한국산 참빗고사리속(우드풀과)의 분류 (Taxonomic study of the genus Diplazium (Woodsiaceae) in Korea)

  • 한국산 우드풀과 참빗고사리속(Diplazium)내 9 분류군간의 유연관계를 밝히기 위하여 동아시아산 20 분류군을 포함해 군내군 29 분류군과 군외군 개고사리속(Athyrium) 3 분류군 등 총 32 분류군을 대상으로 20개의 형태 형질을 바탕을 maximum parsimony tree와 neighbor-joining tree로 분석하였다. 그 결과 제주도 천지연과 남제주군 교래리에 분포하는 참빗고사리속의 큰잔고사리(Diplazium nipponicum)는 열편의 끝이 뾰족해지고, 땅속줄기에서 엽상체가 인접하는 형질에 의하여 암고사리(Diplazium chinense)와 가장 유사하였다. 큰잔고사리는 비늘조각과 열편의 가장자리가 톱니형이고, 소우편이 겹쳐지지 않는 특징이 암고사리와 달랐다. 큰잔고사리와 암고사리는 우편의 끝이 꼬리처럼 길게 뾰족해지는 형질에 의하여 큰개고사리(D. mesosorum) 및 섬잔고사리(D. hachizoense)와 유사하였다. 이들 군은 일본 특산인 D. subtripinnatum와 일본과 대만에 분포하는 D. kawakamii와 동일군을 형성하였다.

국내 분리 흉막폐렴균의 apxIA, IIA, IIIA 유전자 Cloning, 염기서열 분석 및 단백질 발현 (Cloning, Sequencing and Expression of apxIA, IIA, IIIA of Actinobacillus pleuropneumoniae Isolated in Korea)

  • Actinobacillus pleuropneumoniae causes a highly contagious pleuropneumoniae in swine. The bacterium produces several virulence factors such as exotoxin, LPS, capsular polysaccharide, etc. Among them, the exotoxin, called Apx, has been focused as the major virulence factor, and the toxin consists of 4 gene cluster. apx CABD. apxA is the structural gene of toxin and has four different types, I, II, III, and IV. As the first step of development of a new subunit vaccine, the three different types of apxA gene were amplified from A. pleuropneumoniae isolated from Korea by PCR with primer designed based on the N- and C-terminal of the toxin. The sizes of apxIA, IIA and IIIA were 3,073, 2,971 and 3,159bps, respectively. The comparison of whole DNA sequences of apxIA, IIA and IIIA genes with those of the reference strain demonstrated 98%, 99% and 98% homology, respectively. In addition, the phylogenetic analysis was performed based on the amino acid sequences compared with 12 different RTX toxin family using the neighbor-joining method. ApxA proteins of Korean isolates were identical with reference strains in this study. All ApxA proteins were expressed in E. coli with pQE expression vector and identified using Western blot with polyclonal antibodies against culture supernatants of A. pleuropneumoniae serotype 2 or 5. The sizes of each expressed ApxA protein were about 120, 110, 125 kDa (M.W.), respectively. The results obtained in this study could be used for the future study to develop a new vaccine to porcine pleuropneumoniae.

Genetic diversity analysis of Thai indigenous chickens based on complete sequences of mitochondrial DNA D-loop region

  • Objective: Complete mtDNA D-loop sequences of four Thai indigenous chicken varieties, including Pra-dhu-hang-dam (PD), Leung-hang-khao (LK), Chee (CH), and Dang (DA) were explored for genetic diversity and relationships with their potential ancestor and possible associates to address chicken domestication in Thailand. Methods: A total of 220 complete mtDNA D-loop sequences of the four Thai indigenous chicken varieties were obtained by Sanger direct sequencing of polymerase chain reaction amplicons of 1,231 to 1,232 base pair in size. A neighbor-joining dendrogram was constructed with reference complete mtDNA D-loop sequences of Red Junglefowl (RJF) and those different chicken breeds available on National Center for Biotechnology Information database. Genetic diversity indices and neutrality test by Tajima's D test were performed. Genetic differences both within and among populations were estimated using analysis of molecular variance (AMOVA). Pairwise fixation index ($F_{ST}$) was conducted to evaluated genetic relationships between these varieties. Results: Twenty-three identified haplotypes were classified in six haplogroups (A-E and H) with the majority clustered in haplogroup A and B. Each variety was in multiple haplogroups with haplogroups A, B, D, and E being shared by all studied varieties. The averaged haplotype and nucleotide diversities were, respectively 0.8607 and 0.00579 with non-significant Tajima's D values being observed in all populations. Haplogroup distribution was closely related to that of RJF particularly Gallus gallus gallus (G. g. gallus) and G. g. spadiceus. As denoted by AMOVA, the mean diversity was mostly due to within-population variation (90.53%) while between-population variation (9.47%) accounted for much less. By pairwise $F_{ST}$, LK was most closely related to DA ($F_{ST}=0.00879$) while DA was farthest from CH ($F_{ST}=0.24882$). Conclusion: All 4 Thai indigenous chickens are in close relationship with their potential ancestor, the RJF. A contribution of shared, multiple maternal lineages was in the nature of these varieties, which have been domesticated under neutral selection.

Genetic Structure of Mongolian Goat Populations Using Microsatellite Loci Analysis

  • We studied genetic diversity and relationships among Mongolian goat populations on the basis of microsatellite DNA polymorphisms. DNA samples from eight populations (Bayandelger, Ulgii Red, Zavkhan Buural, Sumber, Zalaajinst White, Erchim Black, Dorgon, and Gobi Gurvan Saikhan) from geographically distinct areas of Mongolia were analyzed by using 10 microsatellite DNA markers. Since the 10 markers were highly polymorphic, the genetic characteristics of these native goat populations could be estimated. Genetic diversity within populations, as estimated by the expected heterozygosities, was high, ranging from 0.719 to 0.746, but genetic differentiation between populations was low, representing only 1.7% of the total genetic variation. The results suggest that Mongolian native goat populations still have a semi-wild genetic structure reflecting traditional Mongolian nomadism and the short history of artificial selection. The genetic relationships among the populations were not clear in the neighbor-joining tree generated from the modified Cavalli-Sforza chord genetic distances. By using principal components analysis, the five core populations of Mongolian native goats (Bayandelger, Ulgii Red, Zavkhan Buural, Sumber, and Dorgon) and the populations crossed with Russian breeds (Zalaajinst White, Erchim Black, and Gobi Gurvan Saikhan) were distinguished. There was no correlation between genetic relationships among the populations and the geographical distribution of the populations.

가다랑어 (Katsuwonus pelamis) 자어의 국내 출현 (Occurrence of skipjack tuna (Katsuwonus pelamis) larvae in Korean waters)

  • Three specimens of larvae (5.2~7.8 mm in standard length (SL)), of the family Scombridae, were collected from the southeastern waters off Jeju Island in August, 2014. These specimens were identified to Katsuwonus pelamis have melanophores distributed on the 1st dorsal-fin spines and post ventral margin on caudal peduncle. An analysis of the cytochrome oxidase submit I (COI) sequences (652 base pairs) of mitochondrial DNA showed that our specimens must be K. pelamis, because their sequences were concordant with those of the adult K. pelamis (genetic distance= 0.000~0.002), followed by Auxis rochei (genetic distance= 0.061~0.063) and Euthynnus affinis (genetic distance= 0.077~0.079). During the larval stage, K. pelamis differed from Thunnus spp. species in melanophores distribution period of 1st dorsal-fin spines, lower jaws and ventral margin on caudal peduncle. In conclusion, occurrence of K. pelamis larvae means Korean waters are potential spawning ground of K. pelamis.

농촌주민의 지역사회 만족도와 지역공동체 의식 (Rural Resident′s Community Life Satisfaction and Consciousness of Rural Community)

  • The objective of this study was to analyze the evaluation of the rural resident's on their community at view of community life satisfaction. The specific purposes of this study were to identify the degrees of community life satisfaction, consciousness of rural community according to the individual variables. The data were gathered from 1,870 rural residents by the structured interview in the 187 communities sampled by multi-staged cluster stratified sampling method in November, 2003. The major findings of this study were as follows; first, the degree of the rural residents'community life satisfaction was generally low, especially the rural residents were dissatisfied with the opportunities of leisure and education environment in their communities, and by half were satisfied with housing environment. Second, as consciousness of rural community, the degree of rural resident's joining at community meeting was high, and openness to the outside residents' was high. third, the most important factor for consciousness of rural community were community organization, good neighbor. As result of this study, the following conclusion and recommend can be drawn. First, the level of rural resident's community life satisfaction is overall low. Especially it is needed policy efforts and priority investment for advancing satisfaction on leisure and education environment in rural community. Second, to foster rural resident's community consciousness, community orgaruzatlon Is first of all supported and activated. Through viability of these community organization, rural resident's participation in community welfare will be enhanced

인간 태아의 뇌로부터 만들어진 cDNA library에서 내생 레트로바이러스 HERV-W LTR의 클로닝 및 분자계통분류 (Molecular Cloning and Phylogeny of the Human Endogenous Retrovirus HERV-W LTR Family in cDNA Library of Human Fetal Brain)

  • Long terminal repeats(LTRs) of the human endogenous retrovirus(HERV) heve been found to be coexpresed with genes located nearby. It has been suggested that the LTR elements have contributed to the genetic variation of human genome connected to various diseases. Recently, HERV-W family was identified in the cerebrospinal fluids and brains of individuals with schizophrenia. Using cHNA library derived from human fetal brain, we performed PCR amplification and identified seven new HERV-W LTR elements. Those LTR elements showed a high degree of sequence similarity(98∼99%) with HERV-W (AF072500). A phylogentic tree obtained by the neighbor-joining method revealed that seven new HERV-W LTR elements(FB-1, 2, 4, 8, 9, 10, 12) were closely related to the AX000960, AF072504, and AF072506 from Gen Bank database. Our data suggest that several copy numbers of the HERV-W LTR elements are expressed in human feta brain and may contribute to an understanding of biological function connected to neuropsychiatric diseases.

Morphological and Molecular Analyses of $Anabaena$ $variabilis$ and $Trichormus$ $variabilis$ (Cyanobacteria) from Korea

  • This study characterizes three $Anabaena$ strains and 5 $Trichormus$ strains isolated from Korean waters and 3 $Anabaena$ $flos-aquae$ strains procured from the UTEX based on morphological features and molecular analyses. The $Anabaena$ and $Trichormus$ isolates were morphologically assigned to $A.$ $variabilis$ K$\ddot{u}$tzing and $T.$ $variabilis$(K$\ddot{u}$tzing ex Bornet et Flahault) Kom$\acute{a}$rek et Anagnostidis, respectively. The $Anabaena$ and $Trichormus$ strains differed significantly in the mean length of their vegetative cells. The 16S rRNA genes from the $Anabaena$ strains showed a 100% identity to that from $A.$ $variabilis$ ATCC 29413, while the 16S rRNA genes from the $Trichormus$ strains showed a 99.9% identity to that from $T.$ $variabilis$ GREIFSWALD. The overall topology was in agreement for the 16S rRNA gene and $cpcBA$-IGS trees in the both tree-constructing methods. In a neighbor-joining tree based on the 16S rRNA gene, the 3 $Anabaena$ strains were asso-ciated with $A.$ $variabilis$, the 5 $Trichormus$ strains with $T.$ $variabilis$, and the 3 $Anabaena$ (UTEX) strains were with $Nostoc$. To date, this is the first report on $A.$ $variabilis$ and $T.$ $variabilis$ strains originating from Korea.

Internal transcribed spacer (ITS) region의 염기서열 분석에 의한 보길도산 황칠나무의 분자 계통학적 연구 (Phylogenetic Analysis of Dendropanax morbifera Using Nuclear Ribosomal DNA Internal Transcribed Spacer (ITS) Region Sequences)

  • 보길도에서 자라고 있는 황칠나무(Dendropanax morbifera)를 구입하여, 캘러스로 유도한 후, ribosomal DNA(nrDNA)의 internal transcribed spacer (ITS) region의 염기서열을 결정하였다 보길도의 황칠나무(Dendropanax morbifera)의 ITS region의 염기서열을 분석한 결과, 총 689염기를 결정하였다. 결정된 689염기 중에서 ITS1은 222 개염기, 5.8S rDNA는 160염기, ITS2는 233염기인 것으로 판명되었다. GenBank의 BLAST 프로그램(http://www.ncbi.nlm.nih.BLAST)을 사용하여 GenBank/EMBL/DDBJ에 등록되어 있는 Dendropanax 속 33의 염기서열을 수집한 후 multiple alignment를 수행한 결과, 유사도는 99.7%(D. chevalieri)에서 92.6%(Dendropanax arboreus)로 나타났으며, 일본황칠나무(D. trifidus)와는 유사도가 99.4%로 판명되었다.

Platynosomum fastosum (Trematoda: Dicrocoeliidae) from Cats in Vietnam: Morphological Redescription and Molecular Phylogenetics

  • The present study was performed to reveal the morphological characteristics and molecular phylogenetic position of Platynosomum fastosum Kossack, 1910. A total 167 specimens of P. fastosum were collected in 8 (4.9%) out of 163 sets of gall-bladders and bile ducts of cats. The number of worms was 1-105 per infected cat. This species was characterized by having a long and slender body, slightly larger ventral sucker than the oral sucker, indistinct prepharynx, small pharynx, short esophagus, bifurcation midway between 2 suckers, and ceca extending to the posterior end of the body. The length of the partial sequences of ITS1 and 5.8S rDNA of P. fastosum were 990 bp, GC-rich. AT/GC ratio was 0.9, there were 9 polymorphic sites, and intraspecific variations ranged from 0.1% to 0.9%. Phylogenetic analyses by neighbor-joining phylogram inferred from ITS1 rDNA sequences revealed that the genetic distance between P. fastosum specimens ranged from 0.3 to 1.5% while the smallest interspecific distance among dicrocoeliid species was 20.9 %. The redescription and genetic characters of P. fastosum are taxonomically important to recognize future different species of the genus Platynosomum showing high intraspecific and morphological variability.
