  • The effects of carbon and nitrogen sources on the mycelial growth and sporulation of microconidia and chlamydospores of five isolates of Cylindrocarpon destructans (Zinssm.) Scholten causing root rot of Panax ginseng were studied. For the carbon sources, fructose, glucose, maltose, and sucrose in Czapek-Dox broth showed good mycelial growth of 178∼201 mg in dry weight compared with 64 mg of the control. The best carbon sources tested for conidial formation were sucrose and maltose with 2.75 and 3.03 log conidia/ml, respectively. For the nitrogen sources, aspartic acid, NaNO3, KNO3, arginine, threonine, and leucine increased mycelial growth of the fungi to 208∼231 mg in dry weight without significant difference (p=0.05) among them. Meanwhile the growth with cystine was poor (26.3 mg dry weight), and no conidium and chlamydospore were formed. Maximum microconidial formation was observed in the media with NaNO3 and KNO3 as 3.37 and 3.35 log conidia/ml, and for the chlamydospore formation the (NH4)2SO4-containing medium and the nitrogen-absent medium were the best as 3.40 and 3.57 log chlamydospores/ml, respectively. No conidium was found in the medium without nitrogen sources, in which chlamydospore formation increased 6 times more than in the nitrogen-amended medium. However, deletion of carbon source in the medium did not affect on the formation of conidia and chlamydospores of C. destructans.

Mycelial Growth of Edible Ectomycorrhizal Fungi According to Nitrogen Sources

  • Ectomycorrhizal fungi are associated with plants roots and acquire significant amounts of nitrogen sources from the soil. For artificial cultivation, mass production of ectomycorrhizal fungi in liquid media is required. We studied the edible ectomycorrhizal mushrooms Hygrophorus russula, Ramaria fumigata, Sarcodon aspratus, and Tricholoma matsutake. All strains except S. aspratus NIFoS 2031 grew generally well on modified Melin-Norkran's (MMN) medium compared to on other media. All strains analyzed in this study showed significantly higher growth on organic nitrogen. Specifically, two strains of H. russula significantly responded to both tryptone and neopeptone media. Among different species and strains, there were clear differences in the capacity to grow on animal-based organic nitrogen sources.

Assimilation of Peptides and Amino Acids and Dissimilation of Lactate During Submerged Pure Cultures of Penicillium camembertii and Geotrichum candidum

  • The behavior of Penicillium camembertii and Geotrichum candidum growing in submerged pure cultures on simple (glutamate) or complex (peptones) substrates as nitrogen and carbon sources and lactate as a second carbon source was examined. Similar to the behavior previously recorded on a simple substrate (glutamate), a clear differentiation between the carbon source and the energy source was also shown on peptones and lactate during P. camembertii growth, since throughout growth, lactate was only dissimilated, viz., used for energy supply by oxidation into $CO_2$, whereas peptides and amino acids from peptones were used for carbon (and nitrogen) assimilation. Because of its deaminating activity, G candidum preferred peptides and amino acids to lactate as energy sources, in addition to being assimilated as carbon and nitrogen sources. From this, on peptones and lactate, G candidum grew faster than P. camembertii (0.19 and 0.08 g/l/h, respectively) by assimilating the most readily utilizable peptides and amino acids; however, owing to its lower proteolytic activity, the maximum biomass was lower than that of P. camembertii (3.7 and 5.5 g/l, respectively), for which continuous proteolysis and assimilation of peptides were shown.

  • This study aimed to investigate growth rate and nutrient removal efficiency of Chlorella vulgaris according to nitrogen sources and frequency of pH adjustment. Nitrogen and phosphorus removal efficiencies were evaluated in the three different conditions using $NO_3{^-}$, $NH_4{^+}$ as a sole nitrogen source and mixed condition. Initial nutrient concentrations in artificial wastewater were 30 mg-N/L and 3 mg-P/L similar to secondary wastewater effluent. When nitrogen source was $NO_3{^-}$, there was no inhibition on the growth of C. vulgaris with adjusting pH every 24 hr while growth inhibition occurred with $NH_4{^+}$ caused by pH drop. N, P removal efficiencies were no significant depending on the nitrogen sources. As pH was adjusted to 7 by pH-stat, growth rate and nutrient removal efficiencies were increased compared to adjusting pH every 24 hr, however, growth rate and nutrient removal efficiencies were no significant depending on the nitrogen sources.

Effects of Carbon and Nitrogen Sources on Fatty Acid Contents and Composition in the Green Microalga, Chlorella sp. 227

  • In order to investigate and generalize the effects of carbon and nitrogen sources on the growth of and lipid production in Chlorella sp. 227, several nutritional combinations consisting of different carbon and nitrogen sources and concentrations were given to the media for cultivation of Chlorella sp. 227, respectively. The growth rate and lipid content were affected largely by concentration rather than by sources. The maximum specific growth was negatively affected by low concentrations of carbon and nitrogen. There is a maximum allowable inorganic carbon concentration (less than 500~1,000 mM bicarbonate) in autotrophic culture, but the maximum lipid content per gram dry cell weight (g DCW) was little affected by the concentration of inorganic carbon within the concentration. The lipid content per g DCW was increased when the microalga was cultured with the addition of glucose and bicarbonate (mixotrophic) at a fixed nitrogen concentration and with the lowest nitrogen concentration (0.2 mM), relatively. Considering that lipid contents per g DCW increased in those conditions, it suggests that a high ratio of carbon to nitrogen in culture media promotes lipid accumulation in the cells. Interestingly, a significant increase of the oleic acid amount to total fatty acids was observed in those conditions. These results showed the possibility to induce lipid production of high quality and content per g DCW by modifying the cultivation conditions.

Effects of Some Amino Acids on Ammonia Secretion and Extracellular Protease Activity by Three Oomycetes in Synthetic Medium with or without Glucose

  • The effects of different concentrations of three amino acids as carbon and or nitrogen sources on mycelial dry weights, changes in pH values of synthetic medium, ammonia secretion and extracellular protease activity by three zoosporic fungi, pathogens of fish and shellfish, were studied. As compared with the control, the addition of isoleucine and aspartic acid as nitrogen sources were generally stimulative for mycelial dry weight production whereas phenylalanine was inhibitory irrespective to the tested fungal species. When amino acids served as carbon and nitrogen sources, the mycelial dry weights of the three fungi were increased (mostly non-significantly) relative to untreated control but weights were decreased as the concentrations of the three amino acids raised. The addition of individual amino acids as carbon and nitrogen sources to the medium significantly increased pH values of the medium comparable to the control. The addition of each of the three amino acids as carbon and nitrogen sources to the medium significantly induced ammonia secretion by the three species of zoosporic fungi. Ammonia secretion in synthetic medium amended with amino acids as nitrogen source raised by the three zoosporic fungi relative to untreated control except in case of Achlya racemosa treated with isoleucine. Extracellular protease activity was almost promoted in case of Achlya proliferoides and Saprolegnia furcata cultures treated with isoleucine and aspartic acid individually in presence of glucose and vice versa in case of phenylalanine. However, extracellular protease activity of A. racemosa decreased compared with the control at various concentrations of isoleucine and both phenylalanine and aspartic acid assumed inconsistent effects. Extracellular protease activity of the three zoosporic fungi in the medium devoid of glucose varied depending upon zoosporic fungal species, the tested amino acid and the applied concentrations. The values of protease activity were approximately less two folds than that obtained in presence of glucose.

Effects of nitrogen sources on cell growth and biochemical composition of marine chlorophyte Tetraselmis sp. for lipid production

  • Nitrogen is one of the most critical nutrients affecting cell growth and biochemical composition of microalgae, ultimately determining the lipid or carbohydrate productivity for biofuels. In order to investigate the effect of nitrogen sources on the cell growth and biochemical composition of the marine microalga Tetraselmis sp., nine different N sources, including NaNO3, KNO3, NH4NO3, NH4HCO3, NH4Cl, CH3COONH4, urea, glycine, and yeast extract were compared at the given concentration of 8.82 mM. Higher biomass concentration was achieved under organic nitrogen sources, such as yeast extract (2.23 g L−1) and glycine (1.62 g L−1), compared to nitrate- (1.45 g L−1) or ammonium-N (0.98 g L−1). All ammonium sources showed an inhibition of cell growth, but accumulated higher lipids, showing a maximum content of 28.3% in ammonium bicarbonate. When Tetraselmis sp. was cultivated using yeast extract, the highest lipid productivity of 36.0 mg L−1 d−1 was achieved, followed by glycine 21.5 mg L−1 d−1 and nitrate 19.9 mg L−1 d−1. Ammonium bicarbonate resulted in the lowest lipid productivity of 14.4 mg L−1 d−1. The major fatty acids in Tetraselmis sp. were palmitic, oleic, linoleic and linolenic acids, regardless of the nutritional compositions, indicating the suitability of this species for biodiesel production.

팔당호 내 식물플랑크톤 안정동위원소 분석을 통한 유기물 기원 평가 (Evaluation of Organic Matter Sources of Phytoplankton in Paldang Reservoir using Stable Isotope Analysis)

  • The organic matter sources of phytoplankton and related environmental factors influencing algal bloom in Paldang reservoir were studied using nitrogen and carbon isotope ratio(${\delta}^{15}N$, ${\delta}^{13}C$). Phytoplankton samples for stable isotope analysis were collected from four points in reservoir using a plankton net. Physicochemical water quality, algal taxa and hydrological data were collected from published monitoring material. Phytoplankton samples were analyzed by IRMS. CN ratio of each sample was very similar to that of phytoplankton from literature cited. ${\delta}^{15}N$ of each sample was decreased during July. Mixing and dilution of nitrogen sources due to increment of influx by concentrated rainfall were considered as the main reason for the decline of ${\delta}^{15}N$. Based on analyzed ${\delta}^{15}N$ value of each sample, nitrogen source of Bughan river sample was presumed to come from soil. The nitrogen sources of Namhan river and Kyeongan stream samples seemed to be sewage or animal waste. Low ${\delta}^{15}N$ value in August (2012) seemed to be influenced by isotope fractionation due to the blooming of nitrogen-fixation blue-green algae (Anabaena spp.). Variation in ${\delta}^{15}N$ values particularly by blue-green algal bloom was considered the important factor for estimating the organic matter sources of phytoplankton.

The Schizosaccharomyces pombe Gene Encoding ${gamma}-Glutamyl$ Transpeptidase I Is Regulated by Non-fermentable Carbon Sources and Nitrogen Starvation

  • In our previous study, the first structural gene (GGTI) encoding ${\gamma}-glutamyl$ transpeptidase was cloned and characterized from the fission yeast Schizosaccharomyces pombe, and its transcription, using the GGTI-lacZ fusion gene, containing the 1,085 bp upstream region from the translational initiation point, was found to be enhanced by sodium nitroprusside and L-buthionine-(S,R)-sulfoximine (BSO). In the present work, regulation of the GGTI gene was further elucidated. Non-fermentable carbon sources, such as acetate and ethanol, markedly enhanced the synthesis of ${beta}-galactosidase$ from the GGTI-lacZ fusion gene. However, its induction by non-fermentable carbon sources appeared to be independent of the presence of the Pap1 protein. Nitrogen starvation also gave rise to induction of GGTI gene expression in a Pap1-independent manner. The three additional fusion plasmids, carrying 754, 421 and 156 bp regions, were constructed. The sequence responsible for the induction by non-fermentable carbon sources and nitrogen starvation was identified to exist within a -421 bp region of the GGTI gene. Taken together, the S. pombe GGTI gene is regulated by non-fermentable carbon sources and nitrogen starvation.

Candida bombiocola로 부터 미생물 계면활성제 생산시 관여 인자에 관한 연구 (Effects of Culturing Parameters on the Production of Microbial Biosurfactant from Candida bombiocola)

  • 본 연구에서는 C. bombiocola에서 생성되는 미생물 계면활성제인 Sophorolipid의 생산관여인자 중, 질소원의 종류에 따른 영향과 C/N비율에 따른 생성량의 변화를 관찰하였다. 유기질소원이 비교적 놓은 생성량을 보였으며 그 중 pepton의 경우가 80g/L에 해당하는 높은 생성량을 보였다. C/N의 비율의 변화에 의한 Sophorolipid의 생성량의 변화 양상이 질소원의 종류에 따라 각기 다르게 나타났다. 또한 탄소원의 50%을 배양 이틀후 공급하는 방법으로 Sophorolipid가 약 30% 증가됨을 알 수 있다.
