ADP-rubosylation may be involved in the process of macrophage activation. Nitric oxide (NO) has emerged as an important intracellular and interacellular regulatory molecule with function as diverse as vasodilation, neural communication or host defense. NO is derived from the oxidation of the terminal guanidino nitrogen atom of L-arginine by the NADPH -dependent enzyme, nitric oxide synthase (NOS) which is one of the three different isomers in mammalian tissues. Since NO can exert protective or regulatory functions in the cell at a low concentration while toxic effects at higher concentrations, its role may be tightly regulated in the cell. Therefore, this paper was focused on signal transduction pathway of NO synthesis, role of endogenous TGF-$\beta$ in NO production. effect of NO on superoxide formation. Costimulation of murine peritoneal macrophages with interferon-gamma (IFN-γ) and phorbol 12-myristate 13-acetate (PMA) increased both NO secretion and mRNA expression of inducible nitric oxide synthase (iNOS) when PMA abolished costimulation. Pretreatmnet of the cells with PMA abolished costimuation effects due to the depletion of protein kinase C (PKC) activities . The involvement of PKC in NO secretion could be further confirmed by PKC inhibitor, stauroprine, and phorbol ester derivative, phorbol 12,13-didecanoate. Addition of actinomycine D in IFN-γ plus PMA stimulated cells inhibited both NO secretion and mRNA expression of iNOS indication that PMA stabilizes mRNA of iNOS . Exogenous TGF-$\beta$ reduced NO secretion in IFN -γ stimulated murine macrophages. However addition of antisense oligodeoxynucleotide (ODN) to TGF-$\beta$ to this system recovered the ability of NO production and inhibited mRNA expression of TGF-$\beta$. ACAS interactive laser cytometry analysis showed that transportation of FITC -labeled antisense ODN complementary to TGF-$\beta$ mRNA could be observed within 5 min and reached maximal intensity in 30 min in the murine macrophage cells. NO released by activated macrophages inhibits superoxide formation in the same cells . This inhibition nay be related on NO-induced auto -adenosine diphosphate (ADP) -ribosylation . In addition, ADP-ribosylation may be involved in the process of macrophage activation .
Rehmanniae radix preparata is the root of Rehmanniae glutinosa Liboschitz var. purpurea Makino which has been classified into Scrophulariaceae. Rehmanniae radix preparata has been used for the treatment of diabetes, for the relief of the pain, and for the anti-oxidative action. In this study, the effect of the aqueous extract of Rehmanniae radix preparata on lipopolysaccharide-induced inflammation was investigated by using 3-(4,5-dimethylthiazol- 2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay, reverse transcription-polymerase chain reaction (RT-PCR), Western blot, prostaglandin E2 immunoassay, and nitric oxide (NO) detection in mouse BV2 microglial cells. In the present results, the aqueous extract of Rehmanniae radix preparata suppressed prostaglandin E2 (PGE2) synthesis and nitric oxide production by inhibiting the lipopolysaccharide-stimulated expressions of cyclooxygenase-2 (COX-2) and inducible nitric oxide synthase (iNOS) mRNA and protein in mouse BV2 cells. These results show that Rehmanniae radix preparata exerts anti-inflammatory effect probably by suppressing of COX-2 and iNOS expressions.
연구 배경 : 기도의 신경성 염증에서 산화질소가 관여하는 것으로 알려져 있으나, 그 역할에 대해서는 논란이 많다. 본 연구는 기도 신경성 염증에 관여하는 산화질소의 역할을 보다 명확히 밝히고자 하였다. 방법 150-350gm의 백서를 이용하여 기도의 신경성 염증에서 신경단백질 수용체 차단제인 FK224와 산화질소 합성효소 억제제인 $N^{\omega}$-nitro-L-arginine (L-NNA) 의 혈장유출에 대한 효과를 먼저 확인하고, 기도 신경성 염증에 관여하는 산화질소가 기도의 신경말단에서만 유리되는 지 또는 신경성 염증에서 유리된 신경 단백질로 인하여 다른 폐장 조직 세포에서도 산화질소가 유리되는 지를 규명하기 위하여 산화질소 합성효소의 종류와 그 분포를 polyclonal anti-NOS antibody에 대한 면역화학효소법으로 확인하여 다음과 같은 결과를 얻었다. 결과 : 백서의 기도 신경성 염증에서 신경단백질 수용체 차단제인 FK224는 혈장유출을 억제시키며 산화질소 합성효소 억제제인 L-NNA는 혈장유출을 증가시켰다(P<0.05). 기도 신경성 염증유발시 조직내 염증세포의 침윤은 증가되었으며, FK224로 전처치시 조직내의 염증세포의 침윤을 억제시켰다(P<0.05). 염증을 유발하는 것으로 알려진 유도형 산화질소 합성효소(iNOS)의 활성도는 침착된 염증세포에서만 유의하게 증가하였다(P<0.05). 염증을 억제하는 것으로 알려진 산화질소를 생성하는 구성형 산화질소 합성효소(cNOS)인 eNOS의 활성노는 혈관내피세포에서 증가하였으나 의미는 없었고, bNOS의 활성도는 신경성 염증에서 신경세포에서만 증가되었으며, FK224에 의해서도 bNOS의 활성도는 억제되지 않았다. 결론 : 기도의 신경성 염증에서 조직내 염증세포가 증가되며 iNOS에서 생성되는 산화질소가 주로 혈장유출에 관여하는 것으로 사료된다. FK224의 전처치는 염증세포의 조직내 침윤을 억제시키며, iNOS 의 활성도도 감소시켜 기도 혈장유출을 억제시키는 것으로 생각된다. 또한 기도의 신경성 염증에서 NANC신경에서도 산화질소가 유리됨을 알 수 있었으며, 기도 신경성 염증에서 산화질소 합성효소 억제제인 L-NNA로 혈장유출이 증가되는 것은 bNOS에서 유리되는 산화질소의 생성을 L-NNA가 억제시킬 수 있으므로 산화질소 합성효소 억제제가 기도 신경성 염증의 혈장유출을 증가시키는 데에 bNOS가 일부 작용할 것으로 생각되는 바이다.
In macrophages, nitric oxide (NO) is released as an inflammatory mediator and has been proposed to be an important modulator of many pathophysiological conditions including inflammation. In this study, we have examined the inhibition effects of NO production by 85% methanol extract of the flower of Pueraria thunbergiana (PF) in mouse macrophages. Extract of PF (1, 10, 100 ${\mu}g/ml$) inhibited NO production, inducible NO synthase and cyclooxygenase-2 expression in interferon-g and lipopolysaccharide-stimulated mouse peritoneal macrophages and it had no cytotoxicity. These data suggest that 85% methanol extract of PF might be useful in controlling macrophages mediated inflammatory disease.
연구배경: 산화질소(${\cdot}NO$)는 여러 세포에서 산화질소 합성효소(NOS)에 의해서 생산되며 다양한 병태생리과정에 관여한다. 여러 cytokine들이 iNOS의 발현을 촉진시키고 산화질소 생산을 증가시킴으로써 염증반응을 증폭시키고 세포와 조직손상을 초래한다고 알려진 바, 과산화수소($H_2O_2$)가 세포내 NOS의 발현과 산화질소형성에 미치는 영향을 알아보고자 하였다. 방법: 마우스 대식세포주 RAW264.7에 여러 가지 cytokine과 세균 내독소 (LPS)로 자극을 준 세포군 이에 더하여 $H_2O_2$, NOS 억제제 (L-NAME) 및 항산화제 (catalase)등을 사용하여 세포를 자극한 후 생성된 산화질소 산화물의 농도를 측정하고 Northern analysis로 iNOS mRNA의 발현정도를 보아 다음과 같은 성적을 얻었다. 결과: Cytokine과 LPS 자극군에서 대조군보다 ${\cdot}NO$ 생산이 높았고, 이 자극군에 $H_2O_2$를 추가로 자극하였을 때 ${\cdot}NO$생산이 2 배 이상 유의하게 높았다. Cytokine 자극군에서 $H_2O_2$의 자극 농도에 따른 ${\cdot}NO$생산은 $H_2O_2$의 농도가 증가할수록 유의하게 증가하였다. LPS와 IFN-$\gamma$ 자극군에서 L-NAME을 같이 자극시에 ${\cdot}NO$의 양은 L-NAME의 농도증가에 따라 유의하게 감소하였고, Cytokine 및 $H_2O_2$자극군에서도 추가로 자극한 L-NAME 의 농도증가에 따라 ${\cdot}NO$의 양은 유의하게 감소하였다. Cytokine과 $H_2O_2$ 자극균에 catalase를 같이 자극 하였을 때 ${\cdot}NO$의 양은 유의하게 감소했고, Mercaptoethanol과 phenanthroline을 전처치하고 LPS와 IFN-$\gamma$ 및 $H_2O_2$로 자극한 군에서 이들의 전처치한 농도가 높을수록 ${\cdot}NO$의 양은 유의하게 Cytokine자극군과 IFN-$\gamma$, LPS 자극군에 $H_2O_2$를 추가 자극 후 Northern analysis 결과 $H_2O_2$는 iNOS mRNA 발현을 현저히 증가시켰다. 결론: 이상의 결과로 과산화수소가 cytokine과 내독소 등으로 자극된 마우스 대식세포에서 산화질소생산에 유의한 증폭효과를 나타냈고, iNOS mRNA 의 발현도 증가시켰음을 확인할 수 있었다.
The overproduction of nitric oxide (NO) by inducible nitric oxide synthase (iNOS) is known to be responsible for vasodilation and hypotension observed in septic shock and inflammation. Inhibitors of iNOS, thus, may be useful candidates for the treatment of inflammatory diseases accompanied by overproduction of NO. In the course of screening oriental anti-inflammatory herbs for the inhibitory activity of NO synthesis, a crude methanolic extract of Curcuma zedoaria exhibited significant activity. The activity-guided fractionation and repetitive chromatographic procedures with the EtOAc soluble fraction allowed us to isolate three active compounds. They were identified as 1,7-bis (4-hydroxyphenyl)-1,4,6-heptatrien-3-one (1), procurcumenol (2) and epiprocurcumenol (3) by spectral data analyses. Their concentrations for the 50% inhibition of NO production $(IC_{50})$ in lipopolysaccharide (LPS)-activated macrophages were 8, 75, 77 ${\mu}M$, respectively. Compound 1 showed the most potent inhibitory activity for NO production in LPS-activated macrophages, while the epimeric isomers, compound 2 and 3 showed weak and similar potency. Inhibition of NO synthesis by compound 1 was very weak when activated macrophages were treated with 1 after iNOS induction. In the immunoblot analysis, compound 1 suppressed the expression of iNOS in a dose-dependent manner. In summary, 1,7-bis (4-hydroxyphenyl)-1,4,6-heptatrien-3-one from Curcuma zedoaria inhibited NO production in LPS-activated macrophages through suppression of iNOS expression. These results imply that the traditional use of C. zedoaria rhizome as anti-inflammatory drug may be explained at least in part, by inhibition of NO production.
Oxytocin is a neuropeptide produced primarily in the hypothalamus and plays an important role in the regulation of mammalian birth and lactation. It has been shown that oxytocin has important cardiovascular protective effects. Here we investigated the effects of oxytocin on vascular reactivity and underlying the mechanisms in human umbilical vein endothelial cells (HUVECs) in vitro and in rat aorta ex vivo. Oxytocin increased phospho-eNOS (Ser 1177) and phospho-Akt (Ser 473) expression in HUVECs in vitro and the aorta of rat ex vivo. Wortmannin, a specific inhibitor of phosphatidylinositol 3-kinase (PI3K), inhibited oxytocin-induced Akt and eNOS phosphorylation. In the rat aortic rings, oxytocin induced a biphasic vascular reactivity: oxytocin at low dose (10-9-10-8 M) initiated a vasorelaxation followed by a vasoconstriction at high dose (10-7 M). L-NAME (a nitric oxide synthase inhibitor), endothelium removal or wortmannin abolished oxytocin-induced vasorelaxation, and slightly enhanced oxytocin-induced vasoconstriction. Atosiban, an oxytocin/vasopressin 1a receptor inhibitor, totally blocked oxytocin-induced relaxation and vasoconstriction. PD98059 (ERK1/2 inhibitor) partially inhibited oxytocin-induced vasoconstriction. Oxytocin also increased aortic phospho-ERK1/2 expression, which was reduced by either atosiban or PD98059, suggesting that oxytocin-induced vasoconstriction was partially mediated by oxytocin/V1aR activation of ERK1/2. The present study demonstrates that oxytocin can activate different signaling pathways to cause vasorelaxation or vasoconstriction. Oxytocin stimulation of PI3K/eNOS-derived nitric oxide may participate in maintenance of cardiovascular homeostasis, and different vascular reactivities to low or high dose of oxytocin suggest that oxytocin may have different regulatory effects on vascular tone under physiological or pathophysiological conditions.
It has been generally accepted that glutamate mediates the ischemic brain damage, excitotoxicity, and induces release of neurotransmitters, including norepinephrine(NE), in ischemic milieu. In the present study, the role of nitric oxide(NO) in the ischemia-induced $[^3H]norepinephrine([^3H]NE)$ release from cortex slices of the rat was examined. Ischemia, deprivation of oxygen and glucose from $Mg^{2+}-free$ artificial cerebrospinal fluid, induced significant release of $[^3H]NE$ from cortex slices. This ischemia-induced $[^3H]NE$ release was significantly attenuated by glutamatergic neurotransmission modifiers. $N^G-nitro-L-arginine$ methyl ester(L-NAME), $N^G-monomethyl-L-arginine$ (L-NMMA) or 7-nitroindazole, nitric oxide synthase inhibitors attenuated the ischemia-evoked $[^3H]NE$ release. Hemoglobin, a NO chelator, and 5, 5- dimethyl-L-pyrroline-N-oxide(DMPO), an electron spin trap, inhibited $[^3H]NE$ release dose-dependently. Ischemia-evoked $[^3H]NE$ release was inhibited by methylene blue, a soluble guanylate cyclase inhibitor, and potentiated by 8-bromo-cGMP, a cell permeable cGMP analog, zaprinast, a cGMP phosphodiesterase inhibitor, and S-nitroso-N-acetylpenicillamine (SNAP), a nitric oxide generator. These results suggest that the ischemia-evoked $[^3H]NE$ release is mediated by NMDA receptors, and activation of NO system is involved.
산화질소는 생물체내에서 생리적이나 병리학적으로 중요한 역할을 한다고 알려져 있으며, 특히 침샘조직에서 침분비작용과 샘혈류 조절에 중요한 인자의 하나로 관여함이 알려져 있다. 산화질소합성효소 (nitric oxide synthase, NOS)는 동위효소로서 내피산화질소합성효소 (endothelial NOS, eNOS), 신경산화질소합성효소 (neuronal NOS, nNOS)와 유도산화질소합성효소 (inducible NOS, iNOS)가 있으며, 세포내에서 내인성산화질소를 합성한다고 알려져 있다. 그러나 산화질소합성효소의 세포내 분포에 관한 전자현미경적 연구는 매우 드물며, 흰쥐 침샘에서의 산화질소생산효소(NOS)에 대한 전자현미경적 연구는 없었다. 흰쥐 침샘에서 NOS의 세포내 분포를 규명하기 위하여 면역전자현미경방법을 이용한 금입자표지법을 시행하여 아래와 같은 결과를 얻었다. eNOS에 양성 면역반응을 보이는 구조는 침샘의 분비세포 중 장액세포에 있는 전자밀도가 높은 분비과립이었으며, 점액분비세포의 점액분비과립에서는 비교적 약한 면역반응성이 관찰되었다. 즉 턱밑샘과 혀밑샘을 구성하고 있는 두 종류의 분비세포 중 장액세포의 분비과립에 금입자가 비교적 많이 표지되었으며, 점액세포의 분비과립에서는 적은 수의 금입자가 관찰되었고, 침샘의 소엽속관 (intralobular duct)의 전자밀도가 높은 분비과립에서도 금입자가 표지된 것이 관찰되었다. 귀밑샘에서도 장액세포의 분비과립과 소엽속관의 분비과립에 금입자가 표지되었다. nNOS의 양성 면역반응은 턱밑샘에서 점액세포의 분비과립에서만 약간의 금입자가 관찰되었으며, 턱밑샘, 혀밑샘 및 귀밑샘의 분비세포와 분비관세포에서는 iNOS에 대한 양성 면역반응이 관찰되지 않았다. 흰쥐 침샘에서 산화질소합성효소 중 eNOS는 침샘분비세포의 분비과립에 존재하며, 특히 전자밀도가 높은 장액성분비과립에 주로 분포하고 있으며, 분비관 중에서 소엽속관에도 분포하고 있는 것이 관찰되었으나, 다른 동위효소인 nNOS와 iNOS는 거의 관찰되지 않았다. 산화질소합성효소가 흰쥐 침샘분비세포의 분비과립과 소엽속관의 분비과립에 분포하고 있는 것으로 보아 침샘에서 산화질소가 침의 생산과 분비에 중요한 역할을 담당하는 것으로 생각된다.

① 당 사이트는 회원이 서비스 이용내용에 있어서 본 약관 제 11조 내용을 위반하거나, 다음 각 호에 해당하는

- 기타 정상적인 서비스 운영에 방해가 될 경우
② 상기 이용제한 규정에 따라 서비스를 이용하는 회원에게 서비스 이용에 대하여 별도 공지 없이 서비스 이용의
일시정지, 이용계약 해지 할 수 있습니다.
제 17 조 (전자우편주소 수집 금지)
회원은 전자우편주소 추출기 등을 이용하여 전자우편주소를 수집 또는 제3자에게 제공할 수 없습니다.
제 6 장 손해배상 및 기타사항
제 18 조 (손해배상)
당 사이트는 무료로 제공되는 서비스와 관련하여 회원에게 어떠한 손해가 발생하더라도 당 사이트가 고의 또는 과실로 인한 손해발생을 제외하고는 이에 대하여 책임을 부담하지 아니합니다.
제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
