The dornor, 2 years old, 20kg and mixed breed, was bred naturally on day 1 and day 3 of estrus and eight gastrulae were collected by flushing the uterus of the donor after laparatomy on day 13 after the second mating. The embryos were frozen by programmable freezer and preserved for about 3 months in liquid nitrogen. Another bitch in natural estrus, 2 years old, 30kg, mixed breed, was selected as the recipient and the frozen embryos(8 gastrulae) were thawed and each 4 embryos were transferred into upper partr of left and right uterine horn, respectively, on day 13 after the proper mating day determined by vaginal smear. The ecipient delivered 6 offspring 48 days after embryo transfer. Of 6 puppies, one was still birth and two puppies died one month after birth. Parentage test was performed by DNA analysis using microsatellite sequences for 3 puppiers, the recipient, the donor, the male dog bred with the donor, and the male dog raised near to the recipient. The markers selected for the test were CXX 873(133-157 base pair) and CXX 894(141-165 base pair). Using primers manufactured according to the markers, the blood samples were processed for polymerase chain reaction and the PCR products were treated for electrophoresis. The three puppies showed identical band to that of recipient, consequently, it was concluded that the puppies were offspring of the recipient mated naturally by the male dog, not the offspring by embryo transfer.
To monitor the reproductive patterns of endangered common chimpanzee including annual reproductive cycle, amenorrhea, breeding season, and pregnancy diagnosis, Time-Resolved Fluorescence Immuno Assay (TR-FIA) was used to trace MRH (estradiol, progesterone, testosterone) and human chorionic gonadotropin (HCG). In result of this research, age was not the important factor in determining the reproduction capability in common chimpanzee; it was rather greatly influenced by the combination of various factors such as individual's fertilizing ability, presence of obstetrical diseases, the pattern of mating behavior, and the mental communication under the introduction of a mating partner. This research will play an important role in operating conservation project for common chimpanzees and can be extended also for shedding new light on understanding human menopause and obstetrical diseases.
The aim of the present study was to investigate the ovulation rate and its relationship to fertilization ability in Landrace, Durock and Crossbred pigs. Gilts were natural mated at a body weight of at least 120 kg under the same hormone treatment. Embryos were surgically collected 1 day after natural mating (Day 0). Embryos derived from in vivo-fertilized oocytes were cultured in medium PZM-3. The ovaries were examined and the pathological findings were recorded. The number of corpus hemorrhagicum was counted, and was assumed to equal the ovulation rate. There was no difference in the number of corpus hemorrhagicum (20.4, 28.8 and 23.2) and ovulation (13.5, 26.8 and 17.2) in the Landrace, Durock and Crossbred pigs. The two pronucleus formation was 76.0, 80.0 and 86.9%. The Day-7 embryos had blastocyst rates of 68.0, 75.0 and 73.9%. There was no difference in the number of total cells and apoptotic cells. In the future, more studies require determining relationships between ovulation and fertilization rate in different species of pigs.
The objective of this study was to examine the reproductive characteristics of cloned miniature piglets produced from surrogate domestic pigs. Somatic cell nuclear transfer (SCNT) miniature pig embryos were transferred into domestic pigs. As controls, domestic pigs of the same breed with surrogates for SCNT embryos and miniature pigs of the same breed with the somatic cell donor were bred by artificial insemination and natural mating, respectively. Surrogate domestic pigs that farrowed cloned miniature piglets had a significantly longer gestation length (118.1 days) than conventionally bred domestic (115.4 days) and miniature (115.5 days) pigs. Furthermore, the birth weight of cloned miniature piglets produced from domestic pigs (743 g) was significantly greater than that of miniature piglets produced by natural breeding (623 g). Also, cloned miniature piglets had a significantly lower weaning rate (49.7%) than conventionally produced domestic (91.5%) and miniature (100%) piglets. No differences were observed between female and male cloned piglets in gestation length, litter size, birth weight, or weaning rate. Our results demonstrate that gestation length is extended in domestic pigs that are transferred with SCNT miniature pig embryos and that cloned miniature piglets have increased birth weight and high pre-weaning mortality.
To clarify taxonomic status of the two sibling species, Moroco oxycephalus and M. lagowskii reproductive isolation mechanisms were investigated at sympatric area located in Kansung-up, Kosung-gun, Kangwon-do, Korea. Genetic analysis was performed to reveal mating system and intensity of Hybridization between the two species. The frequencies of hybrids were increased since 1989, and then the observed hybrid frequencies ($H_O$) did not significantly differ from the expected hybrid ($H_E$) in 1998 and 1999. However, based on histological analysis of two parents and their hybrid s gonads, the hybridizations between M. oxycephalus and M. lagowskii produced mostly fertile females but sterile males in accordance with Haldane s rule. Although it was suspected that pre- and postmating isolation mechanisms were affected between the two species, M. oxycephalus and M. lagowskii seemed to be strongly isolated with microhabitat at sympatry until 1997. Since 1998, hybrid frequencies were increased by habitat disturbance. However, their hybrid frequencies would be reduced by postmating isolation mechanisms. Therefore, the two species are considered to be distinct species recently diverged.
The Yeonsan Ogye (YO) chicken is a natural heritage of Korea, characterized by black feathers, skin, bones, eyes, and comb. The purebred of YO population has been reared under the natural mating system with no systematic selection and breeding plan. The purpose of this study was to identify the genetic diversity and find the optimal number of population sub-division using 12 polymorphic microsatellite (MS) markers to construct a pedigree-based breeding plan for the YO population. A total of 509 YO birds were used for this study. Genetic diversity and population structure analysis were conducted based on the MS marker genotype information. The overall average polymorphic information content value and expected heterozygosity of the population were 0.586, and 0.642, respectively. The K-mean cluster analysis based on the genetic distance result confirmed that the current YO population can be divided into three ancestry groups. Individuals in each group were evaluated based on their genetic distance to identify the potential candidates for a future breeding plan. This study concludes that a future breeding plan with known pedigree information of selected founder animals, which holds high genetic diversity, could be the best strategy to ensure the conservation of the Korean YO chicken population.
Changes of control efficacy of chemical to potato late blight caused by Phytophthora infestans in potato fields from 2001 to 2004 were examined. Control efficacy of metalaxyl was suddenly decreased from 100% in 2002 to 50% in 2004 and that of dimethomorph also was similar to those of metalaxyl. However, the control efficacy of ethaboxam no great change. Both A1 and A2 mating type isolates were isolated from 2001 to 2004 in several areas in Korea. The majority of the P. infestans isolates were A1 mating type. Total 939 isolates of P. infestans obtained from several areas in Korea from 2001 to 2004 were examined for changes of sensitivity to metalaxyl. Frequencies of metalaxyl resistance isolates were gradually increased from 17% in 2001 to 84.2% in 2004, but isolation frequencies of metalaxyl sensitive and intermediate resistant isolate were decreased. Cause of decreasing control efficacy of metalaxyl was thought by increase of resistance isolates in A1 mating type population according to increasing metalaxyl use. Most isolates were grown at 0.5 ${\mu}g/ml of dimethomorph and isolates grown at 1 ${\mu}g/ml of dimethomorph were approximately 10.2-22.9%. However, no isolate was able to grow at 5.0 ${\mu}g/ml. Based on these results, minimum inhibitory concentrations (MIC) of dimethomorph to P. infestans were determined to be 0.5-1.0 ${\mu}g/ml. Our results indicated that the reason decreasing control efficacy of dimethomorph was not caused by occurrence of resistant isolates. About 5% and 12.1% isolates among the total isolates collected in 2003 and 2004 were grown on V-8 juice rye agar containing 1.0 ${\mu}g/ml ethaboxam. The 2.1 and 25.4% isolates had MICs of 0.2-0.4 ${\mu}g/ml, and MIC values of 87.9% and 74.3% isolates were less than 0.2 ${\mu}g/ml concentrations of ethaboxam. Therefore, resistance development by P. infestans to ethaboxam is not likely to occur in the natural condition.
The development of efficient method for the production of transgenic mice has been investigated in our laboratory. This study was conducted to develop the artificial insemination in the mouse. Spermatozoa were collected from the cauda epididymis of ICR males(age:12~15 weeks, Body weight : 30g) and artificially inseminated into the intrauterine via cervix of hormone-primed ICR females(age: 6~8 weeks, body weight: 25g) using the capillary tube, 200~300 $\mu$m in inner diameter. The effect of concentration of sperm(80$\times$104, 40$\times$104, 20$\times$104, 10$\times$104, 5$\times$104, 3$\times$104, 1$\times$104/20${mu}ell$. The artificial insemination was succeeded but fertilization rate was very low(5~15%) compared to the natural mating and 59 normal youngs born from 60 females. Therefore, our findings suggest that it is possible to produce the great number of mice from the same orgin of male by artificial inseminatin. However, the lower pregnancy rate has to be solved to used broadly the artificial insemination in mouse.
The spatial distributions of allelic frequencies and ecological traits by randomization were studied in the natural population of two species (Pinus rigida and Pyrola japonica). Both species showed significant positive spatial autocorrelation as measured by Moran's I. In P. rigida, the genetic similarity was shown in individuals within up to a scale of 18 m distance and this is partly due to combination of pollen and seed dispersal by wind or men. In P. japonica, significant spatial autocorrelation was consisted of a scale of 8 m intervals. These population structure in the distribution of allelic frequencies is related to mating systems such as outcrossing and vegetative spread. The results also indicate that positive species associations between P. rigida and P. japonica can occur when both species select the same habitat or require the same environmental conditions.
Calcium ions play an important role in the establishment and maintenance of pregnancy, but molecular and cellular regulatory mechanisms of calcium ion action in the uterine endometrium are not fully understood in pigs. Previously, we have shown that calcium regulatory molecules, transient receptor potential vanilloid type 5 (TRPV6) and calbindin-D9k (S100G), are expressed in the uterine endometrium during the estrous cycle and pregnancy in a pregnancy status- and stage-specific manner, and that estrogen of conceptus origin increases endometrial TRPV6 expression. However, regulation of S100G expression in the uterine endometrium and conceptus expression of S100G has been not determined during early pregnancy. Thus, we investigated regulation of S100G expression by estrogen and interleukin-$1{\beta}$ (IL1B) in the uterine endometrium and conceptus expression of S100G during early pregnancy in pigs. We obtained uterine endometrial tissues from day (D) 12 of the estrous cycle and treated with combinations of steroid hormones, estradiol-$17{\beta}$ ($E_2$) and progesterone ($P_4$), and increasing doses of IL1B. Real-time RT-PCR analysis showed that $E_2$ and IL1B increased S100G mRNA levels in the uterine endometrium, and conceptuses expressed S100G mRNA during early pregnancy, as determined by RT-PCR analysis. To determine if endometrial expression of S100G mRNA during the implantation period was affected by the somatic cell nuclear transfer (SCNT) procedure, we compared S100G mRNA levels in the uterine endometrium from gilts with SCNT-derived conceptuses with those from gilts with conceptuses derived from natural mating on D12 of pregnancy. Real-time RT-PCR analysis showed that levels of S100G mRNA in the uterine endometrium from gilts carrying SCNT-derived conceptuses was significantly lower than those from gilts carrying conceptuses derived from natural mating. These results showed that S100G expression in the uterine endometrium was regulated by estrogen and IL1B of conceptus origin, and affected by the SCNT procedure during early pregnancy. These suggest that conceptus signals regulate S100G, an intracellular calcium transport protein, for the establishment of pregnancy in pigs.
