Synergistic Interactions of Schizostatin Identified from Schizophyllum commune with Demethylation Inhibitor Fungicides

  • Botrytis cinerea, which causes gray mold disease in more than 200 plant species, is an economically important pathogen that is mainly controlled by synthetic fungicides. Synergistic fungicide mixtures can help reduce fungicide residues in the environment and mitigate the development of fungicide-resistant strains. In this study, we screened microbial culture extracts on Botrytis cinerea to identify an antifungal synergist for tebuconazole. Among the 4,006 microbial extracts screened in this study, the culture extract from Schizophyllum commune displayed the most enhanced activity with a sub-lethal dosage of tebuconazole, and the active ingredient was identified as schizostatin. In combination with 5 ㎍/ml tebuconazole, schizostatin (1 ㎍/ml) showed disease control efficacy against gray mold on tomato leaf similar to that achieved with 20 ㎍/ml tebuconazole treatment alone. Interestingly, schizostatin showed demethylation inhibitor (DMI)-specific synergistic interactions in the crossed-paper strip assay using commercial fungicides. In a checkerboard assay with schizostatin and DMIs, the fractional inhibitory concentration values were 0.0938-0.375. To assess the molecular mechanisms underlying this synergism, the transcription levels of the ergosterol biosynthetic genes were observed in response to DMIs, schizostatin, and their mixtures. Treatment with DMIs increased the erg11 (the target gene of DMI fungicides) expression level 15.4-56.6-fold. However, treatment with a mixture of schizostatin and DMIs evidently reverted erg11 transcription levels to the pre-DMI treatment levels. These results show the potential of schizostatin as a natural antifungal synergist that can reduce the dose of DMIs applied in the field without compromising the disease control efficacy of the fungicides.

  • HIV-1의 envelope glycoprotein은 중화항체에 의한 체액성 면역반응의 중요한 target으로 surface glycoprotein인 gp120과 transmembrane glycoprotein인 gp41로 이루어져 있다. gp120과 gp41의 ectodomain으로 이루어진 gp140 유전자를 PCR의 방법으로 증폭하고 Semliki Forest virus(SFV) 유래 expression system을 이용하여 mammalian 세포에서 발현하였다. 발현된 gp140은 natural HIV-1에서와 같이 oligomer를 형성하였다. 발현된 gp140을 정제하여 BALB/c 마우스에 접종하여 항체가 형성되었음을 확인하였다.

Inhibitory Effects of Flavonoids Isolated from Leaves of Petasites japonicus on $\beta$-Secretase (BACE1)

  • The deposition of the amyloid $\beta}$ ($A{\beta}$)-peptide following proteolytic processing of amyloid precursor protein (APP) by $\beta$-secretase (BACE1) and $\gamma$-secretase is critical feature in the progress of Alzheimer's disease (AD). Consequently, BACE1, a key enzyme in the production of $A{\beta}$, is a prime target for therapeutic intervention in AD. In the course of searching for BACE1 inhibitors from natural sources, the ethyl acetate fraction of Petasites japonicus showed potent inhibitory activity. Two BACE1 inhibitors quercetin (QC) and kaempferol 3-O-(6"-acetyl)-$\beta$-glucopyranoside (KAG) were isolated from P. japonicus by activity-guided purification. QC, in particular, non-competitively attenuated BACE1 activity with $IC_{50}$ value of $2.1{\times}10^{-6}\;M$ and $K_i$ value of $3.7{\times}10^{-6}\;M$. Both compounds exhibited less inhibition of $\alpha$-secreatase (TACE) and other serine proteases including chymotrypsin, trypsin, and elastase, suggesting that they ere relatively specific and selective inhibitors to BACE1. Furthermore, both compounds significantly reduced the extracellular $A{\beta}$ secretion in $APP_{695}$-transfected B103 cells.

Expression Patterns of Growth Related Genes in Juvenile Red Spotted Grouper (Epinephelus akaara) with Different Growth Performance after Size Grading

  • Fish shows great difference in growth rate between individuals during larval development and early growth. This difference seriously reduces the production efficiency in fish culture. Growth hormone (GH)/Insulin-like growth factor 1 (IGF1) system is said to play some pivotal roles in fish growth. In this study, we investigated differences of GH, IGF1 and GHR gene expressions in juvenile red spotted grouper (Epinephelus akaara) with different growth performance. Red spotted groupers were reared under the same environmental condition (water temperature $24{\pm}1^{\circ}C$, natural light) for 96 days after hatching. They were divided into 3 groups by size (fast growing, middle growing and slow growing groups: FGG, MGG, and SGG, respectively). RNA was extracted from the brain, liver and muscle tissues from each group, and target gene expression was examined by real-time PCR. In the brain with pituitary gland, expression of GH gene in FGG was significantly higher than the expression in SGG, but the expression of IGF1 and GHR genes in the muscle was highest in SGG. Difference of GHR and IGF1 mRNA in the liver between groups with different growth performance was less clear than that in other tissues, although level of IGF1 mRNA was higher in SGG than in MGG. These results suggest that hormonal governing of growth is not the same in fast growing and slow growing fish, and size grading could cause a shift of hormonal state and growth pattern in this species.

Anti-obesity Activity of Ethanol Extract from Bitter Melon in Mice Fed High-Fat Diet

  • In many cases, obesity is associated with metabolic disorders. Recently, natural compounds that may be beneficial for improving obesity have received increasing attention. Bitter melon has received attention as a diabetes treatment. $NAD^+$-dependent deacetylase (Sirtuin 1, SIRT1) has emerged as a novel therapeutic target for metabolic diseases. In this study, ethanol extract of bitter melon (BME) suppressed adipocyte differentiation and significantly increased the expression of SIRT1 in fully differentiated 3T3-L1 cells. Moreover, it enhanced the activation of AMP-activated protein kinase (AMPK). In high-fat diet (HFD)-fed induced-obesity mice, BME suppressed HFD-induced increases in body weight and white adipose tissue (WAT) weight. BME also increased the expression of SIRT1 and suppressed peroxisome proliferator-activated receptor and sterol regulatory element binding protein 1 expressions of WAT from HFD-fed mice. These findings suggest that BME prevents obesity by activating the SIRT1 and AMPK pathway and that it may be a useful dietary supplement for preventing obesity.

Isoflavones Extracted from Sophorae fructus Upregulate IGF-1 and TGF-$\beta$ and Inhibit Osteoclastogenesis in Rat Born Marrow Cells

  • Isoflavones have been a central subject in research on the natural phytoestrogens found in Leguminosae. Their effects on bone formation and remodeling are important in that they can act like estrogen by binding on estrogen receptors on the target cell surface. We, therefore, believed that isoflavones may help in the treatment of patients with estrogen deficiency disease such as estrogen replacement therapy (ERT) for osteoporosis. As commonly known, osteoporosis is one of the hormonal deficiency diseases, especially in menopausal women. When estrogen is no longer produced in the body a remarkable bone remodeling process occurs, and the associated events are regulated by growth factors in the osteoblast lineage. In the present study, we investigated whether isoflavones (Isocal) extracted from Sophorae fructus affect the growth factors IGF-I and TGF-$\beta$ that have been known to be related with bone formation. In the study, we found that the active control (PIII) effectively enhanced the level of nitric oxide (NO) and growth factors, and thereby inhibited osteoclastogenesis. The most efficient concentration was $10^{-8}$% within five days, whereas the comparative control (soybean isoflavone) was not as effective even at a lower concentration. In conclusion, the products which contain enriched glucosidic isoflavone and nutrient supplements such as shark cartilage and calcium can be used for osteoporosis therapy by enhancing the production of IGF-I and TGF-$\beta$. Furthermore, the NO produced through endothelial constitutive NO synthase (ecNOS) may playa role in inhibiting bone reabsorption.

Analyzing the Effect of Management Strategies on Gum Talha Yield from Acacia Seyal, South Kordofan, Sudan

  • The present study was carried out from September 2007 to February 2008 in Umfakarin natural forest reserve, South Kordofan, Sudan. The objective was to analyze the effect of different management strategies on yield of gum talha from Acacia seyal. A total of 493 single target trees were selected, based on their diameters, and assigned to tapping treatments in three different stand densities (making a total of nine treatments per stand density). The treatments are as follows: tapping date with three levels (first of October, 15 October and first of November) and two levels of local tapping tools (sonki, and makmak). Untapped trees were used as control. The first picking of gum was started fifteen days after tapping while the subsequent pickings were done in intervals of fifteen days. Yield per tree throughout the season was obtained by summing up the gum yield from all pickings. Yield throughout the season (from first to the last picking) were analyzed. General linear model (GLM) was used to test the effect of different tapping treatments on the yield of gum talha. Post hoc test after analysis of variance (ANOVA) based on Scheffe test was performed to examine the differences in gum yield as a result of different management strategies. The results showed that tapping has a significant influence on gum yield. Analysis of pick-to-pick yield indicated that only three treatments in dense stand density showed a decreasing pattern while the rest of treatments either have constant or unclear patterns. The results of the present study were based on a single season data and that may underscore the real effect of Acacia seyal stands' management strategies on gum talha yield. Conducting gum yield experiments in permanent trial plots are highly recommended in order to analyze gum yield of seasonal time series.

Model reduction techniques for high-rise buildings and its reduced-order controller with an improved BT method

  • An AMD control system is usually built based on the original model of a target building. As a result, the fact leads a large calculation workload exists. Therefore, the orders of a structural model should be reduced appropriately. Among various model-reduction methods, a suitable reduced-order model is important to high-rise buildings. Meanwhile, a partial structural information is discarded directly in the model-reduction process, which leads to the accuracy reduction of its controller design. In this paper, an optimal technique is selected through comparing several common model-reduction methods. Then, considering the dynamic characteristics of a high-rise building, an improved balanced truncation (BT) method is proposed for establishing its reduced-order model. The abandoned structural information, including natural frequencies, damping ratios and modal information of the original model, is reconsidered. Based on the improved reduced-order model, a new reduced-order controller is designed by a regional pole-placement method. A high-rise building with an AMD system is regarded as an example, in which the energy distribution, the control effects and the control parameters are used as the indexes to analyze the performance of the improved reduced-order controller. To verify its effectiveness, the proposed methodology is also applied to a four-storey experimental frame. The results demonstrate that the new controller has a stable control performance and a relatively short calculation time, which provides good potential for structural vibration control of high-rise buildings.

  • 기계번역 품질 예측(Quality Estimation, QE)은 정답 문장(Reference sentence) 없이도 기계번역 결과의 질을 평가할 수 있으며, 활용도가 높다는 점에서 그 필요성이 대두되고 있다. Conference on machine translation(WMT)에서 매년 이와 관련한 shared task가 열리고 있고 최근에는 대용량 데이터 기반 Pretrained language model(PLM)을 적용한 연구들이 주로 진행되고 있다. 본 논문에서는 기계번역 품질 예측 task에 대한 설명 및 연구 동향에 대한 전반적인 survey를 진행했고, 최근 자주 활용되는 PLM의 특징들에 대해 정리하였다. 더불어 아직 활용된 바가 없는 multilingual BART 모델을 이용하여 기존 연구들인 XLM, multilingual BERT, XLM-RoBERTa와 의 비교 실험 및 분석을 진행하였다. 실험 결과 어떤 사전 학습된 다중언어 모델이 QE에 적용했을 때 가장 효과적인지 확인하였을 뿐 아니라 multilingual BART 모델의 QE 태스크 적용 가능성을 확인했다.

기후변화가 마로해의 김 양식에 미치는 영향 및 대응방안 (Effects of Climate Change on Purple Laver Farming in Maro-hae (Jindo-gun and Haenam-gun), Republic of Korea and Countermeasures)

  • Global warming affects critical natural resources, one of which is the oceans that occupy 70% of the total cover of the earth. In other words, ocean warming is a subset of global warming which needs to be addressed urgently. Purple laver (pyropia spp.) is one of the most vulnerable items to climate change although it is a major export product of Korean fisheries. The purpose of this study is to analyze the causality of how climate change caused by global warming affects the increase or decrease of PLP (purple laver production). The target area for analysis was set to Maro-hae between Jindo-gun and Haenam-gun. We selected marine environmental factors and meteorologic factors that could affect PLP as variables, as well as co-integration tests to determine long-term balance, and the Granger causticity tests. As a result, PLP and marine environmental factors WT (water temperature), pH, and DO confirmed that long-term equilibrium relationships were established, respectively. However, there is only causality with WT and it is confirmed that there is only a correlation between pH and DO (dissolved oxygen). There was no long-term equilibrium relationship between PLP and HDD (heating degree days) and there is a causal effect that HDD affects PLP; however, it was less clear than that of WT. The relationship between PLP and RF (rainfall), WS (wind speed), SS (percentage of sunshine), and FF (farm facilities) was all balanced in the long term, and causality exists. Based on the results of the analysis, policy proposals were made.
