Taxus chinensis로부터 파클리탁셀 정제를 위한 음압 캐비테이션 아세톤-펜테인 분별침전 (Negative Pressure Cavitation Acetone-Pentane Fractional Precipitation for the Purification of Paclitaxel from Taxus chinensis)

  • 본 연구에서는 음압 캐비테이션 아세톤-펜테인 분별침전으로 Taxus chinensis로부터 파클리탁셀의 침전 효율을 획기적으로 개선하였다. 음압 -200 mmHg에서 침전할 경우 짧은 조업 시간(5분)에 대부분의 파클리탁셀을 회수(>99.9%) 할 수 있었다. 침전 속도 상수는 대조군 대비 1.512~5.073배(-50~-200 mmHg) 증가하였다. 음압으로 활성화에너지는 -3737~-6536 J/mol 감소하였으며, 이로 인해 침전 속도가 증가하였다. 또한 음압 도입으로 침전물 크기는 5.3배 감소하였으며, 파클리탁셀의 확산 계수는 7.0배 증가하였다.

시청각 동영상에 의해 유발된 부정적 감성에 따른 자율신경계 반응 (ANS responses in Negative Emotions Induced by Audio-visual Film Clips)

  • 부정적 감성은 인간의 생존에 기여한다는 점에서 중요한 기능을 한다. 본 연구에서는 시청각 동영상자극을 이용하여 부정적 감성을 유발하고 각 부정적 감성에 따른 자율신경계 반응을 구분하고자 하였다. 실험에는 성인 남녀 30명(22.6세${\pm}$1.24, 남: 15명, 여: 15명)이 참여하였다 예비실험을 통해 2분 길이의 동영상 자극을 실험자극으로 선정하였다. 실험참여자가 선정된 동영상자극을 시청하는 동안 피부전기활동(EDA), 심전도(ECG)가 측정되었고, 하나의 자극제시가 끝난 후에 실험참여자들에게 각 감성자극에 의해 경험되는 감성에 대한 심리평가를 실시하였다. 심리반응 분석결과, 각 부정적 감성 자극은 표적감성을 적절하고 효과적으로 유발하는 것으로 나타났다. 자율신경계 반응분석결과, 각각의 감성자극들은 서로 구분되는 자율신경계 활동패턴을 유발하였다. 그리고 공포자극은 다른 유형의 부정적 감성자극보다 피부전기활동과 심전도의 지표들에서 높은 교감신경계의 활성화를 유발하였다. 본 연구는 각 부정적 감성에 대한 자율신경계 반응을 세분화하였다는 데 의의가 있다.

Effect of flexion degrees in elbow joint on muscle activation of the extensor carpi radialis and biceps brachii muscles in healthy young adults

  • Objective: Chronic lateral epicondylitis is a condition which becomes sore and tender on the lateral side of the elbow joint damaged from overuse and repetitive use of the extensor muscles of the forearm. The purpose of this study was to investigate the effects of flexion degrees in the elbow joint on extensor carpi radialis longus and brevis and biceps brachii muscles in individuals with healthy young adults. The main purpose of this study was to suggest the feasibility of optimal elbow angle during therapeutic eccentric exercise with resistance for strengthening of wrist extensors. Design: Cross-sectional study. Methods: Thirty health young adults (male 15, female 15) participated in this study. This study measured muscle activation in four different conditions of elbow flexion, $0^{\circ}$, $30^{\circ}$, $60^{\circ}$, and $90^{\circ}$ during eccentric exercise with weight loading in wrist extensors, extensor carpi radialis longus and brevis and biceps brachii muscles using surface electromyography. Results: The muscle activation of extensor carpi radialis showed a negative relationship with the degrees of elbow joint flexion. With increasing elbow flexion angles, the ECRL muscle activation amount was significantly lower (p<0.05). In contrast, the muscle activation of the ECRB muscle activation amount was significanlty higher (p<0.05). Conclusions: This study suggests that the eccentric exercise of wrist extension with selected activation of wrist extensor muscles according to elbow flexion positions, and suggests that the extensor carpi radialis longus and brevis will need to be strengthened for preventing and treating chronic lateral epicondylitis regardless of degrees of elbow joint flexion.

(E)-1-(2-(2-nitrovinyl)phenyl)pyrrolidine inhibits Inducible Nitric Oxide Synthase Expression in RAW264.7 Macrophages Stimulated with Lipopolysaccharide

  • Toll-like receptors (TLRs) play an important role for host defense against invading pathogens. TLR4 has been identified as the receptor for lipopolysaccharide (LPS), which is a cell wall component of gram-negative bacteria. The activation of TLR4 signaling by LPS leads to the activation of NF-${\kappa}B$ and the expression of pro-inflammatory gene products such as cytokines, inducible nitric oxide synthase (iNOS), and cyclooxygenase-2 (COX-2). To evaluate the therapeutic potential of (E)-1-(2-(2-nitrovinyl)phenyl)pyrrolidine (NVPP), previously synthesized in our laboratory, NF-${\kappa}B$ activation and iNOS and COX-2 expression induced by LPS were examined. NVPP inhibited the activation of NF-${\kappa}B$ induced by LPS. NVPP also suppressed the iNOS expression induced by LPS but it did not suppress COX-2 expression induced by LPS. These results suggest that NVPP has the specific mechanism for anti-inflammatory responses.

Estrogen Pretreatment of Organotypic Hippocampal Slices Protects Neurons against Oxygen-Glucose Deprivation with Akt Activation

  • In several experimental models, estrogens protect neurons against ischemic insults. However, the recent clinical studies of hormone replacement showed negative results to prevent stroke. Therefore, optimal models to study estrogen replacement for neuroprotection are needed before its clinical ap-plication. Organotypic hippocampal slice under oxygen-glucose deprivation (OGD) has been established as a model of cerebral ischemia and has advantages to study drug effects. We investigated whether estrogen protected CAI neurons and affected activation of Akt (pAkt) in CAI region under OGD. Thus, rat hippocampal slices on day 7 of culture were treated with $17-{\beta}$ estradiol (E, 1 nM) for 7 days before 30 min OGD, and cell death of CAI neurons was quantified by propidium iodide (PI) staining and expression of pAkt was studied by Western blot and immunofluorescence. PI intensity in slices treated with E was significantly reduced 72 hour after OGD compared to that of non-treated slices (p < 0.05). E pretreatment also increased the expression of pAkt 72 hour after OGD compared to that of no treatment (p<0.01). These data suggest that estrogen pretreatment may rescue neurons from ischemic insults through the activation of Akt and also indicate that our model would be a useful alternative method to study the mechanisms and effects of estrogen replacement treatment for neuroprotection.

Transient activation of the MAP kinase signaling pathway by the forward signaling of EphA4 in PC12 cells

  • In the present study, we demonstrate that ephrin-A5 is able to induce a transient increase of MAP kinase activity in PC12 cells. However, the effects of ephrin-A5 on the MAP kinase signaling pathway are about three-fold less than that of EGF. In addition, we demonstrate that EphA4 is the only Eph member expressed in PC12 cells, and that tyrosine phosphorylation induced by ephrin-A5 treatment is consistent with the magnitude and longevity of MAP kinase activation. Experiments using the Ras dominant negative mutant N17Ras reveal that Ras plays a pivotal role in ephrin-A5-induced MAP kinase activation in PC12 cells. Importantly, we found that the EphA4 receptor is rapidly internalized by endocytosis upon engagement of ephrin-A5, leading to a subsequent reduction in the MAP kinase activation. Together, these data suggest a novel regulatory mechanism of differential Ras-MAP kinase signaling kineticsexhibited by the forward signaling of EphA4 in PC12 cells.

개인의 고유한 전전두엽의 비대칭성이 음향감성에 미치는 영향 (Influence of Intrinsic Prefrontal Cortex Asymmetry on Sound-induced Affects)

  • This study is aimed to inspect how the different sensitivities in Behavioral activation system (BAS) and Behavioral inhibition system (BIS) modulate on the properties of physiological responses stimulated by positive or negative affective sound. We measured the electroencephalogram (EEG) of 32 students, consisted of four groups depending on the BAS and BIS sensitivities, during listening to meditation music or noise. The EEG was recorded at Fp1 and Fp2 sites. After listening to music or noise, subjects reported the affect induced by the sound. For EEG, the power in the alpha band at Fp2, especially in the alpha-2 band (9.0-11.0 Hz) increased during the subjects listening to music, while the power at Fp1 increased during noise. During listening to meditation music, there is a tendency that the left-sided activation in prefrontal cortex (PFC) is positively correlated with the difference of BAS(Z)-BIS(Z). During listening to noise, there is a tendency that the right-sided activation in PFC is dominant in case any of the sensitivity of BAS or BIS is high. From these results we suggest that the physiological responses of different individuals in BAS and BIS react differently under the same emotionally provocative challenge.

압력변화에 따른 ${\beta}$-피콜린과 염화벤조일류의 반응에 대한 연구 (A Study for the Reaction of ${\beta}$-Picoline with p-Substituted Benzoyl Chlorides by Pressure)

  • 10$^{\circ}C$와 20$^{\circ}C$에서 ${\beta}$-피콜린과 치환된 벤조일클로라이드류의 반응을 압력변화에 따라 아세토니트릴용매내에서 전기전도도법으로 연구하였다. 유사 1차속도상수와 2차속도상수로부터 여러 가지 활성화파라미터들(${\Delta}V{\neq}$, ${\Delta}{\beta}{\neq}$, ${\Delta}H{\neq}$, ${\Delta}S{\neq}$ and ${\Delta}G{\neq}$)을 구하고, Hammett ${\rho}$값의 압력의존성을 설명하였다. ${\Delta}V{\neq}$, ${\Delta}{\beta}{\neq}$${\Delta}S{\neq}$ 는 모두 음의 값을 나타내었고, 압력증가에 따라 속도상수와 Hammett ${\rho}$값은 증가하며, 결합형성이 더욱 촉진된 반응메카니즘으로 진행됨을 알 수 있었다.

염화벤조일류와 피리딘과의 반응에 대한 압력의 영향 (The Effect of Pressure on the Reaction of p-Substituted Benzoyl Chlorides with Pyridine)

  • Acetonitrile 용매내에서 pyridine과 치환된 benzoyl chloride류의 반응을 압력변화(1-1000 bar)에 따라 속도론적으로 연구하였다. 속도상수로부터 활성화파라미터들(Δ$V^{\neq}$, ${\Delta}{\beta}^{\neq}$, ${\Delta}H^{\neq}$, ${\Delta}S^{\neq}$ and ${\Delta}G^{\neq}$)을 구하였다. 압력이 증가함에 따라 속도상수는 증가하고 ${\Delta}V^{\neq}$, ${\Delta}{\beta}^{\neq}$, 및 ${\Delta}S^{\neq}$는 모두 음의 값을 나타내었으며, 치환기효과와 종합하여 반응메카니즘을 고찰한 결과 전체적인 반응은 $S_N2$ 반응메카니즘으로 진행되나, 치환체와 압력변화에 따라 전이상태 구조에 약간의 변화가 있으리라 생각된다.

fMRI를 이용한 조해혈(照海穴)(KI6) 자침(刺針)이 대뇌피질(大腦皮質) 운동영역(運動領域)의 활성변화(活性變化)에 관(關)한 연구(硏究) (The New Finding on BOLD Response of Motor Acupoint KI6(照海) by fMRI)

  • Objective : Recent studies Suggested that there is a strong correlation between acupuncture stimulation and its related cortical activation. Anther study showed that either positive or negative BOLD effects could be observed depending on anatomical structure in acupuncture. Material and Methods : 1) Subjects and paradigms : Two separate stimulation paradigms were performed on five healthy (aged 22-23 yrs) in this study. First, the paradigm of acupuncture stimulation was that the acupuncture needle was inserted in acupoints KI6(照海), which is located in lateral side of the foot and then continuously twisted(補瀉를 除外한 捻轉法) for 70 seconds for 10 cycles of activation. During rest period (70 seconds), the needle was completed removed from acupoint. Total 60 cycles were performed and 10 images were obtained per cycle. Second, non-acupoint was randomly selected and the same paradigm was performed as acupoint stimulation. The stimulation protocol comprised 10 cycles of alternating activation and rest (10 images per cycle). Total 60 cycles were performed and each cycle take about 1.5 sec for motor task. Subjects take an at least 15 minutes break before starting anther paradigm. Conclusion : In this study, I investigated a new acupoint KI6(照海) which was known as motor - related acupoint and obtained an evidence that the stimulation of KI6(照海) resulted in negative BOLD response to stimulation.
