난과식물의 생약학적 연구 (Pharmacognostical Studies on Orchidaceae Plants)

  • In order to estimate accurate originality of the important crude drugs, Gastrodia, Dendrobium, Bletilla and Cremastra spp. were investigated comparing their morphological, anatomical and physicochemical characteristics and antibacterial, antifungal activities. The results of the studies as follows: 1. In morphological and anatomical studies, Korean Gastrodia contained more mucilage duct and symbiotic fungi than imported Gastrodia. Korean Dendrobium contained starch grains but without starch grains in the imported Dendrobium from China enclosed with thicken silicified wall. The corm of Cremasta appendiculata contained raphides of bundles with mucilage while the bulbs of Tulipa edulis contained several various starch grains form. 2. In physiological and TLC analysis, crude drugs in Orchidaceae contained common constituents with fluorescence and much mucilage. 3. The EtOH extracts of Gastrodia, Dendrobium, Bletilla, Cremastra showed antibacterial activities against B. subtilis and E. coli (Dendrobium>Gastrodia>Bletilla>Cremastra). But, no antifungal activities against C. albicans, A. niger were observed.

Antifungal Activity of Plumbagin Purified from Leaves of Nepenthes ventricosa x maxima against Phytopathogenic Fungi

  • A kind of naphthoquinone, plumbagin was purified and identified from the leaves of Nepenthes $ventricosa\;{\times}\;maxima$ through solvent extraction, silica gel column chromatography, and recrystallization. The yield (0.51%) was higher than that of the root of Plumbago scandens (0.26%), P. capensis (0.15%), and N. thorelii (0.092%). It exhibited antifungal activity against all plant pathogenic fungi tested, Alternaria alternata, Aspergillus niger, Bipolaris oryzae, Fusarium oxysporum, Phytophthora capsici, Rhizoctonia solani, Rhizopus stolonifer var. stolonifer and Sclerotinia sclerotiorum. The minimum inhibitory concentration values ranged from about 4.8 to $56.6\;{\mu}g/ml$ against the above eight fungi and R. solani was the most sensitive.

  • Glucoamylase from the culture filtrate of Aspergillus nicer was purified by ammonium sulfate precipitation, aceton precipitation, DEAE-cellulose ion exchange chromatography and Sephadex G-50 gel fillration. Glucoamylase was secreted into the medium upon growth on glucose, sucrose or a variety of other hexose sugars or hexose sugar polymers and little or no glucoamylase activity was found when glycerol or xylose was used as the carbon source. The optimum pH and temperature (or the maximum enzyme activity were found to be 5.0 and $50^{\circ}C$, respectively. The enzyme was considerably thermostable, for no loss of activity was observed when the enzyme was preincubated at $60^{\circ}C$ for 30 min. The enzyme activity was inhibited by 20 mM of $Hg^{2+}$, $Fe^{2+}$. The km value for starch was 0.045%.

고(문)서에 서식하는 미생물의 특성에 관한 연구 (Studies on the Characteristics of Microorganisms Isolated fro]m Ancient Archives)

  • Habitative microorganisms were isolated from the floated air and surface of record materials of library with ancient archives. The major ecological fungi from them were isolated and identified as Aspergillus niger, Aspergillus oryzae, Neurospora sitophile, Mucor mucedo, Mucor rouxii, Penicilliun notatum, Rhizopus delema, Rhizopus nigricans, Thamnidium elegans, and Tricoderma viridae. When the cellulase activity of fungi isolated from ancient archives and documents was analyzed, Mucor rouxii and Penicilliun notatum showed the highest avicelase and filter paper activity, to 18.089 and 2.819 units, respectively, showing destructive ability of old archives and documents. Whereas, Mucor mucedo revealed the highest CMCase activity of exoglucanase to 7.044 units.

pH가 곰팡이의 지질 축적과 지방산 조성에 미치는 영향 (Effect of pH on Lipid Accumulation and Fatty Acid Composition of Molds)

  • Effect of initial PH on the lipid accumulation and fatty acid composition of some species of mold were investigated. The maximum lipid content content produced by Aspergillus niger var. macrosporus was 17.5% at PH 3.5, by Aspergillus fumigatus 23.5% at PH 3.5 by Penicillium spinulosum 12.0% at PH 4.0 and by Penicillium notatum 7.3% at PH 4.0. The major fatty acids were palmitic, stearic, oleic and linoleic acid in all experimental molds. At PH from 4.5 to 6.0, the proportion of linoleic acid was increased and those of palmitic and oleic acid were decreased with rising in PH, therefore, degree of lipid unsaturation was increased in all experimental molds.

몇몇 음식 부식 균류의 방사선 감수성에 대한 연구 (Radiation Sensitivity of Some Food Decay Fungi)

  • Five species of food decay fungl, Aspergillus flavus, Asp. uiger, Penicillum sp., Botrytis cinerea, and Rhizopus stolonifer, were examined for their radio sensitivity in several suspension media. Asp. flavus, Asp. niger and Penicillum sp. have almost the same sensitivity toward gamma rays. with D value in the range of 30 to 35 K rad, whereas Botrytis cinerea has a D value of approximately 55K rad and Rhizopus stolonifer, the most re4sistant fungus studied, has a crease in their radioresistnace when compared with spores irradiated in water. Asp. flavus and penicillium sp. spores irradiated in citrate buffer at pH3-7 showed almost no change in their radisensitiity with pH, but Botryis cinerea spores showed a distinct decrease in their radioresistnace at pH 6 and 7. Penicillum sp. spores irradiated in sucrose solutions showed no sinificant change in their radioresistance. Botrytis cinerea spores displayed a higher radioresistance when they were irradiated in sucrose solution than in water.

두유 응고효소 생산 곰팡이의 분리 및 동정 (Isolation and Identification of the Fungi Producing a Soybean Milk Clotting Enzyme)

  • Twenty-five fungal strains producing an extracellular soybean milk clotting enzyme were isolated from 146 soil samples, and identified as 11 species belonging to six genera of Aspergillus oryzae (5 strains), Aspergillus flavus (2 strains), Aspergillus parasiticus (1 strain), Aspergillus tamarii (2 strains), Aspergillus niger (4 strains), Aspergillus fumigatus (2 strains), Mucor hiemalis (2 strains), Wallemia sebi (4 strains), Scopulariopsis condida (1 strain), Fusarium redolens(1 strain) and Verticillum lecanii (1 strain). Among them, Aspergillus oryzae 020 and Aspergillus tamarii 287 showed relatively high soybean milk clotting activity. The coagulabilities of the enzyme from representative strains of those species decreased as the pH of soybean milk increased from 6.0 to 7.0 The optimum temperature for soybean milk clotting enzymes of those strains were 65$^{\circ}C$.

한국산 Prunus 속 종자의 항균성과 청산배당체에 관한 연구 (A Comparative Study on the Antimicrobial Activities of the Seeds of Prunus Species)

  • HPLC analysis of three Prunus species, Armeniacae Semen. Persicae Semen, and Mume Semen, showed that 24. 06 mg/g, 5. 79 mg/g and 3/ 10 mg/g of amygdalin in the MeOH extract and 3. 59 mg/g, 5. 41 mg/g and 13. 48 mg/g of benzaldehyde in the MeOH extract hydrolyzed with ${\beta}-glucosidase$ were contained respectively. The MeOH extract of Mume Semen showed strong antibacterial activities against two species of bacteria (E. coli and B. subtilis) while that of Armeniacae Semen showed mild, and that of Persicae Semen showed no effect. Against A. niger, a strong antifungal activity was observed with Armeniacae Semen and only mild activities with Persicae Semen and Mume Semen. None oh the three inhibited the growth of S. cervisiae. The above results may possibly be suggestive of the correlationships between the contents of amygdalin and benzaldehyde in the test extracts with the antimicrobial potencies.

Influence of Garlic and Red Pepper on the Microflora of Kajami Shik-hae

  • Kajami Shik-hae processing consists of the fermentation of low salted Kajami (6% NaCl maximum) coated with a vegetable mixture, composed with cooked millet, red pepper, garlic and ginger. Lactic bacteria are the main component of the microflora. In order to determine their eventual selective role on the microflora the antimicrobial activity of garlic and red pepper was tested with some strain of bacteria and molds isolated from Shik-hae and Shik-hae raw materials. And the influence of their concentration in Kajami Shik-hae on the microflora was also checked. At the concentration of 10% garlic have no inhibitory activity against lactic bacteria but on strains of Bacillus, Micrococcus and Aspergillus niger. At the concentration of 20% red pepper showed a slight inhibitory activity on two strains of Bacillus. These results shows that red pepper and garlic are not only flavoring ingredients but they might play an important role in the control of the microflora growth and composition during Kajami Shik-hae fermentation.

  • Water extract, and methanol extract, its chloroform and hexane fractions, and estragole from Agastache rugosa O. Kuntze were tested to find the inhibition effect on the growth of several microorganisms. The organisms used were Escherichia coli ATCC 1129, Staphylococcus aureus IAM 1011, Vibrio parahaemolyticus WP, Bacillus subtilis ATCC6633, Aspergillus oryzae KFCC 890, Aspergillus niger KCCM 11240. Water and methanol extracts at the concentration of 0.5%, and chloroform and hexane fraction at the concentration of 0.05% inhibited the growth of microorganisms from 1/5 to 2/3 of the control group. Estragole identified from the hexane faction as a major component, its authentic compound completely inhibited the growth of Vibrio parahaemolyticus completely at the concentration of 0.03%, and the other bacteria were at 0.05%.
