered that the possibility and application of fashion goods through the developed samples made of Korean traditional paper.

Effects of Different Sanitizers on the Quality of 'Tah Tasai' Chinese Cabbage (Brassica campestris var. narinosa) Baby Leaves (살균소독제가 다채 어린잎채소(Brassica campestris var. narinosa)의 수확 후 품질에 미치는 영향)

  • The demand of packaged baby leaves has been increased for its convenient use as fresh-cut produce. This investigation was aimed to explore the effects of different sanitizers on the quality parameters of 'Tah Tasai' Chinese cabbage (Brassica campestris var. narinosa) baby leaves. Thirteen days old baby leaves were harvested and washed in tap water (TW), 100 ppm chlorine solution (Cl), 2 ppm ozonated water ($O_3$), 15 ppm chlorine dioxide solution ($ClO_2$) and washing with 0.2% citric acid solution followed by 50% ethanol spray (CA+Et). The samples were then packaged in 50 ${\mu}m$ polyethylene bags and stored at $5^{\circ}C$ for 10 days. Off-odor of packaged baby leaves was not detected during storage. There was no significant difference in color parameters among the treatments. Samples treated with $O_3$ showed substantially higher electrolyte leakage throughout the storage. This treatment also rendered a higher accumulation of $CO_2$ in the packages. Samples treated with Cl and CA+Et maintained good overall visual quality with higher scores compared to that of $O_3$ and $ClO_2$. Although Cl treatment showed lower number of total aerobic count at the beginning of storage, citric acid in combination with ethanol treatment was more effective until the end of storage. The combined treatment also showed comparatively lower coliform plate count. This result indicates that citric acid wash followed by ethanol spray could be an alternative to chlorine for environment friendly sanitization of baby leaves.

The Dyeability and Antimicrobial Activity of Cotton Fabric Dyed with Sophora Radix Extracts on Skin Microorganisms (고삼 추출액를 이용한 염색 면포의 염색성와 피부 미생물 억제효과)

  • The aim of this study was to elucidate dyeability and antimicrobial activity of cotton fabrics dyed with Sophora Radix extracts according to various mordants. Dyes were extracted from Sophora Radix using ethanol. Then, cotton fabrics were dyed with extracts two times by post-mordanting method in which the extract was 60% (owf, the mordant was 3% (owf), L.R was 1:20, the temperature was 60~7$0^{\circ}C$, the time of dyeing was 60min., and the time of mordanting was 60min.. The dyeability was evaluated by surface color and color fastness. The skin microorganism was evaluated on S. sureus, B. subtilis, S. epidermidis, P. acnes, P. aeruginosa, E. coli, A. niger, C. albicans and T. mentagrophytes. The results are as follows; 1. When mordants were treated, surface color was 3.3Y to 0.1 GY in H (hue) value which indicated greenish yellow to yellow 2. The color fastness to perspiration, dry-cleaning, rubbing, and washing stain fabric showed 4~5 degree. The color fastness to light was improved to 4 degree by treatment of mordants. The color fastness to washing was 2 degree which was somewhat poor. 3. Cotton dyed with ethanol extracts was excellent on S. aureus, B. subtilis, S. epidermidis and p. antis. But that showed poor antibacterial activities on P. aeruginosa and E. coli such as gram negative baterials 4. Antibacterial activity of cotton fabrics dyed didn't be improved by treatment of mordant 5. Antifungal activity of cotton dyed with ethanol extracts was excellent on T. mentagrophytes. Especially, on T. mentagrophytes there was no growth of fungus during 72 hours in cotton dyed mordanting with SnCl$_2$.$_2$$H_2O$.

Effect of Blanching Time on Changes of Hazardous Factors in Leafy Vegetables Treated by Pesticides (데치는 시간이 침투성 및 비침투성 농약 처리 엽채류의 위해성 요인 변화에 미치는 영향)

  • This study was conducted to investigate the effect of blanching time on changes of hazardous factors in leafy vegetables treated by systemic and non-systemic pesticides. Changes of hazardous factors in leafy vegetables treated by systemic and non-systemic pesticides showed the same trend. In changes of microbial counts in leafy vegetables according to blanching time, the total plate count was significantly decreased by blanching and total coliforms were not detected in any of the vegetables. In traceable metal contents of fresh vegetables before washing, the levels in spinach, chard and whorled mallow were 0.042, 0.040 and 0.032 ppm for Pb, 0.020, 0.023 and 0.019 ppm for Cd, and 0.029, 0.034 and 0.030 ppm for As, respectively. Therefore, the levels of Pb, Cd and As in leafy vegetables were very traceable and in terms of food safety there was no problem. On the other hand, the traceable metal contents in leafy vegetables did not show significant difference by blanching and blanching time. Nitrate contents in fresh vegetables before washing were detected at traceable levels. The nitrate contents in spinach, chard and whorled mallow were significantly decreased by $22\%,\;17\%\;and\;14\%$ after blanching time of 5 min, 9 min and 10 min, respectively.

Rapid Staining Using the Shorr Method for Intraoperative Peritoneal Washing Cytology in Advanced Gastric Cancer: a Pilot Study from a Single Institution

  • Purpose: Intraoperative peritoneal washing cytology (PWC) is used to determine treatment strategies for gastric cancer with suspected serosal invasion. However, a standard staining method for intraoperative PWC remains to be established. We evaluated the feasibility of a rapid and simple staining method using Shorr's stain for intraoperative PWC in advanced gastric cancer. Materials and Methods: Between November 2012 and December 2014, 77 patients with clinical T3 or higher gastric cancer were enrolled. The sensitivity, specificity, and concordance between the Shorr staining method and conventional Papanicolaou (Pap) staining with carcinoembryonic antigen (CEA) immunohistochemistry (IHC) were analyzed. Results: Intraoperative PWC was performed laparoscopically in 69 patients (89.6%). The average time of the procedure was 8.3 minutes, and the average amount of aspirated fluids was 83.3 mL. The average time for Shorr staining and pathologic review was 21.0 minutes. Of the 77 patients, 16 (20.7%) had positive cytology and 7 (9.1%) showed atypical findings; sensitivity and specificity were 73.6% and 98.2% for the Shorr method, and 78.9% and 98.2% for the Pap method with CEA IHC, respectively. Concordance of diagnosis between the 2 methods was observed in 90.9% of cases (weighted ${\kappa}$ statistic=0.875) and most disagreements in diagnoses occurred in atypical findings (6/7). In overall survival, there was no significant difference in C-index between the 2 methods (0.459 in Shorr method vs. 0.458 in Pap with CEA IHC method, P=0.987). Conclusions: Shorr staining could be a rapid and reliable method for intraoperative PWC in advanced gastric cancer.
