Effects of Dietary Xanthophylls and See Weed By-Products on Growth Performance, Color and Antioxidant Properties in Broiler Chicks (Xanthophylls과 해조 부산물 첨가 급여가 육계의 사양성적, 육색 및 항산화 특성에 미치는 영향)

  • This experiment was conducted to investigate the effects of dietary pigment sources on the performance, color and antioxidant properties in broiler chick. Experimental diet was formulated to have isocalories and isonitrogen during the experiment period. Total xanthophylls content in the experimental diet was formulated to have 30ppm. Experimental trials were done for five weeks with six treatment groups; T1 (Control), T2 (Olo Glo, natural yellow pigment), T3 (Kern Glo, natural red pigment), T4 (canthaxanthin, synthetic red pigment), T5 (asthaxanthine, natural red pigment), and T6 (seaweed by-products). Body weight gain and feed intake were significantly lower (p<0.05) in T6 group than in other treatments. Mortality was lower in T2, T3 and T4 than in control, but higher (p<0.05) in T5 and T6. The sources of pigments did not have any effects on the dressed carcass and abdominal fat pad (p>0.05). The gizzard weight was significantly lower in T6 (p<0.05) than in others. Pigmentation of leg skin was significantly lower (p<0.05) in control and T6. Effects of dietary pigments was greater with red pigments than with yellow pigments, and those were also greater with natural pigments than with synthetic ones. The peroxide value (POV), thiobarbituric acid reactive substances (TBARS) and pH values of chicken meat were increased (p<0.05) in all treatments at 12 day storage, and was higher (p<0.05) in pigments supplementation group. No differences of CIE L$\^$*/(lightness) and b$\^$*/(yellowness) were not found by storage days and xanthophylls sources. The a$\^$*/(redness) after 12 day storage was significantly (p<0.05) decreased in all treatments, but those of T4 and T5 were higher than those of others. These results showed that feeding of xanthophylls sources to chick could improve color intensity and inhibit lipid oxidation of leg meat.

  • Carotenoids are isoprenoids with a long polyene chain containing 3 to 15 conjugated double bonds, which determines their absorption spectrum. They typically consist of a $C_{40}$ hydrocarbon backbone often modified by different oxygen-containing functional groups, to yield cyclic or acyclic xanthophylls. Much work has also been focused on the identification, production, and utilization of natural sources of carotenoid (plants, microorganisms and crustacean by-products) as an alternative to the synthetic pigment which currently covers most of the world markets. Nevertheless, only a few carotenoids (${\beta}-carotene$, lycopene, astaxanthin, canthaxanthin, and lutein) can be produced commercially by fermentation or isolation from the small number of abundant natural sources. The market and demand for carotenoids is anticipated to increase dramatically with the discovery that carotenoids exhibit significant anti-carcinogenic activities and play an important role in the prevention of chronic diseases. The increasing importance of carotenoids in the feed, nutraceutical food and pharmaceutical markets has renewed by efforts to find ways of producing additional carotenoid structures in useful quantities. Because microorganisms and plants synthesize hundreds of different complex chemical carotenoid structures and a number of carotenoid biosynthetic pathways have been elucidated on a molecular level, metabolic and genetic engineering of microorganisms can provide a means towards economic production of carotenoid structures that are otherwise inaccessible. The aim of this article is to review our current understanding of carotenoid formation, to explain the perceived benefits of carotenoid in the diet and review the efforts that have been made to increase carotenoid in certain microorganisms.

The Study on the Ion Water Characteristics of Raw Water in the Domestic Natural Mineral Water (국내 유통 중인 먹는샘물 원수의 이온류 수질 특성에 관한 연구)

  • The goal of this study is to provide basic data to establish a foundation for the provision of safe drinkable water. The raw water of natural mineral water was analyzed to determine the quantities of anions (F-, Cl-, NO3-N-, and SO42- ) and cations (Ca2+, K+, Mg2+, and Na+) during the former and latter half of 2016. Analysis of the current quality of the raw water of natural mineral water among domestic manufacturers showed average anions contents of 0.46mg/L of fluorine, 8mg/L of chlorine ion, 1.5mg/L of nitrate nitrogen, and 12mg/L of sulfate ion. While the fluorine content was greater than the water quality criterion of 2.0mg/L at four points, the fluorine level was overall stable. The average cations contents included 21.3mg/L of calcium, 1.0mg/L of potassium, 3.4mg/L of magnesium, and 9.6mg/L of sodium. The chemical characteristics were compared among the major ions, and the results are presented in a piper diagram. The content ratio of cations was in the order of Ca2+> Na+>Mg2+>K+, whereas that of anions was in the order of SO42->Cl->NO3-N->F-. While the cations were slightly scattered, the anions were generally concentrated except for at a few points. The Ca-Na-HCO3 type was dominant overall in water sources from diorite, gneiss, and granite, while the Na-Mg-Ca-HCO3-Cl type was dominant in basalt sources. Mineral water manufacturers source their water under various conditions, including in-hole casing, excavation depth, and contact state of bedrock; even within the same rocky area, some differences in the water quality type can occur. When the depth of the water source was taken into account, the mean anions contents of F-, Cl-, NO3-N-, and SO42- were similar, with no significant differences according to depth. Of the cations, K+ and Na+ showed no significant differences across all the tubular wells, whereas Ca2+ and Mg2+ decreased in content with depth.

Occurrence of an invertase producing strain of Aspergillus niger LP5 isolated from longan pollen and its application in longan syrup production to feed honey bees (Apis mellifera L.)

  • Background: In northern Thailand, the longan flower is the principal nectar source for honey production. Microorganisms play a critical function in the agricultural ecology. The morphological characteristics of fungal species found in longan pollen were studied. Aspergillus spp. were found to be invertase-producing strains and were employed in the longan syrup production process. The purpose of this study was to evaluate the effects of invertase-added longan syrup on the adult honey bee population numbers that were fed by this syrup for 16 weeks. Results: Different fungal species were found in longan pollen samples. Aspergillus was the main genus, with three predominant sections: Nigri, Flavi, and Terrei. Other isolated species were Trichoderma spp., Rhizopus spp., Neurospora spp., Chaetomium spp., Fusarium spp. and Penicillium spp. However, Aspergillus spp. is the only fungal species that produces the enzyme invertase. The invertase-producing strains belonging to the Aspergillus section Nigri were found to be A. niger LP5 with an optimum activity at pH 6.0 and 60℃. When A. niger LP5 invertase was used for longan syrup processing, the highest levels of glucose (3.45%) and fructose (2.08%) were found in invertase added longan syrup (C), while fresh (A) and boiled longan syrup (B) had lower contents of both sugars. The sucrose content was detected in (A) at 4.25%, while (B) and (C) were at 4.02% and 3.08%, respectively. An appropriate amount of sugar to feed and maintain the honey bee population was considered. The data showed no statistically significant differences between the two selected forms of longan syrup compared to the sugar syrup examined by the adult honey bee population. Conclusions: The main species of isolated fungi from longan pollen were Aspergillus spp. The discovery of an invertase-producing strain of A. niger LP5 has enabled its application for enzyme utilization in the invert sugar preparation process. The adult worker bee populations fed by longan syrup from both boiled and invertase-added sources showed an increasing trend. Artificial syrup made from longan fruit to feed honey bees when natural food sources are limited can be applied.

Characterization of human cardiac mesenchymal stromal cells and their extracellular vesicles comparing with human bone marrow derived mesenchymal stem cells

  • Cardiac regeneration with adult stem-cell (ASC) therapy is a promising field to address advanced cardiovascular diseases. In addition, extracellular vesicles (EVs) from ASCs have been implicated in acting as paracrine factors to improve cardiac functions in ASC therapy. In our work, we isolated human cardiac mesenchymal stromal cells (h-CMSCs) by means of three-dimensional organ culture (3D culture) during ex vivo expansion of cardiac tissue, to compare the functional efficacy with human bone-marrow derived mesenchymal stem cells (h-BM-MSCs), one of the actively studied ASCs. We characterized the h-CMSCs as CD90low, c-kitnegative, CD105positive phenotype and these cells express NANOG, SOX2, and GATA4. To identify the more effective type of EVs for angiogenesis among the different sources of ASCs, we isolated EVs which were derived from CMSCs with either normoxic or hypoxic condition and BM-MSCs. Our in vitro tube-formation results demonstrated that the angiogenic effects of EVs from hypoxia-treated CMSCs (CMSC-Hpx EVs) were greater than the well-known effects of EVs from BM-MSCs (BM-MSC EVs), and these were even comparable to human vascular endothelial growth factor (hVEGF), a potent angiogenic factor. Therefore, we present here that CD90lowc-kitnegativeCD105positive CMSCs under hypoxic conditions secrete functionally superior EVs for in vitro angiogenesis. Our findings will allow more insights on understanding myocardial repair.

  • Natural gas fuel has known to be very promising in terms of abundancy and economic value. Therefore it is widely treated as research topics in a variety field of production, storage and utilization. Natural gas has become one of the major sources for the power generation by using internal combustion engines(ICE). Development of natural gas fuel injection device should be preceded to realize a reliable natural gas fuel supply system for a MW class power generation reciprocating ICE. In this research, an injection valve which consists of solenoid and body part with a moving plate was designed and its dynamic performance was experimented in the engine-like environment. Displacement length and diameter of an armature and diameter of a solenoid coil were tested at former study. In this research the effect of materials of solenoid core, size of main housing inlet and supply gas pressure are examined.

Mixture Proportioning Approach for Low-CO2 Lightweight Aggregate Concrete based on the Replacement Level of Natural Sand (천연모래 치환율에 기반한 저탄소 경량골재 콘크리트 배합설계 모델)

  • The purpose of this study is to propose a mixture proportioning approach based on the replacement level of natural sand for reducing $CO_2$ emissions from artificial lightweight aggregate concrete(LWAC) production. To assess the effect of natural sand on the reduction of $CO_2$ emissions and compressive strength of LWAC, a total of 379 specimens compiled from different sources were analyzed. Based on the non-linear regression analysis using the database and the previous mixture proportioning method proposed by Yang et al., simple equations were derived to determine the concrete mixture proportioning and the replacement level of natural sand for achieving the targeted performances(compressive strength, initial slump, air content, and $CO_2$ reduction ratio) of concrete. Furthermore, the proposed equations are practically applicable to straightforward determination of the $CO_2$ emissions from the provided mixture proportions of LWAC.

Effects of Supplemental Macroalgae and Spirulina in the Diets on Growth Performance in Juvenile Abalone(Haliotis discus hannai) (참전복 사료의 해조류 및 spirulina 첨가 효과)

  • A 20-week growth trial was conducted in flow-through aquarium system to investigate the effects of supplemental macroalgae and spirulina in the diets on growth and body cmposition in juvenile abalone (Haliotis discus hannai). Four replicate groups of the abalone averaging 65mg were fed one of ten isonitrogenous (34%) and isolipidic (7.5%) diets containing 8.1% Ulva, 7.5% Undaria, 11% Laminara, 11% Sargassum, spirulina (5, 10 and 15%), or dried Undaria powder (10 and 20%). In addition, these formulated diets were compared with natural food(dried Undaria). Survival rate of abalone were not significantly affected by the different dietary macoralgae sources, spirulina or dried Undaria powder levels (P>0.05). Weight gain and soft body weight of abalone fed the diet containing Sargassum was significantly higher (P<0.05) than those of abalone fed the diets containing Laminaria, 20% dried Undaria powder and natural food. Survival rate, weight gain, soft body weight and shell length of abalone fed natural food were lowest (P<0.05) among all diet. Moisture, protein and lipid contents of soft body were not influenced by experimental diets except natural food. Lipid content of abalones fed natural food was significantly lower than those of abalone fed other diets (P<0.05). These data indicate that abalone can more efficiently utilize Sargassum than Ulva, Undaria, Laminaria or spirulina.

Direct Identification of Vibrio vulnificus by PCR Targeting Elastase Gene

  • A PCR assay for the rapid detection of Vibrio vulnificus strains was developed using a virulence gene for elastase found in various Vibrio species. The DNA sequences in the elastase gene facilitated the identification of a species-specific probe for pathogenic V. vulnificus strains from both clinical and environmental sources. Using an elastase gene-based PCR reaction, a species-specific 507-bp PCR product was visualized by agarose gel electrophoresis. Three different DNA extraction methods were then compared to improve the simplicity and rapidity of detection. A PCR assay using the conventional DNA extraction or boiling method was able to detect as few as 25 V. vulnificus cells, making the detection limits at least 1-log-scale lower than that for the EDT A-treated DNA extraction method. In particular, the boiling method, which does not require purification of the chromosomal DNA, was very effective in terms of simple and rapid detection. Meanwhile, the detection limit in a mixed bacterial culture that included other bacteria, such as Escherichia coli or Bacillus subtilis, was two V. vulnificus cells, which was 1-log-scale lower than that for the control. Accordingly, when coupled with a new DNA extraction method, the elastase gene-based PCR can provide a rapid, specific, and sensitive method for identifying V. vulnificus in clinical and environmental samples.

On Efficient Implementation of the Regeneration Projects for the Zones Vulnerable to Natural Disasters (자연재해위험지구 정비사업의 효율적인 추진방안 연구)

  • The purpose of this study is to suggest some effective ways to implement the regeneration projects for the zones vulnerable to natural disasters which have been delayed due to shortage of the financial resources, and thereby complete the disaster prevention projects as early as possible and thus secure the financial resources for the projects continuously. For this purpose, the researcher reviewed the statutory grounds and designation procedures or the zones vulnerable to natural disasters, and thereupon, examined and analyzed the regeneration projects being implemented in Gyeonggi-do in view of contents by zone, history of damages, annual investment plans and financial resources for investments. In addition, the researcher examined the criteria for reserve of the disaster management fund and its uses and investigated the fund sources and expenditures and thereby, addressed the problems of the statutory reserve and suggested their solutions. All in all, the researcher suggested that the disaster management fund should be invested in regeneration projects for the zones vulnerable to natural disasters in order to remove the disaster risks earlier and that the uses of the fund should be adequately deregulated with a reserve fund ceiling system to secure the financial resources for the disaster prevention projects continuously.
