Comparative study on the cellular activities of osteoblast-like cells and new bone formation of anorganic bone mineral coated with tetra-cell adhesion molecules and synthetic cell binding peptide

  • Purpose: We have previously reported that tetra-cell adhesion molecule (T-CAM) markedly enhanced the differentiation of osteoblast-like cells grown on anorganic bone mineral (ABM). T-CAM comprises recombinant peptides containing the Arg- Gly-Asp (RGD) sequence in the tenth type III domain, Pro-His-Ser-Arg-Asn (PHSRN) sequence in the ninth type III domain of fibronectin (FN), and the Glu-Pro-Asp-Ilu-Met (EPDIM) and Tyr-His (YH) sequence in the fourth fas-1 domain of ${\beta}$ig-h3. Therefore, the purpose of this study was to evaluate the cellular activity of osteoblast-like cells and the new bone formation on ABM coated with T-CAM, while comparing the results with those of synthetic cell binding peptide (PepGen P-15). Methods: To analyze the cell viability, 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide assay was performed, andto analyze gene expression, northernblot was performed. Mineral nodule formations were evaluated using alizarin red stain. The new bone formations of each group were evaluated using histologic observation and histomorphometrc analysis. Results: Expression of alkaline phosphatase mRNA was similar in all groups on days 10 and 20. The highest expression of osteopontin mRNA was observed in the group cultured with ABM/P-15, followed by those with ABM/T-CAM and ABM on days 20 and 30. Little difference was seen in the level of expression of collagen type I mRNA on the ABM, ABM/T-CAM, and ABM/P-15 cultured on day 20. There were similar growth and proliferation patterns for the ABM/T-CAM and ABM/P-15. The halo of red stain consistent with $Ca^{2+}$ deposition was wider and denser around ABM/T-CAM and ABM/P-15 particles than around the ABM particles. The ABM/T-CAM group seemed to have bone forming bioactivity similar to that of ABM/P-15. A complete bony bridge was seen in two thirds of the defects in the ABM/T-CAM and ABM/P-15 groups. Conclusions: ABM/T-CAM, which seemed to have bone forming bioactivity similar to ABM/P-15, was considered to serve as effective tissue-engineered bone graft material.

Assessment of Bond Characteristics between New and Old Concrete in Various Mixtures and Joint Conditions (배합 및 접합면 처리에 따른 신·구 콘크리트의 부착특성 평가)

  • Although the construction joints of a concrete structure are properly treated with some measures, leakage has frequently occurred. A series of tests on the bond characteristics between new and old concrete were carried out in this study, assuming that the leakage at the construction joints has certain relationship with the bond of concrete. To assess the bond characteristics under various conditions, a number of specimens were made that have an interface between new and old concrete and bond strength, flexural strength and splitting tensile strength were measured. Main test variables are type and amount of mineral admixtures, treatment method of the interface and type of waterstops. In addition, the effects of placing interval between the concrete and of the age of the strength tests were investigated. The test results showed a slightly increased bond strength when applying mineral admixtures, which can be attributed to the interface filled with the calcium silicate hydrate that is formed by pozzolanic reaction. On the other hand, the bond strength was higher when the interface was treated rough and dry, and the roughness of a waterstop affected the bond capacity of the waterstop. Also, an assessment is required that considers the type of strength test because the bond strength varied according to the test methods.

『Bonchojeonghwa(本草精華)』, Medical Historical Approach to Bibliographic Notes (『본초정화(本草精華)』의 해제(解題)에 관한 역사학적(醫史學的) 접근)

  • The currently existing "Bonchojeonghwa (本草精華)" is a manuscript without the preface and the epilogue, composed of 2 books in 2 volumes. This book is a quintessence of knowledge on science of medicinal ingredients (medicinal phytology I herbal science) as well as an trial of new development in Chosun medical science. I.e. this book includes surprising change representing medical science in Chosun dynasty as a single publication on science of medicinal ingredients. It holds a value essential to clinician as a specialized book in medicinal ingredients, and Includes richer content on medicinal ingredients than any other books published before. In addition, it is away from boring list-up of superfluous knowledge as seen in "Bonchokangmok(本草綱目)" published in China, and well summarizes essential knowledge which can be used within a range of medicines available in Korea. This book has an outstanding structure that can be even used in today's textbook on science of medicinal ingredients, as it has clear theory, system and classification. Because it handles essential learning points prior to prescription to disease, it is possible to configure new prescription and adjustment of medicinal materials. Moreover, this book can play a good role for linguistic study at the time of publication, because it describes many drugs in Hangul in many parts of the book. "Bonchojeonghwa" includes a variety of animals, plants and mineral resources in Korea, like "Bonchokangmok" which was recently listed in UNESCO. As such, it has a significance in natural history as well as pharmacy in Korean Medicine. It has various academic relationships all in biologic & abiologic aspects. It has importance in sharing future biological resources, building up international potential, setting up the standard for biologic species under IMF system, and becoming a base for resource diplomacy. We should not only see it as a book on medicinal ingredients in terms of Oriental Medicine, but also make an prudent approach to it in terms of study strengthening Korea's national competitiveness. After bibliographical reviewing on the features & characteristics of the only existing copy of "Bonchojeonghwa" housed in Kyujanggak(奎章閣) of Seoul National University, the followings are noted. First, "Bonchojeonghwa" is a specialized book on medicinal ingredients voluntarily made by private hands to distribute knowledge on drugs in the desolate situation after Imjinoeran (Japanese Invasion in 1592), without waiting for governmental help. Second, it raised accessibility and practicality by new editing. Third, it classified 990 different kinds of drugs into plant, animal, and mineral at large, and dassified more in detail into 15 'Bu' and 48 'Ryu' at 258 pages. Fourth, the publication of this book is estimated to be around 1625~1633, at the time of Injo's reign in 17th century. Fifth, it contains the existing & up-to-date knowledge at the time of publication, and it is possible to see the supply-demand situation by Hangul descriptions in 149 places in the book. By the fact that there are many linguistic evidences of 17th century, explains well when the book was published.

New site classification system and design response spectra in Korean seismic code

  • A new site classification system and site coefficients based on local site conditions in Korea were developed and implemented as a part of minimum design load requirements for general seismic design. The new site classification system adopted bedrock depth and average shear wave velocity of soil above the bedrock as parameters for site classification. These code provisions were passed through a public hearing process before it was enacted. The public hearing process recommended to modify the naming of site classes and adjust the amplification factors so that the level of short-period amplification is suitable for economical seismic design. In this paper, the new code provisions were assessed using dynamic centrifuge tests and by comparing the design response spectra (DRS) with records from 2016 Gyeongju earthquake, the largest earthquake in history of instrumental seismic observation in Korea. The dynamic centrifuge tests were performed to simulate the representative Korean site conditions, such as shallow depth to bedrock and short-period amplification characteristics, and the results corroborated with the new DRS. The Gyeongju earthquake records also showed good agreement with the DRS. In summary, the new code provisions are reliable for representing the site amplification characteristic of shallow bedrock condition in Korea.

The Changing Patterns of Demand-Supply and Role of Mineral Resources in Economic Growth during Industrialization of the Republic of Korea (한국공업화과정(韓國工業化過程)에서의 광물자원(鑛物資源)의 수급구조변화(需給構造變化)와 경제성장(經濟成長)에 있어서의 역할(役割))

  • A total of 12 mineral commodities significant in domestic output, economy and/or strategy of the Republic of Korea are chosen to examine the structural changes in production and demand-supply of these minerals during the last two decades of her industrialization. These include iron and manganese ores as the raw materials for iron and steel making, copper, zinc and tungsten ores among other non-ferrous metallic minerals, limestone (cement), kaolin, talc, pyrophyllite and graphite among other non-metallic minerals, and anthracite coal as the only domestic source of fossil energy. These are reviewed historically in time-series based on the statistical data which are tabulated and graphed in terms of domestic output, export, import, apparent demand-supply, its increasing rate, and self-sufficiency rate of each commodity. The increasing rates of demand-supply (IRDS) of some more important commodities are compared with those of Gross Domestic Production (GDP) and Economic Growth Rate (EGR) to evaluate how the IRDS contributed to the GDP and EGR. The major results revealed are as follows: Among the 12 commodities, the domestic output of 8 commodities appeared to have grown with steady upward trends: they are ores of lead, zinc and tungsten, limestone (cement), kaolin, talc, pyrophyllite and anthracite coal. Two commodities, ores of iron and copper, continued with unchanging or slightly declining trends and varied fluctuations, in spite of their cardinal importance to the heavy industry and strategy of Korea. The remaining two, graphite and manganese ore, have gradualy declined in domestic output in which the former has still enough resource potential but the latter has not and virtually ceased its domestic output. Trade patterns for mineral commodities in the Republic of Korea during the last two decades have changed greatly, being marked by a shift from mineral-exporting to mineral importing, mainly because of increasing consumption of mineral raw materials for industrialization rather than beceuse of decreasing output of domestic mineral commodities in quantity. In terms of trade patterns, the 12 commodities concerned in this study can be classified into the following four groups. The 1st group - ores of lead and tungsten have only been exported without imports. The 2nd group - amorphous graphite, and pyrophyllite have mainly been exported but partly been imported. The 3rd group - kaolin, talc and crystalline graphite have equally been exported and imported, but quantity of imports have rapidly been increased with time. The 4th group - ores of iron, manganese and zinc have shifted from exports to imports during the industrialization, particularly owing to the initiation of iron and steel making by the Pohang Iron and Steel Company in the middle 1970' s and the new establishment of the Onsan Zinc Refinery in the late 1970' s. All of the 12 commodities under considerations were far above 100% in self-sufficiency rate before or in the early 1960' s. Recently, however, most of them have been declined to below 100% except for those of limestone (cement) and pyrophyllite. It is particularly serious to identify that the self-sufficiency rates of the three important metallic minerals, iron, copper and manganese ores in 1982 appeared to be 5.1%, 0.5%, and 0.01%, respectively. The average self-sufficiency rate of the total domestic minerals produced in 1982 was 14.4% (in value) for that year. Mining industry appeared to be extremely high in its intermediate demand rate whereas its intermediate input rate to be quite low indicating that mineral raw materials have been exerted strong forward linkage effects upon the other industries rather than backward linkage effects. In comparing the curves of increasing rates of demand-supply of several major minerals - iron ore, manganese ore, copper ore, limestone (cement), kaolin, and anthracite coal - with those of Gross Domestic Production and Economic Growth Rate drawn on every graph, it is clearly shown that the curves of increasing rates of demand-supply comprise around 6 to 7 periods of cycles which roughly harmonious with those of the curves of GDP and EGR, except for the curve of anthracite coal of which the configuration seems to have resulted from the (artificial) government's mineral policy rather than from economic free market mechanism. The harmonic feature of these curves well suggests that the increasing rates of demand-supply of major minerals have been significantly contributed to the GDP and EGR. In addition, the wider amplitudes of the iron, manganese and copper curves than those of the limestone (cement) and kaolin curves indicate that the contribution of the former, metallic commodities, has been greater than that of the latter, non-metallic commodities.

Development Status and Research Direction in the Mineral Carbonation Technology Using Steel Slag (제철 슬래그를 이용한 광물 탄산화 기술의 개발 현황과 연구 방향)

  • In the present paper, we investigated the development status of precipitated calcium carbonate (PCC) production using steel slag, which is one of mineral carbonation (MC) technologies, from the standpoint of $CO_2$ utilization. Principle, feature, and global and domestic development status of the mineral carbonation technology were discussed together with the overview of the production method and market of PCC. Mineral carbonation is known as stable and environmentally-friendly technology enabling economical treatment of industrials wastes. Typically, PCC is produced by the reaction of $CO_2$ with supernatant solution after Ca extraction from steel slag followed by the separation of solid and liquid. The development status of MC using steel slag is at the pilot stage (Slag2PCC at Aalto University), and there remains the process economics improvement for commercialization. Key technologies for the further development are efficient extraction of Ca ions from steel slag including impurities removal, valorization of PCC via shape and size control, usage development and value-addition of residual slag, and optimization of reaction conditions for continuous process setup, etc.

Effects of the decoction water on the extraction of the bioactive compounds from rhubarb (전탕용수의 종류에 따른 대황 성분의 추출효과)

  • The quality of decoction water would be important for the preparation of herbal medicines. Four types of decoction water, S1, S2, S3 and S4, three types of mineral water from Jeju island and one type of tap water from Seoul region were evaluated. The contents of minerals in decoction water were analysed by ICP-AES and ICP-MS. There was significant difference in the mineral contents between the types of decoction water. The bioactive components from rhubarb were extracted with four types of water and the relationship between mineral contents in water and the extracted amounts of components was evaluated. The total extraction was calculated by evaporating water and weighing the residue. The bioactive components in rhubarb were determined by HPLC method. Kruskall-Wallis rank sum tests were used. Multiple factor analysis was used to analyze the relation between the contents of mineral and total extraction or bioactive components in the decocted solutions. Not only the total extraction but also the amount of extracted bioactive components showed statistically significant relationship with the contents of minerals and anions in decoction water.

Stratigraphy, Lithology and Diagenetic Mineral Facies of the Tertiary Yeonil Group (제 3기 연일층군의 층서, 암상 및 속성 광물상)

  • In the Heunghae area, genetic relationships among sedimentary facies, lithology, stratigraphy and diagenetic mineral facies of the Yeonil Group, are discussed. Conglomerate and sandstone of lower to middle parts of the Yeonil Group contain considerable amounts of volcaniclastic sediments, which were derived from the Tertiary volcanics exposed in the western margins of the sedimentary basin. A new stratigraphic division of the Yeonil Group into the Chunbuk and Pohang Formations is proposed on the basis of sedimentary facies, lithologic characteristics including volcaniclastic feature, and the presence of a key bed of siliceous mudstone overlying the Chunbuk Formation. Diagenetic mineral facies largely depend on the lithology and composition of sediments. Heulandite, smectite, calcite, and opal-CT are commonly found as diagenetic minerals in the Yeonil Group. Among these authigenic minerals, heulandite occurs as the coarse- grained main cement in conglomerates and sandstones of the Chunbuk Formation. Formation of the zeolite cement is favored by partial volcaniclastic lithology of the Chunbuk Formation. Smectite composition and diagenetic mineral facies such as heulandite and opal-CT may reflect that the Yeoil Group has undergone a shallow rial temperature ranging $40{\~}60^{\circ}C$.

Construction of Open-source Program Platform for Efficient Numerical Analysis and Its Case Study (효율적 수치해석을 위한 오픈소스 프로그램 기반 해석 플랫폼 구축 및 사례 연구)

  • This study constructed a new simulation platform, including mesh generation process, numerical simulation, and post-processing for results analysis based on exploration data to perform real-scale numerical analysis considering the actual geological structure efficiently. To build the simulation platform, we applied for open-source programs. The source code is open to be available for code modification according to the researcher's needs and compatibility with various numerical simulation programs. First, a three-dimensional model(3D) is acquired based on the exploration data obtained using a drone. Then, the domain's mesh density was adjusted to an interpretable level using Blender, the free and open-source 3D creation suite. The next step is to create a 3D numerical model by creating a tetrahedral volume mesh inside the domain using Gmsh, a finite element mesh generation program. To use the mesh information obtained through Gmsh in a numerical simulation program, a converting process to conform to the program's mesh creation protocol is required. We applied a Python code for the procedure. After we completed the stability analysis, we have created various visualization of the study using ParaView, another open-source visualization and data analysis program. We successfully performed a preliminary stability analysis on the full-scale Dokdo model based on drone-acquired data to confirm the usefulness of the proposed platform. The proposed simulation platform in this study can be of various analysis processes in future research.

Setting and Compressive Strength Development of Hihg Flowing Concrete (고유동 콘크리트의 응결특성과 압축강도 발현)

  • High flowing concrete has been made using a combination of different cementitious material. The use of supplementary cementitious material like ground granulated slag is not only interesting from an economical point of view but also from a mechanical and rheological point of view. In the case of high strength concrete, relation between the maturity and compressive strength development of high strength concrete is aproximated by appling gompertz curve and suggested new estimating method. It is the aim of this study to analysis the effect of different types of mineral fine power on the setting and compressive strength development of high flowing concrete.
