Synthesis of Two Nitro Analogs of Tranylcypromine: Relations of Aromatic Substitution of Nitro Groups to MAO-Inhibitory Activity

  • Two new nitro analogs of tranylcypromine, (E)-2-(p-nitrophenyl)cyclopropylamine ((E)-p-NTCP) and (E)-2-(m-nitrophenyl)cyclopropylamine ((E)-m-NTCP) were synthesized in order to examine the effect of aromatic nitro substitution on the MAO-inhibitory activity of 2-phenylcyclopropylamines. The compounds were obtained by treating t-butyl (E)-2-(p-nitrophenyl) cyclopropanecarbamate and t-butyl (E)-2-(m-nitrophenyl)cyclopropanecarbamate with p-toluenesulfonic acid in $CH_3$CN. Inhibitions of rat brain mitochondrial MAO-A and B by the compounds were examined using serotonin and benzylamine as the substrate at both in vitro and ex vivo levels. It was found from in vitro measurements that (E)-p-NTCP at $6.0{\times}10^{-5}M$ elicited merely 22.5% inhibition against MAO-B without any effect on MAO-A. In contrast, (E)-m-NTCP showed fair degrees of inhibitions of MAO-A and B with $IC_{50}$ values, $2.5{\times}10^{-7}M\;and\;1.4{\times}10^{-6}M$, respectively. It was also noted from (E)-m-NTCP that m-nitro substitution caused a shift of selectivity of the inhibition toward MAO-A. According to ex vivo measurements at 1.5, 3, 6, and 12 hr following the administration of a dose of 0.015 mmol/kg, i.p. to the rats, the inhibition percents of MAO-A by (E)-m-NTCP were 58.6, 63.7 63.6, and 46.6%, slightly lower than those observed by tranylcypromine. Whereas, (E)-m-NTCP at the same dose level did not show significant inhibitions against both MAO-A and MAO-B. Possible reasons for the difference in potencies between (E)-m-NTCP and (E)-p-NTCP were sought in relation to differing electron withdrawing effects of m- and p-substituents which will influence electron density of the side chain amino functions and the partitions.

  • Approximate rate and stoichiometry of the reaction of ten compounds which contain functional group such as nitrile, nitro, halogen and one of these functional group together with a carbonyl group by the two phase reduction were tested at room temperature. Nitrile, nitro and halogen were all inert under these condition. Therefore selective reduction of carbonyl group in the presence of these group were examined. Thus m-nitrobenzaldehyde, m-nitroacetophenone, p-bromoacetophenone and p-cyanobenzaldehyde were reduced to corresponding alcohols in excellent yields, 95∼100 %.

Simultaneous Utilization of Two Different Pathways in Degradation of 2,4,6-Trinitrotoluene by White Rot Fungus Irpex lacteus

  • This study confirmed that white rot fungus Irpex lacteus was able to metabolize 2,4,6-trinitrotoluene (TNT) with two different initial transformations. In one metabolic pathway of TNT a nitro group was removed from the aromatic ring of TNT. Hydride-Meisenheimer complexes of TNT (H/sup -/-TNT), colored dark redo were confirmed as the intermediate in this transformation by comparison with the synthetic compounds. 2,4-Dinitrotoluene as a following metabolic product was detected, and nitrite produced by denitration of $H^-$-TNT supported this transformation. In the other TNT pathway, nitro groups in TNT were successively reduced to amino groups via hydroxylamines. Hydroxylamino-dinitrotoluenes and amino-dinitrotoluenes were identified as the intermediates. The activity of a membrane-associated aromatic nitroreductase was detected in the cell-free extract of I. lacteus. This enzyme catalyzed the nitro group reduction of TNT with NADPH as a cofactor, Enzyme activity was not observed in the presence of molecular oxygen.

  • A palladium-catalyzed C-H arylation reaction of nitroimidazoles and nitropyrazoles was developed using aryl bromides as arene donors. The electron-withdrawing effect of the nitro group allows for direct C-H arylation reactions of the nitro diazoles with high regioselectivity under mild conditions. The new C-H arylation approach is thus complementary to nucleophilic substitution reactions, enabling the preparation of complex nitroazole compounds.

  • Structure-activity relationship studies of allylamine type of antimycotics were carried out to evaluate the effect of naphthyl and methyl portion of naftifine. Compounds with 3,4-difluorophenyl (2a-5a), 4-hydroxyphenyl (2b-5b), 3-nitro­phenyl (2c-5c), 4-chlorobenzothiazoly (2d-5d) and 5-methylfurfural (2e-5e) instead of naphthyl group, and with hydrogen (3a-3e), methyl (4a-4e) and ethyl (5a-5e) in the place of methyl in naftifine were synthesized and tested for their in vitro anti-fungal activities against five different fungi. Fourteen compounds (3a, 4a, 5a, 3b, 4b, 5b, 3c, 4c, 5c, 3d, 4d, 3e, 4e and 5e) showed significant anti-fungal activities against T. mentagrophytes. (E)-N-(3-Phenyl-2-propenyl)-3,4-difluoro-ben­zenemethaneamine(3a), (E)_N_(3_phenyl_2_propenyl)_4_hydroxy_benzenemethaneamine(3b) and (E)-N-ethyl-N-(3-phenyl­2-propenyl)-3-nitro-benzenemethaneamine (5c) displayed moderate anti-fungal activities against all five different fungi.

Chemical Lithography by Surface-Induced Photoreaction of Nitro Compounds

  • Searching for systems of self-assembled monolayers (SAMs) that can be used as templates for chemical lithography, we found that nitro groups on aromatic SAMs are selectively converted on Ag to amino groups by irradiation with a visible laser. 4-nitrobenzenethiol on Ag was thus converted to 4-aminobenzenethiol by irradiating it with an $Ar^+$ laser. This was evident from surface-enhanced Raman scattering (SERS) as well as from a coupling reaction forming amide bonds. The surface-induced photoreaction allowed us to prepare patterned binary monolayers on Ag that showed different chemical reactivities. Using the binary monolayers as a lithographic template, we induced site-specific chemical reactions, such as the selective growth of biominerals on either the nitro- or amine-terminated regions by adjusting the crystal-growth conditions. We also demonstrated that patterned, amine-terminated monolayers can be fabricated even on gold by using silver nanoparticles as photoreducing catalysts.

Another Evidence for Nitric Oxide as Mediator of Relaxation of Isolated Rabbit and Human Corpus Cavernosum

  • To prove the hypothesis that NO- and N $O_2$-carrying molecules potentiate photorelaxation by generating NO, investigation was carried out using isolated rabbit and human corpus cavernosum. Corporal smooth muscle, in the presence or absence of endothelium, relaxed only slightly upon ultraviolet light (366 nm) irradiation. But, NO-and/or N $O_2$-containing compounds such as streptozotocin and $N^{G}$-nitro-L-arginine methyl ester significantly (p<0.01) enhanced photorelaxation in this tissue. In addition, $N^{G}$-nitro-D-arginine methyl ester, known to lack inhibitory action on NO synthase, showed concentration-dependent potentiation of the photorelaxation. Oxygen radical generating system via copper+ascorbic acid and guanylate cyclase inhibitor, methylene blue, significantly (p<0.05) inhibited the streptozotocin-potentiated photorelaxation. Nitrite was accumulated by photolysis of streptozotocin, $N^{G}$-nitro-L-arginine methyl ester and $N^{G}$-nitro-D-arginine methyl ester, in a concentration and exposure time dependent manner. These observations indicate that NO is a potent relaxant of rabbit and human corpus cavernosum and further support the hypothesis that NO is released by photolysis from NO- and N $O_2$-carrying molecules.lecules.

Changes in Concentration Levels of Polycyclic Aromatic Compounds Associated with Airborne Particulate Matter in Downtown Tokyo after Introducing Government Diesel Vehicle Controls

  • The effectiveness of the government regulation on tail-pipe emission for diesel vehicles issued in 2003 in Tokyo was evaluated in this study. Variations in annual average concentrations of polycyclic aromatic hydrocarbons (PAHs) and nitro-PAHs associated with airborne particulate matter were investigated in connection with the variation in airborne elemental carbon (EC) concentration in downtown Tokyo, Japan in 2006-2007 and in 1997-1998. The annual average concentrations of EC, seven different PAHs, and 1-nitropyrene were found to have decreased significantly from 1997-1998 to 2006-2007. The most prominent decrease in atmospheric concentration was observed for 1-nitropyrene, which is a representative nitro-PAH originating from diesel vehicles. This indicated that the government control has worked to considerably reduce both atmospheric mutagens and airborne particulate matter. In contrast, the concentrations of two nitro-PAHs, 2-nitrofluoranthene and 2-nitropyrene, remained the same. These nitro-PAHs are known to be formed by atmospheric nitration of their parent PAHs, and this result suggested factors other than the concentration of parent PAHs and $NO_2$ affects the degree of atmospheric formation of nitro-PAHs.
