  • Solid-liquid phase change (i.e. melting or solidification) occurs in a number of situations of practical interest. Some common examples include the melting of edible oil, metallurgical process such as casting and welding, and materials science applications such as crystal growth. Therefore, due to the practical importance of the subject, there have been a large number of experimental and numerical studies of problems involving phase change during the past few decades. Also, this study presented the effective way to enhance phase change heat transfer.

  • Heat transfer phenomena during inward melting and solidification process of the phase change material were studied expertimentally. The phase change medium was 99% pure n-docosane paraffin ($C_{22}H_{46}$). The solid-liquid interface motion during phase change was recorded photographically. Measurements were made on the temperature, the solid-liquid interface, the melted or frozen mass and the various energy components stored or extracted from the cylinder wall. For melting, the experimental results reaffirmed the dominant role played by the conduction at an early stage, by the natural convection at longer time. For solidification, natural convection effects in the superheated liquid were modest and were confined to short freezing time. Although the latent energy is the largest contributor to the total stored or extracted energy, the aggregate sensible energies can make a significant contribution, especially at large cylinder wall superheating or subcooling, large initial phase change material subcooling or superheating.

박슬라브 주형에 따른 용강내의 상변화현상에 대한 수치적 해석 (Numerical Prediction of Phase Change within the Molten Steel with Thin Slab Casting)

  • A numerical analysis has been performed on the two-dimensional rectangular gallium melting problem using the enthalpy method. The major advantage of this method is that the physical domain is discretized with fixed grids without transforming variables and the interface conditions of phase change are accounted for the definition of suitable source terms in the governing equations. But in the fixed method, there is some ambiguity in defining the porosity constant which has no physical interpretation. If the velocity correction is included in the momentum equation, for the appropriate range of porosity constant, the realistic predictions are obtained. The object of the present work is to predict the phase change within the molten steel with thin riser slab using the modified enthalpy-porosity method. The computational procedures for predicting velocity and temperature are based on the finite volume method and the non-staggered grid system. The influence of natural convection on the melting process is considered. A comparison with the experimental results shows that the modified method is better than the previous one.

  • Natural convection heat transfer from a horizontal ice cylinder immersed in quiescent cold pure water was studied experimentally. The experiment was conducted for the ambient water temperatures ranging from $2.0^{\cric}C$ to $10.0^{\circ}C$. The flow fields around an ice cylinder and its melting shapes were visualized and local Nusselt numbers obtained. Especially, its attention was focused on the density maximum effects and stagnation point Nusselt number. From the visualized photographs of flow fields, three distinct flow patterns were observed with the ambient water temperature variation. The melting shapes of ice cylinder are various in shape with flow patterns. Steady state upflow was occured at the range of $2.0^{\circ}C \leq T_{\infty} \leq 4.6^{\circ}C$ and steady state downflow was occured at $T_{\infty} \geq 6.0^{\circ}C$. In the range of $4.7^{\circ}C < T_{\infty} < 6.0^{\circ}C$, three-dimensional unsteady state flow was observed. Especially, the melting shapes of ice cylinder have formed the several spiral flutes for the temperatures ranging from $5.5^{\circ}C$ to $5.8^{\circ}C$. For upflow regime, the maximum stagnation point Nusselt number exists at $T_{\infty} = 2.5^{\circ}C$ and as the ambient water temperature increases the Nusselt number decreases. At ambient water temperature of about $5.7^{\circ}C$, Nusselt number shows its minimum value.

Fixed-Grid Simulation of Convection-Dominated Melting in a Rectangular Cavity

  • Numerical solutions for the convection-dominated melting in a rectangular cavity are presented. The enthalpy-porosity model is employed as the mathematical model. This model is applied in conjunction with the EIT method to detect boundary movement in a phase changing environment. The absorption and evolution of latent heat during the phase change is dealt with by the enthalpy-based energy equation. This seems to be more efficient than resolving the temperature-based energy equation. The velocity switch-off, which is required when solid changes into liquid, is modeled by the porous medium assumption. For efficiency and simplicity of the solutions procedure, this paper proposes a simple algorithm, which iterates the temperature and the liquid fraction of the cells comprising the front layer. The numerical results agree reasonably well with the experimental data and other previous works using the transformed-grid system.

  • This study presents experimental work on phase change heat transfer, in order to increase heat transfer rate, ultrasonic vibrations were introduced. Solid-liquid phase change occurs in a number of situations of practical interest. This study reveal that ultrasonic vibrations accompany the effects like agitation, acoustic streaming, cavitation, and oscillating fluid motion. Such effects are a prime mechanism in the overall melting process when ultrasonic vibrations are applied. Some common examples include the melting of edible oil, metallurgical process such as casting and welding, and materials science applications such as crystal growth. Therefore, this study presented the effective way to enhance phase change heat transfer.

Complex Detection Between Transcription Regulator and Promoter DNA by UV Spectroscopic Method

  • UV spectrophotometer was used to detect protein-DNA complex from DNA melting profile under constant temperature increase. Melting temperature (Tm) was $43^{\circ}C$ in copA duplex DNA alone. In the presence of Proteus mirabilis transcription regulator protein (PMTR) protein at 0.2 and 0.4 ${\mu}M$, Tm's were $45{\pm}0.5$ and $47.6{\pm}0.6^{\circ}C$, respectively. According to fluorescence polarization and gel shift assay. PMTR:copA complex was detected by the retarded migration on gel and the dissociation constant ($K_d$) was $(9.2{\pm}2.8){\times}10^{-9}M$.

  • Urea-SCR system, which converts nitrogen oxides to nitrogen and water in the presence of a reducing agent, usually AdBlue urea solution, is known as one of the powerful NOx reduction systems for mobile as well as stationary applications. For its consistent and reliable operation in mobile applications, such various problems as transient injection, ammonia slip, and freezing in cold weather have to be resolved. In this work, therefore, numerical study on three-dimensional unsteady heating problems were analyzed to understand the melting and heat transfer characteristics such as urea liquid volume fraction, temperature profiles and generated natural convection behavior in urea solution by using the commercial software Fluent 6.3. After validating by comparing numerical and experimental data with pure gallium melting phenomena, numerical experiment for urea melting is conducted with three different coolant heating models named CH1, 2, and 3, respectively. Finally, it can be found that the CH3 model, in which more coolant is concentrated on the lower part of the urea tank, has relatively better melting capability than others in terms of urea quantity of $1{\ell}$ for start-up schedule.

  • The collapse of ice shelves is a process that can severely increase the rise of global sea-levels through the reduction of the buttressing effect of ice shelves and the consequent acceleration of the ice flow of ice sheets. In recent years, the West Antarctic ice shelves in the Amundsen Sea, whose buttressing effect is essential for a great part of the West Antarctic ice sheet, have been experiencing the most rapid melting and thinning in the world. The melting of the West Antarctic ice shelves is caused primarily by heat transported by Circumpolar Deep Water (CDW). For this reason, it is important to investigate ice-ocean interactions that could influence the melting of ice shelves and evaluate the stability of West Antarctic ice shelves. A lot of researchers have been actively investigating the West Antarctic ice shelves in the Amundsen Sea. High-impact journals have recognized the importance of and published studies on ice-ocean interactions occurring near and under the ice shelves as well as the connections among ice shelves. However, in situ observations are limited due to extreme weather and sea-ice conditions near the ice shelves; therefore, many scientific questions remain unanswered. This study introduces the characteristics of the Amundsen Sea and investigate the past and latest research issues in this region. This study also gives suggestions regarding important scientific questions and directions for future research that should help early-career scientists take the lead in future research on the melting dynamics of the West Antarctic ice shelves in the Amundsen Sea.

Synthesis and base pairing properties of DNA-RNA heteroduplex containing 5-hydroxyuridine

  • 5-Hydroxyuridine (5-OHU) is a major lesion of uridine and cytosine produced in RNA by various chemical oxidants. To elucidate its biochemical and biophysical effects on RNA replication, the site-specifically modified oligoribonucleotides containing 5-OHU were synthesized with C5-hydroxy-5'-ODMTr-2'-TBDMS-uridine phosphoramidite using automated solid phase synthesis. The base-pairing properties of nucleotides opposite 5-OHU in 24 mer oligoribonulcleotides with dNTP were studied using three reverse transcriptases (Super-$Script^{TM}II$-, AMV-, MMLV-RT) in cDNA synthesis. Adenine as well as guanine was incorporated preferentially by all reverse transcriptases. In the UV-melting temperature experiment, the results from the relative stabilities of the base pairs were A : 5-OHU > G : 5-OHU > T : 5-OHU $\approx$ C : 5-OHU. Circular Dichroism (CD) studies showed that DNA-RNA containing 5- OHU heteroduplexes exhibit a similar conformation between the A-type RNA and B-type DNA. These results suggest that 5- OHU from oxidative damage was mainly influenced by adenine mismatch.
