A Study on the Physiological Effects and Dyeing Properties of the Extract of Fermented (Part I) (발효쪽 추출물의 생리적 기능과 염색특성(제1보))

  • The purpose of this study was to investigate the antimicrobial activity, antimutagenic and anticancer effects and dyeing properties of the fermented indigo extract. The physiological effects of natural color extracts from colorant plants(gardenia, beet and indigo) were studied. The methanol extract of indigo showed an inhibitory effect on the growth of E. coli and Staph. aureus, and also showed a strong antimicrobial effect on Trich. mentagrophytes compared to others. The methanol extract of indigo showed antimutagenic activities against aflatoxin B1(AFB1) in the Ames test using Salmonella typhimurium TA 100. The proliferation of Clone M-3 mouse melanoma cells and A431 human epidermoid carcinoma cells was inhibited by the methanol extract of indigo. So we decided to use natural indigo for dyeing the fabrics because of those effects. Dried indigo leaves were fermented at variouss temperature and the fermented indigo was reduced by using alkaline(NaOH, Ca(OH)2) and glucose to dye the fabrics. The values of K/S fermented indigo showed the highest value when it was fermented at 3$0^{\circ}C$. The indigo fermented at 3$0^{\circ}C$ had the greatest number of total bacterial counts and we identified one of the main microorganisms as Aspergillus niger. This microorganism was responsible for the indigo fermentation and accelerated indigo fermentation. So it can be supposed to reduce the fermentation period of indigo by inoculating Aspergillus niger into the indigo leaves at 3$0^{\circ}C$.

Antimicrobial activities of Monascus koji extracts (식품유해균에 대한 홍국 추출물의 항균활성)

  • Currently, natural food colorants and preservatives are being used for their general health benefits. Monascus koji, the product of certain fungi that grow on rice grains, has been added to many foods for coloring and preservation. In this study, the antimicrobial activities of Monascus koji ethanol extracts were investigated. Six Monascus strains (M. araneosus KFRI 00371, M. kaoliang ATCC 46597, M. pilosus IFO 4520, M. purpureus IFO 4482, M. ruber IFO 32318 and M. sp. ATCC 16437) were selected based on their relative intensity of red pigment. Two Monascus extracts, M. kaoliang ATCC 46597 and M. purpureus IFO 4482, displayed antimicrobial activities against Bacillus subtilis, B. cereus, Micrococcus luteus, Staphylococcus aureus and Salmonella typhimurium in concentration-dependent manners. The two extracts showed their strongest antimicrobial activity against S. typhimurium, a cause of food poisoning. Therefore, these results suggest that Monascus koji could be used as a natural food colorant and preservative.

Dyeing Properties and Colour Fastness of Cotton and Silk Fabrics Dyed with Cassia tora L. Extract

  • A natural colorant was extracted from Cassia tara L. using buffer solutions (pH: 2-11) as extractants. The dyeing solution (Cassia tara L. extract) extracted using pH 9 buffer solution was found to give the highest K/S values of dyed fabrics. Cotton and silk fabrics were dyed with Cassia tara L. extract at $60^{\circ}C$ for 60 min with pre-treatment of various metal salts as mordants. It was found that Cassia tara L. extract was polygenetic dyestuffs and its major components were anthraquinones. Studies have been made on the effects of the kind of mordant on dyeing properties and colour fastnesses of cotton and silk fabrics. The K/S of cotton fabrics increased in the order of the dyeing using $FeSO_4 >CuSO_4> ZnSO_4> MnSO_4\cong Al_2(SO_4)_3 > NiSo_4 > none$, however, the K/S of silk fabrics increased in the order of the dyeing using $FeSO_4 > CuSO_4 > ZnSO_4\cong Al_2(SO_4)_3 > MnSO_4\cong NiSO_4 > none$. It was found that the K/S values of dyed fabrics were largely affected by the colour difference $(\DeltaE)$ between mordanted fabric and control fabric. However, they were not depended on the content of mordanted metal ion of the fabrics. Mordants $FeSO_4$ and CuSO_4$ for cotton fabric, $FeSO_4,\; CuSO_4,\; and\; Al_2(S0_4)_3$ for silk fabric were found to give good light fastness (rating 4).

Natural Dyeing of Cationic-modified New Rayon (cocell) Fabric - Gallnut- (양이온화 뉴레이온(코셀) 직물의 천연염색에 관한 연구 - 오배자를 중심으로 -)

  • This study investigated the substantivity of anionic dyes for cationic-modified new rayon (cocell) fabric treated with cationic agent (CA), 3-(Chloro-2-hydroxypropyl)-trimethylammonium chloride (CA). We also investigate the dyeability of cationic-modified new rayon fabric after dyeing with gallut. CA was converted in an aqueous solution of sodium hydroxide into epoxypropyl trimethylammonium chloride. Treating with this epoxy reagent modified the hydroxyl groups of the new rayon fabric into the trimethylammonium group through ether linkage. The introduction of new cationic sites into new rayon fabric by pretreating with cationic agent improved the substantivity of the Gallnut dye with the new rayon dyebath. The degree of the cationization of cationic-modified new rayon and cotton fabric was evaluated by nitrogen (N) content. This study extracted the colorant of gallnut with hot water at $90^{\circ}C$ and 120 min. Cationic-modified new rayon fabric dyed with extracted solution from gallnut according to concentration of gallnut, dyeing temperature, dyeing time and concentration of cationic agent. Dyeability (K/S) was obtained by CCM observation after dyeing with gallut solution. In addition, fastness to washing and light were also investigated. The degree of crystallinity of new rayon and cotton fabric were 42.15% and 54.94%, respectively. N (%) content of cationic-modified new rayon was higher than the cationic-modified cotton. Dyeability (K/S) increased significantly with the increasing concentration of CA and gallut.

Antioxidant activity of hot-water extracts and floral waters from natural plant pigments (천연색소 함유 식물추출물과 플로럴 워터의 항산화 활성)

  • The antioxidant potentials of ten kinds of medicinal plants used as natural dyeing agents were tested. Among the plant extracts and floral waters that were examined in this study, the antioxidant activities of the DPPH and ABTS radical scavengers increased with increasing amounts of the extracts. The hot-water extracts from Aphis chinensis, Caesalpinia sappan L., Rumex crispus L., and Areca catechu effectively inhibited the DPPH and ABTS radicals at concentration below 0.1 mg/mL. The floral water obtained through steam distillation from Areca catechu, Rubia akane Nakai, Coptis chinensis, and Phellodendron amurense showed relatively valid antioxidant activities. In particular, the natural colorants extracted from Rumex crispus L., Areca catechu, and Aphis chinensis effectively suppressed the photogenerated singlet oxygen induced by a photosensitizer in in-vitro assay systems. The concentrations ($IC_{50}$) of the hot-water Aphis chinensis extract required to exert a 50% reduction effect on DPPH, ABTS, and singlet oxygen were found to be 8.5, 8.0, and $210{\mu}g/mL$, respectively. The study results suggest that extracts derived from naturally occurring colorants as well as floral waters could be used as natural antioxidants in the food and health-care industries.

Changes in chemical properties, antioxidant activities, and cytotoxicity of turmeric pigments by thermal process (가열처리에 의한 심황색소의 화학적 특성, 산화방지 활성 및 세포독성 변화)

  • Turmeric oleoresin, extracted from the rhizome of Curcuma longa L., is a widely-used natural food colorant. Curcuminoids, the major pigments in turmeric, which include curcumin, demethoxycurcumin (DMC), and bisdemethoxycurcumin (BMC), possess various physiological activities. In the present study, changes in the chemical properties, antioxidant activities, and cytotoxicity of turmeric pigments upon heating were investigated. Color intensity of turmeric was significantly reduced after heating. Residual levels of curcumin, DMC, and BMC after 15 min of heating at $95^{\circ}C$ were 11.9, 37.4, and 77.3% respectively. Scavenging activities of turmeric against 2,2'-azobis-3-ethyl-benz-thiazoline-6-sulfonic acid (ABTS), 2,2-azobis (2-amidinopropane) hydrochloride (AAPH) peroxyl radicals, and nitrite were significantly enhanced after heating, while 2,2-diphenyl-1-picryl-hydrazyl (DPPH) radical scavenging activity remained unaffected. Generation of $H_2O_2$ from turmeric was increased via thermal decomposition. Cytotoxicity of turmeric pigments against colon cancer and normal intestinal cells was reduced significantly after heating. The results indicate that thermal processing affects chemical properties and bioactivities of turmeric pigments. These effects should be considered during the processing of foods containing turmeric pigments.

Natural Dyeing of Rayon Fabric using Loess (황토를 이용한 인견직물의 천연염색)

  • This study dyed rayon fabric using loess as a natural colorant. To obtain the optimal dyeing conditions, various dyeing conditions were examined (temperature, pH, time, and concentration). The color fastness was evaluated using standard washing and rubbing fastness tests. The results were as follows: The loess powder particle size ranged from 0.4 to $1.7{\mu}m$ with a distribution range of 1.1 to $1.4{\mu}m$, representing a fine and uniform manufactured loess powder. The loess component analysis showed a large amount of silicon dioxide and aluminum oxide. TheFT-IR spectra showed that the ammonium group in the rayon fabric produced N-H banding at $1,540cm^{-1}$. The highest K/S value for the rayon fabric was obtained when the pH was 8.0, and this value increased rapidly with a longer dyeing time and when increasing the loess concentration to 30% (w/v). Pre-treatment with a soybean solution produced the highest K/S value for the rayon fabric with a loess concentration of 30% (w/v). The SEM analysis showed a higher amount of loess adhered to the rayon fabric surface when increasing the loess concentration. However, pre-treatment with a cationic agent and soybean solution resulted in a much higher attachment of loess to the fabric surface. Thus, the experimental results showed that using a cationized fabric and pre-treatment with a soybean solution are more effective when dyeing rayon fabric with loess than when using only loess.

Carotenoid Contents of Yellow Sweet Potatoes (황색고구마의 Carotenoid 색소 함량)

  • Twelve varieties of yellow sweet potatoes were measured for carotenoids content to assess their potential as a source of natural food colorant. Benihayato variety had the highest content of carotenoids (20.2 mg/100 g fr wt) but other varieties ranged with $4.6{\sim}16.7\;mg/100\;g$ fr wt. Hunter's +a-values (redness) increased linearly $(R^{2}=0.826)$ with carotenoids content of yellow sweet potatoes. Absorption spectrum of carotenoids extracted from the Benihayato variety was similar to that of standard ${\beta}-carotene$ indicating that carotenoids in yellow sweet potato are mostly composed of ${\beta}-carotene$.

Effect of Chitosan and Mordant Treatments on the Color Change of Cotton and Nylon Fabrics Dyed using Rhusjara ica (면과 나일론 직물의 오배자 염색 시 Chitosan 처리와 매염이 색상에 미치는 영향)

  • The effect of high purity chitosan was studied on its application to the natural dyeing using Rhusjara ica. In the dyeing experiment, examinations were introduced on the difference between the chitosan treated fabrics and chitosan untreated fabrics, and on the difference according to the type of mordanting agents and the mordanting methods. Dyeing was carried out for the chitosan treated and untreated fabrics under the conditions of non-mordanting and Al, Sn, Fe mordanting. As a result, we found that, in the case of cotton, chitosan untreated fabrics were not dyed enough, and the dyeing effect was not developed even though the mordanting was introduced. However, the chitosan treated fabrics developed excellent dyeing result even in the non-mordanting case. Through the results of cotton fabric dyeing, it was presumed that the dyeing affinity of the chitosan toward the Rhusjara ica, which prefer specific dyestuff (mainly black color), is selectively high. In the case of nylon, the dyeing effect is not developed easily under all conditions regardless of the chitosan treatment or the mordant treatment. It was inferred that a certain portion of the Rhusjara ica dyestuff made direct bonding with the functional elements in the nylon molecules without the help of the mordant. Through the results of nylon fabrics, it was presumed that some functional elements of the nylon molecules had selective affinity toward the specific colorant (mainly yellow color) of the Rhusjara ica.

Effects of Gamma Irradiation on Physicochemical Properties of Red Beet and Stability of Betalain in the Red Beet (Beta vulgaris L.) (감마선 조사가 비트(Beta vulgaris L.)의 물리화학적 특성 및 Betalain 색소 안정성에 미치는 영향)

  • The physicochemical properties of red beets and the stability of betalain pigments irradiated at 2.5, 5, 10 and 30 kGy were evaluated. Betalain extraction yield of irradiated red beets was not significantly different from non-irradiated red beet. The red beet irradiated at 5kGy showed high optical density value in betacyanin and betaxanthin. In Hunter's color value lightness (L), redness (a) and yellowness (b) decreased with increment of irradiation dose. All red beets showed no difference in pH. The red beets irradiated above 10 kGy had poor textural property quality but antioxidant activity of betalain was not significantly changed by gamma irradiation. Considering natural colorant and textural quality, the gamma irradiation above 10kGy was an undesirable technique for red beets.
