  • Two lignanes, Comp.-I, mp 108-1$0^{\circ}C$ and Comp.-II 50-52$^{\circ}$were isolated from Korea ginseng extract by repeated column chromatographic purification. Comp.-I was identified as gomisin-N and Comp.-II as gomisin-A by spectrometric analysis, both of which have already been described as the anti-hepatotoxic lignan components of Schizandra chinesis Bail.

Purification and Characterization of Biosurfactant from Tsukamurella sp. 26A

  • A biosurfactant produced by Tsukamurella sp. 26A was purified by procedures including acid precipitation, ethylacetate extraction, and adsorption chromatography. The purified biosurfactant reduced the surface tension of water from 72 mN/m to 30 mN/m at a concentration of 250 mg/l, whereas the minimum interfacial tension against n-hexadecane was lowered to 1.5 mN/m at a concentration of 40 mg/i. The compound stabilized oil-in-water emulsions with a variety of commercial oils and had strong emulsification and stabilization activities when compared to those of commercial emulsifiers and stabilizers. Surface tension was stable over a broad range of pH (2-12) and temperature ($100^{\circ}C$, 3h). The biosurfactant was identified as glycolipid having a hydrophilic moiety of trehalose.

  • The process of biological treatment of organic wastewater is principally associated with those of self-purification in the natural water environment. The treatment system has e intensive function of stabilizing wastewater more effectively than in natural water, which is like natural water concentrated in a small space. Biological treatment of wastewater involves activated sludge and various modified process, trickling filter, rotating disk, oxidation ditch, etc. for aerobic decomposition and anaerobic processes such as anaerobic decomposition and methane fermentation. The basic characteristic of these processes is the use of mixed culture for the conversion of pollutants. This review forcuses on the various kinds of microorganisms related to each treatment processes. Kinetic analysis of the activated sludge process is discussed in order to understand the basis of control and maintenance of the biological treatment process.

  • An extracellular protease was purified to homogeneity from the culture supernatant of psychrotrophic bacteria Bacillus sp. JH 108 using procedures including ammonium sulfate fractionation, anion exchange chromatography, gel filtration chromatography, and cation exchange chromatography. The enzyme exhibited a molecular weight of 36 kDa, an optimum pH of 8 to 9, and optimum temperature of $60^{\circ}C$. The enzyme preferentially hydrolyzed leucine at the N-terminus of peptides and thus can be classified as an aminopeptidase. It was strongly inhibited by metal chelating agents such as EDTA and l, l0-phenanthroline. The activity lost by EDTA was restored with $Zn^{2+}{\;}or{\;}Co^{2+}$. These divalent cations also stimulated the native enzyme. This suggests that the enzyme is a metalloprotease acting as a leucine aminopeptidase.

  • We constructed an efficient T-vector, pTQEST216T that employed an engineered esterase as an indicator for direct cloning of PCR products. After ligation of the XcmI-digested vector with PCR products, this cloning system could easily discriminate positive clones owing to insertional inactivation of the esterase reporter. Additionally, PCR products were efficiently cloned into this vector without the gel purification steps, owing to the well-designed multi-cloning site that was in-frame fused at the circularly permutated gap of the reporter.

  • Five crystalline substances, which are positive to$L_{IEBERMAN}-B_{URCHARD}$ reaction, were isolated from the unsaponifiable fraction of the fresh leaves of Betula latifolia $K_{OMAROV}$ (Betulaceae)by silica gel column chromatographic purification. Compound $A\;(C_{29}H_{50}O,\;mp\;136^{\circ}), \;B\;(C_{30}H_{52}O_3,\;mp\;165^{\circ}), \;C\;(C_{30}H_{52}O_4,\;mp\;237^{\circ}), \;D\;(C_{30}H_{52}O_3,\;mp\;196^{\circ})\;and\; E\;(C_{30}H_{52}O_4,\;mp\;121^{\circ})$ were isolated. Compound B was characterized as a new tetracyclic triterpenoid. Compounds A, C and D were identified as ${\beta}-sitosterol$, betulatriterpene C, and betulafolienetriol, respectively.

Purification and Characterization of Superoxide Dismutase from Pseudomonas polycolor

  • Superoxide dismutase (SOD) was purified from Pseudomonas polycolor to an electrophoretically homogeneous state and partially characterized. SOD was purified by ammonium sulfate fractionation, column chromatography on DEAE-Sephadex A-50, phenyl-Toyopearl 650 M, and gel filtration on Sephadex G-100. The molecular weight and subunit molecular weight of the purified enzyme were estimated to be 40, 000 and 20, 000, respectively. The purified enzyme remained stable at pH 9.0~11.0, $25^{\circ}C$ for 40 hr, but rapidly became inactive below 9.0. SOD was stable up to $45^{\circ}C$ at pH 9.0 with about 80% relative activity, but rapidly became inactive at temperature above that. The enzyme was insensitive to cyanide and fluoride, and sensitive to hydrogen peroxide and azide. The results suggest that the enzyme be an iron-containing SOD.

  • This study was conducted to find out the effect of natural zeolite addition on the purification of livestock farming wastewater and the application of zeolite used for purification of livestock farming wastewater on growth of sudangrass. In a semi-continuous process it was found that COD, $NH_4-N$, $NO_3-N$ and T-N contents in the effluent solution decreased with the treatment of natural zeolite. The zeolite addition was effective on the purification of livestock farming wastewater. By the results of application for sudangrass cultivation, the accumulative N-uptake contents by sudangrass were lower than that of control(35kg/10a) at early growth stage, but were higher at the late growth stage.
