수질 오염물질 배출저감을 위한 완충식생 복원 모델 개발 (Development and Application of a Model for Restoring a Vegetation Belt to Buffer Pollutant Discharge)

  • In order to improve water quality in the Paldang Lake, a riparian vegetation belt, treatment wetland, and artificial floating island were designed for introduction in the upland field, the estuary of tributaries, and the section of water facing mountainous land, respectively. We synthesized vegetation information collected from a reference river and found that herbaceous, shrubby, and tree vegetation zones tended to be dominated by Phragmites japonica, Phalaris arundinacea, etc.; Salix gracilistyla, S. integra, etc.; and S. koreensis, S. subfragilis, and Morus alba, respectively. In our plan, the herbaceous vegetation zone, which is established on floodplains with a high frequency of disturbance, will be left in its natural state. A shrubby vegetation zone will be created by imitating the species composition of the reference river in the ecotone between floodplain and embankment. A tree vegetation zone will be created by imitating species composition on the embankment slope. In the treatment wetland, we plan to create emerged and softwood plant zones by imitating the species composition of the Zizania latifolia community, the Typha orientalis community, the P. communis community, the S. integra community, and the S. koreensis community. The floating island will be created by restoring Z. latifolia and T. orientalis for water purification purposes.

Inhibitory Effects of Flavonoids Isolated from Leaves of Petasites japonicus on $\beta$-Secretase (BACE1)

  • The deposition of the amyloid $\beta}$ ($A{\beta}$)-peptide following proteolytic processing of amyloid precursor protein (APP) by $\beta$-secretase (BACE1) and $\gamma$-secretase is critical feature in the progress of Alzheimer's disease (AD). Consequently, BACE1, a key enzyme in the production of $A{\beta}$, is a prime target for therapeutic intervention in AD. In the course of searching for BACE1 inhibitors from natural sources, the ethyl acetate fraction of Petasites japonicus showed potent inhibitory activity. Two BACE1 inhibitors quercetin (QC) and kaempferol 3-O-(6"-acetyl)-$\beta$-glucopyranoside (KAG) were isolated from P. japonicus by activity-guided purification. QC, in particular, non-competitively attenuated BACE1 activity with $IC_{50}$ value of $2.1{\times}10^{-6}\;M$ and $K_i$ value of $3.7{\times}10^{-6}\;M$. Both compounds exhibited less inhibition of $\alpha$-secreatase (TACE) and other serine proteases including chymotrypsin, trypsin, and elastase, suggesting that they ere relatively specific and selective inhibitors to BACE1. Furthermore, both compounds significantly reduced the extracellular $A{\beta}$ secretion in $APP_{695}$-transfected B103 cells.

  • 최근 친환경 재료와 구조물에 대한 관심이 증대되면서 친환경 재료인 황토에 대한 연구가 활발히 진행되고 있다. 황토는 건설 및 마감 재료로 사용된 전통적인 친환경 건설 재료 중의 하나이다. 황토는 높은 축열성, 습도 조절 능력, 자정력, 탈취 및 항균성능, 인체에 유익한 다량의 원적외선이 방사되는 점 등 건설 재료로서 많은 장점을 갖고 있다. 그러나, 황토는 낮은 강도특성과 건조수축 균열 특성 때문에 현대 건설에 사용되지 않고 있는 실정이다. 최근 연구자들의 연구결과에 따르면, 황토는 플라이 애쉬나 포졸란 같은 천연 포졸란 재료로 사용될 수 있다. 본 논문에서는 황토와 슬래그, 재생 PET 섬유가 혼입된 콘크리트의 역학적 성질과 구조적인 휨 거동 실험을 수행하였다. 실험 결과 황토콘크리트의 압축강도와 탄성계수는 기준 시멘트 콘크리트 시편보다 상대적으로 낮게 평가되었으며, 철근 콘크리트 보의 휨 거동은 거의 유사하게 나타났다.

[ $\beta$ ]-Secretase (BACE1) Inhibitors from Pomegranate (Punica granatum) Husk

  • In the course of screening for anti-dementia agents from natural products, two $\beta$-secretase (BACE1) inhibitors were isolated from the husk of pomegranate (Punica granatum) by activity-guided purification. They were identified as ellagic acid and punicalagin with $IC_{50}$ values of 3.9 $\times$$10^{-6}$ and 4.1$\times$$10^{-7}$ M and Ki values of 2.4$\times$$10^{-5}$and 5.9$\times$$10^{-7}$ M, respectively. The compounds were non-competitive inhibitors with a substrate in the Dixon plot. Ellagic acid and punicalagin were less inhibitory to $\alpha$-secretase (TACE) and other serine proteases such as chymotrypsin, trypsin, and elastase, thus indicating that they were relatively specific inhibitors of BACE1.

Herbivory effects and growth rate of invasive species, Pomacea canaliculata on different macrophytes species

  • Wetland ecosystems act as natural freshwater purification systems, but their rich biodiversity is being threatened with the introduction of the non-native freshwater snail, Pomacea canaliculata. This study was conducted to measure the herbivory effects and growth rate of P. canaliculata on common macrophytes: Ipomoea aquatica, Ipomoea batatas, Pandanus amaryllifolius and Cucurma longa. In separate experiments, the macrophyte species were served as the snails' food as individual species and simultaneously. In the individual treatment, the growth pattern and rate were based on the snails' weight (mg/snail/day; n = 9) while the individual feeding consumption (mg/snail/day) was calculated from the leftover food. In the simultaneous treatment, the herbivory effects were evaluated as the feeding preference (%) from observations every two hours, while the total feeding consumption (mg) was calculated based on the food remaining after a 12-hour experiment (3 replicates: total n = 27). The results indicated that the growth pattern was significant for snails grazing on I. aquatica but not when other macrophyte species were eaten. The individual feeding consumption was higher when using I. aquatica than P. amaryllifolius but the growth rate for snails grazing on I. aquatica and P. amaryllifolius did not differ significantly. Meanwhile, the consumption of C. longa deterred the snails' growth rate. Although the snails consumed all the macrophytes in the individual experiment, when given the species simultaneously, the feeding preference and total feeding consumption were directed significantly more toward I. aquatica than P. amaryfollius and C. longa. We conclude that P. canaliculata is a generalist feeder given a limited choice of food but tends to show a strong feeding preference after being introduced to more food choices. These findings indicate that the introduction of P. canaliculata into wetland ecosystems may increase the herbivory effects on macrophytes, making these ecosystems vulnerable to the impact of eutrophication and biodiversity reduction.

Polarity affects the antioxidant and antimicrobial activities of jellyfish (Acromitus hardenbergi) extracts

  • Jellyfish is an emerging aquaculture species, farmed for Oriental cuisines and nutraceutical ingredients. This study aimed to examine antioxidative and antimicrobial potentials of various fractions of the jellyfish, Acromitus hardenbergi. The bell and oral arms of the jellyfish were sequentially extracted with petroleum ether (PE), dichloromethane (DCM), chloroform (CHCl3), methanol (MeOH), and water (H2O) to extract its bioactive in an increasing polarity gradient. Test fractions were assayed for antiradical activities using electron spin resonance spectrometry, β-carotene-linoleate model and Folin-Ciocalteu assay; and antimicrobial activity against 2 Gram-negative bacteria, 4 Gram-positive bacteria and 2 fungal species using the disc diffusion assay. All fractions were also subjected to Fourier Transform Infrared (FTIR) analysis to identify types of functional groups present. It was found that the hydrophilic extracts (H2O fractions) possessed the most effective radical scavenging activity (p < 0.05) while the lipophilic extracts (PE fractions) the most active antimicrobial activity, especially against Gram-positive bacteria (p < 0.05). Total oxidation substrates content was found to be highest in the PE fractions of jellyfish bell and oral arms (p < 0.05). FTIR data showed that the H2O and MeOH fractions contains similar functional groups including -OH, -C=O, -N-H and -S=O groups, while the PE, DCM, and CHCl3 fractions, the -CH3, -COOH groups. This study showed that A. hardenbergi contains antioxidants and antimicrobials, thereby supporting the traditional claim of the jellyfish as an anti-aging and health-promoting functional food. Bioassay-guided fractionation approach serves as a critical milestone for the strategic screening, purification, and elucidation of therapeutically significant actives from jellyfish.

Identification and Characterization of a Bacteriocin from the Newly Isolated Bacillus subtilis HD15 with Inhibitory Effects against Bacillus cereus

  • Natural antimicrobial substances are needed as alternatives to synthetic antimicrobials to protect against foodborne pathogens. In this study, a bacteriocin-producing bacterium, Bacillus subtilis HD15, was isolated from doenjang, a traditional Korean fermented soybean paste. We sequenced the complete genome of B. subtilis HD15. This genome size was 4,173,431 bp with a G + C content of of 43.58%, 4,305 genes, and 4,222 protein-coding genes with predicted functions, including a subtilosin A gene cluster. The bacteriocin was purified by ammonium sulfate precipitation, Diethylaminoethanol-Sepharose chromatography, and Sephacryl gel filtration, with 12.4-fold purification and 26.2% yield, respectively. The purified protein had a molecular weight of 3.6 kDa. The N-terminal amino acid sequence showed the highest similarity to Bacillus subtilis 168 subtilosin A (78%) but only 68% similarity to B. tequilensis subtilosin proteins, indicating that the antimicrobial substance isolated from B. subtilis HD15 is a novel bacteriocin related to subtilosin A. The purified protein from B. subtilis HD15 exhibited high antimicrobial activity against Listeria monocytogenes and Bacillus cereus. It showed stable activity in the range 0-70℃ and pH 2-10 and was completely inhibited by protease, proteinase K, and pronase E treatment, suggesting that it is a proteinaceous substance. These findings support the potential industrial applications of the novel bacteriocin purified from B. subtilis HD15.

Quantitative Analysis and Enantiomeric Separation of Ephedra Alkaloids in Ma Huang Related Products by HPLC-DAD and UPLC-MS/MS

  • Ephedra is a genus of the Ephedraceae family and is found in temperate regions, such as Central Asia and Europe. Among the various ephedra species, Ma Huang (Ephedra herb) is derived from the aerial parts of Ephedra sinica S tapf, Ephedra equisetina Bunge, and Ephedra intermedia Schrenk & C.A. Mey. Ma Huang contains various ephedra alkaloids, including (-)-ephedrine, (+)-pseudoephedrine, (-)-norephedrine, (+)-norpseudoephedrine, (-)-methylephedrine, and (+)-methylpseudoephedrine, which are found naturally as single enantiomers, although they can be prepared as racemates. Although the use of Ma Huang in foods is prohibited in Korea, products containing Ma Huang can be imported, and so it is necessary to develop a suitable analytical technique for the detection of Ma Huang in foods. Herein, we report the development of analytical methods for the detection of ephedra alkaloids in products containing Ma Huang. Following sample purification by solid phase extraction, quantitative analysis was performed using ultra-performance liquid chromatography-triple quadrupole mass spectrometry (UPLC-MS/MS). Additionally, the enantiomers were successfully separated using HPLC-DAD. We successfully analyzed various food samples, where the ephedra alkaloids were qualitatively and quantitatively determined, and the enantiomers were separated. It is expected that these methods may contribute toward preventing the distribution of illegal products containing Ma Huang.

잘피 게바다말로부터 폴리페놀 물질(MP-1)의 분리 및 동정 (Identification of Polyphenol Substances (MP-1) from Seagrass, Phyllospadix Japonica Makino)

  • BACKGROUND: Phyllospadix japonica Makino is a perennial plant belonging to the family Zosteraceae. This species is native to the eastern coast of the Korean Peninsula, and it is found attached to rocks on the seashore. As with all seagrass species, P. japonica is also known to play a major role in protecting the marine environment, and studies on its physiological activities suggestive of its antioxidant, antibacterial and anti-inflammatory potentials have been reported. In this study, purification and structural analysis were performed to identify the polyphenol substances derived from P. japonica. METHODS AND RESULTS: An polyphenol substance MP-1 was purified from the 70% aqueous methanol extract of P. japonica using Diaion® HP-20 column chromatography, ODS column chromatography, and medium-pressure liquid chromatography (MPLC). The purified MP-1 was identified as rosmarinic acid having a molecular weight of 360 and a molecular formula of C18H16O8 through electrospray ionization (ESI)-mass and nuclear magnetic resonance (NMR) spectroscopic analysis. CONCLUSION(S): This study highlights the processes used for the identification of the polyphenol substance derived from P. japonica. Rosmarinic acid, the polyphenol derived from P. japonica identified by this study, is a kind of bioactive substance mainly present in plants. These findings provide an important starting point and are valuable for future studies on bioactive substances in seagrass.
