Serological Analysis of Sonchus Yellow Net Virus Proteins in Infected Nicotiana edwardsonii Leaf Tissues (Sonchus Yellow Net Virus에 감염된 Nicotiana edwardsonii 잎으로부터의 바이러스 단백질의 혈청학적 분석)

  • Antibodies were raised against fusion proteins of the N-terminus and a region containing the GDNQ (Gly-Asp-Asn-Gln) polymerase motif of the L (polymerase) protein of sonchus yellow net virus (SYNV). Immunoblot analyses using these antibodies revealed the presence of the L protein in purified SYNV preparations and in nuclear extracts from infected tobacco. The serological analyses and detection in a polyacrylamide gels suggested that the L protein is present in at least a 20 fold lower abundance than the G, N, M1 and M2 proteins, and has size corresponding to a molecular weight of over 200 kDa as predicted from nucleotide sequence data. Electron microscopy with gold-labelled antibodies was used to localize the N, M2, and G proteins of SYNV in thin sections of infected tissue. When sections of SYNV-infected tissue were treated with antisera against total SYNV proteins and N protein, gold label could be detected in both the viroplasms and in virus particles. With the anti-M2 protein antiserum, the gold label was strongly localized in the viroplasms but only limited labelling of the virus particle sonly. Limited labelling of the L protein was observed in the viroplasms and the virus particles, presumably because of the low abundance of L protein in the tissues.

  • The reaction of seven cultivars of Nicotiana tabacum to eight naturally occurring strains of potato virus Y from tobacco and one from potato was determined by mechanical inoculations in greenhouse tests. Dixie Bright 244-2, McNair 3D, and Golden Stock Penish were highly tolerant to three mild strains, two from the United States and one from Korea, and to four severe strains, one each from the United States, West Germany, South Africa, and Korea. They also had some tolerance to a severe strain from Child and one from United States. Virus concentration in infected leaf tissue was virus strain-and cultivar-dependent.

A Super-Absorbent Polymer Combination Promotes Bacterial Aggressiveness Uncoupled from the Epiphytic Population

  • Plant leaf surface is an important niche for diverse epiphytic microbes, including bacteria and fungi. Plant leaf surface plays a critical frontline defense against pathogen infections. The objective of our study was to evaluate the effectiveness of a starch-based super-absorbent polymer(SAP) combination, which enhances water potential and nutrient availability to plant leaves. We evaluated the effect of SAP on the maintenance of bacterial populations. In order to monitor bacterial populations in situ, a SAP mixture containing Pseudomonas syringae pv. tabaci that expressed recombinant green fluorescent protein(GFPuv) was spray-challenged onto whole leaves of Nicotiana benthamiana. The SAP combination treatment enhanced bacterial robustness, as indicated by disease severity and incidence. Unexpectedly, bacterial numbers were not significantly different between leaves treated with the SAP combination and those treated with water alone. Furthermore, young leaves treated with the SAP combination had more severe symptoms and a greater number of bacterial spots caused by primary and secondary infections compared to young leaves treated with the water control. In contrast, bacterial cell numbers did not statistically differ between the two groups, which indicated that measurement of viable GFP-based bacterial spots may provide a more sensitive methodology for assessing virulence of bacterial pathogens than methods that require dilution plating following maceration of bacterial-inoculated leaf tissue. Our study suggests that the SAP combination successfully increased bacterial aggressiveness, which could either be used to promote the ability of biological agents to control weedy plants or increase the robustness of saprophytic epiphytes against competition from potentially harmful microbes.

Effects of Phytohormone and Activated Carbon on the Growth and Rooting of Teratoma Shoot Induced from Crown Gall Callus in Nicotiana tabacum cv. NC 2326 (연초 Crown Gall Callus유래 Teratoma Shoot의 생육과 발근에 미치는 식물호르몬과 활성탄의 영향)

  • These studies were conducted to investigate effects of phytohormone and activated carbon on the growth and rooting of teratoma shoots induced from Nicotiana tabacum cv. NC2326 transformed by Aerobacterium tumefaciens C58. GA was effective for shoot elongation and reduction of multiple shoots from teratoma shoot, however, leaves of teratoma shoot cultured on the medium with GA were pointed. ABA was also effective in promoting shoot elongation, but was not for reduction of multiple shoots. Teratoma shoot cultured on the medium with 1 n activated carbon promoted shoot elongation and inhibited the number of shoots differentiated, but was grown as abnormal shoot. Addition of 1% activated carbon and 0.5mg/l BA to culture media was effective for shoot elongation and reduction of multiple shoot and for formation of round leaves as normal leaves. Though these shoots were inoculated on the rooting medium, they could not from roots but formed multiple shoots. Boric acid, myo-inositol and sucrose were also ineffective on the rooting of teratoma shoots.

Protoplast Isolation and Fusion of Nicotiana glauca and Solanum tuberose Transformed by Selectable Marker Genes (표지유전자로 형질전환된 연초와 감자로부터 원형질제의 유리 및 융합)

  • Protoplasts were isolated from mesophyll of tobacco(Nicotiana glauca) transformed with kanamycin-resistant gene (NPT II gene) and potato hairy root callus containing Ri plasmid of Agrobacterium rhiEogenes, and protoplasm fusion was made between the isolated protoplasts. The transgenic tobacco leaf tissue could grow on the media containing high concentrations of kanamycin, but not on the phytohormone-free media. On the other hand, the potato hairy root calli could be cultured on the phytohormone-free media but not on media containing more than 40 ㎍/ml kanamycin. In these conditions, the viability of both protoplasts were above 90%, These selection markers were used for the selection of protoplasts fused between the two, i.e. protoplast fusion was detected using selection media containing 100㎍/ml kanamycin and with no phytohormone. The mixture of 1.0% cellulase, 0.3% macerozyme, and 0.7M mannitol was best for the maximum protoplast production for tobacco, and that of 2.0% cellulase, 2.0% macerozyme, 1.0% dricelase, and 0.5M mannitol for potato. Both tobacco mesophyll and potato callus protoplasts were fused by using PEG solution on the selectable medium. Cell walls were regenerated after 5 days in this medium, and colonies were alive until 4 weeks after cultural, but died after 6 weeks.

  • In situ recovery of recombinant human granulocyte-macrophage colony stimulating factor (hGM-CSF) was performed in transgenic Nicotiana tabacum cell suspension cultures. Aqueous two-phase systems (ATPS) were used to utilize its biocompatibility. Transgenic plant cells could be grown up to 15.7 g/L in normal medium and 18.6 g/L in ATPS composed of 6% (w/w) polyethylene glycol (PEG) 20,000 and 10% (w/w) dextran 2,000,000. It was proved that the A TPS was not harmful to cell growth. In addition, It is expected to recover hGM-CSF simultaneously with cell growth. Using this method, maximum hGM-CSF concentration at 1.64 ng/mL was obtained on day 3.

  • Transgenic Nicotiana tabacum cells were cultivated for the production of murine granulocyte macrophage-colony stimulating factor (mGM-CSF) in both a stirred tank bioreactor and an airlift bioreactor with draft tube. Cell growth and mGM-CSF production in the airlift bioreactor were found to be better than those achieved in the stirred tank bioreactor. In the airlift bioreactor, 9.0g/L of cells and 2.2ng/mL of mGM-CSF were obtained (11.0g/L and 2.4ng/mL, respectively in shake flasks). Although the lag period was prolonged and mGM-CSF production was lowered by 33% in the stirred thank bioreactor as compared to the control culture, the maximum cell density was increased up to 12.0g/L due to better mixing by agitation at the higher cell density.

  • Nicotiana Tabacum L., cv. NC2326, Burley2l and Sohyang seeds were coated with different five materials - zeolite, serpentine, coal briquet ash, serpentine+zeolite and coal briquet ash+zeolite - to investigate the effects of coating on germination. The coating materials had different characteristics and gave varied effects on germination. Among three tobacco varieties, coated seeds of Burley2l showed the most sensitive responses in germination rate of decreasing and delaying at low temperature on seedbed and laboratory tests.

  • Field test were conducted to select the best method for soil phosphorus test and to obtain the critical levels of each method for P fertilizer recommendations based on tobacco production. Truog method was recognized to be the mort suitable one for soil P test. N. Carolina, Bray No.1 and Olsen methods were followed in order. Bray No. 2 and Lancaster methods showed the least satisfaction. The critical levels of available P by lancaster, Bray No. 1, N. Carolina,01sen, and Truog methods were 55ppm, 32ppm, 21ppm, 19ppm, and 22ppm, respectively. Below the critical level a large yield response can be expected with 10kg $P_2O_5$/10a, above critical level little or no response is expected. In soils the more the initial available p, the more available P was carried over, while the more the total amount of silt and clay, the less the residual P was found. The increase in residual P by 10kg of $P_2O_5$ was greater than that of 5kg/10a. The chemical composition and the price of the cured leaf were not affected by the $P_2O_5$ application, indicating that the increase in the rate of $P_2O_5$ application would not exert on the quality of tobacco.

Correlation Coefficient of Leaf Breakdown with Some Agronomic Characters in Flue- Cured Tobacco (Nicotiana tabacum L.) (황색종 연초(Nicotiana tabacum L.)에서 엽탈락과 농경적 형질과의 상관)

  • This study was conducted to obtain the breeding information for the leaf breakdown, and correlation coefficients between leaf breakdown and some agronomic characters in flue-cured tobacco at Taegu Experiment Station, Korea Ginseng & Tobacco Research Institute. NC 95 and NC 2326 were used as parents. The bulk method procedure was used from the F2 to the F4 generations, 54 plants were selected among F4, and 11 lines were selected among 54 F5 which based on high resistance to bacterial wilt compared with NC2326. The entries were composed of parents and 11 F6 advanced lines, and designed in randomized block with 3 replications. Plant seedlings were transplanted in 16 April. The number of breakdown leaf were investigated at flowering stage, 18 June. There were significant difference between NC 95 and NC 2326 for the number of breakdown leaf, number and weight of ground suckers, days to flower, yield and resistance to bacterial wilt, and similar results were appeared among lines. Estimated heritability in the broad-sense ranged from 59.3% to 87.4% for all characters. Most of genetic correlations were higher than phenotypic coefficients between characters. Number of breakdown leaf was positively correlated with days to flower, the number and weight of ground suckers, while it was negative to yield. One line among 11 had few breakdown leaf, low producted ground suckers and high resistance to bacterial wilt. Above results suggest that the number and weight of ground suckers can be use as a selection criterion for leaf breakdown even under the condition without leaf breakdown.
