약독 담배 모자이크 바이러스 I. 약독바이러스의 유기 및 특성 (Studies on Mild Mutants of Tabacco Mosaic Virus I. Induction of Mild Mutants and Their Characteristics)

  • TMV-OM계통을 접종하여 고온처리시킨 Samsun으로부터 3종의 약독변이계통 Tg 5272, Tw 227 및 Tw 333을 유기시켰다. 이들 약독바이러스는 담배 및 고추에 가벼운 mottling 증상이나 무증징으로 감염되어 TMV-OM에 의한 모자이크, 기형, 위축현상 등을 나타내지 않고 건전식물과 비교하여 생육에 거의 차이를 보이지 않았다. 또한 국부병반기주에 감염되었을 때에도 원주에 비해 전반적으로 미약한 반응을 나타냈고, 특히 Tw 227 및 Tw 333은 N. glutinosa, D. stramonium 및 G. globosa에서 매우 특이한 병징을 나타냈다. 약독바이러스의 물리적 성질 및 TMV-OM 항혈청에 대한 혈청학적 성질의 차는 인정되지 않았다. 한편 약독바이러스를 둔화하였을 때, 약독바이러스의 농도는 TMV-OM에 비해 매우 낮았으나, 이들 바이러스의 자외선흡수 및 전기영동패턴에는 차이를 보이지 않았다.

그라디오러스에 발생하는 BYMV와 CMV에 관한 연구 (Bean Yellow Mosaic Virus and Cucumber Mosaic Virus Causing Mosaic Disease on Gladiolus in Korea)

  • 그라디오러스에 발생하는 바이러스병을 포장에서 조사한 결과 이 병율이 $43.3\%$이었다. BYMV는 명아주에 엽맥괴사병징, 강남콩에 심한 모자익 병징 그리고 잠두에 모자익 병길을 일으켰다. CMV는 명아주에 국부병반, 담배와 오이에 모자익 병징을 일으켰다. 분류 동정된 두 바이러스는 모두 복숭아 혹진딧물에 의하여 전염되었다. 바이러스를 순화하여 흡광도를 측정한 결과 두 바이러스 모두 260nm에서 최고의 흡광도를 나타내었다. 한천내확산법을 이용한 항혈청과의 반응 결과 SDS를 처리한 BYMY와 CMV는 각각 그 바이러스의 항혈청과 뚜렷한 반응대를 형성하였다. 바이러스 입자의 진자현미경검경 과파 BYMV는 길이가 750nm였으며, CMV는 직경이 30nm이었다.

Acibenzolar-S-Methyl(ASM)-Induced Resistance against Tobamoviruses Involves Induction of RNA-Dependent RNA Polymerase(RdRp) and Alternative Oxidase(AOX) Genes

  • Tobamoviruses are the major viral pathogens of tomato and bell pepper. The preliminary results showed that Acibenzolar-Smethyl(ASM; S-methylbenzo(1,2,3) thiadiazole-7-carbothiate) pre-treatment to tomato and tobacco plants reduces the concentration of Tomato mosaic tobamovirus(ToMV) and Tobacco mosaic tobamovirus(TMV) in tomato and bell pepper seedlings, respectively. Pre-treatment of the indicator plant(Nicotiana glutinosa) with the ASM followed by challenge inoculation with tobamoviruses produced a reduced number and size of local lesions(67 and 79% protection over control to TMV and ToMV inoculation, respectively). In order to understand the mechanism of resistance the gene expression profiles of antiviral genes was examined. RT-PCR products showed higher expression of two viral resistance genes viz., alternative oxidase(AOX) and RNA dependent RNA polymerase(RdRp) in the upper leaves of the ASM-treated tomato plants challenge inoculation with ToMV. Further, the viral concentration was also quantified in the upper leaves by reverse transcription PCR using specific primer for movement protein of ToMV, as well as ELISA by using antisera against tobamoviruses. The results provided additional evidence that ASM pre-treatment reduced the viral movement to upper leaves. The results suggest that expressions of viral resistance genes in the host are the key component in the resistance against ToMV in the inducer-treated tomato plants.

Characteristics of Cucumber mosaic virus isolated from Zea mays in Korea

  • A virus causing mottle and stunt symptom on Zea mays was observed around Ulleng-do, Korea and identified as Cucumber mosaic virus (CMV-ZM) based upon biological, serological, and molecular characteristics. In host range studies, the CMV-ZM isolate produced local lesions on Datura stramonium, Vigna unguiculata, Cucurbita moschata, Chenopodium amaranticolor, Ch. quinoa, whereas this isolate produced systemic mosaic on Nicotiana tabacum cv. 'Xanthi-nc', Capsicum annuum, Solanum lycopersicum, Solanum melongena, Cucurbita pepo, and Z. mays. In addition, chlorotic local rings on inoculated leaves along with severe mosaic, malformation, and fern leaf symptoms on upper systemic leaves were shown in N. glutinosa plants. Complete nucleotide sequences of each genomic RNA segment was determined and compared to those of the other CMV strains. Comparison of the nucleotide sequence of 1a open reading frame (ORF) revealed approximately 89.2-92.4% sequence identity with each CMV subgroup IA and IB strain, while showing only 78% sequence identity with CMV subgroup II. Nucleotide sequence analysis of RNA2 ORFs revealed 85.3-97.6% sequence identity with subgroup I. In ORFs of RNA3, levels of nucleotide sequence identities were higher than 92-99.2% with CMV subgroup I and lower than 82% with CMV isolates of subgroup II. These results suggest that CMV-ZM isolate is more closely related to subgroup I than subgroup II and therefore, CMV-ZM isolate might be classified into as CMV subgroup I based on biological and molecular analysis.

Variation in the Pathogenicity of Lily Isolates of Cucumber mosaic virus

  • Two isolates of Cucumber mosaic virus (CMV) originated from lily plants, named Ly2-CMV and Ly8-CMV, were compared with their pathological features in several host plants. Ly2-CMV and Ly8-CMV could induce systemic mosaic symptom in Nicotiana benthamiana, but Ly2-CMV could not systemically infect tomato and cucumber plants that have been used for CMV-propagative hosts. While Fny-CMV used as a control infected systemically the same host plants, producing typical CMV symptoms. Ly8-CMV could infect systemically two species of tobacco (N. tabacum cv. Xanthi-nc and N. glutinosa) and zucchini squash (Curcubita pepo), but Ly2 failed systemic infection on these plants. As resulted from tissue-print immunoblot assay, different kinetics of systemic movement between Ly2-CMV and Ly8-CMV were crucial for systemic infection in tobacco (cv. Xanthi-nc). Sequence analysis of full-length genome of two lily isolates showed Ly2 and Ly8 belonged to subgroup IA of CMV. The lily isolates shared overall 98 % sequence identity in their genomes. Coat protein, 3a protein, and 2b protein involved in virus movement was highly conserved in genomes of the isolates Ly2 and Ly8. Although there is the low frequency of recombinants and reassortants in natural CMV population, phylogenetic analysis of each viral protein among a number of CMV isolates suggested that genetic variation in a defined population of CMV lily isolates was stochastically produced.

Inhibitory effects of the extract from Quercus dentata gallnut against plant virus infection

  • Pepper mild mosaic virus (PMMoV) and cucumber mosaic virus (CMV) are important pathogens in various vegetable crops worldwide. We have found that methanol extracts of Quercus dentate (Oaimyo Oak) gal/nut strongly inhibit PMMoV and CMV infection. Based on this result, the inhibitor named as "KN0912" formulated from the extract of Q. dentate gallnut was tested for its inhibitory effects on PMMoV or CMV infection to each local lesion host plant (Nicotiana glutinosa; PMMoV, Chenopodium amaranticolor; CMV). Pre-treatment effect of KN0912 against infections of each virus to local host plant was measured to be $75.1{\pm}0.5{\sim}97.5{\pm}1.5%$ to PMMoV and $70.6{\pm}2.2{\sim}99.0{\pm}1.0%$ to CMV in 1~10mg/ml conc. and the absorption effect of the antiviral composition of KN0912 to the inside of tobacco leaves tissue, was inhibited by 55.7% to PMMoV and 63.8% to CMV. The persistence of KN0912 treatment was maintained until after the 3 days high inhibitory effect by 98% to PMMoV and by 95.1% to CMV. Inhibitory effects on systemic host plants of KN0912 were measured to be 80~90% to PMMoV and 60~75% to CMV. From the change of morphological characteristics of PMMoV particles under EM, we are tentatively suggested that one mode of action of KN0912 is inactivation due to the destruction of virus particles.

무병종서 생산을 위한 감자X바이러스 및 엽권바이러스에 관한 연구 (Studies on the Potato Virus X and Potato Leaf Roll Virus for Disease-free Seed Potato Production)

  • A series of experiment was carried out to study on the production of disease-free seed potatoes at the Alpine Experiment Station from 1960 to 1968, which initiated a study of comparison on degeneration of plain warm region and high altitude products and the effect of latent potato virus X (PVX) and potato leaf roll virus(PLRV) on degeneration. Particular observations were made on some aspect of the nature of potato virus disease and its control such as concentrations of PVX, range of host plants, physical properties such as concentrations of PYX, range of host plants, physical properties and carrying effect of insects, by investigating 9 different areas of the main potato producing regions (Kimhae, Taegu, Choongju, Taejoen, Suwon, Kwangju, Chonju, Cheju and Chinju). Highly purified anti-serum was separated and tested for control of the virus disease and also various method of prevention and control of PLRV were observed, using cultivation of sprouted seed tubers, early harvesting method, and systemic chemicals. The results obtained are summarized as follows; 1. Potato yield in the plain region decreased by 32.8~66.3% in the first year cultivation of seed potatoes from colder region, and the rate of virus infection was 92.9 to 95.4%. 2. Plants of three families including, 20 species were susceptible to the PVX, and among the plants Salvia officinalis of a habits only was the carrier while the symptom of Digitalis purpurea of Screphulariaceae was masked. Necrosis and ring spot was occurred in most pJants of the Solanaceae and ring spot symptom also was observed in Nicotiana tabacum L. var. White Burley and in N. glutinosa. 3. The 8$C_2$ strain of virus had the following physical properties; thermal inactivation point, 68-$72^{\circ}C$ : dilution inactivation point, above 1, 000, 000 dilution: ageing in vitro, 240-360 days: and ageing in dry plant tissue, 30 days. 4. Myzus persicae and Oxya spp. did not transmit the 8$C_2$ strain of potato virus. 5. Virus was purified through the ammonium sulphate isolating method, and higher titer value, 1/2048 was obtained through anti-serum test. 6. Inhibition Chenopodiacae on the virus infection of potato was remarkable, and inhibition of local lesion host also was observed. 7, By earlier planting of sprouted seed tubers, growth period could be prolonged by 10 to 12 days. 8. Earlier harvest decreased much the rate of virus infection of seed potatoes. 9. According to the results of aphid control trial using systemic soil insecticides at Kangnung and Taekwanlyung, PSP 204, Disyston and Thimet was effective to aphid control. In particular, control effect of twice treatments of PSP 204 was great. 10. Treatmental effect of those chemicals lasted about 60-70 days. However, single foliar application of emulsified chemicals was not effective to potato virus control. 11. The effect of PSP 204, Disyston, and Thimet on the control of potato leaf roll virus was great, particularly in the case of two treatments of PSP 204, at Kangnung as well as at Taekwanlyung. Higher negative correlationship between the control effect of potato leaf roll virus and potato yield was observed showing the value r=-0.85 at Kangnung, and r=-0.87 at Taekwanlyung. 12. Differences in the control effects among PSP 204, Disyston, and Thimet was not noticed.

동부(Vigna sesquipedalis)에 발생하는 Blackeye Cowpea Mosaic Virus와 Cucumber Mosaic Virus에 관한 연구 (Blackeye Cowpea Mosaic Virus and Cucumber Mosaic Virus Causing Mosaic Disease on Asparagus Bean (Vigna sesquipedalis) in Korea)

  • 모자이크, 엽맥록대, 퇴록반점 등의 병징을 나타내고 있는 동부 (Vigna sesquipedalis)를 진주에서 채집하였다. 전자현미경에 의해 2종의 바이러스가 분리되어 이들의 기주범위, 조즙액중의 부활성화한계, 감염엽세포에 있어서 입자의 소재양식에 의하여 2종의 바이러스를 동정하였다. Blackeye cowpea mosaic virus (BICMV). 이 바이러스는 즙액접종에 의해 2과 7종의 검정식물에 감염이 확인되었다. 그러나 전신감염은 두과식물에 한했다. 내열성은 $50-60^{\circ}C$, 내희석성은 $10^{-4}-10^{-5}$, 내보존성은 1-6일이었다. 바이러스 입자의 형태는 길이 약 750nm의 사상형이다. 병엽 절편의 전자현미경적 관찰에서는 엽육세포의 세포질에서 사상 입자와 풍차상 및 환상의 봉입체가 확인되었다. Cucumber mosaic virus(CM V), 이 바이러스는 즙액접종에 의해 3과 9종의 검정식물에 감염이 확인되었다. 다른 CM V계와 달리 두과식물에도 전신감염을 했다. 이 바이러스 임자의 형태는 직경약 30nm의 입자이다. 감염엽의 전자현미경적 관찰에서는 엽육세포의 세포질 및 액포내에서 바이러스 입자의 응집상이 확인되었다.

목질진흙버섯(Phellinus linteus) 균사체 추출물 제제 PLM-WE1의 식물 바이러스에 대한 감염억제 효과 및 활성성분의 동정 (Inhibitory Effects of PLM-WE1 Formulated from Extract of Phellinus linteus Mycelium against Plant Viruses Infection and Identification of Active Compound)

  • 목질진흙버섯(Phellinus linteus)을 액체 배양한 균사체의 열수 추출물을 이용하여 PMMoV 및 CMV에 대한 감염억제효과를 검정한 결과, PMMoV 및 CMV에 높은 감염억제효과가 발견되었다. 이 결과를 토대로, P. linteus 균사체의 열수추출물을 주성분으로 제제화한 PLM-WE1의 효능 평가를 위하여, PMMoV 및 CMV의 국부감염 기주식물에서 PLM-WE1의 바이러스 접종전 처리효과, 엽이면 처리시의 감염억제효과 및 PLM-WE1의 지속효과 실험을 수행하였다. 그 결과, PLM-WE1(처리농도 10 mg/ml)의 바이러스 접종전 처리시의 감염억제율은 PMMoV 99.2%와 CMV 80.3%이었으며, 본 제제(농도 5 mg/ml)의 엽이면 처리시의 감염억제율은 PMMoV 45.0%와 CMV 41.9%, 약효지속성은 처리 3일 후까지 PMMoV 75%와 CMV 62% 수준으로 감염억제 효과를 보였다. 바이러스 전신감염기 주식물을 이용한 감염억제효과는 PMMoV 75~85%와 CMV 75%이었다. PLM-WE1제제의 바이러스 감염억제 기작을 해석하기 위하여, 정제한 PMMoV에 제제를 혼합하여 전자현미경으로 관찰한 결과, 바이러스 입자의 형태적 변화는 관찰되지 않아, PLM-WE1의 바이러스 감염억제기작은 바이러스입자의 응집(aggregation) 작용과는 다른 기작에 의한 것임이 추정되었다. 목질진흙버섯 균사체로부터 식물바이러스 감염억제 성분의 분리 및 동정을 위하여 유기용매분획, 산 가수분해 및 에탄올 분별침전을 실시하여 고활성 분획을 수득하였다. 이 분획물을 $^1H$-NMR 및 $^{13}C$-NMR을 이용하여 분석한 결과, $\alpha$- 또는 $\beta$-glucan이 결합된 다당류(polysaccharide)로 확인되었다.

칸나에서 분리한 Cucumber mosaic virus의 특성 (Characterization of an Isolate of Cucumber mosaic virus Isolated from Canna generalis Bailey)

  • 전형적인 줄무늬 모자이크 증상을 나타낸 칸나로부터 Cucumber mosaic virus(CMV)의 한 계통(Can-CMV)을 분리하고, 특성을 조사하였다. Can-CMV는 대부분의 전신감염 기주에서 병징이 발현되지 않았고, 이들 기주의 접종엽 및 상엽에서 RT-PCR로 바이러스의 감염여부를 조사한 결과, N. benthamiana와 N. glutinosa에서만 바이러스가 검출되었으며, 다른 기주로부터는 확인되지 않았다. 한편 Chenopodium amaranticolor의 접종엽에 발현된 국부 괴사병반은 대조로 접종한 Fny-CMV나 LS-CMV에 의해서 형성된 병반보다 매우 작은 크기의 반점을 형성하는 특성을 보였다. 또한 Vigna unguiculata의 접종엽에는 Fny-CMV나 LS-CMV가 접종 2-3일 후에 뚜렷한 괴사병반을 형성하는데 반해서, Can-CMV는 접종 4-5일 후에 윤곽이 확실치 않은 퇴록병반을 형성하였다. Can-CMV에 감염된 N. benthamiana로부터 추출한 dsRNA는 4종의 밴드가 검출되었으며, 이들 분자의 크기 및 종수는 Fny-CMV나 LS-CMV와 차이를 나타내지 않았다. Can-CMV의 항원은 Fny-CMV의 항혈청에 대해서 한 종의 뚜렷한 침강선을 형성하였으며, Fny-CMV의 항원에 의해서 형성된 침강선과 융합하였고, LS-CMV의 침강선과는 분지선을 형성함으로서, 혈청학적으로 서브그룹 I에 속하는 계통으로 판단되었다. 또한 Can-CMV에 감염된 N. benthamiana로부터 RNA를 추출하여 CMV-specific 프라이머를 이용한 CMV-RNA3의 외피단백질 유전자를 포함하는 3'영역에 대해서 RT-PCR을 실시하였다. Can-CMV는 Fny-CMV나 LS-CMV와 마찬가지로 예상되었던 약 950bp크기의 cDNA가 증폭되었으며, 이 cDNA를 EcoRI으로 처리하였을 때에는 절단되지 않았고, MspI에서는 595bp 및 350bp의 절편으로 절단되었다. 이와 같은 결과는 Fny-CMV의 패턴과 일치하였으며, 이러한 결과로부터 Can-CMV가 서브그룹 IA에 속하는 계통으로 확인되었다. 이상과 같은 결과들로부터, Can-CMV는 앞으로 바이러스와 기주의 다양한 상호관계를 이해하는데 있어서 중요한 병원학적 성질을 가지고 있는 것으로 생각되었다.
