한국산 Raja속 어류의 분류학적 연구

  • Electrophoretic analysis was performed to determine the interspecific genetic relationships of the genus Raja collected in Korea waters. We confirmed 5 species [Raja (Okamejei) meerdervoortii Bleeker, R.(O).acutispina Ishiyama, R.(O) kenojei Muller et Henle, Raja (Dipturus) kwangtungensis Chu, and R.(D.) pulchra Liu] inhabting in Korea. As results of starch gel electrophoresis, 24 genetic loci were obtained from 13 enzymic and non-enzymic proteins, and allele frequencies of each locus were calculated . Even thought their various dorsal color patterns, the mean genetic similarity among 4 conspecific populations of R.(O) kenojei showed highly close genetic relationships (S=0.966) R.(O) kenojei and R.(D) kwangtungensis which belong to different subgenus , revealed subspecific level of differentiation (S=0.829). R. (Diptuuus ) kwangtungensis was genetically much closer to Raja(Okamejei) species than to Raja (Dipturus) species group. R.(O) acutispina and R.(D) kwangtungensis were unrecorded species to Korea.

Anti-allergic effect of Cortex Mori (상백피(桑白皮)의 항알레르기성 효과)

  • Moraceae comprise a large family of sixty genera and neary 1,400 specieses, including important groups such as Artocarpus, Morus, and Ficus. In particular, Morus(mulberry) is a small genus of tree and shrubs found in temperate and subtropical regions of the Northern hemishere and has been widely cultivated in China and Korea, In addition, the root bark of mulberry tree have been used as an antiphlogic, diuretic, and expectorant in white medicine, and the crude drug is known as "Sangbaikpi" in Korea. Recently, some papers have been published reporting the hypotensive effect, antiviral effect, antifungal effect, inhibitory effect of cAMP-phosphodiesterase, and anticancer effect of this extract. Little is known about that Cortex mori could have been an antiallergic effect. The purpose of this study was the development of an antiallergic agent with an antiallergic effect from Cortex mori. For this, several in vivo and in vitro experimental models were used. Results are 1) Cortex mori inhibited the compound 48/80-induced degranu-lation, histamine release and calcium uptake of rat peritoneal mast cells, 2) compound 48/80-induced anaphylactic shock and cutaneous reaction were significantly inhibited by pretreatment of Cortex mori, and 3) Cortex mori inhibited the ovalbumin-induced late astmatic reaction. From the above results it is suggested that Cortex mori has some substances with an antiallergic activity. Our final purpose of this study is to develope the new drug with an antiallergic activity from Cortex mori

Shrink-Wrapped Boundary Face Algorithm for Surface Reconstruction from Unorganized 3D Points (비정렬 3차원 측정점으로부터의 표면 재구성을 위한 경계면 축소포장 알고리즘)

  • A new surface reconstruction scheme for approximating the surface from a set of unorganized 3D points is proposed. Our method, called shrink-wrapped boundary face (SWBF) algorithm, produces the final surface by iteratively shrinking the initial mesh generated from the definition of the boundary faces. Proposed method surmounts the genus-0 spherical topology restriction of previous shrink-wrapping based mesh generation technique, and can be applicable to any kind of surface topology. Furthermore, SWBF is much faster than the previous one since it requires only local nearest-point-search in the shrinking process. According to experiments, it is proved to be very robust and efficient for mesh generation from unorganized points cloud.

Isolation and Genetic Study of Hantavirus from Apodemus peninsulae Captured in Yeuncheon-gun, Kyunggi-do (경기도에서 채집한 Apodemus peninsulae에서 한탄바이러스 분리와 유전학적 연구)

  • Hantaviruses are distributed in rodent population world-widely even in geographical areas where hemorrhagic fever with renal syndrome (HFRS) has not been reported. Various species of Family Muridae and Arvicolidae serve as the natural reservoirs of hantaviruses. Hantaan virus, Seoul virus, Puumala virus, Prospect HII virus, Sin Nombre virus and New York virus are members of genus Hantavirus and isolated from lungs of A. agrarius, R. norvegicus, C. glareolus, M. pennsylvanicus, P. maniculatus and P. leucopus respectively. This experiment was intended to find the distribution of hantavirus infection among wild rodents and isolate the hantavirus from lung tissue of seropositve Apodemus peninsulae, and compared the nucleotide and amino acid sequences with prototype of hantaan virus 76-118 strain. Hantaviral sequences were amplified from lung tissues of A. peninsulae by reverse-transcriptase polymerase chain reaction. Alignment and comparison of the 324 nucleotide of G2 region of M-genomic segment diverged 4.6% and 0% at the nucleotide and amino acid levels, and complete N protein-coding region of S-genomic segment diverged 3.7% and 1.4% nucleotide and amino acid levels, respectively. This is the report to spill-over on the hantaan virus from A. agrarius to A. peninsulae in Korea.

Molecular Classification of Commercial Spirulina Strains and Identification of Their Sulfolipid Biosynthesis Genes

  • Cyanobacterial strains of the genus Spirulina have recently been identified as an excellent source of sulfolipids, some of which possess anti-HIV properties. Thus, to investigate the distribution of sufolipid biosynthesis pathways in Spirulina, a genetic screening/phylogentic study was performed. Five different strains of Spirulina [Spirulina (Jiangmen), Spirulina sp., S. platensis, S. maxima, and Spirulina seawater] sourced from different locations were initially classified via 16S rDNA sequencing, and then screened for the presence of the sulfolipid biosynthesis genes sqdB and sqdX via a PCR. To assess the suitability of these strains for human consumption and safe therapeutic use, the strains were also screened for the presence of genes encoding nonribosomal peptide synthases (NRPSs) and polyketide synthases (PKSs), which are often associated with toxin pathways in cyanobacteria. The results of the 16S rDNA analysis and phylogenetic study indicated that Spirulina sp. is closely related to Halospirulina, whereas the other four Spirulina strains are closely related to Arthrospira. Homologs of sqdB and sqdX were identified in Spirulina (Jiangmen), Spirulina sp., S. platensis, and the Spirulina seawater. None of the Spirulina strains screened in this study tested positive for NRPS or PKS genes, suggesting that these strains do not produce NRP or PK toxins.

Detection of Polyhydroxyalkanoate-Accumulating Bacteria from Domestic Wastewater Treatment Plant Using Highly Sensitive PCR Primers

  • Polyhydroxyalkanoate (PHA) is a class of biodegradable plastics that have great potential applications in the near future. In this study, the micro-biodiversity and productivity of PHA-accumulating bacteria in activated sludge from a domestic wastewater treatment plant were investigated. A previously reported primer set and a self-designed primer set (phaCF1BO/phaCR2BO) were both used to amplify the PHA synthase (phaC) gene of isolated colonies. The new primers demonstrated higher sensitivity for phaC, and combining the PCR results of the two primer sets was able to widen the range of detected genera and raise the sensitivity to nearly 90%. Results showed that 85.3% of the identified bacteria were Gram-negative, with Ralstonia as the dominant genus, and 14.7% were Gram-positive. In addition, Zoogloea and Rhizobium contained the highest amounts of intracellular PHA. It is apparent that glucose was a better carbon source than pentone or tryptone for promoting PHA production in Micrococcus. Two different classes, class I and class II, of phaC were detected from alphaproteobacteria, betaproteobacteria, and gammaproteobacteria, indicating the wide diversity of PHA-accumulating bacteria in this particular sampling site. Simultaneous wastewater treatment and PHA production is promising by adopting the high PHA-accumulating bacteria isolated from activated sludge.

Development and Characterization of New Microsatellite Markers for the Oyster Mushroom (Pleurotus ostreatus)

  • We developed and characterized 36 polymorphic microsatellite markers for the oyster mushroom (Pleurotus ostreatus). In total, 169 alleles were identified with an average of 4.7 alleles per locus. Values for observed ($H_o$) and expected ($H_E$) heterozygosities ranged from 0.027 to 0.946 and from 0.027 to 0.810, respectively. Nineteen loci deviated from Hardy-Weinberg equilibrium. Significant (P<0.05) excess heterozygosity was observed at nine loci. Linkage disequilibrium (LD) was significant (P<0.05) between pairs of locus alleles. Cluster analysis revealed that five species of genus Pleurotus made a distinct group, and the individual cultivars were grouped into major five groups from G-1 to G-5. The diverse cultivars of P. ostreatus were discriminated and the other four species revealed a different section in the UPGMA tree. These microsatellite markers proved to be very useful tools for genetic studies, including assessment of the diversity and population structure of P. ostreatus.

Phylogenetic Differentiation of Streptomyces spp. Isolated from Potato Scab Lesions in Jeju Island of Korea on the Basis of 16S rRNA Gene Sequences (제주도 감자 더뎅이병징에서 분리된 Streptomyces spp.의 16S rRNA 유전자 염기서열 분석)

  • Potato scab is prevalent in all potato-growing areas of Jeju Island and causes economically significant losses. Streptomyces species are known as pathogens of potato scab. In this study, we analyzed the 16S rRNA sequences of Streptomyces spp, which are isolated from potato scab lesions in Jeju Island, and constructed 16S rRNA phylogenetic tree. All isolates were clearly differentiated into the genus Streptomyces, and the tree also showed that new scab-causing Streptomyces spp or not yet named species of Streptomyces are existed in Jeju Island, Korea.

Allexivirus Transmitted by Eriophyid Mites in Garlic Plants

  • Viruses in garlic plants (Allium sativum L.) have accumulated and evolved over generations, resulting in serious consequences for the garlic trade around the world. These viral epidemics are also known to be caused by aphids and eriophyid mites (Aceria tulipae) carrying Potyviruses, Carlaviruses, and Allexiviruses. However, little is known about viral epidemics in garlic plants caused by eriophyid mites. Therefore, this study investigated the infection of garlic plants with Allexiviruses by eriophyid mites. When healthy garlic plants were cocultured with eriophyid mites, the leaves of the garlic plants developed yellow mosaic strips and became distorted. In extracts from the eriophyid mites, Allexiviruses were observed using immunosorbent electron microscopy (ISEM). From an immunoblot analysis, coat proteins against an Allexivirus garlic-virus antiserum were clearly identified in purified extracts from collected viral-infected garlic plants, eriophyid mites, and garlic plants infected by eriophyid mites. A new strain of GarV-B was isolated and named GarV-B Korea isolate 1 (GarV-B1). The ORF1 and ORF2 in GarV-B1 contained a typical viral helicase, RNA-directed RNA polymerase (RdRp), and triple gene block protein (TGBp) for viral movement between cells. The newly identified GarV-B1 was phylogenetically grouped with GarV-C and GarV-X in the Allexivirus genus. All the results in this study demonstrated that eriophyid mites are a transmitter insect species for Allexiviruses.

Notes on Korean Strobilomycetaceae (III) -On Austroboletus and the Key to Genera of the Strobilomycetaceae- (한국산 귀신그물버섯과에 대한 고찰(III) -남방그물버섯속과 귀신그물버섯과의 속 검색표-)

  • To study the species diversity of the genus Austroboletus in Korea, 12 specimens were collected from 1982 to 1996 at 5 locations including Odaesan. Collected specimens were observed and three species among them were identified and described by observing macroscopic and microscopic characters, one of which was confirmed new to Korea.: Austroboletus subvirens (Hongo) Wolfe. The three species are as follows; Austroboletus fusisporus (Kawam. ap. Imaze. & Hongo) Wolfe, A. gracilis (Peck) Wolfe and A. subvirens (Hongo) Wolfe and registered here with the key to Genera of the Strobilomycetaceae in Korea.
