추출용매 변화에 따른 바나바(Lagerstroemia speciosa Pers.) 잎 추출물의 항당뇨 효과 (Anti-diabetic Effects of Banaba Leaf Extracts (Lagerstroemia speciosa Pers.) through Solvents)

  • 본 연구는 바나바 잎 추출 용매에 따른 항당뇨 효과에 관한 것으로 세포 독성 실험을 통하여 인체에 대한 독성이 없음을 확인하였고 $\beta$-cell 인슐린 분비 실험을 통하여 항당뇨 효과를 알아보았다. 또한 당뇨 유발쥐를 이용한 인슐린 분비 실험, 체중변화 측정, 혈중 및 혈청의 glucose 농도, 총콜레스테롤 수치, HDL-cholesterol 농도, triglyceride 농도, free fatty acid 농도, langerhans islet에서의 인슐린 분비 의 확인을 통해 혈당조절 효과를 비교하였다. 물 혹은 에탄올을 단독으로 이용하여 추출한 추출물이나 70%, 90% 에탄올로 추출한 추출물 보다 열수로 먼저 추출한 후 에탄올로 다시 추출하여 혼합안 추출물이 항당뇨 효과가 더 뛰어남을 알 수 있었다. 이는 친수성(hydrophilic) 유효성분들과 소수성(hydrophobic) 유효성분들의 추출 유무나 함량에 의한 것이다. 특히, 많은 문헌에 언급된 코로소린산이라는 단일 성분(unique)에 의한 항당뇨 효과라기보다는 친수성 및 소수성 폴리페놀 및 항산화성분과 같은 유효성분들이 조합되어(mixture) 항당뇨 효과에 영향을 미치는 것으로 사료된다.

보중익기탕의 T세포 증식 유도 효과 (Effect of Bu-Zhong-Yi-Qi-Tang on Proliferation of T Cells)

  • 먼저 OVA항원에 대해 특이적으로 증식반응을 나타내는 T세포주를 수립하였고, 수림된 세포주는 세포 표면 단백질이 CD4$^{+}$CD8$^{-}$이며 IL-2와 IFN-${\gamma}$를 분비하는 Type I에 속하는 보조 T세포(Thl)인 것을 확인하였다. 보중익기탕의 total 분획은 OVA항원에 대해 특이적으로 반응하는 Thl세포의 증식반응을 증가시키는 효과를 나타내지 않았으며 고농도에서 오히려 증식반응을 억제하였다. 그러나, 보중익기탕의 polysaccharide 분획은 전반적 인 농도에서 T세포의 증식반응을 유의하게 증가시키는 효과가 있는 것으로 나타났다. 보중익기탕의 polysaccharide 분획을 첨가하였을 때 T세포의 IL-2 분비량은 대조군보다 약간 적었지만, IFN-${\gamma}$ 분비량은 대조군보다 증가하였다. 그리고, 분비된 IL-2와 결합하는 T세포의 IL-3 수용체 발현양도 증가하는 것으로 나타났다. 또한, 항원제시세포의 MHC class II의 발현양도 증가시켰다. 이상의 결과로 보중익기탕의 polysaccharide분획은 T세포의 IL-2수용체 발현양을 증가시키고, 항원제시세포의 MHC classs II의 발현양을 증가시켜서 T세포의 증식반응을 증가시키는것으로 생각된다 그리고 보중익기탕이 생체 면역반응에 미치는 보다 정확한 효과를 평가하기 위해서는 직접 살아있는 실험동물에 투여하는 in vivo 실험이 필요하다.

  • Objective : In the present study, we examined whether Canavalia gladiata D.C. (CG) and Arctium lappa L., Redix (AL) mixture (CGAL), their components, lupeol and chicoric acid, regulate immune system and suppress the tumor in vitro and in vivo. Methods : LPS-induced reactive oxygen species (ROS) and nitric oxide (NO) were measured after treatment with CG extract (CGE), CGAL, lupeol, chicoric acid and lupeol and chicoric acid mixture (lupeol+CA) in Raw264.7 cell. To determine the effect of CGE on immune responses, immune cell population and IgG production were assessed in mice. To investigate the effect of CGAL and their component on anti-tumor activity, tumor volume and weight were measured, cell cycles and immune cell population were analyzed in MC38 injected tumor bearing mice. Also, NK cell activity was determined in splenocyte isolated from tumor bearing mice. Results : CGE, CGAL, lupeol, chicoric acid and lupeol+CA decreased the LPS-induced ROS and NO production without cell toxicity in RAW264.7 cells. CGE increased the immune cell populations of $CD4^+T$, $CD8^+T$ and macrophages in various immune organ of mice. In tumor bearing mice, CGAL, lupeol, chicoric acid and lupeol+CA suppressed tumor volume and weight. In cell cycle analysis, they decreased the percentages of S phase. In addition, CGAL, lupeol, chicoric acid and lupeol+CA immune cell populations of $CD4^+T$, $CD8^+Tcell$, NK cell and macrophage in tumor as well as NK cell activity. Conclusion : CGAL and its compounds may enhance immune responses and suppress tumor growth, and may be capable of developing health functional foods.

Effects of Surface-Applied Dairy Slurry on Herbage Yield and Stand Persistence : I. Orchardgrass, Reed Canarygrass and Alfalfa-Grass Mixtures

  • Comparative studies of the effects of rates and frequency of application of dairy slurry on herbage yield and stand persistence of alfalfa and various forage grasses have not previously been conducted. The results being reported here are part of a larger study having a primary objective of comparing the effectiveness of alfalfa (Medicago sativa L.), various grasses and alfalfa-grass mixtures for utilizing nutrients from applied dairy slurry. The objectives of this part of the study were to evaluate the effects of various rates and frequencies of application of slurry on herbage yield and stand persistence of orchardgrass (Dactylis glomerata L.), reed canarygrass (Phalaris arundinacea L.), and alfalfa-orchanrdgreass and alfalfa-reed canarygrass mixtures managed as a 4-cutting management system. A randomized complete block design with treatments in a split plot arrangement with four replicates was used. The main plots consisted of 9 fertility treatments: 7 slurry rate and time of application treatments, one inorganic fertilizer treatment, and an unfertilized control. The sub-plots consisted of the two grasses and two alfalfa-grass mixture mentioned above. Slurry was composed from stored solids scraped from the alleyways of a free-stall housing barn and water added to form a slurry having about 8% solids. Manure was pumped from a liquid spreader tank into 10.4 L garden water cans for manual application to the plots. Herbage yields within species were generally unaffected by various rates of application in the first production year. Herbage yields of grasses and alfalfa-grass mixtures the second year were generally not affected by frequency of application for the same rate of slurry applied. Slurry application resulted in greater herbage yield increases in grasses than alfalfa-grass mixtures in the 4-cutting management system. In general, herbage dry matter yields of grasses from the dairy slurry treatments equaled or exceeded yields from the inorganic fertilizer treatment. Stand ratings of grasses and alfalfa-grass mixtures were not changed by manure application rates. In this study, the highest rate of slurry ($967kg\;total\;N\;ha^{-1}$ in 1995 plus $2,014kg\;N\;ha^{-1}$ in 1996) was not detrimental to herbage yields or stand persistence of any of the species. It was concluded that applying dairy slurry to these cool-season grasses and alfalfa-grass mixtures managed in a 4-cutting system is an acceptable practice from the standpoint of herbage yield and satnd persistence and by doing so the utilization of inorganic fertilizers can be reduced.

This paper compares the effects of rates and frequencies of application of dairy slurry on herbage yield and stand persistence of alfalfa, orchardgrass, tall fescue (Festuca arundinacea Schreb.), and alfalfa-orchardgrass mixture managed as a 5-cutting system. A randomized complete block design with treatments in a split plot arrangement with four replicates was used. The sub-plots were the forage species. Manure used for the study was composed from stored solids scraped from the alleyways of a free-stall dairy barn. Water was added to from a slurry having about 8 % solids. Slurry was pumped from the liquid spreader tank into 10.4 L garden watering cans for manual application to the plots. Herbage yields of alfalfa, tall fescue, and alfalfa-orchardgrass were generally not affected by slurry application rates and were not significantly different from the inorganic fertilizer treatment. Tall fescue significantly outyielded all other forage species at all manure and the inorganic fertilizer treatments in the second year when rainfall during the growing season was unusually high. Grasses generally had a greater response to manure applications than alfalfa and alfalfa-orchardgrass. Increasing rates of manure did not increase herbage yields of alfalfa and alfalfa-orchardgrass. Herbage yields within each species were not affected by frequency of application of the same total rate. Stand ratings of alfalfa, orcahrdgrass and alfalfa-orchardgrass were significantly lower for the very high manure application rate compared to the control treatment. Based upon the results of this study, multiple annual applications of slurry manure can be made onto these species at rates up to $1,700kg\;total\;N\;ha^{-1}\;yr^{-1}$ without detrimental effects on herbage yield and stand persistence.

Effect of D-glucose feeding on mortality induced by sepsis

  • Sepsis is the life-threatening response to infection which can lead to tissue damage, organ failure, and death. In the current study, the effect of orally administered D-glucose on the mortality and the blood glucose level induced by D-Galactosamine (GaLN)/lipopolysaccharide (LPS)-induced sepsis was examined in ICR mice. After various amounts of D-glucose (from 1 to 8 g/kg) were orally fed, sepsis was induced by injecting intraperitoneally (i.p.) the mixture of GaLN /LPS. Oral pre-treatment with D-glucose dose-dependently increased the blood glucose level and caused a reduction of sepsis-induced mortality. The oral post-treatment with D-glucose (8 g/kg) up to 3 h caused an elevation of the blood glucose level and protected the mortality observed in sepsis model. However, D-glucose post-treated at 6, 9, or 12 h after sepsis induction did not affect the mortality and the blood glucose level induced by sepsis. Furthermore, the intrathecal (i.t.) pretreatment once with pertussis toxin (PTX; $0.1{\mu}g/5ml$) for 6 days caused a reduction of D-glucose-induced protection of mortality and hyperglycemia. Furthermore, once the hypoglycemic state is continued up to 6 h after sepsis initiated, sepsis-induced mortality could not be reversed by D-glucose fed orally. Based on these findings, it is assumed that the hypoglycemic duration between 3 and 6 h after the sepsis induction may be a critical time of period for the survival. D-glucose-induced protective effect against sepsis-induced mortality appears to be mediated via activating PTX-sensitive G-proteins in the spinal cord. Finally, the production of hyperglycemic state may be critical for the survival against the sepsis-induced mortality.

표식유전자를 이용한 담배와 감자의 원형질체 융합 (Protoplast Fusion of Nicotiana glauca and Solanum tuberosum Using Selectable Marker Genes)

  • 표식 유전자를 이용하여 체세포 잡종체를 선발하기 위한 연구의 일환으로 감자조직에는 T-DNA를 도입하여 식물호르몬 무첨가 배지에서도 생장가능한 형질전환체을 획득하고, 담배조직에는 NPT H gene을 도입하여 kanamycin에 대해서 저항성을 나타내는 형질전환체를 획득하여 각각의 특성구명과 원형질체를 유리하여 도입된 유전자 marker를 이용해서 융합을 시도한 바 그 결과는 다음과 같다. 1. 감자 괴경에서 Agrobacterium tumefaciens Ach5와 A. rhizogenes ATCC15834를 접종하여 crown gall tumor 및 hairy root를 유기하였으며 이러한 tumor조직은 식물 호르몬 무첨가 배지에서 생장이 가능하였다. 2. 감자에서 유기된 hairy root로부터 callus 형성은 2.4-D 2mg/1 첨가된 MS 배지에서 가장 양호하였으며 casein hydrolysate 1g/1가 첨가하면 유연 한 callus의 증식이 더욱 왕성하였다. 3. Activated charcoal이 0.5~2.0g/1 첨가된 배지에서는 crown gall tumor callus의 절단면이 갈변되는 것을 방지 할 수 있어 생존률을 높일 수 있었으나 hairy root에서는 갈변되어 고사되었다. 4. 2, 4-D 2mg/1와 casein hydrolysate 1g/1를 첨가한 배지에 hairy root callus를 현탁배의한 결과 양호한 많은 callus 덩어리들을 단시간에 얻을 수 있었다. 5. $Tri^-$parental mating으로 NPTII gene이 coding되어있는 binary vector인 pGA643을 wild type 및 disarmid된 Agrobacterium 내에 도입하여 Agrobacterium tumefaciens Ach5/pGA643, A.tumefaciens $A_4T$/pGA643, A. tumefaciens LBA4404/pGA643를 획득하였다. 이 세 개의 conjugant를 사용하여 0.7% agarose gel 상에서 pGA643을 확인하였다. 6. pGA643이 도입왼 Agrobacterium tumefaciens LBA4404와 담배 조직과 동시배양하여 kanamycin $100\mug$/ml 첨가된 배지에 생존하는 callus를 선발하였으며 동일배지에서 callus의 증식이 가능하였다. 7.형질전환된 담배 callus로부터 식물체 형성은 BA 2mg/1를 첨가한 배지에서 가능하였다. 8. 재분화된 담배의 엽조직은 kanamycin.이 $1000\mug$/ml 첨가된 MS 배지에서도 왕성히 callus가 유기되어, 이는 재분화체에서도 NPTII gene이 그대로 유지되고 있음을 확인할 수 있었다. 9. 담배의 정상 shoot와 형질전환된 shoot를 kanamycin이 $100\mug$/ml이 함유된 MS배지에 기내삽목한 결과, 정상 shoot는 발근이 되지 않고 황화 되었으나 형질전환된 shoot는 발근이되었으며 정상적으로 생장을 하였다. 10. 감자의 T-DNA가 도입된 현탁배양 callus는 cellulase 2%, macerozyme 2%, dricelase 1%에서 양호하게 유리되었다. 11. Osmoticum으로서 mannitol 농도 0.8M에서 담배와 감자의 두 조직 모두 원형질체유리가 가장 효과적이었다. 생존력은 T-DNA가 도입된 감자의 hairy root를 callus로 탈분화 시킨 후 현탁배양한 callus가 mannitol 0.5M에서 97%를 나타냈고, 그리고 NPTII gene이 도입된 담배의 엽조직은 mannitol 0.7M에서 94%로 최고를 타나냈다. 12. 원형질체 융합은 PEG solution 처리 15분 후부터 관찰되기 시작하여 20분에 완전히 융합되었고, 융합된 원형질체는 선발 marker인 호르몬 무첨가 및 kanamycin 첨가배지에서 배양 5일 후에 세포벽이 재생되었으며 4주일 후부터 colony들이 관찰되었다.

하계 대한해협 저층냉수의 물리.화학적인 특성 및 기원 (Physicochemical Properties and the Origin of Summer Bottom Cold Waters in the Korea Strait)

  • Hydrographic survey in the Korea Strait has long history that has begun in August 1917 at the Busan - Tsushima cross section, still continues to date. However, chemical properties of bottom cold water found exclusively in the western channel of the Korea Strait during summer did not receive much scientific attention. The aim of the study is to decipher the enigmatic origin of the Korea Strait Bottom Cold Water (KSBCW) in terms of chemical properties. The physicochemical properties of the KSBCW are extracted from the CREAHS II hydrographic data. OMP method was applied to analyze origin of the KSBCW quantitatively. The KSBCW is well defined by low temperature below $10^{\circ}C$. The cold waters exhibited the local presence near the coast at about 120m depth with a thickness of 20m to 30m. The cold water was characterized by relatively cold, saline and higher chemical concentrations than adjacent waters. The KSBCW seems to have different origin kom that of the coastal upwelled waters at the Ulgi-Gampo because it is saline, denser and contains considerably less dissolved oxygen than upwelled waters. The physicochemical properties are reported to have noticeable annual variations which suggest the complex origin of the KSBCW. OMP analysis show that the KSBCW is a mixture of three water types; TMW (24%), ESIW (36%) and ESPW (40%). Relationship between the KSBCW and the east Sea circulation is traced by mapping the water masses that have similar T, S and DO of KSBCW. The result showed that the KSBCW is most possibly an extension of southward flowing coastal intermediate waters. Front these results, we expect that the monitoring KSBCW will provide us valuable information about the East Sea circulation.

  • Today, every culture has taken on the compromise form by means of the cultural difference, variety, and pluralism according to the internationalization and the advance so that it has been developed toward the half-blooded and multilayered the aspect. In accordance with this current of the times, this thesis observed the feminism found in the multiculturism of the end of the 20th century, the third world, and technology with the side of the tendency of modern fashion, considered the hybride phenomenon which is pulling down the wall between culture and genre due to the social diversification. and predicted the fashion trend do 21st century serf on it. Multiculturism is the movement that began to arise in the academic world of America and the literary world form the close of 1980's in accepting the variety of culture and regarding the culture with the more balanced and wide view and just as it is, it means the attitude of accepting one or more cultures of variely and the position of taking interest in the culture of minority race not the culture of a governing race. It is the fashion of feminism adapts dualism like unisex, androgynous look, etc of bisexual lendency in the 1980's, it shows new style with crossover of liberal sense because there is not the difference of sex in fashion. The eco-feminism pursues the natural sexuality not being instrumental and dismantling and expressed it in the Gender expression of an integrated human being. The trend of ethnic fashion in the close of 20th century is that the element of hippie is working so strongly. By adding embroidery of Oriental style, accessories of Indian style, feathers, beads, a hempen hood to the ethnic costumes of Asia and Latin, is shows the figure of ethnic hippie. As the cycler fashion is the future clothes through technology of computer, it uses a cool glass material bringing up the image of a spacesuit in order to expresses cyber image through artificial color combination of sheen colors, Though this techno-color fashion has established the fresh stimulation and the innovative aspect with ultramodern materials and image of futurism, it transmits a hope of estranged people and the natural elements. Hybride means a cross and mixture of animal and plant in Korean and is also called fusion. The phenomenon of hybrid predicts to comes the period of a cross and variation because something completely new comes into the world by contamination, mixture and compromise through meeting something different each other and it has on advantage of developing something existing to one more stage. It is prospected that in the society of 21st century, the borderline of traditional gender will be disappeared, variety and individuality will determine the individual behavior, and the masculine value will be substituted by feminine value. In the society giving priority to feminine value, a fashion stuck closely to women is what must reflect lives of woman under the proposition of woman's beauty, being on original function. So, it is considered that a fashion with added convenience and practicality having the function which is easy to put on, comfortable to act, able to express solves so much, and able to show various appearances according to T.P.O will get into the spotlight.

Brassinolide와 Auxin류(類)의 식물(植物) 생장조절제(生長調節劑)가 무우 자엽병(子葉柄)의 굴절(屈折)에 미치는 영향(影響) (Effect of Brassinolide and Auxin Type Plant Growth Regulators on Radish Petiole Curvature)

  • HBR[(22s, 23s)-homobrassinolide]과 auxin류(類)의 생장조절제(生長調節劑) 단독(單獨) 또는 혼용(混用) 처리시(處理時) 무우 자엽병(子葉柄)의 상편생장(上偏生長)(굴절(屈折))에 어떻게 影響(影響)을 마치는가를 구명(究明)하여 얻어진 결과(結果)를 요약(要約)하연 다음과 같다. 1. HBR 단독처리(單獨處理)는 농도(濃度)에 관계(關係)없이 무우 자엽병(子葉柄) 굴절(屈折)에 영향(影響)하지 않았다. 2. 천연식물생장조절제(天然植物生長調節劑) IAA, IBA, NAA의 굴절효과(屆折效果)는 짧았고 IAA는 처리후(處理後) 4(시간(時間)), IBA는 12~24 시간(時間), NAA는 12 시간(時間) 지나면 감소(減少)하였다. 이 가운데 NAA 100ppm 처리구(處理區)에서 가장 높았고 IAA 처리(處理)에서 전반적으로 가장 높았고 IAA 처리(處理)에서 전반적으로 가장 효과(效果)가 낮았다. 3. 합성(合成) auxin 류(類)인 MCP나 2, 4-D의 굴절효과(屈折效果)는 대체로 36시간(時間) 지속(持續)되었으며 고농도(高濃度)인 100 ppm에서 무처리(無處理)에 3배(倍) 이상(以上) 증대(增大)시켰다. 30ppm 이하(以下)의 저농도(低濃度)에서는 효과(效果)도 작고 농도간(濃度間) 차(差)도 작았다. 4. NAA와 HBR의 혼용(混用)은 NAA 100ppm 처리(處理)에서 HBR 100 ppm 단독(單獨)보다 약(約) 3배(倍) 증대(增大)시켰으나 30 ppm 이하(以下)의 농도(濃度)에서는 큰 차이(差異)가 없었다. 5. HBR에 MCP 3ppm의 혼용(混用)은 HBR 단독처리(單獨處理)에 비(比)하여 굴절각(屈折角)을 약(約) 3배(倍) 증대(增大)시켰다. HBR과 2.4-D의 혼용(混用)은 농도(濃度)가 증가(增加)할수록 굴절각(屈折角)의 반응(反應)이 크게 나타났으며 100 ppm 혼용(混用)에서 HBR 단독처리(單獨處理)에 비(比)하여 약(約) 8배(倍) 증가(增加)시켰다.
