The effects of cecectomy on nitrogen utilization and nitrogen excretion were examined in single comb white leghorn (SCWL) cockerels fed a 5% protein diet supplied with urea. The cecectomy tended to increase nitrogen balance and nitrogen utilization and significantly decreased uric acid excretion (p < 0.01). Urea and ammonia excretion tended to be about 60% increased and decreased by cecectomy in SCWL cockerels, respectively, but blood ammonia, urea and uric acid concentrations were not affected. The results are in good agreement with those obtained previously in cecum-ligated chickens. It is concluded that the improvement of nitrogen utilization and decreases in urinary uric acid excretion in cecectomized chickens do not result from the modification of cecal fermentation.
In this review, we discuss the ways in which our understanding of the controls of nitrogen use efficiency applied to crop improvement has been increased through the development of molecular physiology studies using transgenic plants or mutants with modified capacities for nitrogen uptake, assimilation and recycling. More recently, exploiting crop genetic variability through quantitative trait loci and candidate gene detection has opened new perspectives toward the identification of key structural or regulatory elements involved in the control of nitrogen metabolism for improving crop productivity. All together these studies strongly suggest that in the near future nitrogen use efficiency can be improved both by marker-assisted selection and genetic engineering, thus having the most promise for the practical application of increasing the capacity of a wide range of economically important species to take up and utilize nitrogen more efficiently.
This study was performed to investigate the effect of dietary levels on protein metabolism in eight healthy Korean adult females. The 20-day metabolic study consisted of 2 day adaptation period and three 6-day experimental. Three experimental diets were low protein (LP : protein 44g), high protein(HP : protein 85g) and high animal protein (HAP : protein-84g). The apparent absorption and balance on nitrogen were significantly higher in high protein than in low protein diet. Nitrogen, absorption rate was about 75% for low protein and about 85% for high protein intake. The mean values of nitrogen balance were -1.28% for low protein and 0.78% for high protein diet. All the subjects were in negative nitrogen balance at the low protein intake while they were in positive nitrogen balance at the high protein intake. The mean daily urinary nitrogen excretion increased with increased level of protein intake. Urea nitrogen was the largest part of the urinary nitrogen. The ratio of urea nitrogen to total urinary nitrogen increased significantly for 79 to 85% as protein intake was doubled.
This study was conducted to obtain accurate data on the intake, digestibility and nitrogen balance of protein in Korean college women. Subjects were 8 female college students, aged from 21 to 23, and maintained their menu and life patterns regular during a 4- week study. The same amount of diet that the subjects had consumed, and feces and urine were collected and measured to extract their nitrogen content by Kjeldahl method. From this data, apparent digestibility and the body nitrogen balance were estimated by determing daily protein intake and excretion. The daily protein intake was 56.9$\pm$1.4g and daily fecal protein loss was 6.3$\pm$0.2g. The apparent digestibility of protein was 89.6$\pm$0.7$\%$. The daily nitrogen intake measured by Kjeldahl method was 9.43$\pm$0.2g. The urinary nitrogen excretion was 7.64$\pm$0.23g and fecal nitrogen excretion was 1.02$\pm$0.03g. An in vitro study was conducted to determine the effect of nitrate-nitrogen used as a sole dietary nitrogen source on ruminal fermentation characteristics and microbial nitrogen (MN) synthesis. Three treatment diets were formulated with different nitrogen sources to contain 13% CP and termed i) nitrate-N diet (NND), ii) urea-N diet (UND), used as negative control, and iii) tryptone-N diet (TND), used as positive control. The results of 24-h incubations showed that nitrate-N disappeared to background concentrations and was not detectable in microbial cells. The NND treatment decreased net $CH_4$ production, but also decreased net $CO_2$ production and increased net $H_2$ production. Total VFA concentration was lower (p<0.05) for NND than TND. Suppression of $CO_2$ production and total VFA concentration may be linked to increased concentration of $H_2$. The MN synthesis was greater (p<0.001) for NND than UND or TND (5.74 vs. 3.31 or 3.34 mg/40 ml, respectively). Nitrate addition diminished methane production as expected, but also increased MN synthesis.
A study was conducted to investigate nitrogen balance and to estimate daily nitrogen requirement in 43 Korean female college students students maintaining their usual diet and activity levels. Nitrogen intake and excretion were measured in two separate peroids about one month apart, each period lasting for 3 days. Nitrogen intake was assessed by duplicate portion analysis of diet, and N excretion in faces and urine were measured during the study period. Mean daily nitrogen intake level was 129.3mg/kg B.W and the apparent digestibility of nitrogen was 76%. Mean daily urinary nitrogen excretion was 113.5mg/kg BW. 895 of total nitrogen intake. Mean daily nitrogen balance of subjects was -14.5mg/kg BW. Mean daily requirements of nitrogen for 0 balance, calculated by regression analysis of N balance and energy-adjusted N intake. were 1) 197.mg/kg B.W with the present energy intake level of the study subjects. 2) 157mg/kg B.W when energy intake is sufficient to maintain energy balance, and 30 130mg/kg B.W. when energy intake is Korean RDA level for moderate activity. When energy intake level is sufficient to meet their requirement, daily protein requirement for 0 balance is about 1.0g/kg B.W. The results of this study indicate that nitrogen intake level of young female college students is not sufficient to meet their requirements, and they should increase protein intake together with increase in energy intake.
The effect of formaldehyde treatment of solvent and mechanical extracted cottonseed meal on the performance, digestibility and nitrogen balance was assessed in lambs. Four total mixed rations viz., A, B, C and D containing 40% untreated and treated solvent and mechanical extracted meal were prepared. Sixteen male lambs with average BW of 20-22 kg were randomly allocated to experimental rations and were fed individually during ninety days growth trial. The treatment of solvent extracted cottonseed meal resulted in a linear decrease in ruminal protein degradation. Maximum decrease (64%) in protein degradation was observed at 4 h incubation time with 0.3% formaldehyde treatment. Highest daily BW gain was observed in lambs fed on rations Band D compared to lambs fed on rations A and C. Daily BW gain was higher on rations having 0.3% for fromaldehyde treated cottonseed meals. Higher DM digestibility was observed on ration D compared to other rations. Higher (p<0.05) CP and CF digestibility was observed on rations Band D compared to rations A and C. Nitrogen retention as % age of nitrogen intake was (p<0.05) higher for lambs fed rations B and D compared to rations A and C. Similar pattern was observed for nitrogen retention as percent of nitrogen absorbed. The present study suggested that oil extraction methods of cottonseed did not alter their meal utilization in lambs, however, formaldehyde (0.3%) treatment of meals enhanced its efficiency for growth, digestibility and nitrogen balance in lambs.
The analysis of nail nitrogen content reflected recent nutritional status. Therefore, estimation of nail nitrogen content has been previously reported in a small group of neonates ana school children. Nail samples were obtained from 52 economically middle and 21 low classes. The contents of nitrogen in their fingernails were determinded by micro-Kjeldahl method. Mean nail nitrogen content was $133.1{\pm}8mg/g$ in the middle class which is significantly higher than that of the low class $122.1{\pm}8mg/g$. The nitrogen contents of nails obtained in summer were found by analysis of variance and student's t-test to he lower than those obtained in winter.
