  • Maximal production of heteropolysaccharide-7(PS-7) with a batch culture for 48 hr was $10.0\;g/{\ell}$ and its conversion rate from 2% (w/v) glucose to PS-7 was 50%. After substitution of media, production of PS-7 continued and reached its maximal production. The highest productivity occurred when the fresh medium, which contained all ingredients, for the production of PS-7 was substituted. Higher production of PS-7 was maintained at a dilution rate of 0.0125, which was established as the optimal dilution rate for the production of PS-7 by B. indica HS-2001.

  • The aim of this study is to identify the role of this natural treatment system, which consists of Oenanthe javanica and wasted concrete as bio-media. Therefore, it was designed to experiment water quality, BOD, SS, T-N, T-P to recognize the efficiency of treatment system with one biofilter tank using wasted concrete and vegetation bed using Oenanthe javanica. It was also designed to compare two different biofilter reactors, which are air lift and upflow methods. In the result, it was demonstrated that upflow method is more efficient system to control water quality of polluted streams. The vegetation bed using Oenanthe javanica has the treatment efficiency of 41 % (BOD), 52 % (COD), 60% (SS), 36 % (T-P), 70 % ($NH_4-N$). It was therefore proved that removal rates of nutrients are not so good except $NH_4-N$ concentration with nitrification.

  • The present study has been perromed to investigate the optimal conditions for protoplast formation and regeneration of oleandomycin-producing Streptomyces antibioticus (S. antibioticus) KCTC 1081. Mycelia were grown in YME medium containing 0.2% (w/v) glycine and converted into the protoplast by incubating at 35.deg.C for 60 minutes in protoplast buffer (P buffer) containing 4 mg/ml lysozyme. The reversion of protoplasts to the normal filamentous state was examined by the growth on various synthetic agar media. A high reversion rate was obtained by incubating the protoplasts on a hypertonic agar medium containing 20 mM $Mg^{++}$, 5 mM $Ca^{++}$ and 0.3 M sucrose at 28.deg.C for 5 days. From these experiments, we established the improved regeneration medium and a protocol which supports higher and more consistent levels of regeneration of S. antibioticus protoplasts. The regenerant showed an increased antimicrobial activity compared with that of the initial strain.n.n.

  • The rate of growth and production of bioactive substances from Rehmannia glutinosa Liboschitz (Scrophulariaceae) were studied with the variation on the constituents of the culture media. The best growth was observed from MS basal medium containing 3.0 ppm NAA and 2.0 ppm kinetin. Carbohydrates (fructose, glucose and sucrose), phytosterols(${\beta}-sitosterol$, campesterol and stigmasterol) and carotenoid like substances were identified by GC-MS and TLC from the callus mass. However, catalpol was not detected from both solid and cell suspension cultures containing geraniol. Callus cultured Rehmannia glutinosa in the MS basal medium containing 0.1 ppm NAA and 0.1 ppm kinetin become differentiated to root.

Study on the adsorption of berberine hydrochloride (Berberine hydrochloride의 흡착에 관한 연구)

  • When a large quantity of insoluble drugs was added into a solution containing a small amount of soluble drugs, the latter was adsorbed to the former. Their adsorption was expected to alter the drug activity. Berberine hydrochloride (BH) was selected as a soluble drug and tested with frequently combined insoluble drugs and antacids for their adsorption phenomena in prufied water, gastric and intetinal fluid test solutions, respectively. The adsorption isotherms of kaolin and natural anuminum silicate with BH in the three media fitted the Langmuir (LM) equation, and that of talc in purified water and gastric fluid fitted it, but in intestinal fluid it fitted the Freudlich quation. The adsorption isotherm of aluminum hydroxide fitted the LM equation only in intestinal fluid. The degree of adsorption of BH in purified water and gastric fluid is in the following order: magnesium trisilicate, kaolin, natural aluminum silicate and talc; in intestinal fluid: magnesium trisilicate, kaolin, talc, natural aluminum silicate and aluminum hydroxide. Magnesium stearate did not adsorb BH.

  • In order to study the effects of $Ca^{++}$ and dbcAMP on sperm motility, cauda epididyal spermatozoa of mouse were incubated in the media containing the various concentrations of $Ca^{++}$ and dbcAMP, and Its motility were observed and analyzed. The rate(%) of the sperm with forward motility which may be fertile, reached at the equilibrium state in 30-60 min. after the beginning of incubation. $Ca^{++}$ requirement for the maintenance of sperm motility obviously appeared in 3hrs. after the beginning of incubation. In the medium containing 0.01mM $Ca^{++}$, the rate of sperm forward motility was highest(30.6-40.6%), whereas in 10mM $Ca^{++}$ medium, the rate was rather reduced because of a severe agglutination phenomenon. On the other hand, 1mM dbcAMP in the medium was significantly effective for the maintenance of sperm forward motility.

  • When enterobacterium, Escherichia coli B was cultivated with normal media in the presence of $0.2{\sim}0.6%$ Palmitoylcarnitine and $0.05{\sim}0.2%$ Ginseng Saponin, maximum population growth of the bacteria was presented 71% and 31%, respectively. Such a result, in vitro test, was concluded from the result that both detergents stimulated $C^{14}$-glucose, $C^{14}$-alanine and $C^{14}$-phosphatidylethanolamine uptake into the membrane of cells. The pre-treatment of cells with different amounts of Palmitoylcarnitine from $0.005{\sim}0.05{\mu}$ moles represented a significant increase of uptake, 33% of $C^{14}$-glucose, 129% of $C^{14}$-alanine and 158% of phosphatidylethanolamine at the concentration of $0.05{\mu}$ moles of Palmitoylcarnitine. On the other hand, the result of $C^{-2}%$ Saponin treatment showed the maximum value of uptake, 17% of $C^{14}$-glucose and 112% of $C^{14}$-alanine. In case of $C^{14}$-phosphatidylethanolamine, the maximum uptake showed 25% of increase at the concentration of $C^{14}$% Saponin.

Effect of the Photosensitizer on the Photo refractive Effect Using a Low $T_g$ Sol-Gel Glass

  • We prepared the photorefractive sol-gel glass based on organic-inorganic hybrid materials containing a charge transporting molecule, second-order nonlinear optical (NLO) chromophore, photosensitizer, and plasticizer. Carbazole and 2-{ 4-[(2-hydroxy-ethyl)-methyl-amino]-benzylidene}-malononitrile were reacted with isocyanato-triethoxy silane and the functionalized silanes were employed to fabricate the efficient photorefractive media induding 2,4,7-trinitrot1uorenone (TNF) to form a charge transfer complex. The prepared sol-gel glass samples showed a large net gain coefficient and high diffraction efficiency at a certain composition. As the concentration of photosensitizer increased, the photorefractive properties were enhanced due to an increment of charge carrier density. Dynamic behavior of the diffraction efficiency was also investigated with the concentration of the photosensitizer.

Strength properties of composite clay balls containing additives from industry wastes as new filter media in water treatment

  • Pebble matrix filtration (PMF) is a water treatment technology that can remove suspended solids in highly turbid surface water during heavy storms. PMF typically uses sand and natural pebbles as filter media. Hand-made clay pebbles (balls) can be used as alternatives to natural pebbles in PMF treatment plants, where natural pebbles are not readily available. Since the high turbidity is a seasonal problem that occurs during heavy rains, the use of newly developed composite clay balls instead of pure clay balls have the advantage of removing other pollutants such as natural organic matter (NOM) during other times. Only the strength properties of composite clay balls are described here as the pollutant removal is beyond the scope of this paper. These new composite clay balls must be able to withstand dead and live loads under dry and saturated conditions in a filter assembly. Absence of a standard ball preparation process and expected strength properties of composite clay balls were the main reasons behind the present study. Five different raw materials from industry wastes: Red Mud (RM), Water Treatment Alum Sludge (S), Shredded Paper (SP), Saw Dust (SD), and Sugar Mulch (SM) were added to common clay brick mix (BM) in different proportions. In an effort to minimize costs, in this study clay balls were fired to $1100^{\circ}C$ at a local brick factory together with their bricks. A comprehensive experimental program was performed to evaluate crushing strength of composite hand-made clay balls, using uniaxial compression test to establish the best material combination on the basis of strength properties for designing sustainable filter media for water treatment plants. Performance at both construction and operating stages were considered by analyzing both strength properties under fully dry conditions and strength degradation after saturation in a water bath. The BM-75% as the main component produced optimum combination in terms of workability and strength. With the material combination of BM-75% and additives-25%, the use of Red Mud and water treatment sludge as additives produced the highest and lowest strength of composite clay balls, with a failure load of 5.4 kN and 1.4 kN respectively. However, this lower value of 1.4 kN is much higher than the effective load on each clay ball of 0.04 kN in a typical filter assembly (safety factor of 35), therefore, can still be used as a suitable filter material for enhanced pollutant removal.

  • Due to media's characteristics, the methods of acting expression in stage differ from that in media. However, actors playing in either genre use the same internalization as the basis of their performance. The acting arising from actors' internalization is the same in stage acting and media acting. Nevertheless, media acting, by its genre characteristics, requires a more delicate and natural acting than that in stage acting. Such is because media acting shows a far more detailed and amplified performance that is shown on screen. The purpose of this research is to examine how to differentiate the expression characteristics of media acting from the expression methods of stage acting. Through this research, I attempt to present the expression methods of media acting that can show real life acting reflecting media's distinct characteristics, with the aim of benefiting students who practice media acting or professional actors. Diverse expression methods presented by training of media acting are expected to enable actors engaged in this field to produce excellent outcomes through acting training to successfully bring out a realistic and convincing performance through internalization. And together with the opportunity to build delicate and detailed acting styles that are required in media acting, acting styles that are differentiated from those in stage acting will also be discussed.
