Development of cellulose nano beads based a rapid detection kit to detect staphylococcal enterotoxin B

  • Staphylococcal enterotoxin is a very common cause of food poisoning. Conventional detection methods for the toxin including enzyme-linked immunosorbent assays (ELISAs), chemiluminescence (ECL), and polymerase chain reaction (PCR) assays require a lot of time, efforts, and expert technicians. Lateral flow strip kits have shown great potential for the rapid detection of foodborne pathogens. The lateral flow strip kit is widely used in clinical settings because it is easy to use, fast, and cost effective. A typical lateral flow strip kit uses colloidal gold to generate a visual signal. However, the lateral flow strip kit based on colloidal gold has limited sensitivity to fulfill food safety regulation requirements. This study was performed to develop a rapid test kit for pathogenic staphylococcal enterotoxin B (SEB) in food samples. The rapid detection kit was fabricated based on a nitrocellulose lateral-flow strip. Cellulose nano beads and SEB antibodies were used as the tag and receptor, respectively, to improve the detection performance. Manually spotted SEB antibody and anti-rabbit antibody on the surface of the nitrocellulose membrane were used as test and control spots, respectively. The feasibility of the rapid test kit to detect SEB in samples was evaluated. The sensitivity of the kit was 10 ng/mL SEB spiked in PBS. Additionally, the rapid test kit could detect 1 ng/mL of SEB in chicken meat extract.

Assessment of Emitted Volatile Organic Compounds, Metals and Characteristic of Particle in Commercial 3D Printing Service Workplace (실제 3D 프린팅 작업장에서 발생하는 공기 중 유기화합물, 금속 및 입자특성 평가)

  • Objectives: 3D printing technologies have become widely developed and are increasingly being used for a variety of purposes. Recently, the evaluation of 3D printing operations has been conducted through chamber test studies, and actual workplace studies have yet to be completed. Therefore, the objective of this study was to determine the emission of volatile organic compounds(VOCs), metals, and particles from printing operations at a workplace. This included monitoring conducted at a commercial 3D printing service workplace where the processes involved material extrusion, material jetting, binder jetting, vat photo polymerization, and powder bed fusion. Methods: Area samples were collected with using a Tenax TA tube for VOC emission and MCE filter for metals in the workplace. For particle monitoring, Mini Particle Samplers(MPS) were also placed in the printer, indoor work area, and outdoor area. The objective was to analyze and identify particles' size, morphology, and chemical composition using transmission electron microscopy with energy dispersive spectroscopy(TEM-EDS) in the workplace. Results: The monitoring revealed that the concentration of VOCs and metals generated during the 3D printing process was low. However, it also revealed that within the 3D printing area, the highest concentration of total volatile organic compounds(TVOC) was 4,164 ppb at the vat photopolymerization 3D printing workplace, and the lowest was 148 ppb at the material extrusion 3D printing workplace. For the metals monitoring, chromium, which, is carcinogenic for humans, was detected in the workplace. As a characteristic of the particles, nano-sized particles were also found during the monitoring, but most of them were agglomerated with large and small particles. Conclusions: Based on the monitoring conducted at the commercial 3D printing operation, the results revealed that the concentration of VOCs and metals in the workplace were within Korea's occupational exposure limits. However, due to the emission of nano-sized particles during 3D printing operations, it was recommended that the exposure to VOCs and metals in the workplace should be minimized out of concern for workers' health. It was also shown that the characteristics of particles emitted from 3D printing operations may spread widely within an indoor workplace.

Behavior Evaluation of Aluminium oxide through Measurement of Mass Concentration under Laboratory Environment (연구실 환경에서의 질량농도 측정을 통한 Aluminium oxide의 거동 평가)

  • With the development of nanotechnology, the amount of nanomaterials increases and the problems of environment and the toxic property associated with it have become a social problem. But regulations and laws of nanomaterials have not yet been established. The purpose of this study is to utilize as the database of safety guidelines for research activities' workers associated with nanomaterials to conduct a behavior evaluation of aluminium oxide, which is most widely used in thirteen kinds of WPMN manufactured nanomaterials in the country. The experiment proceeds in the chamber reduced to 1/6 size of the actual laboratory, the test method was performed in NIOSH 0500. As the results of the study, the mass concentration was in inverse proportion to the particle size of the nanomaterials. And the mass concentration during the operation of ventilation equipment was reduced to about 1/8 times. In the future, it can be utilized as the database of safety guidelines for research activities' workers associated with nanomaterials. However, in order to increase the reliability of the study, the experiment of the mass concentration by particle size and Condensation Particle Counters will be needed additionally.

Effect of Nano Particles on the Hathcing rate of Artemia sp. Cyst (알테미아(Artemia sp.) Cyst 부화율에 미치는 나노입자의 영향)

  • 9 kinds of nanoparticle used for this study was a particle with the size of less than 100 nm of diameter, and Artemia sp. cyst examined what kind a influence to have upon the process hatched out in nauplius. 82% hatched in nauplius at the opposition ward where a nanoparticle wasn't added after 24 time course. AGZ020, Nano silver, P-25, Sb and SnO nanoparticle showed hatching rate of 18%, 20%, 13%, 50% and 0% respectively by the 20mg/L density, and it became clear that a harmful effect is big, but I had a harmful effect compared with the opposition ward by 75%, 60%, 73% and 73% respectively by Ag-$TiO_2$, In, Sn and Zn nanoparticle, but a feeble thing was known relatively compared with AGZ020, Nano silver, P-25, Sb and SnO nanoparticle. The difference was mused this with the ingredient a nanoparticle has. Ag is included 2% and AGZ020, Nano silver and P-25 nanoparticle are used widely as anti-fungus agent, and the SnO nanoparticle which became combination is a light catalyst pill, and oxygen is used for a Sn particle. This and others, a possibility that use is generalized and flows into aquatic environment in sequence the home electronics, functionality cosmetics, anti-fungus agent and a light catalyst pill at present becomes high for nanoparticles and others. The anxiety which has an influence on the ecology world in the water with this can be generated, so I'd have to study the potential danger a nanoparticle has continuously.

  • 21세기 신기술은 나노기술(Nano Technology,NT)을 통해서 이뤄질 것으로 보인다. 로마를 통하지 않으면 어디로도 갈 수 없었던 것처럼 작게 생각하지 않으면 첨단기술에 접근할 수 없는 상황이 펼쳐지고 있다. 이전에 재료공학자ㆍ기계공학자 불리던 연구자들이 미지 세계 개척자로서 작은 것들을 새롭게 창조하는 '나노의 방주'에 서둘러 들어서고 있다. 나노의 방주에는 화학뿐만 아니라 물리ㆍ건축ㆍ공학 등이 자유롭게 넘나든다. 이들은 혜성처럼 등장한 나노라는 화두를 중심에 놓고 서로 영향을 주고받으며 기술의 향방을 결정하고 있다. 나노기술이 열려라 참깨식의 마술적 주문으로 작용하는 셈이다. 전능의 신 노릇을 하는 나노는 새로운 것을 만들어낼 뿐 아니라 다른 기술에 놀라운 능력을 부여하기도 한다. 종교인들이 신을 통해 새롭게 태어나듯 일상적 기술이 나노를 통하면 황금알을 낳는 신기술로 거듭나는 것이다. 그 이름에서도 알 수 있듯이 한국나노의료기는 현실보다는 미래지향적인 전략적 기업으로 발전을 거듭하고 있었다

Establishments of Fabrication and Evaluation Methods for Innovative SiC Fiber Reinforced SiC Matrix Composites

  • Based on the improvement in reinforcing SiC fibers and the utilization of very fine nano-SiC powders, the well known liquid phase sintering (LPS) process was drastically improved to become a new process called the Nano Infiltration and Transient Eutectic Phase (NITE) Process. Laboratory scale NITE-SiC/SiC composites demonstrated excellent mechanical properties, thermal conductivity, hermeticity and microstructure stability which made them attractive for not only energy application but many other industrial applications. For the real deployments of these materials, mass production system and evaluation methods, together with the design code and safety assurance systems are essential. The current efforts to establish these bases were introduced.

SWIR 이미지 센서 기술개발 동향 및 응용현황

  • Imaging in the Short Wave Infrared (SWIR) provides several advantages over the visible and near-infrared regions: enhanced image resolution in in foggy or dusty environments, deep tissue penetration, surveillance capabilities with eye-safe lasers, assessment of food quality and safety. Commercially available SWIR imagers are fabricated by integrating expensive epitaxial grown III-V compound semiconductor sensors with Si-based readout integrated circuits(ROIC) by indium bump bonding Infrared image sensors made of solution-processed quantum dots have recently emerged as candidates for next-generation SWIR imagers. They combine ease of processing, tunable optoelectronic properties, facile integration with Si-based ROIC and good performance. Here, we review recent research and development trends of various application fields of SWIR image sensors and nano-materials capable of absorption and emission of SWIR band. With SWIR sensible nano-materials, new type of SWIR image sensor can replace current high price SWIR imagers.

  • Recent rapidly growth in nanotechnolgies is promised novel benefits through the exploitation of their unique industrial and biomedical applications. In addition, these nanomaterials and nano-consumer products have increased in quantity per year, and thus their uncontrolled release into the environment is anticipated to grow dramatically in future. Many papers for cytotoxicity of nanomaterials have been already reported, and thus government supports has funded to various research topics for nanosafety. Herein, we analyzed the importance and performance of nanosafety researches and tried to show the research direction where we have to go.

Mechanical Properties Evaluation of Natural and Synthetic Rubber (천연 및 합성고무의 기계적 물성 평가)

  • Mechanical properties of rubber material are very important in design procedure to assure the safety and reliability of the rubber components. In this paper, the material test and accelerated heat aging test were carried out. In order to investigate the effects of heat-aging on the material properties, hardness, elongation, stress-strain curves and dynamic characteristics were obtained from various test conditions. Also, rubber material coefficients were determined by both the uniaxical and equi-biaxial tensile tests.

Safety evaluation of bacteriophages for application as sanitizers (박테리오파지의 살균소독제 응용을 위한 안전성 평가)

  • To evaluate the safety of bacteriophages for application of sanitizer, endotoxin content and cell cytotoxicity of two Escherichia coli and four Staphylococcus aureus phages were determined. Endotoxin ratio was determined by the Limulus amebocyte lysate (LAL) assay as a test for representative biological endotoxin content. The average endotoxin average content of the 9 log PFU/mL lysate was 18.6 EU/mL and that of the 10 log PFU/mL lysate was 5.9 EU/mL, suggesting that the phage lysate was not suitable for clinical applications, but suitable for food pathogen control applications. To confirm the cell cytotoxicity of the phage lysates, MTT assay was performed using Raw 264.7 cells treated with 9 log PFU/mL phages. Results of the assay indicated that the phage lysates did not significantly decrease the cell viability (p>0.05). These results indicated that bacteriophages would be suitable as a food safety sanitizer.
