  • There is a many kinds with nondestructive testing such as RT and UT representatively. Referred before two testing methods there is a limit which is spatial such as nuclear pipe, small vessel, sealing up vessel. So a new technique needs to overcome the limit which is spatial. shearography will be able to overcome the limit which is spatial. This paper introducing shearography which was known as non-contact full-field testing method and It is an interferometric technique for measurement of surface deformation such as displacement or displacement gradient. Also, a research about internal defect of the flange welding zone was accomplished. About variation with method pressurized with the Gaseous Nitrogen. Phase map where is various were measured according to changing a sheared direction, size of crack and loaded pressure. Consequently, crack quantitatively to be detected qualitatively was measured by using shearography.

Estuarine Behavior and Flux of Nutrients in the Seomjin River Estuary (섬진강 하구역에서 영양염의 하구내 거동과 플럭스)

  • In order to estimate the nutrient flux of the Seomjin River into the coastal waters of South Sea, and to understand the estuarine reactions during mixing between river water and seawater, we collected surface water along the salinity gradient in the Seomjin River estuary from Mar. 1999 to Apr. 2001. We found that nitrate and silicate were delivered by fluvial input, while phosphate was, supplied from disposed wastes in the Gwangyang Bay. Mean annual flux of dissolved inorganic nitrogen (DIN), phosphate and silicate into the Gwangyang Bay was estimated 10.9 molesㆍsec$^{-1}$(4,820 tonnesㆍyr$^{-1}$), 0.07 molesㆍsec$^{-1}$(68 tonnesㆍyr$^{-1}$), 13.3 molesㆍsec$^{-1}$(11,747 tonnesㆍy$^{-1}$), respectively. An evident removal of phosphate, silicate and ammonium at the mid-salinity zone during the dry season was attributed to the active uptake of phytoplankton, and consequently nutrient flux into the Gwangyang Bay was low. Whereas, during the flood season in summer, conservative or additional distribution of the nutrients was observed in the estuary. As a rsult nutrient flux into the Gwangyang Bay was maintained high. High concentrations of chlorophyll a and the active removal of nutrient during the dry season at the mid-salinity zone suggest that nutrient distribution in the Seomjin River estuary was mainly controlled by biological processes and nutrient fluxes into the Gwangyang Bay might be significantly modified of by the primary production.

Effects of Rare Earth Metal Addition on the Cavitation Erosion-Corrosion Resistance of Super Duplex Stainless Steels

  • Austenitic stainless steels such as AISI 316L have been used in equipment in which fluid flows at high speeds which can induce cavitation erosion on metallic surfaces due to the collapse of cavities, where the collapse is caused by the sudden change of local pressure within the liquid. Usually AISI 316L is susceptible to cavitation erosion. This research focuses on developing a better material to replace the AISI 316L used in equipment with high speed fluid flow, such as impellers. The effects of Rare Earth Metal (REM) additions on the cavitation erosion-corrosion resistance of duplex stainless steels were studied using metallographic examination, the potentiodynamic anodic polarization test, the tensile test, the X-ray diffraction test and the ultrasonic cavitation erosion test. The experimental alloys were found to have superior mechanical properties due to interstitial solid solution strengthening, by adding high nitrogen (0,4%), as well as by the refinement of phases and grains induced by fine REM oxides and oxy-sulfides. Corrosion resistance decreases in a gentle gradient as the REM content increases. However, REM containing alloys show superior corrosion resistance compared with that of other commercial alloys (SAF 2507, AISI 316L). Owing to their excellent mechanical properties and corrosion resistance, the alloys containing REM have high cavitation erosion-corrosion resistance.

Trait-based algal community assembly associated with Pectinatella magnifica (Bryozoa, Phylactolaemata)

  • Habitat-forming species increase spatial complexity and alter local environmental conditions, often facilitating the assembly of plants and animals. We conducted a trait-based approach to algal assemblages associated with the freshwater bryozoan, Pectinatella magnifica. Association with algae leads to the inner bodies of the bryozoans being colored green; this is frequently observed in the large rivers of South Korea. We collected the green-colored gelatinous matrices and phytoplankton from waterbodies of the two main rivers in South Korea. Algal assemblages within the colonies and in the waterbodies were compared using the three diversity indices (richness, diversity, and dominance), and the composition of functional groups (FGs) and morphologically based functional groups (MBFGs) between the colonies within and outside of P. magnifica colonies. The most dominant and common species within the colonies were Oscillatoria kawamurae and Pseudanabaena catenata, both of which were assigned to the same FG (codon S1). Of the algal assemblages within the colonies, the dominance was higher, while the richness and diversity were lower, than those in the waterbodies. There was variation in the compositions of FGs and MBFGs in the waterbodies outside the colonies. Total nitrogen and orthophosphate led to dominance, and were significant factors for the variation in FGs in the waterbodies, whereas there were no such significant factors within the colonies. This trait-based approach to the community structure of associated algae provides the status and habitat gradient of these communities, which are stable, isolated, and consistent with the overgrowth of shade-adapted tychoplanktonic cyanobacteria.

Transient thermal stress of CFRP propellant tank depending on charging speed of cryogenic fluid

  • In order to increase thrust of the space launch vehicle, liquid oxygen as an oxidizer and kerosene or liquid hydrogen as a fuel are generally used. The oxidizer tank and fuel tanks are manufactured by composite materials such as CFRP (Carbon Fiber Reinforced Plastic) to increase pay load. The thermal stress of the cryogenic propellant tank should be considered because it has large temperature gradient. In this study, to confirm the design integrity of the oxidizer tank of liquid oxygen, a numerical analysis was conducted on the thermal stress and temperature distribution of the tank for various charging speed of the cryogenic fluid from 100 ~ 900 LPM taking into account the evaporation rate of the liquid nitrogen by convective heat transfer outside the tank and boiling heat transfer inside the tank. The thermal stress was also calculated coupled with the temperature distribution of the CFRP tank. Based on the analysis results, the charging speed of the LN2 can majorly affects the charging time and the resultant thermal stress.

Change in Influent Concentration of Domestic Wastewater from Separated Sewer and Biological Nitrogen and Phosphorus Removal of a Full Scale Air-vent SBR (분류식 하수관거로의 전환시 유입하수의 성상 변화 및 선회와류식 SBR공법의 처리 특성)

  • This study was carried out to investigate change in influent concentration of domestic wastewater flowed from a newly constructed separate sewer system (SSS) and biological nutrients removal efficiency of a full scale Air-vent sequential batch reactor (SBR, $600m^3/d$). The average concentration of $BOD_5$, SS, T-N and T-P from SSS were 246.5 mg/L, 231.6 mg/L, 42.974 mg/L, 5.360 mg/L, respectively which corresponds to 2.2times, 1.2times, 1.8times and 2.1times higher than those from the conventional combined sewer system (CSS). The removal efficiency of $BOD_5$, SS, T-N, and T-P for the Air-vent SBR operated with influent from SSS averaged 99.1%, 99.0%, 91.2%, and 93.5%, respectively. Especially the respective nitrogen and phosphorus removal was 15% greater than that of the SBR operated with influent from CSS. Simultaneous nitrification and denitrification (SND) was observed in an aerobic reactor(II) as a result of DO concentration gradient developed along the depth by the Air-vent system. In order to achieve T-N removal greater than 90%, the C/N ratio should be over 6.0 and the difference between $BOD_5$ loading and nitrogen loading rate be over 100 kg/day (0.130 kg $T-N/m^3{\cdot}d$). Even with high influent T-P concentration of 5.360 mg/L from SSS (compared with 2.465 mg/L from CSS) T-P removal achieved 93.5% which was 15.5% higher than that of the SBR with influent from CSS. This is probably due to high influent $BOD_5$ concentration from SSS that could provide soluble carbon source to release phosphorus at anaerobic condition. In order to achieve T-P removal greater than 90%, the difference between $BOD_5$ loading and phosphorus loading rate should be over 100 kg /day (0.130 kg $T-N/m^3{\cdot}d$).

The effects of temperature on the growth rate and nitrogen content of invasive Gracilaria vermiculophylla and native Gracilaria tikvahiae from Long Island Sound, USA

  • The red alga Gracilaria vermiculophylla, a species native to the waters of Korea and Japan, has invaded marine coastal areas of Europe and the Americas, thriving in conditions that differ from those of its native habitat. In recent years, G. vermiculophylla has been discovered in the Long Island Sound (LIS) estuary growing alongside the native congener Gracilaria tikvahiae. The goal of this study was to determine whether the two strains of G. vermiculophylla from different regions of the world have evolved genetic differences (i.e., ecotypic differentiation) or if the physiological performance of the strains simply reflects phenotypic plasticity. Two strains of G. vermiculophylla (isolated in Korea and LIS) and a strain of the LIS native G. tikvahiae were grown for four weeks under temperatures ranging from 20 to $34^{\circ}C$ using a temperature gradient table (all other environmental conditions were kept constant). At the end of each week, wet weight of each sample was recorded, and thalli were reduced to the original stocking density of $1gL^{-1}$ (excess biomass was preserved for tissue carbon and nitrogen analysis). Generally, the growth rates of Korean G. vermiculophylla > LIS G. vermiculophylla > G. tikvahiae. After one week of growth G. tikvahiae grew 9.1, 12.0, 9.4, and 0.2% $d^{-1}$, at temperatures of 20, 24, 29, and $34^{\circ}C$, respectively, while G. vermiculophylla (LIS) grew 6.6, 6.2, 5.7, and 3.6% $d^{-1}$. G. vermiculophylla (Korea) grew 15.4, 22.9, 23.2, and 10.1% $d^{-1}$, much higher than the two strains currently inhabiting the LIS. On average, the LIS G. vermiculophylla strain contained 4-5% DW N, while the Korean strain and G. tikvahiae had more modest levels of 2-3% N DW. However, tissue N content declined as temperature increased in LIS and Korean G. vermiculophylla. The non-native haplotype may have evolved genetic differences resulting in lower growth capacity while concentrating significantly more nitrogen, giving the non-native a competitive advantage.

Bio-protective potential of lactic acid bacteria: Effect of Lactobacillus sakei and Lactobacillus curvatus on changes of the microbial community in vacuum-packaged chilled beef

  • Objective: This study was to determine the bacterial diversity and monitor the community dynamic changes during storage of vacuum-packaged sliced raw beef as affected by Lactobacillus sakei and Lactobacillus curvatus. Methods: L. sakei and L. curvatus were separately incubated in vacuumed-packaged raw beef as bio-protective cultures to inhibit the naturally contaminating microbial load. Dynamic changes of the microbial diversity of inoculated or non-inoculated (control) samples were monitored at $4^{\circ}C$ for 0 to 38 days, using polymerase chain reaction-denaturing gradient gel electrophoresis (PCR-DGGE). Results: The DGGE profiles of DNA directly extracted from non-inoculated control samples highlighted the order of appearance of spoilage bacteria during storage, showing that Enterbacteriaceae and Pseudomonas fragi emerged early, then Brochothrix thermosphacta shared the dominant position, and finally, Pseudomonas putida showed up became predominant. Compared with control, the inoculation of either L. sakei or L. curvatus significantly lowered the complexity of microbial diversity and inhibited the growth of spoilage bacteria (p<0.05). Interestingly, we also found that the dominant position of L. curvatus was replaced by indigenous L. sakei after 13 d for L. curvatus-inoculated samples. Plate counts on selective agars further showed that inoculation with L. sakei or L. curvatus obviously reduced the viable counts of Enterbacteraceae, Pseudomonas spp. and B. thermosphacta during later storage (p<0.05), with L. sakei exerting greater inhibitory effect. Inoculation with both bio-protective cultures also significantly decreased the total volatile basic nitrogen values of stored samples (p<0.05). Conclusion: Taken together, the results proved the benefits of inoculation with lactic acid bacteria especially L. sakei as a potential way to inhibit growth of spoilage-related bacteria and improve the shelf life of vacuum-packaged raw beef.

The Effects of Soybean Cultivation on Soil Microorganism Activity (콩 재배가 토양 미생물 군집 활성도에 미치는 영향)

  • BACKGROUND: For sustainable agriculture, there are various agricultural practices including low input. Over the last few decades high input of chemical fertilizer and compounds results in environmental pollution and deterioration of soil fertility. Soybean (Glycine max L.) is well known eco-friendly crop due to their symbionts. Soybean has a relationship with nitrogen fixation bacteria called rhizobia. In this research work, we investigated effects of soybean cultivation on soil microorganism activities. METHODS AND RESULTS: Experiments were conducted in pots and potato cultivation was used as reference. Soil chemical properties were analyzed considering soil nutrient over cropping period. For the soil microbial community analysis, dehydrogenase activity analysis (DHA) analyzed along with denaturing gradient gel electrophoresis. The results showed that higher soil organic matter in the soybean cultivation soil than in the potato cultivation soil. Available $P_2O_5$ concentration increased gradually in both pots but showed higher value in the potato cultivation soil. DHA value implying microbial activities showed higher value in the soybean cultivation soil over all cropping period. CONCLUSION: The cause of high microbial activity in the soybean cultivation soil was considered to the effects of some specific microorganisms related to soybean cultivation. Therefore, the availability of soybean cultivation for sustainable agriculture should be encouraged in terms of microorganism community activity in soil.

Properties and Activities of Nireogenase System of Azospirillum amazonensa Kp1 (Azospirillum amazonense Kp1의 질소고정효소계의 활성 및 특성)

  • The maximum nitrogen fixation activity of the associative, microaerobic and acid tolerant bacteria, Azospirillum amazonense Kp1 was obtained with 0.2Kpa of $O_{2}$ and showed a reversible inhibition by the higher concentrations. Ammonium treatment caused a gradual inhibition of the activity up to 350mM. The nitrogenase systems were purified by gradient chromatography on DEAE-52 cellulose, heat treatment and preparative PAGE. The MoFe protein showed molecular weight of 210,000 including two nonidentical subunits with apparent molecular weights of 55,000 and 50,000 and an isoelectricpoint of 5.2 and contained 2, 24 and 28 atoms of Mo, Fe and acid labile S per molecule. The Fe protein revealed molecular weight of 66,000 including two types of subunits with molecular weights of 35,000 and 31,000 and an isoelectric point of 4.6, and contained 4 atoms of Fe and 6 atoms of S per molecule. The maximum specific nitrogenase activity attained 2,200 and 1,700nM $C_2H_4mg^{-1} min^{-1}$, respectively for MoFe and Fe proteins at pH7 and $35^{\circ}C$. The activity was lost after 10 and 30 days under the cold room ($4^{\circ}C$) condition for Fe and MoFe proteins, respectively.
