The Change of Physiological Characteristics as Water Purification Capability by Native Aquatic Plants (자생수생식물의 수질 정화에 따른 생리활성 변화)

  • This study looked into the water quality improvement effect of reed (Phragmites communis), bur reed (Sparaganium stoloniferum), Alisma canaliculatum, and Hydrocharis dubia in order to develop native hydrophytes suitable to natural-type wetlands for ecological restoration. The result confirms that the concentration of wastewater was lowered in most plants indicating a possibility in wastewater treatment. Among four species, reed (Phragmites communis) and bur reed (Sparaganium stoloniferum) were most effective in removing a majority of contaminants. Considering that reed (Phragmites communis) is a species that has been used most often among hydrophytes, bur reed (Sparaganium stoloniferum), which is a native species in Korea, showed equivalent level of outstanding effect. In the physiological and growing activity and its relations to water quality improvement effect, each species appeared differently. Reed (Phragmites communis) and bur reed (Sparaganium stoloniferum) were best also in physiological and growing activity. In the case of Hydrocharis dubia, growth was strong in polluted water environment but its physiological activity was poor, indicating that it was under stress. The growth and physiological activity of Alisma canaliculatum were also poor, indicating that it too is being stressed. However, measuring organic contents in the plants of each species shows that the organic contents of Alisma canaliculatum was high. Alisma canaliculatum is a species effective in removing pollutants in water, although it is under stress. A unique phenomenon in removing water pollution with plants is that each species removed different pollutants. In the case of Alisma canaliculatum, which was relatively poor in removing pollution, it was remarkable in reducing nitrogen and phosphorus. Hydrocharis dubia was also effective in eliminating floating matter.

Extraction of Genistein and Formononetin from Sophoraflavescens Aiton using Ultrasonic wave (초음파를 이용한 고삼에 포함된 Genistein 및 Formononetin의 추출)

  • In this work, we the ettect on extraction amounts and general composition content of phytoestrogen genistein and formononetin extracted from Sophoraflavescens Aiton by various ultrasonic waves(35, 72, and 170 KHz) and extraction time(30, and 60 min) were compared using extraction solvent water 100%. The pretreatment step was composed of ultrasonic waves extraction, filtration, concentration, and membrane filtration. The extracted sample was analyzed by reversed-phase high performance liquid chromatography(RP-HPLC). And the mobile phase applied was linearly changed with A/B of 80/20~65/35 vol% for 60 min(A water/acetic acid, 99.9/0.1 vol%, B acetonitrile/acetic acid, 99.9/0.1 vol%). The experimental results, general composition carbohydrate(0.255 to 0.413%) excepts, other ingredients was confirmed almost similarly. Also, The highest yield of extraction amount 3.17g was obtained by ultrasonic waves with a frequency of 170 KHz and an extraction time of 60 min. This work offers would be useful for chemical and biological studies of natural plants and its products.

Purification of Anti PC-3 Prostate Cancer Agents from Gleditsiae Spina (조각자(皂角刺)에서 PC-3 생장 억제 성분 정제)

  • Objectives : Gleditsiae Spina has the effects of expelling toxins, draining pus, invigorating blood and resolving abscesses. Some clinicians apply the herb for patients suffering from cancer. However, its anti-cancer activities are not well understood. In the present study, anti-tumor agents from Gleditsiae Spina were purified. Methods : The viability of the PC-3 cell line was determined using MTT assay, and the induction of apoptosis by Gleditsiae Spina extract in PC-3 cells was measured by Annexin-V/propidium iodide double staining assay detected by flow cytometry. TLC and HPLC analysis were used to separate and identify the anti-cancer agents. Results : Treatment of the extract resulted in significant decreased cell viability of PC-3 cells in a dose- and time-dependent manner. Dose-dependent apoptotic cell death was also measured by flow cytometry analysis. The anti-cancer agents were successfully separated and identified by using TLC and HPLC analysis and the most potential agent among them was separated from EtOAC fraction. Conclusions : These results might be applied in developing new drugs from natural resources like Korean traditional medicine, and also support the clinical usefulness of herbal medicine.

Determining the priority order of wetland functions (내륙습지 기능의 우선순위 결정)

  • In this study, the functional priority of wetlands was determined through analysis of previous research. To determine relative importance, three processes were performed. First, quantitative values from the case studies were normalized. Second, non-quantitative values were prioritized based on standard criteria. Third, equal weight was applied as long as there was no special consideration regarding a particular value's disproportionate priority in the research. Finally, results were grouped into large, medium, and small classes.In this study, the functions of the medium class were found to be the most significant, in the following order of priority: water supply and ground water recharge; culture and recreation; biodiversity; product; water quality control; flood control; erosion control; moderation of climate change; and provision of biochemical matter. To verify these results, we compared our findings with those of an assessment that used the Rapid Assessment Method (RAM) on the same type of study area. Whereas this comparison indicated some correlations by the culture, water storage, and genetic sustainability functions, it suggested a lack of such relationship by the water purification and flood prevention functions.

  • In this study, we found that it is necessary to use the relatively new resource from seaweed extracts to extract and process pulp and create a local brand that can contribute to the local fishermen, development of new bio material, establishment of natural infrastructure, and acquisition of foreign investment. This study's seaweed pulp separation process is very overwhelming as seaweed is a compound of glycoside and polysaccharide. Nevertheless, we intend to develop a purification process and introduce applied technology to explore a new applied technology of pulp process. Once this technology is fully developed and mass produced, it would contribute to greater exports and increasing income level for the local fishermen. The ultimate goal of this study is to gather technical data from the first and second years of application, apply seaweed pulp to increase bio effect, and develop new functional bio-plastic packaging material, raw material, and samples with special characteristics of high molecules.

Improved Baculovirus Vectors Expressing Barnase Using Promoters from Cotesia plutellae Bracovirus

  • The goal of this study was to create a novel baculovirus expression system that does not require recombinant virus purification steps. Transfection of insect cells with transfer vectors containing barnase under control of the Cotesia plutellae bracovirus (CpBV) promoters ORF3004 or ORF3005 reduced cell growth. Co-transfection with bApGOZA DNA yielded no recombinant viruses and nonrecombinant backgrounds. To further investigate the detrimental effects of barnase on insect cells, two recombinant bacmids harboring the barnase gene under control of the CpBV promoters, namely bAcFast-3004ProBarnase and bAcFast-3005ProBarnase, were constructed. While no viral replication was observed when only the recombinant bacmids were transfected, recombinant viruses were generated when the bacmids were co-transfected with the transfer vector, pAcUWPolh, through substitution of the barnase gene with the native polyhedrin gene by homologous recombination. Moreover, no non-recombinant backgrounds were detected from unpurified recombinant stocks using PCR analysis. These results indicate that CpBV promoters can be used to improve baculovirus expression vectors by means of lethal gene expression under the control of these promoters.

Purification and Characterization of Biopolymer by Bacillus coagulans CE-74 (Bacillus coagulans CE-74가 생산하는 Biopolymer의 분리 및 정제)

  • Screening was performed to isolate biopolymer-producing microorganisms from natural sources. The bacteriological characteristics of this strain and physicochemical properties of the biopolymer produced were investigated. The bacterial strain was identified as a Bacillus coagulans. Crude biopolymer treated with ethanol and actone was purified to fraction I and II by ion exchange chromatography and gel chromatography (Shephadex G-100 and Shepharose CL-2B). Analysis of chemical composition and various color reaction revealed that the polymer is composed of amino acids. It was confirmed that fraction II is a homopolymer of glutamic acid and fraction I is a homopolymer of lysine by analysis data of amino acid analyzer, GC and IR.

Analysis of water purification in the FWS wetland for Agreculture Area (농업지역 내 FWS 인공습지의 수질정화효율 분석)

  • Annually, the scale of agricultural areas in Korea were being reduced as the lands were converted to other land uses. While the rate of productivity were either being maintained or increased, the pollution load from these areas were still greater in magnitude. Although the levels of pollutant concentration released in the agricultural watersheds were minimal, the combined quantities mostly from diffuse sources were high. As a consequence, the Ministry of Environment (MOE) in Korea adopted the use of free water surface (FWS) flow constructed wetlands to reduce the pollutant loadings emitted from agricultural watersheds for the improvement of water quality and protection of aquatic ecosystems. In this study, a constructed wetland treating stream water in an agricultural watershed was monitored since April 2009 subsequent to its completion in December 2008. Satisfactory performance was achieved for TSS, BOD and TP with 26%, 28% and 39% pollutant removal rates, respectively. In addition, the effluent water quality was improved and achieved compliance the national water quality criteria. Results of this study can be useful to establish design parameters and employ proper removal techniques of similar natural treatment systems for future implementation in the country.

Perspectives for the Industrial Use of Bacteriocin in Dairy and Meat Industry (축산업 분야에서의 박테리오신의 산업적 이용 및 향후 전망)

  • More safe and natural food was recently needed by consumers. Antimicrobials including sodium azide, penicillin, and vancomycin were used for therapeutic agents against pathogens such as Listeria monocytogenes, Staphylococcus aureus, Escherichia coli O157:H7 in dairy and meat industry. These antimicrobials and preservatives were prohibited in stock farm and food because they were caused resistant strain and side effects. Bacteriocins are proteinaceous compounds that may present antimicrobial activity towards important food-borne pathogens and spoilage-related microflora. Therefore, bacteriocins were reported as an alternative of antimicrobials. Due to these properties, bacteriocin-producing strains or purified bacteriocins have a great potential of use in biologically based food preservation systems. Despite the growing number of articles regarding on the isolation of bacteriocinogenic strains, genetic determinants for production, purification and biochemical characterization of these inhibitory substances, there are only limited reports of successful application of bacteriocins to dairy and meats. This review describes bacteriocins related to dairy and meat products for the further use.

Strengths and Permeability Properties of Porous Polymer Concrete for Pavement with Different Fillers (충전재 종류에 따른 포장용 포러스 폴리머 콘크리트의 강도 및 투수 특성)

  • Recently, concrete has been made porous and used for sound absorption, water permeation, vegetation and water purification according to void characteristics. Many studies are carried out on the utilization of sewage sludge, fly ash and waste concrete to reduce the environmental load. This study was performed to evaluate the void, strength, relationship between void and strength, permeability and chemical resistance properties of porous polymer concrete for pavement with different fillers. An unsaturated polyester resin was used as a binder, crushed stone and natural sand were used as an aggregate and bottom ash, fly ash and blast furnace slag were used as fillers. The mix proportions were determined to satisfy the requirement for the permeability coefficient, $1{\times}10^{-2}$ cm/s for general permeable cement concrete pavement in Korea. The void ratios of porous polymer concrete with fillers were in the range of $18{\sim}23%$. The compressive strength and flexural load of porous polymer concrete with fillers were in the range of $19{\sim}22$ MPa and $18{\sim}24$ KN, respectively. The permeability coefficients of porous polymer concrete with fillers were in the range of $5.5{\times}10^{-1}{\sim}9.7{\times}10^{-2}$ cm/s. At the sulfuric acid resistance, the weight reduction ratios of porous polymer concrete immersed during 8-week in 5% $H_{2}SO_{4}$ were in the range of $1.08{\sim}3.56%$.
