원충성 질병인 Neospora caninum은 소와 개를 비롯한 여러 동물에서 유ㆍ사산 및 신경증상을 유발하는 질병으로 전 세계적으로 그 피해가 증가하고 있으며, 최근국내에서도 젖소농가를 중심으로 발생보고가 있으나, 한우에 대해서는 거의 역학적조사가 되어있지 않은 바, 본 연구에서는 도축되는 한우, 유ㆍ사산 및 기형송아지를 중심으로 간접형광 항체검사법(IFAT)을 이용한 혈청학적 조사 및 면역조직화학적 방법에 의한 Neospora caninum의 동정을 실시하였다. (중략)
This study was performed to study the sequential clinical and pathologic changes of Korean isolate KBA-2 of Neospora caninum in SCID mice following intraperitoneal infection. Also the results of PCR and in vitro isolation was compared during the study. The infection appears to be disseminated hematogenously, when the infection was chased every 3days up to 21 days following infection. The PCR method was determined to be more effective than in vitro isolation regarding early detection of the organism follwing infection.
Neospora caninum is now recognized as one of the important causes of abortion in cattle worldwide, but there was no report on seroprevalence of antibodies to N. caninum in cattle at Seoul slaughtering center. Serum samples from 610 Korean indigenous cattle (Hanwoo) and 246 Holstein cattle from 452 farms located all over the country, Korea, were tested for N. caninum antibodies using a commercial enzyme-linked immunosorbent assay (ELISA) kit. Seven of 610 (1.1%) Hanwoo and 54 of 246 (22.0%) Holstein cattle were seropositive.
To examine the seroprevalence of Neospora caninum (N. caninum) infections in cattle raised at farms where an abortion had previously occurred in Gongju city and Yeongi-gun of Chungnam Province, 280 head of cattle from six farms were examined by ELISA. Thirty-two pregnant cattle from four farms were examined to determine the abortion rate in N. caninum-infected pregnant cattle and the infection pattern in newborn calves. The six farms where the abortions had previously occurred demonstrated positive reactions to N. caninum. Although the mean seropositive rate of the six farms was 17.5%, the range of seropisitivity was 3.4-57.1% due to the difference in seroprevalence between farms. The general pattern of seropositivity in cows was higher than in heifers. In addition, the abortion rate of N. caninumpositive in 18 out of 32 pregnant dams raised in four farms was 16.7%. The seroprevalence of newborn calves from N. caninum-positive pregnant animals differed according to the farm but was 86.7% overall. In contrast, 14 newborn calves from the N. caninum-negative pregnant cattle showed negative responses. This suggests that N. caninum is endemic and occurs through vertical transmission from chronically-infected dams. Therefore, aborted dams should be discarded.

Protective efficacy of vaccination with Neospora caninum multiple recombinant antigens against N. caninum infection was evaluated in vitro and in vivo. Two major immunodominant surface antigens (NcSAG1 and NcSRS2) and two dense granule proteins (NcDG1 and NcDG2) of N. caninum tachyzoites were expressed in E. coli, respectively. An in vitro neutralization assay using polyclonal antisera raised against each recombinant antigen showed inhibitory effects on the invasion of N. caninum tachyzoites into host cells. Separate groups of gerbils were immunized with the purified recombinant proteins singly or in combinations and animals were then challenged with N. caninum. Following these experimental challenges, the protective efficacy of each vaccination was determined by assessing animal survival rate. All experimental groups showed protective effects of different degrees against experimental infection. The highest protection efficacy was observed for combined vaccination with NcSRS2 and NcDG1. Our results indicate that combined vaccination with the N. caninum recombinant antigens, NcSRS2 and NcDG1, induces the highest protective effect against N. caninum infection in vitro and in vivo.
This study was carried out to investigate the prevalence of Neospora caninum infection in dairy cow and Korean native cattle(KNC), raised in several Chungnam province. To determine the prevalence of antibodies to N caninum, a total of five hundred fifty six sera were analyzed by indirected fluorescent antibody(IFA) test. Five hundred thirty three sera were collected from fifteen dairy herds and twenty three sera were taken from fourteen KNC herds from December 1999 to November 2000. Seropositive ratio of the dairy cattle sera were individually or herdly tested and showed 64.2% and 93.3%, respectively. It was recorded with 78.6% and 47.8% in KNC. The seropositive ratio of dairy cattle was depended on the size of ranch. It was 92.2, 60.7 and 57.9% at the size of less than thirty, thirty to seventy and more than seventy one cattle, respectively However, it was different from the province of Chungnam. The seropositive ratio to N caninum of dairy cattle were 79.5, 53.1, 61.4 and 31.1% at Gongju, Yeongi, Geumsan and Cheongwon, respectively. It showed difference at the growth stage and sex of cattle. The seropositive ratios of N caninum of calf, heifer, premiparous, multiparous(2nd-5th), multiparous (6>th) and bulls confirmed to 25.0, 50.3, 70.3, 71.2, 50.0 and 50.0%, respectively. It was related with brucellosis in cattle. The infected ones with brucellosis were 75.7% of seropositive ratios to N caninum. The results of this study indicated that N caninum infection was widespread in Chungnam province and confirmed existing with brucellosis in cattle.
This survey was carried out to investigate the seroprevalence of Neospora caninum in Korean indigenous cattle that was raised in the southern area of Gyenongbuk and Ulleung-gun. A total of 3,162 cattle from 984 farms were tested for the presence of antibodies against N. caninum using a commercial ELISA (IDEXX Switzerland AG, Liebefeld-bern) test kit. The Seropositive ratio of cattle was 1.38% (40/2,901), 0.77% (2/261) in the southern area of Gyeongbuk and Ulleung-gun, respectively. Regional seropositive rates of farms were 5.31% (11/207), 3.45% (4/116), 1.48% (2/135), 2.13% (2/94), 2.78% (3/108), 3.91% (95/128), 4.17% (4/96), 4.35% (3/69), 6.45% (2/31) at Gumi, Yeongcheon, Gyeongsan, Gunwi, Chengdo, Goryeong, Senogju, Chilgor and Ulleung-gun, respectively. A herd of cattle under 4 years old was 1.4% (28 heads), and above the age of 5 years was 1.1% (14 heads). and in seroprevalenec cattles by herd size farms raising under 100 heads were 39 heads.
톡소포자충과 네오포자충 (Neospora caninum)의 충체 항원으로 톡소포자충 양성 혈청 및 톡소포자충증 환자의 혈청과 ELISA, western blot 및 면역형광법을 실시하였다. ELISA에서는 172명의 톡소포자충 양성 혈청에서 12명 (6.7%)이 두 항원에 모두 반응하였으며, 톡소포자충 음성인 110명의 혈청에서 1명이 네오포자충과 반응하였다. 교차반응을 보인 12 명의 혈청을 western blot으로 확인하였을 때, 톡소포자충 항원과는 다양한 양상으로 반응하였으나, 30 kDa (SAG1)과 22 kDa (SAG2) 항원과 강하게 반응하였다. 네오포자충과의 반응은 급격히 감소하였으나, 세 경우에서 43 kDa 단백질과 반응하였으며, 음성 혈청군의 1명도 43 kDa 단백질과 반응하였다. 면역형광법에서는 모든 양성 혈청이 톡소포자충의 세포막에 표지되었으나, 네오포자충과의 반응은 세포막, 세포 내 소기관, 혹은 둘 다를 표지하였다. 이로써, 톡소포자충과 네오포자충의 항원적 교차반응과 사람 혈청에서 네오포자충에 대한 항체의 존재를 확인하였으며, 네오포자충에 의한 인체감염 가능성에 대해서는 추후 연구가 필요할 것으로 판단된다. 또, N. caninum의 우리말 이름을 네오포자충으로 제안한다.

Congenital $Neospora$$caninum$ infection was diagnosed in two Saanen goat kids from two distinct herds with a history of abortion and weak newborn goat kids in the Southern region of the State of Minas Gerais, Brazil. The first kid was weak at birth, had difficulty to rise and was unable to nurse. Gross lesions of porencephaly and hydrocephalus ex vacuo were seen. Multifocal necrosis, gliosis and non-supurative encephalitis were observed in the brain. Several parasitic cysts with a thick wall that reacted strongly only with polyclonal antiserum to $Neospora$$caninum$ were seen in the cerebral cortex, brain stem and cerebellum. The second kid was born from a $Neospora$$caninum$ seropositive mother that aborted in the last pregnancy. It was born without clinical signs. The diagnosis of neosporosis was based on antibody titer of 1:800 to $N.$$caninum$ by indirect fluorescence antibody test obtained from blood collected before the goat kid ingested the colostrum and $Neospora$$caninum$ DNA was detected by polymerase chain reaction and sequenced from placenta. This is the first report of neosporosis in goats in the southeast region of Brazil.
This study was carried out to investigate an epidemiological state of neosporosis in Korean indigenous cattle in Uljin. Bloody samples were collected from 552 female cattle (337 farms), more than 5 years old, in 10 districts of Uljin, Antibody to N caninum were examined by enzyme linked immunosorbent assay (ELISA). Seroprevalence of individual and farm were 7.6% (42/552) and 8.6% (29/337), respectively. Positive rates by districts was variable $(0%{\sim}14,9%)$, and it was seemed to be a tendency that positive farms gathered with in a small zone. In seroprevalence by herd size farms having between 5-9 heads was top (37%), but there was no significant difference among herd size.
